Effects of Medium Experience on Medium Perception and Communication Process (텍스트매체 사용에 있어서 매체 경험이 매체 인지와 의사소통과정에 미치는 영향)

  • The objective of this study is to examine the media richness theory and the social information processing model by analyzing the effect of media experience on media perception and communication process. To accomplish this objective, a laboratory experiment was conducted. The independent variable was text medium experience and a face-to-face medium was added as a control group. The dependent variables were medium perception and communication process. Medium perception includes perceived richness, medium feeling, task satisfaction, and communication satisfaction. Communication processes were also analyzed to compare each treatment group. The results can be summarized into two facts. First, face-to-face group showed higher perceived richness than text medium group. And experienced text medium group perceived their text medium richer than inexperienced text medium group. Second, experienced text medium groups showed more interactions between subjects than inexperienced text medium group. Experienced text medium group also showed more agreements and meta-communication which could be found in face-to-face group. The result of this study supported media richness theory by finding that face-to-face medium was perceived richer than text medium, And the results also proved social information processing model by comparing experienced text medium group and inexperienced text medium group. The text medium, although thought to be the leanest one, could be perceived richer if users had lots of experience on it.

Growth and Pigment Formation of Genus Monascus on Medium Compositions (배지에 따른 Genus Monascus의 증식도와 색소함량)

  • Growth and pigment formation in Genus Monascus(M. pilosus and M purpureus) related 15 kinds of culture media(Lin medium, SP medium, YM medium, YE medium, GMIN medium, SMO medium, MY medium, GY medium, Nishikawa medium, sucrose medium, stock culture, Mizutani medium, modified Lin medium, Toya medium and rice medium) were investigated. Mizutani medium and Lin medium among 15 kinds of the culture media showed good growth for M. pilosus, M purpureus, fresh mycelium weight cultivated for 10 days at 30$^{\circ}C$ was each 24.5∼26.9 or 15.9∼17.2 g/100 mL. The culture media which showed higher content of pigment production in two fungi were Lin medium(OD: 1.2 ∼ 1.6 and Mizutani medium(OD: 0.8 ∼ 1.0) that showed higher in M. pilosus.

Effects of CT Contrast Medium on the Relaxation Rate of MR Contrast Medium (CT 조영제가 MR 조영제의 이완율에 미치는 영향)

  • In MR, the iodine CT contrast medium reduces the T1 and T2 relaxation times of the substance, resulting in a change in signal intensity. This study aimed to measure the relaxation rate of MR contrast medium with or without diluting CT contrast medium and analyzed the effect of CT contrast medium. Undiluted Gadoteridol solution was diluted with saline to prepare MR contrast medium phantoms with various levels of Gadoteridol concentrations. Moreover, undiluted Iomeprol was mixed with the prepared MR contrast medium phantoms at 1:1 ratio to make MR contrast medium phantoms with containing CT contrast medium for the experiment. T1 and T2 mappings were conducted to quantitatively evaluate the relaxation time and relaxation rate of these phantoms. The results showed that the T1 and T2 relaxation time and relaxation rate of MR contrast medium diluted with CT contrast medium were significantly (p<0.05) shorter than those of MR contrast medium not diluted with CT contrast medium. The results of this study imply that, when MR contrast medium shall be used after injecting CT contrast medium, CT contrast medium should be discharged enough. Moreover, it would be desirable to conduct CT test after taking MRI test in order to reduce the effects of CT contrast medium on MR contrast medium.

  • Eubacterium is one of the predominant bacteria in the human large intestine. currently ES (Eubacterium Selective) medium developed by T. Mitsuoka is commonly used as a selective medium. neomycin sulfate which is one of the selective agents of ES medium inhibited about 50% of the growth of Eubacterium isolated, whereas malidixic acid inhibited only 5% while inhibiting other intestinal bacteria. NES medium which replaced neomycin with nalidixic acid in the ES medium was designed and shown to be better for the isolation and enumeration of Eubacterium sp. than ES medium.

  • Effects of the special media on the mycelium growth in Agaricus campestris has been studied. The results are summarized as follows; 1. The mycelium growth of Agaricus campestris were considerably Stimulated on the Carrot (Dancus carota L.) basal medium which was added 4ml. of carrot extract, Cucumber(Cucumis sativus L.) basal medium added 3ml of cucumber extract, and Radish (Rhaphanus sativus L.) basal medium added 3ml. of radish extract during the culture for 144 hours. 2. The mycelium of Agaricus campestris on the media which was added the several kinds of vegetable extracts shows a lot of growth for 144 hours. The orders are as follows; Carrot basal medium(4ml/100ml)>Cucumber basal medium (3ml/100ml)>Radish basal medium (3ml/100ml)>Lettuce basal medium (2ml/100ml)>Cabbage basal medium (2ml/100ml). However, the Lettuce (Lactuca scariota L.) basal medium and the Cabbage (Brassica chinensis L.) basal medium among these five media are no significant differences.

