Antioxidant and Antibacterial Activities of Lactobacillus-fermented Artemisia annua L. as a Potential Fish Feed Additive (양어 사료첨가제로서의 유산균 발효 개똥쑥의 항산화 및 항균활성)

  • Fermented medical herbs using Lactobacilli have attracted significant attention due to their enhanced biological activities. A traditional medicinal plant, Artemisia annua L., was fermented using a probiotic strain, L. plantarum SK3494. The strain was isolated from Artemisia princeps var. orientalis and molecularly identified through sequence similarities and phylogenetic tree analysis. The antioxidant activity of L. plantarum-fermented A. annua L. (LFA) was determined using the DPPH free radical scavenging assay. Cellular antioxidant activity of LFA was examined using the superoxide radical reduction assay in MAT-C cells. Total polyphenol contents (TPC) and flavonoid contents (TFC) of LFA were determined. The antibacterial activity of LFA against fish pathogens was also determined in this study. The viable cell number (9.38 log10 CFU/ml) and pH (4.1) results showed good adaptive ability of the selected strain during fermentation. LFA was found to have enhanced antioxidant activity compared to non-fermented A. annua L. (NFA) based on the DPPH assay. Cellular antioxidant activity was present in both LFA and NFA. After 24 hr and 48 hr of fermentation, the LFA also showed antibacterial activities against fish pathogens Photobacterium damselae subsp. damselae and Vibrio ichthyoenteri. These results suggest that L. plantarum-fermented A. annua L. may have potential as a feed additive in aquaculture.

Enhancement of Anti-inflammation Effect by Fermentation Process in Aronia melanocarpa (Michx.) Elliott Extract (발효 공정을 통한 아로니아 추출물의 항염증 효능 증진)

  • This study was indicated to enhance the anti-inflammation activities by the fermentation of the fruits of Aronia melanocarpa (Michx.) Elliott. The extracts by 70% ethanol (EE) showed better biological activities than those by hot water (WE) from campared result of the effect of extraction solvents. Then, the extract from 70% ethanol extraction was further fermented by lactic acid, denoted as FEE. For antioxidant activities, the FEE had showed the highest value as 0.832 of reducing powder, in comparison with those of EE and WE. Cytotoxicity of the water extraction (WE) was measured for 12.06% in addition of $1.0mg/m{\ell}$ of FEE. For anti-inflammation activities, NO production from the macrophage, RAW 264.7 was observed as $7.24{\mu}M$ and $8.52{\mu}M$ from FEE and EE, respectively. Prostaglandin $E_2$ ($PGE_2$) production from human fibroblast cell, CCD-986sk, was also estimated for $152pg/m{\ell}$ in addition of $1.0mg/m{\ell}$ of the FEE. The lowest production of both IL-6 and TNF-${\alpha}$ were $3.5pg/m{\ell}$ and $865.5pg/m{\ell}$, respectively in addition of $1.0mg/m{\ell}$ of the FEE, whereas $74.5pg/m{\ell}$ and $982.4pg/m{\ell}$ in treated with same concenrations of the EE. It was also found that the FEE was higher amounts than other extracts through HPLC analysis of the anthocyanins. These results strongly indicate that fermentation process of the lactic acid could enhance anti-inflammation activities of extracts by increasing the amounts of the anthocyanins, especially cyanidin-galactoside. Our results suggest that the application of the fermentation process for other medicinal herbs can be improved their biological activities.

The Study of Safety of Herbal Medicine Including Fuzi(附子) on High Range of LFT, RFT Patients (간, 신장수치 이상 환자에 부자(附子) 배합 처방 투여가 미치는 영향)

