  • Dendritic cells (DCs) are known to professional antigen presenting cell (APC). Due to the role as an effective activator of cytotoxic T Lymphocytes by expressing MHC, adhesion and co-stimulatory molecules, DCs are now widely recognized to play an important role in the immune responses to tumors.We investigated the effect of cultured DCs in murine melanoma pulmonary metastasis model. To follow the metastasis protocol, syngenic melanoma cells were inoculated intra-venously into the mouse (B16F10 into the C57BL/6)8 days prior to the first DC injection (1$\times$106 DCs/ mouse, i.p.) and the autologous tumor cell lysate pulsed-DCs were injected as a therapeutic module twice in two weeks.   • 구강내에서 발생하는 악성 흑색종은 비교적 드문 종양으로 원인에 대해서는 정확히 알려져 있지는 않으나 방사선치료 후에 발생된 양성 흑색종이 악성으로 전환된 보고가 있다. 본원에서 구강내 편평상피세포암으로 국소방사선치료 후 동일한 부위에 6년 후에 악성 흑색종이 발생하였기에 문헌고찰과 함께 보고하는 바이다.

Mouse Melanoma Cell Migration is Dependent on Production of Reactive Oxygen Species under Normoxia Condition

  • Cell migration plays a role in many physiological and pathological processes. Reactive oxygen species (ROS) produced in mammalian cells influence intracellular signaling processes which in turn regulate various biological activities. Here, we investigated whether melanoma cell migration could be controlled by ROS production under normoxia condition. Cell migration was measured by wound healing assay after scratching confluent monolayer of B16F10 mouse melanoma cells. Cell migration was enhanced over 12 h after scratching cells. In addition, we found that ROS production was increased by scratching cells. ERK phosphorylation was also increased by scratching cells but it was decreased by the treatment with ROS scavengers, N-acetylcysteine (NAC). Tumor cell migration was inhibited by the treatment with PD98059, ERK inhibitor, NAC or DPI, well-known ROS scavengers. Tumor cell growth as judged by succinate dehydrogenase activity was inhibited by NAC treatment. When mice were intraperitoneally administered with NAC, the intracellular ROS production was reduced in peripheral blood mononuclear cells. In addition, B16F10 tumor growth was significantly inhibited by in vivo treatment with NAC. Collectively, these findings suggest that tumor cell migration and growth could be controlled by ROS production and its downstream signaling pathways, in vitro and in vivo.

효모에서 분리한 멜라닌 생성 억제 물질의 세포분화 억제 (Inhibition of Melanoma Differentiation by Melanogenesis Inhibitor Isolated from Yeast)

  • 본 연구에서는 효모에서 분리한 melanoston이라고 명명된 멜라닌 생성을 억제하는 물질의 작용 기전을 밝히기 위한 것이다. $\alpha$-MSH를 처리한 B16 melanoma 세포에서 melanoston은 tyrosinase mRNA 발현양을 $10\%$ 정도 저해되는데 그쳤으며 western blotting을 이용한 단백질 측정에서도 이와 비슷한 정도의 단백질 생성 억제를 보였다. 그러나 B16 세포 배양액에 melanoston을 첨가할 경우 세포내 tyrosinase 활성이 $30\%$까지 감소되는 것으로 나타나 melanoston이 tyrosinase inhibitor는 아니지만 세포내 tyrosinase 활성화(activation) 과정을 억제하는 것으로 추측할 수 있었다. 또한 광학 현미경을 이용한 morphology 관찰에서 $\alpha$-MSH를 처리한 세포에서는 많은 dentrite가 형성되면서 세포분화가 일어나는 반면 melanoston를 처리한 경우에는 dendrite가 감소하면서 세포형태가 대조군과 비슷하게 회복되는 것을 알 수 있었다. 또 FITC-anti-tyrosinase-Ab를 이용한 형광염색을 통해서는 $\alpha$-MSH만 처리한 세포에서는 tyrosinase의 분포가 dendrite를 포함한 세포 전체로 퍼져나가는 것을 관찰 할 수 있었고 $\alpha$-MSH와 melanoston을 동시에 처리한 세포에서는 대조군과 비슷하게 tyrosinase가 핵 주변에서만 관찰되어 melanoston이 B16 melanoma 세포의 분화과정에서 이를 억제하는 효과를 주고 있음을 알 수 있었다. 이상의 결과들을 종합해 볼 때 melanoston은 $\alpha$-MSH에 의해 진행되는 B16 세포의 분화를 억제하고 이 과정에서 멜라닌 생성의 주된 효소인 tyrosinase의 활성화를 억제하며 결과적으로는 멜라닌 생성을 저해하는 것으로 사료된다.

전이성 흑색종의 세포학적 소견 (Cytologic Features of Metastatic Melanoma)

  • Metastatic melanoma is often difficult to diagnose morphologically. However, the prognostic and therapeutic decision in cases of metastatic melanoma depends upon the morphologic documentation of the metastatic disease, which may rapidly and accurately be done by cytologic methods such as aspiration cytology of clinically suspicious lesion. Cytological features of 12 metastatic melanoma are described. Evaluation for melanin pigment, background composition, cytoplasmic vacuoles, intranuclear inclusions, bi- or multinucleated cells, eccentric nuclei, nucleoli, and degree of cellular cohesion were made. the presence of melanin pigment, prominent nucleoli, intranuclear cytoplasmic inclusions, lack of cell cohesion, and eccentric nuclei are the important and useful cytologic features in the diagnosis of metastatic melanoma. Bi or multinucleated cells, cytoplasmic vacuoles, and bloody background are also frequently found in smears from metastatic melanoma.

