The Problem on Riboflavin Content Inference of Common Foods for Korean (한국인 상용식품중의 리포블라빈 함량추정에 관한 문제점)

  • In order to study on the riboflavin content of common foods for Korean and the rates of destruction of riboflavin during cooking 26 kinds of the foods were selected and 3 kinds of menu were cooked by standardized method. For each food item and menu riboflavin content was measured by AOAC method. The experimental values of 13 kinds of food such as rice oak mushroom carrot squarsh tangle dried large anchovy apple(Fuji) dried laver ramyun pork soybean curd fried soybean curd and danmugi were almost consistent with food compo-sition values. Whereas those of 12 kinds of foods such as cabbage onion potato kimchi beef sausage dried medium anchovy hair tail soybean paste and egg were considerably different from food composition table values, up to now Alaskan pollack maize loaf bread hamburger bread etc have not been analyzed in food composition table, The rates of retention of riboflavin in menu 1, 2, and 3 cooked by standardized method were 24% 69% 46% respectively. The overally retention rate was in inverse proportion to the time of sunlight exposure during cooking.

  • Most of the surface/interface analysis tools have limited depth profiling c capability in terms of the profiling range and the depth resolution. However, M MEIS can profile the surface and subsurface composition and structure q quantitatively and non-destructively with atomic layer depth resolution. I In this presentation, the MEIS system developed at KRISS will be briefly d described with an introduction on the principle of MEIS. Recent MEIS r results on the surface and interface composition and structural change due to i ion bombardment will be presented for preferential sputtering of T:없Os and d damage depth profiles of SHooD, Pt(l11), and Cu(l1D due to Ar+ ion b bombardment. Direct observation of strained Si lattices and its distribution i in the SHool)-SiCh interface and the initial stage of Co growth on Pt(l11) w will be reported. H surfactant effects on epitaxial growth of Ge on Si(ooD w will be discussed with STM results from SND.

Production of Volatile Oil Components by Cell Culture of Agastache rugosa O. Kuntze

  • To develop systems for economic production of useful essential oil compounds, callus was induced from the seedlings of Agastache rugosa and cultured on MS medium. The volatile oil fraction was extracted from the callus and investigated by mean of GC-MS. The composition of the oil was compared with that of the mother plant. As a result, sixty five compounds including ferruginol were identified in the essential oil fraction. The main component of the oil from the leaves of Agastache rugosa was methyl chavichol (53.6%). Methyl jasmonate and jasmonic acid were added to the culturing cell suspension, separately and the composition of induced oil were compared. The oils from cultured cells treated with jasmonates showed considerably different patterns. Especially, the peak of estragole was found in callus oil after treatment with methyl jasmonate as though the amount was limited to 0.58%. In general, the TIC pattern of GC-MS of the callus oil became more similar to the oil from the leaves after elicitation.

  • This study is to investigate the ethanol tolerance of yeast strains related to fatty acid composition and intracellular ethanol concentration for various fermentation temperatures. The maximum accumulation of ethanol in the cells was decreased by lowering the fermentation temperature, while unsaturated fatty acid content was increased by decreasing the fermentation temperature. Thus, we found that the increase of ethanol accumulation in cells resulted in the decrease of unsaturated fatty acid content. Therefore, it was suggested that the composition of unsaturated fatty acids in the cell membrane be strongly related to the diffusion of ethanol from cell to medium.

  • Recently, every hospital requires the creative and novel design which is different from the conventional concept to deal with various circumstances. Accordingly, various space arrangements are proposed against the past uniform hospital type. But the sufficient studies about medical facilities in small and medium city and countryside still lack, otherwise there is short but continuous accumulation of data about the study for general hospitals. In this sense, this study generalizes the present medical condition in Ansan city and analyzes the spatial composition and area distribution in departments for the purpose of presentation of fundamental data about medical policy and architecture in Ansan city.