  • Objectives : Aconiti Lateralis Preparata Radix(fuzi, 附子) and its class herbs(chaunwu, caowu etc.) are necessary for some clinical conditions, such as cold pain, chilling etc,. But, they has some poison component. And, they have been known to cause liver and kidney injury, and dangerous in the patients who has abnormal range of LFT and RFT. This study shows the consequences for aspartate aminotransferase(AST), alanine aminotransferase(ALT), blood urea nitrate(BUN), and Creatinine were analyzed using samples from patients who took the decoction containing fuzi. Methods : Blood samples for Experiment Group(E) were collected from 63 patients, who took the decoction containing fuzi, admitted into the 6th internal medicine department of Dong Eui Oriental Medical Hospital between from January 2007 to March 2011. In compared to those of experiment group, blood samples as Control Group(C) were collected from 64 patients, who took the decoction not containing fuzi, admitted into the same hospital from January 2009 to April 2011. Results and Conclusions : 1. AST No changed : E 4.17%, C 2.63%, Increased : E 12.5%, C 28.95%, Decreased : E 80.33%, C 68.42% 2. ALT No changed : E2.94%, C5.13%, Increased : E 8.82%, C 20.51%, Decreased : E 88.24%, C 74.36% 3. BUN No changed : E 0%, CG 7.14%, Increased : E 32.25%, CG 14.29% Decreased : EG 67.65%, CG 78.57% 4. Creatinine No changed : EG 5.00%, CG 0%, Increased : EG 35.00%, CG 54.55% Decreased : EG 60.00%, CG 45.45% 5. The results suggest that the decoction containing fuzi isn't harmful AST, ALT, BUN, Cr of the patients who has high range of them.

13 weeks repeated oral dose toxicity studies with LMK02-Jangwonhwan in SD rats (LMK02의 Sprague-Dawley 랫드를 이용한 13 주간 반복 경구투여 독성시험)

  • Objectives : The oriental medicine Jangwonhwan, a boiled extract of 12 medicinal herbs/mushrooms, has been prescribed to patients with cognitive dysfunction, as originally described in the Korean medical text, DonguiBogam(amnesia chapter). Recently, a modified formula of Jangwonhwan (LMK02-Jangwonhwan) consisting of seven medicinal plants/mushrooms, was shown to reduce the ${\beta}$-amyloid deposition in the brain of Tg-APPswe/PS1dE9 mouse model for Alzheimer's disease. The toxicity of LMK02-Jangwonhwan was investigated in SD rats, by a daily oral administration for 13 weeks and NOAEL(No observed adverse effect dose), a definite toxic dose and target organ, as well. Methods : Quality control of the tablet form of LMK02-Jangwonhwan was established by estimating the indicative components, Ginsenoside Rg3 of Red Ginseng and Decursin of Angelicagigas Nakai. The toxicity of LMK02-Jangwonhwan was investigated in 6 week old, specific pathogen free (SPF), Sprageu-Dawley rats by oral administration. Each test group consisted of 10 male and 10 female rats. The groups received doses of 500, 1,000 or 2,000 mg/kg/day of test substance for 13 weeks. The clinical signs, death rate, body weight, food consumption, ophthalmic examination, urinalysis, hematological and serum biochemistry, organ weight and pathological changes were examined and compared with those of the control group. Results : The 13-week repeated oral treatment doses didn't result in any specific symptoms or death. There were no significant changes in the rat's weight and food consumption. Further, ophthalmic examination, urinalysis, hematological, serum biochemistry test and organ weight revealed no significant differences. Conclusions : The no-observed-adverse-effect level(NOAEL) of LMK02 for male and female Sprague-Dawley rats was determined as 2,000mg/kg/day and the target organ wasn't confirmed. Because no significant adverse effects were observed, the target organ could not be determined.

A Clinical study of Taeumin Chongsim Yonja Tang (청심연자탕(淸心蓮子湯) 활용(活用)에 대(對)한 임상적(臨床的) 연구(硏究))

  • 1. Background and Purpose : I intended to be helpful to understand constitutional symptoms of a disease and an application of the prescription through a clinical study of Taeumin Chongsim Yonja Tang. 2. Methods : I studied 125 patients(62 men and 63 women) treated with Taeumin Chongsim Yonja Tang in constitutional clinic of Dongguk Pundang Oriental Hospital. And then I investigated the characteristics of Chongsim Yonja Tang treated patients, remedial values and side effects. 3. Results : Taeumin Chongsim Yonja Tang was used in all age groups both men and women. There were 60 kinds of chief complaints including numbness of hands and feet, headache and fatigue etc. The distributions of remedial values were slightly improved(60.8%), much improved(15.2%), no improved(24.0%) and there was no aggravated patient. Using the prescription, mostly I prescribed only Taeumin Chongsim Yonja Tang without any changing, and added 2-3 herbs to the prescription according to the symptoms of a disease. Taeumin Chongsim Yonja Tang was affected in remedial values by the period of sufferring from illness, treatment and medication. Side effects were diarrhea, indigestion and so on.

