The Screening of the Inhibitory Compounds on Tyrosinase Activity from the Natural Product (고등식물로부터 피부멜라닌 생성에 관여하는 티로시나제 활성 억제물질의 탐색)

  • Tyrosinase is known to accelerate the melanin polymer biosynthesis in melanocyte, so tyrosinase inhibitors hinder the melanin polymer biosynthesis and are useful not only for th e material used in cosmetics as skin-whitening agents but also as the remedy for disturbances in pigmentation. During our search for new melanin biosynthesis inhibitors from natural sources, 130 higher plants were tested for the inhibitory effect against tyrosinase activity by the mushroom tyrosinase assay. Among them, Carex humilis ($IC_{50}$, 10vg/ml), Sophora flavescence ($IC_{50},\;20{\sim}50{mu}g/ml$) and Styrax japonica ($IC_{50},\;10{\mu}g/ml$) inhibited the tyrosinase activity strongly.

  • By screening inhibitory activities on the melanin polymer biosynthesis in B-16 mouse melanoma cell lines, MeOH extract of Phellodendri Cortex was found to have inhibitory effect on melanin polymer biosynthesis. Twelve compounds were isolated from the MeOH extract of P. Cortex. They were identified as obacunone (1), limonin (2), ${\beta}-sitosterol$ (3), bis(2-methylheptyl) phthalate (4), cycloeucalenol (5), berberine (6), palmatine (7), jatrorrhizine (8), syringin (9), umbelliferone (10), rutaecarpine (11) and scopoletin (12) by comparison of their physical and spectral data with those of authentic samples. Among the isolated compounds, berberine (6) and palmatine (7) showed potent inhibitory effect on the melanin polymer biosynthesis in cultured B-16 mouse melanoma cell lines, with Inhibition rate of 96% and 90%, respectively. As a positive control, arbutin exhibited an inhibition rate of 56%.

The Isolation of the Inhibitory Constitutents on Melanin Polymer Formation from the Leaves of Cercis chinensis (박태기나무의 잎으로부터 피부멜라닌 색소생성 억제성분의 분리)

  • Tyrosinase plays an important role in the process of melanin polymer biosynthesis. Therefore, the enzyme inhibitors have been of great concern as cosmetics to have skin-whitening effects on the local hyperpigmentation. During the search for new inhibitory compounds on melanin polymer biosynthesis from natural sources, MeOH extracts of 589 higher plants were tested for the inhibitory effect on tyrosinase activity by the muschroom tyrosinase assay in vitro. Among plants tested, the leaves of Cercis chinensis exhibited potent inhibitory effect on mushroom tyrosinase activity. Subsequently seven active compounds were isolated from the ethyl acetate soluble part of acetone extract of the leaves of C. chinensis by the activity guided fractionation monitoring the inhibitory effect on tyrosinase activity. Their chemical structures were identified as $kaempferol-3-0-{\alpha}-L-rhamnoside$, quercitrin, $myricetin-3-0-{\alpha}-L-rhamnoside$, myricetin-3-0-(2'-O-galloyl)- ${\alpha}$ -L-rhamopyranoside (desmanthin), (-)-epicatechin-3-0-gallate, (-)-epigallocatechin-3-0-gallate, and methyl gallate on the basis of the speculation of spectral data and chemical reaction. Among the flavonol rhamnosides, myricetin-3-0-(2'-O-galloyl)- -L-rhamnoside(desmanthin) showed most potent inhibitory effect on tyrosinase activity and the structure of B-ring in flavonol moiety was related to the activity. (-)-Epigallocatechin-3-O-gallate having pyrogallol group in flavan-3-ol moiety exhibited more potent inhibitory effect than (-)-epicatechin-3-0-gallate having catechol group in flavan-3-ol moiety on mushroom tyrosinase activity.

  • Magnoliae Flos(‘shin-j’). the flower buds of Magnolia kobus, is acrid to taste with a ‘warm’ property. It is a ‘wind-cold’ discutient and nasal decongestant and is principally used in the treatment of nasal congestion with headache, sinusitis and allergic rhinitis. By screening inhibitory activities on the melanin polymer biosysthesis in B-16 mouse melanoma cell lines, methylene chloride extract of Magnoliae Flos was found to have inhibitory effect on melanin polymer biosynthesis. Inhibitory Effect of Enteromorpha linza on the Melanogenesis in B16 Melanoma Cells (파래 추출물의 멜라닌 생성 억제 효과)

  • Melanin is a polymer of phenol which produces hyperpigmentation in the skin. Melanin synthesis is catalyzed by the enzyme tyrosinase. To investigate the whitening activity of the fractions from the ethanol extract of Enteromorpha linza, we studied the inhibitory effect on the tyrosinase activity and melanogenesis in the B16/F1 melanoma cells. The inhibition ratio of tyrosinase activity of ethylacetate fraction from E. linza was as strong as that of kojic acid, a positive control. Also, the melanin production was significantly inhibited by the ethylacetate fraction in a dose dependent manner. The ethylacetate fraction showed the highest activity in the fractions and as strong activity as that of arbutin, a positive control. From these results, we suggest that the E. linza extract might be able to apply to cosmetic and medical fields.