  • Spectroscopic ellipsometry(SE) was employed to characterize the Si/$Si_{1-x}Ge_x$ heterostructure. The dielectric function spectrum of $Si_{1-x}Ge_x$ at an arbitrary x value in the spectral range of $1.5{\sim}4.5\;eV$ was computed by EMA (effective medium approximation) model using the available optical constants measured at a number of fixed x values of Ge composition. The thickness and the Ge composition of $Si_{1-x}Ge_x$ measured by SE was compared with those measured by RBS. DC bias effect on the $E_2$ peak of dielectric function spectra was studied.

Production of Hyaluronic Acid from Streptococcus zooepidemicus (Streptococus zooepidemicus에 의한 히아루론산의 생산)

  • An optimal composition of medium for hyaluronic acid production and some characteristics of its from Streptococcus zooepidemicus were investigated. The hyaluronic acid from S. zooepidemicus was reached maximum level in the BY-medium containing 0.1% beef extract, 0.1% yeast extract, 3.0% glucose, 2.0% peptone, 0.1% NaCl and $0.5%CaCO_3$ (pH 7.5) at $37^{\circ}C$ for 36 hours with shaking. Addition of $CaCO_3$ to the medium was necessary to neulralize the lowered pH which was resulted from hyaluronic acid production. Molecular weights of extracelluar and cellular hyaluronic acid produced by the strain were $1-1.4{\times }10^6$ and $5{\times}10^6$, respectively. The amount of extracellular hyaluronic acid was 91.9% of total hyaluronic acid produced and the vest was all intracellular.

  • As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4\;1.0g, \;sucrose\;30.0g, \;MgSo_4{\cdot}7H_2O\;0.5g, \;FeCl_3{\cdot}6H_2O\;1.0mg\;thiamine\;200{\gamma}g$, and distilled water 1000ml, pH6.5.

  • In 1990s, the necessity of the Korean contemporary medium/small scale gallery as a place of experience rather than that of simple viewing has increased to meet from the requirements of the pluralistic and popularized society. The gallery makes efforts to form a collective street to cope with the influential of the power large scale cultural space as well as finding a novel direction in order to derive the regional sympathy in connection with the culture space of the neighborgood. In this paper, we study the 1990s Korean medium/small scale gallery in two aspects: First, the architectural techniques and elements which are introduces the audience to both interior and exterior spaces constructed by the 1990s gallery architecture, are investigated. The conventional architecture shows the blocking of the neighborhood and the trend of severance when viewing from both the architectural and the civil points. Secondly, we present the design characteristics from the several points of view such as conceptual, spacial, visual angles. The various trends of works in exhibition of the fine arts can provide a means for producing a great deal of variation in recent gallery architecture when viewing from a stage standpoint of exhibition space.

Production of Cellulase and Xylanase by Aspergillus niger KKS

  • A fungal strain capable of producing extracellular cellulase was isolated from farmland. It was identified as Aspergillus niger, and named Aspergillus niger KKS. Production of cellulase and xylanase by the A. niger KKS was studied through a shake-flask culture. The effects of culture conditions such as inoculum size, temperature, pH, and medium composition on the cellulase and xylanase production were examined. The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 7.0, respectively. The optimized medium was composed of 2.0% (w/v) rice straw, 0.5% (w/v) proteose peptone, 0.5% (w/v) $KH_2 PO_4$, 0.05% (w/v) yeast extract, 0.01% (w/v) $CoSO_4 \cdot 7H_2O$, and 0.05% (w/v) $CuSO$_4$\cdot 5H_2O$. When the strain was incubated with the optimized medium, it gave the activities of endoglucanase, $\beta$-glucosidase, $\beta$-xylosidase, xylanase were 3.80, 4.20, 4.00, 80.0 (IU/mL), respectively. Filter paper and cotton activities were 0.68 and 0.045 (IU/mL), respectively. The results of this study show that A. niger KKS is a potential organism with a wide spectrum of enzyme activities, such as those of $\beta$-glucosidase, $\beta$-xylosidase, and xylanase.