Study on Anti-oxidant and Anti-inflammatory Activity of Eggplant-cheongyeolsodokum (가지-청열소독음(淸熱消毒飮)의 항산화 및 항염 효능에 관한 연구)

  • The purpose of this study is verification of the anti-oxidant effect and anti-inflammatory effect of Eggplant-cheongyeolsodokum composed of 8 herbs (Solanum melongena L., Lonicera japonica Thunb., Glycyrrhiza uralensis Fisch., Ligusticum chuanxiong Hort., Angelica gigas Nakai., Coptis deltoidea C. Y. Cheng et Hsiao., Gardenia jasminoides J. Ellis., Forsythia suspensa Vahl) to confirm the possibilities as useful cosmetic material. We used the modified prescription of 'cheongyeolsodokum' contained in Korean traditional medical book 'Donguibogam' as composition of Eggplant-cheongyeolsodokum and their proportions. Eggplant-cheongyeolsodokum were extracted with hot water, 70% ethanol and then powdered. To confirm anti-oxidant effect, we investigated radical scavenging ability (DPPH, $ABTS^+$, superoxide), superoxide dismutase (SOD)-like activity, total polyphenolic contents. Also to confirm anti-inflammatory effect, we investigated inhibition effect of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages, and Inhibition effect of the expression of inflammatory-related proteins (iNOS, COX-2) by western blot analysis. As a result, Eggplant-cheongyeolsodokum showed good anti-oxidant and anti-inflammation effects, we suggest that it can be used as an active ingredient for cosmetics.

Effect of Younnyeniksoobulrodan(延年益壽不老丹) on Antioxidant Capacity in D-galactose induced Aging Rats (연년익수불로단(延年益壽不老丹)이 노화유발 흰쥐의 항산화능에 미치는 영향)

  • Objectives: Younnyeniksoobulrodan(延年益壽不老丹) composed of Polygonum multiflorum THUNB. and some medical herbs is known as formula of senescence delay effect. The purpose of this study is to investigate the effect of Younnyeniksoobulrodan(延年益壽不老丹) on antioxidant enzyme activity such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and liver. Methods: Sprague-Dawley rats divided into 4 gorups, Young group(8 weeks old, N-8), Aging group(18 weeks old, N-18), pathologically induced aging gorup(injected D-galatose 50mg/kg, 1time/day for 6 weeks, CON) and Younnyeniksoobulrodan(延年益壽不老丹) administered group(D-galactose 50mg/kg and Younnyeniksoobulrodan extracts 840.0mg/kg 1time/day for 6 weeks, YIB). Rats were sacrificed and TBARS, SOD, CAT, and GSH-px were measured in rat erythrocytes and liver. Results: Plasma and liver TBARS concentrations of YIB group were significantly lower than those of control. Red blood cell(RBC) SOD activities of YIB group was increased(F=3.445, p=0.033, ANOVA test), and RBC catalase activities of all experimental group were not significantly different. RBC GSH-px activities of YIB group was increased(F=9.365,p=0.0001, ANOVA test). Liver SOD activities of YIB group was higher than those of control(F=4.967, p=0.008, ANOVA test). Liver catalase activities of all experimental group were not significantly different, and liver GSH-px activity of YIB group was significantly higher than that of control(F=3.846, p=0.022,ANOVA test). Conclusions: According to the above results, it is considered that Younnyeniksoobulrodan is effective in inhibiting lipid peroxidation and increasing antioxidative enzyme activities in D-galactose induced aging rat.

