  • Petiole explant of Glehnia littoralis Fr. Schmidt was in vitro cultured MS plant medium(DUCHEFA co.) supplemented with various plant growth regulators and examined to find out their optimum combination and concentration for plantlet regeneration. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on MS plant medium with various kinds of plant growth regulators. Embryogenic calli and adventitious shoot formation were greatly influenced by plant growth regulators. Embryogenic calli induction showed a good response on MS medium supplemented with NAA and BA than others. Especially, combination of 1.0 mg/L NAA and 0.5 mg/L BA on MS medium led to the greatest frequency in adventitious shoot. The results suggest that plant regulator selection be important factor to achieve an efficient regeneration.

  • Various kinds of leaf spots and blights were found in kiwifruit (Actinidia deliciosa) orchards on 2006 in Korea. Disease incidences were quite variable between open-field and rain-proof shelter. Rate of diseased leaves was recorded as about 70% at open-field orchards in late season but use of rain-proof vinyl shelters alleviated the disease incidences by 20%. Angular leaf spots appeared at early infection stage on June and several other symptoms were also recognized as the disease developed afterward. On September, brown leaf blights were the most frequent, followed by grayish brown ring spots, silvering gray leaf blights, zonate leaf blights, dark brown ring spots and angular leaf spots at open-field orchards. Four fungal species were frequently isolated from the disease symptoms. Phomopsis sp. was the most predominant fungus associated with the leaf spot and blight symptoms on kiwifruit, followed by Glomerella cingulata, Alternaria alternata and Pestalo-tiopsis sp. Phomopsis sp. was commonly isolated from angular leaf spots, silvering gray leaf blights, and zonate brown leaf blights. G. cingulata, A. alternata and Pestalotiopsis sp. were isolated from grayish brown ring spots (anthracnose), brown ring spots and zonate dark brown leaf blights. Typical symptoms appeared on the wounded and unwounded leaves, which were inoculated by each of Phomopsis sp., G. cingulata, and Pestalotiopsis sp., but A. alternata caused symptoms only on the wounded leaves.

  • Anthracnose severely occurred on begonia (Begonia semperflorens Link.) nurseries in Gyeongju, Gyeongbuk in July, 2004. More than $80\%$ of begonia seedlings were diseased in the greenhouse surveyed and diseased leaves per plant were $12.1\%$ in average. Yellowish spots occurred on the leaves of begonia as initial symptoms, and they coalesced irregularly to form large brown pleomorphic lesions. Severely infected leaves were defoliated, resulting in abnormal growth of the entire plant. Colletotrichum sp. was repeatedly isolated from the diseased plants and was identified as Colletotrichum acutatum on the basis of the mycological characteristics on potato dextrose agar and RAPD analysis. Pathogenicity of the fungus was also confirmed by artificial inoculation on healthy plants. The optimum temperature for mycelial growth of C. acutatum was around $25^{\circ}C$. The fungus was sensitive to azoxystrobin, bitertanol, diethofencarb-carbendazim, difenoconazole and tebuconazole. This is the first report on the anthracnose of begonia caused by C. acutatum in Korea.

  • Aldose reductase (AR) has been shown to play an important role in the development of the diabetic complications. To discover novel treatments for diabetic complications from natural sources, 59 Chinese herbal medicines have been investigated for inhibitory activities on AR. Among them, 10 herbal medicines, Catalpa fargesii (stem and leaf), Saussurea Laniceps(whole plant), Alnus nepalensis(stem and leaf), Swertia macrosperma (whole plant), Woodfordia fruticosa (stem and leaf), Elsholtzia bodinieri (whole plant), Elsholtzia fruticosa (whole plant), Rosa multiflora (fruit), Nardostachys chinensis (whole plant), Eurya groffii (stem and leaf) exhibited a significant inhibitory activity compared with 3,3-tetramethyleneglutaric acid (TMG) as positive control. Particularly, 4 herbal medicines, C. fargesii (stem and leaf), S. Laniceps (whole plant), A. nepalensis (stem and leaf), S. macrosperma (whole plant) showed two times more potent inhibitory activity than TMG ($5.37{\mu}g/ml$).

  • In 2022, a fungal isolate, designated KNUF-22-049, was obtained from a soil sample collected from a field in Sang-ju, Gyeongsangbuk-do, Korea. The strain exhibited cultural and morphological characteristics, including colony color and size and the shapes and sizes of conidiogenous cells and conidia, that were consistent with those of Apinisia keratinophila. A phylogenetic analysis using the sequences of the internal transcribed spacer regions and the large subunit of the nuclear ribosomal RNA gene confirmed that isolate KNUF-22-049 is most closely related to A. keratinophila CBS 947.73T at a species level. Both the morphological observations and phylogenetic analysis indicated that KNUF-22-049 is indeed an A. keratinophila strain. This study represents the first documentation of Apinisia keratinophila in Korea.

  • Members of the genus Bacillus are known to play an important role in promoting plant growth and protecting plants against phytopathogenic microorganisms. In this study, 21 isolates of Bacillus spp. were obtained from the root micro-ecosystem of Suaeda glauca. Analysis of the 16S rRNA genes indicated that the isolates belong to the species Bacillus amyloliquefaciens, Bacillus velezensis, Bacillus subtilis, Bacillus pumilus, Bacillus aryabhattai and Brevibacterium frigoritolerans. One of the interesting findings of this study is that the four strains B1, B5, B16 and B21 are dominant in rhizosphere soil. Based on gyrA, gyrB, and rpoB gene analyses, B1, B5, and B21 were identified as B. amyloliquefaciens and B16 was identified as B. velezensis. Estimation of antifungal activity showed that the isolate B1 had a significant inhibitory effect on Fusarium verticillioides, B5 and B16 on Colletotrichum capsici (syd.) Butl, and B21 on Rhizoctonia cerealis van der Hoeven. The four strains grew well in medium with 1-10% NaCl, a pH value of 5-8, and promoted the growth of Arabidopsis thaliana. Our results indicate that these strains may be promising agents for the biocontrol and promotion of plant growth and further study of the relevant bacteria will provide a useful reference for the development of microbial resources.

  • In a survey conducted in March 2023, Euphorbia hypogaea plants cultivated within greenhouses in Yongin, Korea exhibited water-soaked areas near the stem base, close to the soil. Subsequent isolation from diseased E. hypogaea led to the identification of a bacterial strain, designated as CBNUMPBL-103. The isolate was identified as Dickeya dadantii through sequencing of the 16s rRNA and phylogenetic analysis. The pathogenicity of the isolate was confirmed by inoculating it into healthy E. hypogaea, resulting in the manifestation of similar symptoms observed during the survey. The re-isolated strain recovered from inoculated plants showed a similar morphology with the inoculated strain. This is the first documentation of D. dadantii causing soft rot of E. hypogaea in Korea.