Effect of Oviductal Fluid and Oviductal Conditioned Medium on Polyspermy and In Vitro Development of Porcine Oocytes (돼지 난관액과 Oviductal Conditioned Medium 이 다정자침입과 체외배발달에 미치는 영향)

  • The objective of this study was to dertermine the effects of oviductal fluid and oviductal conditioned medium on polyspermy and in vitro development of porcine oocytes. The addition of oviductal fluid and oviductal conditioned medium in the prefertilization and fertilization medium significantly decreased polyspermy rates and the mean number of spermatozoa in penetrated eggs(P<0.05). The acrosome reaction rate significantly increased when spematozoa were exposed for 1.5, 3, 4.5h in oviductal fluid and oviductal conditioned medium(P<0.05). When oocytes cultured for 192h, the percentage of oocytes that developed to the morula and blastocyst stage was higher in culture medium with oviductal fluid and oviductal conditioned medium than without oviductal fluid and oviductal conditioned medium(P<0.05). These results indicated that the oviductal secreations will effectively reduce both the polyspermy rates and the mean number of spermatwa in penetrated eggs. And the presence of culture with oviductal fluid and oviductal conditioned medium promotes in vitro development of porcine oocytes.

Morphological Variations in Conidia of Arthrobotrys oligospora on Different Media

  • Most commonly occurring predacious fungus Arthrobotrys oligospora showed great variation in size and shape of conidia on some media. The formation of larger conidia was recorded on beef extract and nutrient agar media. The length of conidia in Richard's YPSS, Sabouraud's, PDA and com meal agar media was of medium size while smaller conidia were produced on Czapek's, Jensen's, Martin's medium. Maximum width of conidia was recorded on YPSS medium followed by Sabouraud's medium. The average size of spores on nematode infested com meal agar medium was slightly increased than those on com meal agar medium.

Effects of culture media conditions on production of eggs fertilized in vitro of embryos derived from ovary of high grade Hanwoo

  • Background: This study was investigated the effects of culture media conditions on production of eggs fertilized in vitro of embryos from ovaries of high grade Korean native cow, Hanwoo. Methods: The IVMD 101 and IVF 100 were used for in vitro maturation of selected Hanwoo oocytes and In vitro embryo culture after in vitro fertilization, respectively. The IVMD 101 and IVD 101 were used for in vitro culture and completely free of serum. Results: The cleavage rates of 2-cell embryos in reference to Hanwoo oocytes were 86.7, 92.9, and 90.1 % in the control group, IVDM101 medium and IVD101 medium, respectively which indicates that the IVDM101 medium and IVD101 medium may result favorable outcomes. The in vitro development rates of blastocysts were 12.4, 38.4 and 32.4 % in the control group, serum free IVMD101 medium and IVD101 medium, respectively. For hatched blastocysts, it was 5.3, 33.9, and 28.6 % in the control group, serum free IVMD101 medium and IVD101 medium, respectively. Hence, more favorable results were expected for the hatched blastocysts in which the IVMD101 medium and IVD101 medium were used than the control group. Average cell numbers of blastocysts were 128.3, 165.7, and 163.6 in the groups of TCM-199 + 10 % FBS medium, IVMD 101 medium, and IVD 101 medium, respectively which clearly show that the IVMD 101 and IVD 101 medium consequence significantly higher cell numbers compared to the control group (i.e., TCM-199 +10 % FBS medium). Pregnancy rate after embryo transfer was 39.6 % when the serum free medium was used which is higher than that of the medium supplemented with serum (32.8 %). In addition, stillbirth rates were 4.9 % in the group of serum free medium whereas it was 13.6 % in the serum supplemented medium (13.6 %). Conclusions: Taken altogether, serum free media, the IVMD 101 and IVD 101 represented more favorable results in the embryo development rate of embryos, cell numbers of blastocyst, and pregnancy rate. Of note, the IVMD 101 medium showed better outcomes hence, it might be a better option for future applications for in vitro culture of bovine embryos.

  • Most of seismic reflection prospecting assumes subsurface formation to be homogeneous media. These models are not capable of estimating small scale heterogeneity which is verified by well log data or drilling core. And those synthetic seismograms by homogeneous media are limited to explain various changes at field data. So we developed a inhomogeneous velocity model which can estimate inhomogeneity of background medium to implement numerical modeling from homogeneous medium and inhomogeneous medium on the model. Background medium using three autocorrelation functions in order to generate inhomogeneous velocity media was according to dominant wavelength of background medium and correlation length of random medium. And then we compared shot gathers. The results show that numerical modeling implemented at inhomogeneous medium depicts complex wave propagation of field data.

Serum Free Medium Development for Recombinant Erythropoietin Production using Novel Cell Line (QT35) (QT35 세포주에서 제조합 에리스로포이에틴 생산을 위한 무혈청 배지의 개발)

  • Human Erythropoietin (EPO) gene is cloned in quail fibrosarcoma cell, QT35. Because molecular weight of EPO is similar to that of serum albumin, cell culture with serum containing medium makes purification of EPO very difficult. Using fractional factorial study, we have developed serum free medium for the recombinant QT35 cell lines, QT N4D4 and QT SY-IMP, which have cloned EPO with glutamine synthetase (GS) gene amplification system and with puromycin selective marker, respectively. Among the seven frequently used medium components, fibronectin, BSA, and EGF were the most important for EPO production. However, sufficient fibronectin supplement to the medium did not make any good attachment of QT35 to culture plate over 3 days. Therefore, to maximize EPO production, we attempted a medium-shift at confluence from serum containing medium to serum free medium(QT SFM6). Using the medium-shift protocol with QT SFM6, nearly the same productivity of EPO was achieved comparing with that without medium-shift. This result was true in both QT35 cell lines in three types of culture, i.e. T flask, microcarrier and roller bottle cultures.
