한약재로부터 Tyrosinase 저해제의 탐색 (Screeing of Tyrosinase Inhibitors from Oriental Herbs)

  • 한약재 174종으로부터 tyrosinase 저해제를 탐색하였다. 한약재는 30일간 methanol에 담가 놓은 후, 그 추출액을 제조하여 사용하였다 Tyrosinase의 활성을 측정하기 위해서는 효소 mushroom Tyrosinase와 기질 L-DOPA, phosphate buffer를 사용하였다. 탐색 결과, 한약재 174종 중 14개종이 tyrosinase를 저해 효과를 보였고, 그중 오배자 추출액이 inhibition activity가 가장 높게 나타났다.

상엽(桑葉) 추출물의 미백활성 및 피부섬유구조 형성에 미치는 효과 (Effect of Mori Follium Extract on the Melanogenesis and Skin Fibril Matrix)

  • Objectives : The Skin is composed of multiple layers, including the epidermis, dermis, and hypodermis. It provides a vital barrier structure that protects vertebrates from external environmental antigens, solvents, ultraviolet light, microorganisms, toxins, and weather conditions. Although several biological effects of Mori Follium have been reported, beneficial effects of Mori Follium in skin health remain unclear. In this study, we prepared water extract of Mori Follium (MLE) and evaluated the effects on melanin accumulation and expression levels of skin fibril-related proteins.Methods : The cytotoxicities of MLE in B16F10 melanoma and human skin fibroblasts (HSF) were examined by MTT assay. Inhibitory effect of MLE on the α-MSH- and IBMX-induced melanosis in B16F10 melanoma was examined. The expression levels of fibronectin, collagen 1α2, and CCN2 in MLE-treated HSF were analyzed by reverse transcription-polymer chain reaction (RT-PCR) and western blotting.Results : The MLE treatment for 24 h did not affect to the B16F10 and HSF at concentrations of 1, 10, 50, 100, 200, 400 and 800 ㎍/ml. The MLE treatment for 72 h significantly and dose dependently suppressed melanin accumulation in B16F10 melanoma. In addition, the MLE treatment up-regulated expression levels of skin fibril-related genes such as fibronectin, collagen 1α2, and CCN2 in HSF. Our western blot analysis revealed MLE-induced up-regulation of skin fibril-related genes required the activation of CCN2 protein.Conclusions : In conclusion, these findings suggest that the MLE could be used in development of cosmetic natural material of maintaining healthy skin.

신이(辛夷)로부터 멜라닌 생성 억제물질의 분리 (Isolation of Melanin Biosynthesis Inhibitory Compounds from the Flowers of Magnolia denudata)

  • The bioassay-guided fractionation of the MeOH extract of the flowers of Magnolia denudata led us to the isolation of six compounds identified as fargesin(1), kobusin(2), aschantin(3), magnolin(4), rel-[7s,8s,8's]-3,4,3',4'-tetra- methoxy-9,7'- dihydroxy-8,8',7.O.9'-lignan(5) and oplodiol(6), respectively. Among the isolated compounds, fargesin(1) showed most potent inhibitory effect on the melanin polymer biosynthesis in cultured B-16 mouse melanoma cell lines$(IC_{50},\;45.7\;{\mu}M)$.

  • 천연추출물로서 미백 재료로 널리 쓰이고 있는 옥용산에 대해서 in vitro 미백활성을 UV 흡수능, tyrosinase 저해활성 그리고 free radical 소거활성을 측정함으로서 검정해 보았다. 옥용산 UV-B와 C 영역에 걸쳐서 높은 자외선 흡수능을 보이는 것으로 관찰이 되었으며 0.8 % 이상의 농도에서 80% 이상의 tyrosinase 저해활성을 가지는 것을 관찰하였다. 또한 조협, 승마, 감송, 정향, 찹쌀분과 옥용산의 추출물은 높은 free radical 소거능을 가지는 것으로 확인되었다.

  • Eight compounds were isolated from the roots of Glycyrrhiza glabra by the tyrosinase inhibitory activity guided fractionation, and their structures were identified as liquiritigenin (1), isoliquiritigenin (2), isoliquiritigenin-2'-O-methyl ether (3), liquiritin (4), isoliquiritin (5), ononin (6), glycycoumarin (7), glycyrol (8) by analysis of spectral data. Compound 3 exhibited the most potent inhibitory effect on mushroom tyrosinase activity ($IC_{50}$, 47 M).

목단피로부터 멜라닌 생성 억제성분의 분리 (Isolation of Inhibitory Components on Tyrosinase Activity from the Bark of Paeonia moutan)

  • The MeOH extract of the bark of Paeonia moutan showed potent inhibitory effect on the mushroom tyrosinase activity in vitro. The activity-guided fractionation of t he MeOH extract resulted in the isolation of three active compounds. The chemical structures of these compounds were elucidated by chemical and spectroscopic evidence as catechin, 1,2,3,6-tetra-O-galloyl-${\beta}$-D-glucose and 1,2,3,4,6-penta-0-galloyl-${\beta}$-D-glucose, respectively. Among them, the inhibitory activity by 1,2,3,6-tetra-galloyl-${\beta}$-D-glucose on mushroom tyrosinase was more potent $(IC_{50}=3.5\;{\mu}M)$ than that of kojic acid $(IC_{50}=8.7\;{\mu}M)$ ,but catechin enhanced the mushroom tyrosinase activity 50% in the concentration of 34.5M.