Effect of Rubi Fructus(Fruit of Rubus Coreanus Miq.) on Antioxidant Capacity in D-galactose Induced Aging Rats (복분자(覆盆子)가 노화유발(老化誘發) 흰쥐의 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objectives: Rubi Fructus(fruit of Rubus coreanus Miq.) composed of Polygonum multiflorum THUNB. and some medical herbs is known as formula of senescence delay effect. The purpose of this study is to investigate the effect of Rubi Fructus(fruit of Rubus coreanus Miq.) on antioxidant enzyme activity such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase(GSH-px) in rat erythrocytes and blood plasma. Methods: Sprague-Dawley rats were divided into 3 groups, Normal group(supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and Rubi Fructus (fruit of Rubus coreanus Miq.) administered group(D-galactose 50mg/kg and Rubi Fructus(fruit of Rubus coreanus Miq.) extracts 85.0mg/200g 1time/day for 6 weeks, BBJ Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, Plasma total lipid, Plasma triglyceride and cholesterol were measured in rat erythrocytes and blood plasma. Results : Plasma TBARS concentrations of all experimental group were not significantly different. Red blood cell(RBC) SOD activities of BBJ group was increased, and RBC catalase activities of all experimental group were not significantly different. RBC GSH-px activities of BBJ group was increased. Plasma total lipid concentration of BBJ group were significantly lower than those of control. Plasma triglyceride, total cholesterol and HDL-cholesterol concentrations of all experimental group were not signi ficantly different. Conclusions: According to the above results, it is considered that Rubi Fructus(fruit of Rubus coreanus Miq.) is effective in inhibiting lipid peroxidation and increasing antioxidative enzyme activities in D-galactose induced aging rat.

Yangkyuksanhwa-Tang Attenuates Ischemic Brain Injury in a Focal Photothrombosis Stroke Model (뇌허혈 마우스모델에서 양격산화탕이 뇌 손상 완화에 미치는 효과)

  • Yangkyuksanhwa-Tang (YKSH), consisting of nine different herbs, is commonly used in Soyangin-type individuals with stroke, based on the Sasang Constitution Theory in Korea. However, no evidence has yet confirmed a beneficial effect of YKSH in ischemic stroke treatment. In this study, we investigated the effects of YKSH on ischemic brain injury in a mouse model of cerebral ischemia. Focal cerebral ischemia in mice was induced by photothrombosis, and behavioral recovery was evaluated. Infarct volume, inflammation, and newly generated cells were evaluated by histology and immunochemistry. YKSH treatment resulted in a significant recovery from the motor impairments induced by focal cerebral ischemia, as determined with wire grip and rotarod tests. YKSH treatment also decreased the infarct volume and the number of cells positive for tumor necrosis factor-${\alpha}$ and myeloperoxidase when compared with a vehicle-treated control group. By contrast, YKSH treatment considerably increased the number of cells positive for glial fibrillary acidic protein and ionized calcium-binding adapter molecule 1, as well as the number of cells doubly positive for Ki67/doublecortin when compared with the vehicle-treated group. These results suggest that YKSH treatment attenuated the infarct size by anti-inflammatory action, astrocyte and microglia activation, and neuronal proliferation, thereby facilitating neurofunctional recovery from a cerebral ischemic assault. YKSH could therefore be a potential treatment for neurofunctional restoration of the injured brains of patients with stroke.

The Inhibitiory Effects of the Scirpi rhizoma on Melanogenesis (삼릉추출물의 멜라닌 생성 저해 효과)

  • To obtain effective and safe depigmenting agents, we investigated the effects of Scirpi rhizoma, a medicine among Chinese herbs, on melanogenesis. Dried S. rhizoma was refluxed with 70% aqueous ethanol and the extract was evaporated to dryness. To determine the effects as a whitening agent, various in vitro tests were performed such as free radical scavenging activity, melanin formation assay, tyrosinase activity and expression of tyrosinase, TRP-1 and TRP-2(western blot and RT-PCR) in B16 melanoma cells. S. rhizoma showed scavenging activities of free radicals and reactive oxygen species (ROS) with the $IC_{50}\;of\;638{\mu}g/mL$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and $21.7{\mu}g/mL$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. S. rhizoma significantly inhibited melanin production in B16 melanoma cells. S. rhizoma treatment(48 h) suppressed the biosynthesis of melanin up to 27% at 100{\mu}g/mL$ and reduced tyrosinase activity up to 31% at $100{\mu}g/mL$ in B16 melanoma cells. S. rhizoma was also able to significantly inhibit tyrosinase and TRP-1 expression in protein and mRNA level. These results suggest that S. rhizoma inhibited melanin biosynthesis by regulating tyrosinase activity and expression in B16 melanoma cells. Therefore, S. rhizoma may be useful as a new antioxidant and whitening agent to inhibit melanogenesis.
