Inhibitory Effect on Melanogenesis of Ponciri Fructus Ethanol Extract (지실(枳實) 에탄올추출물의 멜라닌 형성 억제효과)

  • This study was to investigate the effect of Ponciri Fructus ethanol extract on the melanogenesis. The Ponciri Fructus markedly decreased melanin contents of B16F10 cells at a dose of $80{\mu}g/m{\ell}$. It was observed that the color of cell pellets was totally whitened compared with the control. The treatment of Ponciri Fructus inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis. Regarding protein levels of the melanogenic enzymes, the Ponciri Fructus dose-dependently decreased tyrosinase and TRP-1 protein levels, and tyrosinase and TRP-1 were detected in similar manner. These results suggest that the Ponciri Fructus ethanol extract exerts its depigmenting effects through the suppression of tyrosinase and TRP-1.

New Whitening agent: Kojyl-APPA

  • Exposure of the human skin to UV-light can cause sun-tanning, photoaging and even photo-carcinogenesis. Melanin is important in protecting the skin against UV damage, but excessive or uneven melanin production can lead to the formation of freckles and aged spot. Control of hyperpigmentation is becoming even more important as aged population continues to grow. These needs led us to develop effective and safe depigmenting-agent, kojyl 3-aminopropyl phosphate (kojyl-APPA), called Whitegen. The development of whitegen was based on the fact that phosphate group of 3-aminopropyl phosphate can make kojic acid more compatible to the skin membrane and more stable. Instability of kojic acid has been a problem in cosmetic use. The insertion of phosphoester group has been recognized as a powerful tool to improve such physical properties as solubility and stability, because the phosphodiester residue is well characterized as a non-toxic moiety, having a high affinity for cell membranes. Kojyl-APPA showed no tyrosinase inhibition effect compared to kojic acid in vitro, but showed tyrosinase inhibition effect in situ. It means that kojyl-APPA is converted to kojic acid enzymatically in cells. Kojyl-APPA showed the inhibitory activity on melanin synthesis in mouse melanoma and normal humal melnaocytes and also showed long-lasting stability in comparison with its original form (kojic acid). Kojyl-APPA showed depigmenting effects when applied to UVB-induced hyperpigmentated region of guinea pig skin. Based on these results, kojyl 3-aminopropyl phosphate can be used as a safe and effective ingredient for the brightness and cleanness of skin.

Effect of Mori Follium Extract on the Melanogenesis and Skin Fibril Matrix (상엽(桑葉) 추출물의 미백활성 및 피부섬유구조 형성에 미치는 효과)

  • Objectives : The Skin is composed of multiple layers, including the epidermis, dermis, and hypodermis. It provides a vital barrier structure that protects vertebrates from external environmental antigens, solvents, ultraviolet light, microorganisms, toxins, and weather conditions. Although several biological effects of Mori Follium have been reported, beneficial effects of Mori Follium in skin health remain unclear. In this study, we prepared water extract of Mori Follium (MLE) and evaluated the effects on melanin accumulation and expression levels of skin fibril-related proteins.Methods : The cytotoxicities of MLE in B16F10 melanoma and human skin fibroblasts (HSF) were examined by MTT assay. Inhibitory effect of MLE on the α-MSH- and IBMX-induced melanosis in B16F10 melanoma was examined. The expression levels of fibronectin, collagen 1α2, and CCN2 in MLE-treated HSF were analyzed by reverse transcription-polymer chain reaction (RT-PCR) and western blotting.Results : The MLE treatment for 24 h did not affect to the B16F10 and HSF at concentrations of 1, 10, 50, 100, 200, 400 and 800 ㎍/ml. The MLE treatment for 72 h significantly and dose dependently suppressed melanin accumulation in B16F10 melanoma. In addition, the MLE treatment up-regulated expression levels of skin fibril-related genes such as fibronectin, collagen 1α2, and CCN2 in HSF. Our western blot analysis revealed MLE-induced up-regulation of skin fibril-related genes required the activation of CCN2 protein.Conclusions : In conclusion, these findings suggest that the MLE could be used in development of cosmetic natural material of maintaining healthy skin.

