A Study on Development of the Optimization Algorithms to Find the Seam Tracking (용접선 추적을 위한 최적화 알고리즘 개발에 관한 연구)

  • The Gas Metal Arc(GMA) welding, called Metal Inert Gas(MIG) welding, has been an important component in manufacturing industries. A key technology for robotic welding processes is seam tracking system, which is critical to improve the welding quality and welding capacities. The objectives of this study were to develop the intelligent and cost-effective algorithms for image processing in GMA welding which based on the laser vision sensor. Welding images were captured from the CCD camera and then processed by the proposed algorithm to track the weld joint location. The proposed algorithms that commonly used at the present stage were verified and compared to obtain the optimal one for each step in image processing. Finally, validity of the proposed algorithms was examined by using weld seam images obtained with different welding environments for image processing. The results proved that the proposed algorithm was quite excellent in getting rid of the variable noises to extract the feature points and centerline for seam tracking in GMA welding and could be employed for general industrial application.

Production of Adventitious Root and Analysis of Effective Components from in vitro Culture of Astragalus membranaceus (기내배양을 통한 황기 부정근의 생산과 유효성분 분석)

  • Background : A series of studies were conducted to optimize adventitious root induction in vitro from explants of Astragalus membranaceus using various nutrient media supplemented with plant hormones. Methods and Results : Levels of active components were analyzed from adventitious roots induced under different media conditions. Among the different media conditions, Murashige and Skoog medium supplemented with $1.0mg{\cdot}{\ell}^{-1}$ indole-3-butyric acid resulted in the greatest adventitious root induction rate. The amount of the major active component of the adventitious roots of Ama1, calycosin-7-O-${\beta}$-D-glucopyranoside was higher than that of other adventitious root samples. Conclusions : These results suggest that the adventitious roots of A. membranaceus could be used for the commercial production of medicines.

Biological control of Colletotrichum panacicola on Panax ginseng by Bacillus subtilis HK-CSM-1

  • Background: Biological control of plant pathogens using benign or beneficial microorganisms as antagonistic agents is currently considered to be an important component of integrated pest management in agricultural crops. In this study, we evaluated the potential of Bacillus subtilis strain HK-CSM-1 as a biological control agent against Colletotrichum panacicola. Methods: The potential of B. subtilis HK-CSM-1 as a biological control agent for ginseng anthracnose was assessed. C. panacicola was inoculated to ginseng plants and the incidence and severity of disease was assessed to examine the efficacy of the bacterium as a biological control against C. panacicola. Results: Inoculation of Panax ginseng plants with B. subtilis significantly suppressed the number of disease lesions of C. panacicola and was as effective as the chemical fungicide iminoctadine tris(albesilate). The antifungal activity of B. subtilis against C. panacicola was observed on a co-culture medium. Interestingly, treatment with B. subtilis did not significantly affect the diameter of the lesions, suggesting that the mechanism of protection was through the reduction in the incidence of infection related to the initial events of the infection cycle, including penetration and infection via spore germination and appressorium formation rather than by the inhibition of invasive growth after infection. Conclusion: Our results suggest that B. subtilis HK-CSM-1 can be used as an effective and ecologically friendly biological control agent for anthracnose in P. ginseng.

Sensory Quality Characteristics of Colombia Coffee under Various Processing and Roasting Conditions of Green Beans (콜롬비아 커피 생두의 가공법과 로스팅에 따른 품질 특성)

  • The objective of this study was to investigate the sensory quality characteristics of coffee under various processing and roasting conditions of green beans. Colombia green bean samples were obtained from three processing methods: natural, pulped-natural, and washed methods. The green beans were roasted differently according to Agtron number: light (Agrton #65), medium (Agtron #55), and dark (Agtron #45). Moisture content, density, color value, pH, and total dissolved solid contents (TDS) were measured, and quantitative descriptive analysis (QDA) with 23 sensory attributes was performed for nine coffee samples. Hunter L values were significantly elevated with the degree of processing but reduced with roasting intensity. The pH values of the nine coffee samples were 5.6~6.2 and significantly increased with intensity of roasting. TDS decreased with an increase in processing and increased with roasting intensity. QDA results show that smoky aroma and nutty and chocolate flavor significantly increased while fruit and floral aroma decreased with intensity of processing. The attributes of brownness, oily surface, smoky aroma, bitter and burnt flavor, and greasy and heavy mouthfeel significantly increased while fruity and floral aroma, black tea, sour and umami flavor, pureness, and softness decreased as roasting intensity increased. Principle component analysis showed the overall significant relationships between 23 sensory attributes and nine coffee samples under different processing and roasting conditions.