새로운 Aminoisoquinolinylamide 유도체의 합성 및 흑색종 저해 효능 (Synthesis and Melanoma Inhibitory Activity of New Aminoisoquinolinylamide Derivatives)

  • In this work, a novel series of aminoisoquinolinylamide derivatives 1a-c and 2a-f were synthesized via several reaction steps, starting from 2-methyl-4-nitrobenzonitrile (3) and 1-chloro-5-nitroquinoline (8). And their antiproliferative activities were screened against A375 human melanoma cell line compared to Sorafenib as a reference compound. Among them, compound 1b and 1c exhibited meaningful inhibitory activities. These results demonstrated that aminoisoquinolinylamide scaffold possesses the possibility as the treatment for melanoma.

보골지(補骨脂)의 고량주(高梁酒), 소주(燒酒) 추출물이 B16 melanoma 세포주의 멜라닌 합성에 미치는 영향 (The effcect of Gorynju and a Soju extract Psoraleae fructus on Melanin synthesis of B16 melanoma cells)

  • Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3 ${\mu}g/ml$), in a Soju extract Psoraleae fructus, 1 ${\mu}g/ml$ were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4 ${\mu}g/ml$). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.

가미대황고(加味大黃膏)의 멜라닌세포 활성억제가 자외선 조사로 인한 피부 손상 완화에 미치는 영향 (The effect of Daehwanggogasangbakpi(大黃膏加桑白皮) on the skin damage induced by ultraviolet irradiation)

  • Objective : As a result of increasing amount of ultraviolet ray, skin problems including sunburn, rapid skin aging, melanoma, and even skin cancer continue to rise. In the present study, the effect of oriental herbal extract, Daehwanggo(大黃膏,DH) and Daehwanggogasnagbakpi(大黃膏加桑白皮,DS), as external application, on the skin damage, was investigated. Methods : 30 mice were equally distributed into 3 groups : control, UVB-control and UVB-irradiated and DS-treated group. Also mouse melanoma cell lines were cultured. Tyrosinase inhibition was measured to analyze the UN-protection effect. Melanogenesis in the UV-irradiated melanoma cell lines was compared in DS-treated cell line and control cell line. Sample skin from the ear tissue of the 3 groups were analyzed to observe the inflammatory response, T cell differentiation, apoptosis of keratinocytes. Results : The tyrosinase was more significantly inhibited in the DS group compared to DH group. Antioxidative effects was more prominent in DS group when superoxide dismutase was measured. Both the DS- and DH-treated cell lines showed significantly reduced melanogenesis. The reduction of external skin damage including erythematous papule, eczema, keratinocyte, pyopoiesis was observed in the DS- and DH-treated sample cells. In terms of the effect on the skin damage, sunburn cell, activated skin mast cells, secretion of IL-12, manifestation of HSP70, hyperplasia of epithelial cells, MMP-9 and destruction of the collagen were all significantly improved in the DS-treated sample cells. Melanin cells and the apoptosis in the melanoma cell line were decreased. Conclusion : DH and DS were traditionally applied externally for the scald in the oriental medicine. The present study elucidated the possibility of herbal extracts to be used as ultraviolet protectives. Further investigations are needed to assure the clinical application.

키토산 유도체인 Sulfated N-acetyl Chitosan의 종양전이 억제효과 (Inhibitory Effects of Tumor Metastasis by Chitosan Derivative, of Sulfated N-acetyl Chitosan)

  • 키토산 유도체 인 sulfated N→acetyl chitosan을 합성하여 이 화합물을 쥐에 이식한 B16/BL6 mela noma에 대하여 폐암전이의 억제효과를 조사하였다. 키토산의 황산유도체는 13C-n.m.r.에 의하여 확인한 결과 3, 6, O-disulfate 엄을 알 수 있었다. SuI fated N-acetyl chitosan을 100mg/kg을 투여하였을 때 B16/BL6의 melanoma cells을 가장 효과적 으로 억제하였고 종양 무게의 증식도 대조군에 비해 억제되었다. 폐암의 전이에 있어서 B16/BL6에 sulfated N-acetyl chitosan을 LV. V로 주사하였을 때 B16/BL6의 melanoma의 전이 세포의 수는 자발적인 전이에 있어서도 용량 의존형(20 ~ 1OOmg/kg) 에 따라 줄어들었다. B16/BL6을 접종한 후 sulfated N-acetyl chitosan을 I.V.로 투여했을 때 전이세 포의 수는 현저히 감소하였다. Sulfated N-acetyl chitosan은 laminin의 종양세포에의 세포접착 능력을 부분적으로 억제하였다. 이와 같은 결과는 chitosan의 유도체인 sulfated N-acetyl chitosan이 세포접착 능력이 실험적으로 자발적인 암전이 모델에 효과적으로 작용하였다.