Inhibitory Effect of Ethanol Extract and Juice of the Korean Cherry (Prunus tomentosa Thunberg) on Tyrosinase Activity In vitro (앵두과즙과 Ethanol 추출액의 In vitro에서 Tyrosinase 활성 저해효과)

  • To develop a functional beverage from Korean cherry (Prunus tomentosa Thunberg), inhibition effect of ethanol extract and juice of the korean cherry on melanin systhesis and tyrosinase activity in vitro was investigated. Inhibition ratio of tyrosinase activity increased as the concentration of solid of korean cherry juice increased, and inhibition affect was high in initiation step of enzyme reaction and then gradually decreased. Inhibition ratio of tyrosinase activity was high in the 70% (v/v) ethanol extract of the cherry and the highest in the ethyl acetate fraction of the 70% (v/v) ethanol extract. Ultimatly, the amounts of functional matter (melanin synthesis inhibitor) in the cherry was highest in ethyl acetate fraction of the ethanol extract.

Skin lightening effect of fermented Panax ginseng extract (자연삼 발효 추출물의 미백 활성에 대한 연구)

  • Panax ginseng is known for various pharmacological activities mainly due to saponins. Since minor saponins, generated by the decomposition of major saponins, generally exert higher activities than major saponins, the fermentation may increase the minor saponin contents in ginseng products. In this study, we tested fermented ginseng extract whether or not provide a safe cosmetic ingredient for whitening purpose. In this regard, fermented Ginseng extract was prepared and evaluated the inhibitory activity toward tyrosinase and the melanin synthesis suppression. The safety was tested via cell viability and toxicity test. The skin lightening effect was also evaluated by clinical study. The fermented Ginseng extract exerted higher activities in tyrosine inhibition and in suppressing melanin synthesis compared to Kojic acid and arbutin. In the clinical test, skin lightening effecte of the sample was clearly higher than vehicle or Vitamin C. We thus concluded that the fermented Ginseng extract may provide a safe cosmetic ingredient for skin lightening purpose.

Inhibitory Effect of Aged Black Platycodi Radix Extract on Expression and Activation of Matrix Metalloproteinases in Oxidative-stressed Melanoma Cells (쥐 흑색종 세포에서 산화적 스트레스에 의한 MMPs의 발현과 활성에 대한 흑도라지 추출물의 억제 효과)

  • The root of Playtcodon grandiflorum, called Platycodi radix, has been a favorite edible plant in Asia and contains a large amount of saponins. Melanoma cells (B16F10) were used to investigate the inhibitory effect of aged black Platycodi radix extract (ABPRE) on oxidative stress and matrix metalloproteinases (MMPs). Platycodon radix has been known to have a variety of medicinal effects such as prevention of gastric ulcers, antiallergenic activities, histamine release inhibition, and antioxidant effects. However, the mechanism of its action remains unclear in humans. ABPRE was prepared using ethanol extraction of aged black Platycodi radix. In an antioxidant effect study of ABPRE, it was observed that ABPRE specifically exhibited the scavenging activity of DPPH radical, but did not inhibit the production of malondialdehyde from lipid peroxidation. DNA oxidation was also blocked in the presence of ABPRE. In addition, ABPRE decreased the expression and activation of MMP-2 stimulated by phenazine methosulfate. Furthermore, ABPRE revealed the inhibitory effect on melanin production induced by L-dopa via antioxidant effect and the reduction of tyrosinase expression. Especially, the expression of antioxidant enzymes such as SOD-1 and SOD-2 regulated by Nrf2 was increased in the presence of ABPRE. Therefore, it appears that ABPRE may be a possible chemopreventive agent for the prevention of metastasis related to oxidative stress.

Antimelanogenic Effect of Purpurogallin in Murine Melanoma Cells (마우스 흑색종세포에서 Purpurogallin의 멜라닌 생성 억제 효과)

  • Melanin is one of the most important factors affecting skin color. Melanogenesis is the bioprocess of melanin production by melanocytes in the skin and hair follicles and is mediated by several enzymes, such as tyrosinase, tyrosinase related protein (TRP)-1, and TRP-2. Convenient enzymatic transformation of the simple phenol pyrogallol with polyphenol oxidase originating from pear to an oxidative product, purpurogallin, was efficient. The structure of the pyrogallol oxidation product was identified on the basis of spectroscopic methods. The biotransformation product purpurogallin showed significant inhibitory effects against both melanin synthesis and tyrosinase activity in a dose-dependent manner in B16 melanoma cells. In addition, purpurogallin significantly attenuated melanin production by inhibiting TRP-1, and TRP-2 expression through modulation of their corresponding transcription factors, and microphthalamia- associated transcription factor in B16 cells. Consequently, purpurogallin derived from convenient enzymatic transformation of pyrogallol might be a beneficial material for reducing skin hyperpigmentation.