  • In shipbuilding industry, the manufacturing technology has advanced at an unprecedented pace for the last decade. As a result, many automatic systems for cutting, welding, etc. have been developed and employed in the manufacturing process and accordingly the productivity has been increased drastically. Despite such improvement in the manufacturing technology, however, development of an automatic system for fabricating a curved hull plate remains at the beginning stage since hardware and software for the automation of the curved hull fabrication process should be developed differently depending on the dimensions of plates, forming methods and manufacturing processes of each shipyard. To deal with this problem, it is necessary to create a "plug-in" framework, which can adopt various kinds of hardware and software to construct a full automatic fabrication system. In this paper, a framework for automatic fabrication of curved hull plates is proposed, which consists of four components and related software. In particular the software module for computing fabrication information is developed by using the ooCBD development methodology, which can interface with other hardware and software with minimum effort. Examples of the proposed framework applied to medium and large shipyards are presented.

Some Chromosome Alteranations in the Cultured Chinese Hamster Cells Treated by Steroids (Steroid 물질처리를 받은 Chinese hamster 세포에 있어서의 염색체 이상)

  • This study is concerned with alterations in chromosomes (numbers and morphology) when the culture of Chinese hamster cells (FAF-28 strain) was treated by steroids, testosterone and DOC. 1. In 200 cells of normal untreated cells as control population the chromosome of stemline was decided as which was contained in 158 cells ; that is , in 79 percent of the population. The average chromosome number in above 20 cells observed was calculated as 23.95 with minimum limit at 20 and maximum limit at 70. 2. Many different chromosome numbers, ranging from 19 to 352 were observed in the 200 cells treated by testosterone. The diploid number of 22 showed the peak of variation curve was counted in 71 cells (35.5%) and an average chromosome number of stemline was 22 which was counted in 74 cells (37%). While all of the chromosome number of stemline was 22 which was counted in 74 cells (37%). While all of the chromosome numbers in the 200 cells observed ranged from 20 to 181 , an average chromosome number was also found to be 30.09. 4. The chromosome component in the cultured normal FAF-28 cells with 22 diploid chromosomeswas as follows ; 9a) 2 paris were long and metacentric (LM), (b) 3 pairs were medium length and metacentric (MM), (c) 3 pairs were small and subtelocentric (SS) and (d) 3 pairs were small and metacentric (SM). 5. The twenty cells with 44 chromosomes were selected at random from each cell population treated with testosterone and DOC , so that chromosome idiogram and morphology could be studies. In the twenty cells of the testosterone treated population the average ratio of above four groups, LM ; MM;Ss:SM, was found to be 8.6 : 10.8:13.5:10.7. On the other hand, the average ratio in the same number of cells of the DOC treated one was 7.7 :11.4:12.5:12.7. 6. The five types of the altered chromosomes morphologically in the hundred cells selected at random from each cell population treated by testosterone and DOC were observed (Type I-V). The thirty-one altered chromosomes were found to be in the testosterone treated cell population and the sixteen in DOC treated.

Estimation of Pyrolysis Properties for Fire Propagation Analysis of Furniture Materials (가구소재의 화재전파해석을 위한 열해리 물성 평가)

  • The present study has been conducted to investigate the reaction kinetics and pyrolysis parameters for flame propagation analysis of furniture material components. TGA measurement for component materials such as MDF (medium density fiberboad) panel including coating material, synthetic leather and foam cushion are performed under maximum temperature of $600^{\circ}C$ and heating rate of $10^{\circ}C/min$. The results of TGA have shown that the peak temperature of MDF panel was $324^{\circ}C$ and the initial peak temperature of coating material decreased by $270{\sim}280^{\circ}C$. In the case of synthetic leather and foam materials, the reference temperature and reference rate depend on the type of polymer consisting the sample, the initial kinetic characteristics was classified into 2 categories of about $270^{\circ}C$ and $420^{\circ}C$ of reference temperature for the tested synthetic materials. The present study showed the pyrolysis parameters of reference temperature and reference rate proposed by Lyon to evaluate the pre-exponential factor and activation energy. The present study can contribute to improve the reliability of computational fire analysis and enhance the understanding of fire propagation phenomena based on the thermal properties study of material.

