Recently, arbutin, oil soluble licorice extact (GLY), ascorbyl glucoside (AA2G), and ethyl ascorbyl ether (EAE) have been widely used as functional whitening ingredients. To Investigate which combination between the above agents could be more effective for whitening effect, tyrosinase activity and MSH-induced melanin production in B-16 melanoma cells were investigated. Both GLY and arbutin dose-dependently inhibited purified tyrosinase activity. The inhibitory effects of GLY with AA2A or EAE on Drosinase activity were more potent than those of GLY alone, whereas that of arbutin with other ingreadients did not show those effects. In MSH-induced melanin production in B-16 melanoma cells, the mixture of Gly and EAE more significantly reduced melanin formation than Gly alone. Stability of mixture of GLY, arbutin, AA2A and EAE exposed at the temperature of $25^{\circ}C\;or\;45^{\circ}C$ for 30 days were also investigated. All of the combinations of whitening agents did not show any critical changes in their composition stability. These data suggest that the combination of GLY and vitamin C derivatives such as AA2G and EAE may be useful for the promotion of whitening effect.

Isomaltol glycoside is a hydrophilic furanic glycoside in which the amino acids and sugars of ginseng are thermally denatured during red ginseng production. Various skin whitening tests were conducted on isomaltol glycoside containing a lot of red ginseng extract in order to investigate the skin whitening effect as a cosmetic raw material. We have tested melanin content assay in B16-F10 cells, zebrafish embryo pigmentation assay, mushroom tyrosinase inhibitory activity, western blot analysis to determine skin whitening activity of isomaltol glycosides. In the zebrafish melanin content assay, isomaltol glycoside decreased total melanin content by about 20% and zebrafish tyrosinase activity by about 10% after treatment with 50 and $100{\mu}g/mL$ compared to the untreated control group. Isomaltol glycoside also showed a concentration-dependent decrease in melanin content in B16-F10 melanoma. Furthermore, it increased the expression of MITF phosphorylation factors p-AKT and p-ERK in B16-F10 melanoma and decreased the concentration of MITF. It also inhibited tyrosinase, TRP-1 and TRP-2 expression. The content of isomaltol glycoside was about 3% in the ginseng extract and about 1% in the ginseng root. Thus, isomaltol glycoside is considered as one of the main components that exhibit the whitening activity of ginseng when considered quantitatively as whitening activity.

Vitamin D is a fat-soluble vitamin that is mainly produced in the skin by UV rays. Along with melatonin, it is a representative chronobiotic substance, and the skin plays an important role in distinguishing between day and night. However, vitamin D cannot be used directly in cosmetics because it is a vitamin that acts as a coenzyme and plays a hormonal role in regulating the expression of various types of genes. Therefore, it was to investigate the skin efficacy of provitamin D (7-dehydrocholesterol), a vitamin D precursor that can be used in cosmetics. Our findings reveal that pro vitamin D can effectively inhibit the expression of tyrosinase, the melanin-producing enzyme, thereby attenuating melanin synthesis. This skin tone regulatory effect has been corroborated in vitro using artificial skin models. Additionally, pro vitamin D demonstrated anti-inflammatory properties by suppressing the expression of TNFa and, upon conversion to vitamin D through UV exposure, it was observed to induce the production of the antimicrobial peptide CAMP (LL-37). The inhibitory effect of pro vitamin D on melanin production appears to be a result of it reducing the UV absorption capacity of melanin, thereby inducing the conversion of pro D to vitamin D. Utilizing pro vitamin D in cosmetic formulations could not only modulate skin tone and enhance skin immunity but also expect to contribute to other cutaneous benefits as anti-aging and barrier function improvement with everyday UV exposure. This multifaceted efficacy positions pro vitamin D as a promising ingredient in advancing the formulation of skin care products.
Melanin synthesis is catalyzed by tyrosinase. To investigate the whitening effect of Hizikia fusiformis, fractions from ethanol extract of H. fusiformis were prepared by a systematic fractionation procedure with solvents such as methanol, hexane, butanol, and $H_2O$. The ethanol extract and its fractions were then subjected to evaluate the inhibitory effects on the tyrosinase activity and melanin synthesis in murine B16F10 melanoma cells. The ethanol extract and aqueous fraction exhibited a whitening effect with no cytotoxicity. The ethanol extract showed the highest whitening effect among the samples. The inhibitory effect of $100{\mu}g/ml$ of ethanol extract was higher than that of $10{\mu}g/ml$ of arbutin, but it was lower than that of $10{\mu}g/ml$ of kojic acid. Furthermore, the inhibitory effects of $100{\mu}g/ml$ of methanol, hexane, butanol, and aqueous fractions were similar to those of $10{\mu}g/ml$ of arbutin. The antioxidant activities were examined by comparing the results with that of ascorbic acid as a positive control. The ethanol extract and aqueous fraction showed relatively higher DPPH radical-scavenging activities compared with the other samples. Furthermore, $500{\mu}g/ml$ of ethanol extract and aqueous fraction diminished LPS-induced iNOS expression to 82 and 80%, respectively. These results suggest that ethanol extract and aqueous fraction of H. fusiformis could be used as cosmetic ingredients for whitening and skin protection effects.