  • The purpose of this study was to investigate the effect of the aqueous extract of the Astragalus membranaceous(AM). AM showed inhibitory effect on the tyrosinase activity using L-tyrosine as a substrate. Tyrosinase plays an important role in the process of melanin polymer biosynthesis. In vitro AM extract(1mg/ml) inhibited melanin biosynthesis and are useful for the material used in cosmetics. B16 mouse melanoma cells were cultured in different concentrations. The non-cytotoxicity of the plant extracts was confirmed by MTT assay.

  • To investigate how the aqueous extracts of Epimedium koreanum Nakai (EK) affects pigmentation of skin, the aqueous extract of EK at various concentrations were incubated with 1 $\times$ 10$^{5}$ melanoma cells per well for 72 h. The morphology and number of cell line were not changed, but the aqueous extract of EK increased the tyrosinase activity and the content of melanin polymer in the cell line. These results indicate that the aqueous extract of EK promotes melanogenesis of Bl6 mouse melanoma cell line.

Effect of Rhynchosia Nulubilis Ethanolic Extract on DOPA Oxidation and Melanin Synthesis (서목태 주정 추출물이 DOPA 산화와 멜라닌 합성에 미치는 영향)

  • Melanin is a polymer substance that plays an important role in the determination of hair growth and skin color in vivo. However, melanin, which is over-produced by reactive oxygen species, is known to cause stains, freckles, and hypercholesterolemia, which are associated with aging. Previous studies have shown that polyphosphate, one of the components of Rhynchosia Nulubilis, inhibits skin aging induced by ultraviolet rays. The aim of this study is to investigate the direct effect of Rhynchosia Nulubilis ethanolic extract (RNEE) on melanin synthesis. In this study, RNEE showed no antioxidative effects on scavenging activity of DPPH radical in addition to reducing power. The cytotoxicity of RNEE was increased in a dose-dependent manner in an MTT assay. In addition, RNEE increased tyrosinase activity and melanin synthesis in DOPA-oxidation experiments. RNEE did not promote the conversion L-DOPA into melanin in live cells, but melanin production was promoted in the RNEE-treated group after H2O2 pretreatment compared to the control group in which melanin production was reduced by treatment with H2O2. In addition, RNEE increased the expression level of tyrosinase related protein-2 (TRP-2) and increased the expression level of tyrosinase related protein-1 (TRP-1) at a concentration of $16{\mu}g/ml$. In particular, it was found that RNEE increased the expression level of SOD-3, by which superoxide anion is converted to hydrogen peroxide, higher than the control and ${\alpha}$-MSH used as a positive control at a concentration of more than $16{\mu}g/ml$. The results suggest that RNEE can induce melanogenesis related to black hair.

Positive Effect of Musa paradisiaca Peel Ethanolic Extract on Antioxidant Activity and Melanin Synthesis (바나나 껍질 에탄올 추출물이 멜라닌 합성에 미치는 영향)

  • Aging is accompanied by changes in the body, such as graying hair, wrinkles, and black spots composed of lipid peroxides and proteins. Melanin is a polymer substance produced by an oxidation polymerization reaction from tyrosine, and it determines the color of hair and skin. It has been reported that melanin is synthesized by melanocyte, and its excessive production by reactive oxygen species is associated with aging. The purpose of this study was to determine the direct effects of Musa paradisiaca peel ethanolic extract (MPEE) on antioxidative activity and melanin synthesis. It was observed that the antioxidant activity of MPEE was similar to that of vitamin C, a positive control, in both DPPH radical scavenging assay and reducing power assay. In order to examine cytotoxicity prior to cell experimentation, 3-(4,5-dimethythiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was performed for B16F1 cells. MPEE was not cytotoxic at $32{\mu}g/ml$ or less. In addition, MPEE increased melanin synthesis in live cells in addition to tyrosinase activity and melanin synthesis in dihydroxyphenylalanine (DOPA)-oxidation assay in vitro. Moreover, MPEE increased melanin synthesis in cells aged by pretreatment with $H_2O_2$. The expression levels of tyrosinase-related protein (TRP)-1, TRP-2, and superoxide dismutase (SOD)-2 by western blot analysis were increased in the presence of MPEE. These results suggest that MPEE could promote the melanin synthesis as an antioxidative substance.