Phenol-Rich Compounds Sweet Gel: A Statistically More Effective Antibiotic than Cloxacillin Against Pseudomonas Aeruginosa

  • Objectives: The purpose of this study was to obtain a natural antibiotic from Phenol-rich compounds; for the dressing and the treatment of chronic wounds. Methods: The Phenol-rich compound sweet gel was prepared by blending four natural herbal extracts, Acacia catechu (L.F.), Momia (Shilajit), Castanea sativa, and Ephedra sinica stapf, with combination of a sweet gel medium, including honey, maple saps, Phoenix dactylifera L. (date), pomegranate extract and Azadirachta indica gum as a stabilizer. The combinations were screened by using a well-diffusion assay with cloxacillin as a control. Pseudomonas spp. was tested with our novel antimicrobial compound. The zones of inhibition in agar culture were measured for each individual component and for the compound, and the results were compared with those of the control group which had been treated with cloxacillin. Data were expressed as means ${\pm}$ standard deviations. Quantitative analyses were performed using the paired t-test. Results: The antibiotic effect of the Phenol-rich compound sweet gel was statistically shown to be more significant than that of cloxacillin against Pseudomonas aeruginosa (P < 0.05). Conclusion: Our novel approach to fighting the antibiotic resistance of Pseudomonas proved to be successful. The Phenol-rich compound sweet gel was found to be suitable for use as an alternative medicine and bioactive dressing material, for the treatment of patients with various types of wounds, including burns, venous leg ulcers, ulcers of various etiologies, leg ulcers on the feet of diabetic, unhealed graft sampling sites, abscesses, boils, surgical wounds, necrotic process, post-operative and neonatal wound infection, and should be considered as an alternative to the usual methods of cure.

Study of four weeks repeated-dose toxic test of Sweet Bee Venom in rats Original Articles (Sweet Bee Venom의 rat를 이용한 4주 반복 근육시술 독성시험)

  • Objective: This study was performed to analyse four weeks repeated -dose toxicity of Sweet Bee Venom (SBV-pure melittin, the major component of honey bee venom) in rats. Methods: All experiments were conducted under the regulations of Good Laboratory Practice (GLP) at Biotoxtech Company, a non-clinical study authorized institution. Male and female rats of 5 weeks old were chosen for the pilot study of four weeks repeated-dose toxicity and was injected at the level of 0.56 mg/kg body weight (eighty times higher than the clinical application dosage as the high dosage), followed by 0.28 and 0.14 mg/kg as midium and low dosage, respectively. Equal amount of normal saline was injected as the control group every day for four weeks. Results: 1. No mortality was witnessed in all of the experiment groups. 2. All experiment groups appealed pain sense in the treating time compared to the control group, and side effects such as hyperemia and movement disorder were observed around the area of injection in all experiment groups, and the higher dosage in treatment, the higher occurrence in side effects. 3. Concerning weight measurement, neither male nor female groups showed significant changes compared to the control group. 4. Concerning to the CBC and biochemistry, all experiment groups didn't show any significant changes compared to the control group. 5. Concerning weight measurement of organs, experiment groups didn't show any significant changes compared to the control group. 6. To verify abnormalities of organs and tissues, those such as cerebellum, cerebrum, liver, lung, kidney, and spinal cords were removed and we conducted histologocal observation with H-E staining. Concerning the histologocal observation of liver tissues, some fatty changes were observed around portal vein in 0.56 mg/kg experiment group. But another organs were not detected in any abnormalities. 7. The proper high dosage of SBV for the thirteen weeks repeated test in rats may be 0.28 mg/kg in one time. Conclusion: Above findings suggest that SBV is relatively safe treatment medium. Further studies on the subject should be conducted to yield more concrete evidences.

Macroscopic High-Temperature Structural Analysis Model of Small-Scale PCHE Prototype (II) (소형 PCHE 시제품에 대한 거시적 고온 구조 해석 모델링 (II))

  • The IHX (intermediate heat exchanger) of a VHTR (very high-temperature reactor) is a core component that transfers the high heat generated by the VHTR at $950^{\circ}C$ to a hydrogen production plant. Korea Atomic Energy Research Institute manufactured a small-scale prototype of a PCHE (printed circuit heat exchanger) that was being considered as a candidate for the IHX. In this study, as a part of high-temperature structural integrity evaluation of the small-scale PCHE prototype, we carried out high-temperature structural analysis modeling and macroscopic thermal and elastic structural analysis for the small-scale PCHE prototype under small-scale gas-loop test conditions. The modeling and analysis were performed as a precedent study prior to the performance test in the small-scale gas loop. The results obtained in this study will be compared with the test results for the small-scale PCHE. Moreover, these results will be used in the design of a medium-scale PCHE prototype.