In this study, peptides were isolated from Crassostrea gigas using an ultrasonification process at $40^{\circ}C$. The yield of the peptides was greater than 34%, and their cytotoxicity was found to be less than 22.8% against several cell lines that were treated with the extracts at a dose of 1.0 mg/mL. In addition, the tyrosinase inhibitory and melanin synthesis of the peptides isolated from Crassostrea gigas were also evaluated to determine if they could be used as a potential cosmetic agent. The peptides were found to significantly inhibit the melanin synthesis of the clone M-3 cell line by up to 62.7%. The inhibitory activities of the tyrosinase were observed 34.51% in ascorbic acid, 42.49% in extract with the ultrasonification at $40^{\circ}C$ and 35.37% in $40^{\circ}C$ extract at 1.0 mg/mL concentration, respectively. Finally, when samples were treated with the peptide extracts at a concentration of 0.6 mg/mL, PGE2 expression was significantly decreased. Taken together, these results indicate that Crassostrea gigas may be a source of cosmetic agents capable of improving physiological hyperpigmenting and immuno-modulating skin disorders.

In this study, it studies about Phytosphingosine (PhS) and Phytosphingosine-1-phosphate(PhS1P), and it tries to confirm the effect through anti-inflammatory, anti-melanin, MMP-1 revelation inhibition, and Western blot analysis experiment after grasping toxicity about 3 cells by using B16F10 melanin cell, RAW264.7 macrophage, and HDF fibroblast in order to find out whether it is possible to use as cosmetic material or not by studying biological activity in terns of skin care. As a result of this experiment, it confirmed that toxicity about B16F10, RAW264.7, HDF cell is low, and PhS1P appeared stronger inhibition activity than PhS in anti-inflammatory NO inhibitory activity experiment. MMP-1 revelation was greater in PhS1P, and it confirmed that the mechanism is due to reduction in ERK activity. On the other hands, melanin generation inhibitory activity is better than arbutin, and it confirmed that the mechanism is due to inhibition of revelation of MTF and Tyrosinase. In a nutshell, PhS and PhS1P that are bioactive substance may confirm the possibility to be used as functional cosmetic for wrinkle and skin improvement of whitening cosmetic.
BACKGROUND/OBJECTIVES: Gastrodia elata Blume (GEB), a traditional herbal medicine, has been used to treat a wide range of neurological disorders (e.g., paralysis and stroke) and skin problems (e.g., atopic dermatitis and eczema) in oriental medicine. This study was designed to investigate whether GEB extract inhibits melanogenesis activity in murine B16F10 melanoma. MATERIALS/METHOD: Murine B16F10 cells were treated with 0-5 mg/mL of GEB extract or $400{\mu}g/mL$ arbutin (a positive control) for 72 h after treatment with/without 200 nM alpha-melanocyte stimulating hormone (${\alpha}$-MSH) for 24 h. Melanin concentration, tyrosinase activity, mRNA levels, and protein expression of microphthalmia-associated transcription factor (MITF), tyrosinase, tyrosinase-related protein (Trp)1, and Trp2 were analyzed in ${\alpha}$-MSH-untreated and ${\alpha}$-MSH-treated B16F10 cells. RESULTS: Treatment with 200 nM ${\alpha}$-MSH induced almost 2-fold melanin synthesis and tyrosinase activity along with increased mRNA levels and protein expression of MITF, tyrosinase, Trp1 and Trp2. Irrespective of ${\alpha}$-MSH stimulation, GEB extract at doses of 0.5-5 mg/mL inhibited all these markers for skin whitening in a dose-dependent manner. While lower doses (0.5-1 mg/mL) of GEB extract generally had a tendency to decrease melanogenesis, tyrosinase activity, and mRNA levels and protein expression of MITF, tyrosinase, Trp1, and Trp2, higher doses (2-5 mg/mL) significantly inhibited all these markers in ${\alpha}$-MSH-treated B16F10 cells in a dose-dependent manner. These inhibitory effects of the GEB extract at higher concentrations were similar to those of $400{\mu}g/mL$ arbutin, a well-known depigmenting agent. CONCLUSIONS: These results suggest that GEB displays dose-dependent inhibition of melanin synthesis through the suppression of tyrosinase activity as well as molecular levels of MITF, tyrosinase, Trp1, and Trp2 in murine B16F10 melanoma. Therefore, GEB may be an effective and natural skin-whitening agent for application in the cosmetic industry.