Anti-aging Effects of Prescription Extracts Containing Forsythia viridissima L. (연교를 함유한 처방단 추출물의 항노화 효과)

  • Forsythia fructus has been shown to have antioxidative, anti-inflammatory, antibacterial, anti-aging and whitening effects. This work was carried out to investigate the anti-aging effects of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) on skin. Skin anti-aging effect of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) was evaluated by using antioxidant assay, anti-glycation activity, inhibitory effect of tyrosinase activity, expression of type I procollagen and UVA-induced matrix metalloproteinase-1 on HS68 cells, and reduction of ${\alpha}$-MSH-induced melanin on B16F1 cells. Forsythia viridissima-prescription extracts showed anti-oxidative and anti-glycation activity. The pectinex hydrolysed extract from Yeongyoseungma-tang 75% EtOH extract and Gamiyeongyoseungma-tang 75% EtOH extract increased the type I procollagen synthesis, and decreased the UVA-induced MMP-1 expression on HS68 cells. The pectinex hydrolysed extracts of Hoechunyangkyeok-san 75% EtOH extract had the inhibitory effect of melanin synthesis on B16F1 cells. Based on these results, we suggest that Forsythia viridissima-prescription extracts may be useful as a potential source of functional anti-aging cosmetics.

A Study on the Antioxidant, Whitening and Anti-Inflammatory Activities of Trichosanthis Cucumeroidis Radix Extract (쥐참외뿌리 추출물의 항산화, 미백, 항염증 활성 연구)

  • In this study, the physiological activity effect of trichosanthis cucumeroidis radix extract on antioxidant activity, whitening, and anti-inflammatory activity was checked, and the possibility of its use as a functional material was checked. The purpose of this study was to confirm the antioxidant activity through DPPH radical scavenging activity of trichosanthis cucumeroidis radix extract, whitening activity effect through melanin production inhibition ability for melanin cell B16F10 melanoma cell, and anti-inflammatory activity effect through NO production inhibition ability for macrophage RAW 264.7 cell. As a result of the study, the concentration-dependent DPPH radical scavenging activity of the trichosanthis cucumeroidis radix extract was confirmed, and DPPH radical scavenging activity similar to that of the positive control Ascorbic acid was confirmed. It was confirmed that the melanin production inhibitory activity induced by 100 nM 𝛼-MSH and the NO production inhibitory ability induced by LPS 1 ㎍/mL were significantly suppressed. Accordingly, it is considered that the trichosanthis cucumeroidis radix extract may be used as a functional material having antioxidant, whitening, and anti-inflammatory effects.

The Study on the Whitening Effects of Traditional Chinese Medicines (중의 약재로부터 피부 미백제의 탐색)

  • In order to search for new whitening cosmetic ingredients from Chinese herbal extracts including Chinese herbs and complex prescriptions from TCM (Traditional Chinese Medicine), we screened about 47 TCM extracts collected from China. We tested their inhibitory effects on melanogenesis by using in vitro tyrosinase inhibition assay and B16 melanoma cells. We selected Siphonostegia chinensis and Salvianic miltiorrhiza Bunge. We isolated Danshensu ($\alpha$,3,4-trihydroxybenzenepropanoic acid sodium salt) from Salvianic miltiorrhiza Bunge extract and tested its inhibitory effect on melanin formation in B16-F10 melanoma cells. Danshensu suppressed melanin synthesis up to about 50% at a concentration of $100\;{\mu}g/mL$. Siphonostegia chinensis suppressed melanin synthesis up to about 60% at a concentration of $300\;{\mu}g/mL$. The results showed that these extracts could be used as new natural active ingredients for whitening cosmetics.