To find a novel skin whitening agent, the effect of cordycepin-enriched Cordyceps militaris (CM${\alpha}$) extract fermented by fungi on anti-melanogenesis in B16F0 mouse melanoma cells was investigated. Fermented CM${\alpha}$ was prepared with fungi, including Monascus purpureus (Mp), Aspergillus oryzae (Ao), Aspergillus kawachii (Ak), and Rhizopus oryzae (Ro), respectively. When the content of the phenolics and the flavonoids and the activities of the antioxidant and the mushroom tyrosinase inhibition were measured in the CM fermented by Ak (AkF-CM), the highest content of the phenolics was 46 mg/g dry weight and the highest content of the flavonoids was 0.93 mg/g; the highest activity of the DPPH radical scavenging was 62.74% and the highest activity of the mushroom tyrosinase inhibition was 79.97% CM${\alpha}$CM${\alpha}$. From this result, AkF-CM${\alpha}$ exhibited the highest mushroom tyrosinase inhibitory activity and so it was used in subsequent anti-melanogenesis. B16F0 melanoma cells were treated with 1-10 mg/ml concentrations of AkF-CM${\alpha}$ and 200 ${\mu}M$ arbutin as the positive control. The melanin content and cell viability of the melanoma cells by arbutin treatment decreased to 43% and 92% of the control, respectively. AkF-CM${\alpha}$ treatment at 1, 3, and 5 mg/ml concentrations decreased the extracellular melanin release induced by IBMX treatment by 35%, 45%, and 53%, respectively. AkF-CM${\alpha}$ showed inhibitory activity against both intracellular tyrosinase in melanoma cells and mushroom tyrosinase. AkF-CM${\alpha}$ reduced the protein level of tyrosinase in the IBMX-stimulated cells. These results indicate that AkF-CM${\alpha}$ suppressed the activity and protein content of cellular tyrosinase and decreased the total melanin content in cultured B16F0 melanoma cells.
The intention of this study was to confirm the possible use of an ethanol extracts of Nelumbinis Rhizomatis Nodus (NRN) as a cosmetic material. To this end, we extracted NRN with 70% ethanol and performed biological activity evaluation of whitening efficacy and wrinkle reduction. We performed cellular tyrosinase inhibition and melanin contents assay to check the whitening activity of NRN and carried out a toxicity evaluation of NRN via an MTT assay and the amounts of associated proteins that affect melanin production in a melanoma cell line (B16F10). And collagenase inhibitory assay was performed for the evaluation of anti-wrinkle of samples. In addition, a toxicity evaluation using an MTT assay and matrix metalloprotease (MMP-1) and procollagen synthesis inhibition by NRN were evaluated in a fibroblast cell line (CCD-986sk). Western blot results for the whitening activity evaluation revealed that the levels of two proteins related to melanin production, tyrosinase-related protein-1 (TRP-1) and tyrosinase-related protein-2 (TRP-2), were decreased in a dose-dependent manner. Moreover, collagenase inhibition activity at a concentration of $500{\mu}g/ml$ NRN by measuring epigallocatechin-3-gallate (EGCG) was increased by more than 80% compared to the control group. Meanwhile, procollagen synthesis was reduced by 68.8% in the UVB-induced CCD- 986sk cells group whereas collagen synthesis recovered by 80.2% with $25{\mu}g/ml$ NRN. The MMP-1 expression rate showed 20.2% reduction at $25{\mu}g/ml$. The results of the experiments verified the whitening and wrinkle suppression effects of NRN and confirmed that it could be used as a safe natural cosmetic material in the future.

This study was conducted as follows in order to find out the possibility of using (Chamaecyparis obtusa) by part as a cosmetic material. Chamaecyparis obtusa was separated leaves, bark, wood, and root, and extracted with 99.9% ethanol, and the antioxidant and whitening effects of the sample were confirmed. For this, the following studies were conducted. Antioxidant evaluation was confirmed by 1 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS+) radical scavenging activity. To evaluate the whitening effect, mushroom tyrosinase inhibitory activity evaluation and cytotoxicity evaluation of Chamaecyparis obtusa extract through MTT assay were conducted, and cellular tyrosinase inhibition rate was measured and melanin contents was confirmed. As a result of antioxidant activity, Bark extract among Chamaecyparis obtusa parts showed the best efficacy, and bark in B16F10 melanoma cell showed tyrosinase activity inhibition and melanin contents inhibitory effects. Therefore, it was confirmed that the ethanol extract of Chamaecyparis obtusa was developed as a natural material for safe and excellent whitening functional cosmetics.
