Effect of Medium Composition on In vitro Prothallus Culture of 3 Fern Species in the Family Aspleniaceae (꼬리고사리과(Aspleniaceae) 양치식물 3종 전엽체의 기내배양에 미치는 배지구성물질의 영향)

  • This experiment was conducted to investigate the effects of medium composition, physical condition and growth regulators on prothallus proliferation in 3 fern species belonging to Aspleniaceae (Asplenium incisum, Camptosorus sibiricus and Phyllitis scolopendrium). In all of the 3 species, the effective medium for prothallus proliferation was Murashige and Skoog's basal medium with 1% sucrose and pH 5.8. The optimum $NH_4^+:NO_3^-$ concentration ratio of protahllus proliferation was 20:40mM (1:2 ratio) in A. incisum and P. scolopendrium and 30:30mM (1:1 ratio) in C. sibiricus. However, agar content of medium did not affect the rate of prothallus proliferation. Among the selected 6 kinds of growth regulators (NAA, IAA, 2,4-D, BAP, kinetin and lip), cytokinins generally has a promotive effect, but auxins showed an inhibitory effect on prothallus growth.

  • The purpose of this study is to investigate the teacher's perception of maker education and to suggest the direction of teacher education curriculum for maker education in order to explore the possibility of applying maker education to early childhood education. To this end, a survey was conducted on the perception of maker education for preschool teachers. As a result of the data analysis, the perception of the concept, meaning, and educational environment of maker education were low, while the effectiveness, importance, value, continuous application and development of maker education were highly recognized and the necessity of maker teacher education and the educational environment was suggested. Based on the survey results, the teacher curriculum composition was classified into basic and advanced courses. The basic course included basic contents to understand maker education and maker space. The advanced course included information on how to use various tools, materials and digital media, how to conduct and evaluate maker education to apply maker education. The teacher curriculum composition for maker education can suggest basic directions for developing teacher education programs and strengthening the digital teaching competency of teachers in the future.

Stem Firmness and Flowering Response of Cut Lilies as Influenced by Medium Composition in Box Culture

  • Stem firmness and flowering response of cut lily as influenced by medium composition (Control: Upland soil, Pt: Peatmoss, Pe: Perlite, Ve: Vermiculite, Rrh: Rotted rice-hull, RPt: Russian Peatmoss) were studied. For 'Casa Blanca', plant height and length of flower stalk increased when bulbs were planted in Pt:Rrh:Ve(1:1:1, v/v), and dried leaves of lower part plants decreased by RPt:Pe:Rrh(1:1:1, v/v). In case of 'Marco Polo' plant height and length of flower stalk increased with Pt:Rrh(1:1, v/v) as compared to other treatment, number of leaves and dried leaves increased when bulbs were planted in RPt:Pe:Rrh(1:1:1, v/v) as compared to control. Flowering of 'Casa Blanca' was promoted in Pt:Pe:Ve(1:1:1, v/v) and 'Marco Polo' was accelerated in Pt:Rrh:Ve(1:1:1, v/v). Flower length of 'Casa Blanca' was increased by RPt:Pe:Rrh(1 :1 :1, v/v) as compared with control and 'Marco Polo' was increased when bulbs planted to Pt:Rrh(1 :1, v/v). Flower-bud blasting of two cultivars was increased with Pt as compared with other treatment. Stem firmness of 'Casa Blanca' was increased by Pt:Pe:Ve (1:1 :1, v/v), and especially, stem firmness of upper part plants was increased by Pt:Rrh(1 :1, v/v) in 'Marco Polo' as compared to control plants. but generally, stem firmness of 'Casa Blanca' was not influenced with all cultural media as compared to control.

Control Effects of Micromonospora sp. AW050027 by Media Optimization and Microbial Treatment Against Pine Wood Nematode (Micromonospora sp. AW050027 균주의 배지최적화 및 미생물제제 처리에 의한 소나무재선충 방제효과)

  • Nematicidal activity against pine wood nematode, Bursaphelenchus xylophilus, was tested in the pot and field by the treatment of microbial formulation after media optimization. The optimized media composition was glycerol 10 g, soybean meal 10 g, NaCl 1 g, $CaCO_3$ 2 g, $K_2HPO_4$ 0.125 g per liter and microbial complex formulation was made with liquid and powder type. Most effective antibiotics against symbiotic microorganism with nematode, kanamycin, was added to the formulation. The control effects against pine wood nematodes were checked by pot test and field test. In the result of treatment by trunk injection, five times treatment was more effective than one time and the treatment with the formulation of concentrated culture supernatant was the most effective in the nematicidal activity showing below 10% mortality in pine tree.

  • Recently as the studies on the combined delivery over multiple transmission media are progressed, studies using MMT which is next-generation multimedia transport standard are underway. They are about transmitting video data layered by SHVC through broadcasting network and communication network based on MMT. However, it is difficult to serve synchronized UHD contents in heterogeneous network environment based on MMT, because MMT standard doesn't fully specify the information to indicate the relationship between media. Also, the service that we want to propose in this paper is a broadcasting-based service such as terrestrial broadcasting. Therefore the user must be able to watch various contents continuously. However, the MMT PI(Presentation Information) that controls the spatio-temporal layout of a content has been described as providing only one content. In this paper, we propose the structure of MXD document and MMT PI to provide continuous UHD contents service based on heterogeneous network.

Study on the Revegetation Methods of Phragmites japonica, Miscanthus sacchariflorus, Themeda triandra and pennisetum alopecuroides for the Rehabilitation of Close-to-Nature River (자연형 하천 식생복원을 위한 달뿌리풀, 물억새, 솔새, 수크령의 녹화방법에 관한 연구)

  • This study was carried out to suggest an effective method for the rehabilitation of Close-to-Nature River and artificial wetland. The results on the revegetation methods by seeding and sodding of Phragmites japonica, Miscanthus sacchariflorus, Themeda triandra, and Pennisetum alopecuroides were summarized as follows. Seed germination of Phragmites japonica was 76.3% at $30^{\circ}C$, that of Miscanthus sacchariflorus was 68.7% at $20^{\circ}C$, that of Themeda triandra germinated 52.3% at $25^{\circ}C$, $30^{\circ}C$ constant temperature and that of Pennisetum alopecuroides germinated 86.7% at $30^{\circ}C/20^{\circ}C$ alternating temperature. Seed germination of Pennisetum alopecuroides exceeded 80% at $20^{\circ}C$, $25^{\circ}C$, $30^{\circ}C$ constant temperature and $25^{\circ}C/15^{\circ}C$ alternating temperature. At 60 days after seeding, the ground coverage of Phragmites japonica, Miscanthus sacchariflorus, Themeda triandra, and Pennisetum alopecuroides reached 81%, 81 %, 74%, and 86% respectively in the soil media of vermiculite and peatmoss(1 : 1 by volume). In the results of sod experiment, Phragmites japonica, Miscanthus sacchariflorus, Themeda triandra, and Pennisetum alopecuroides were formed sod completely in soil composition type of vermiculite and peatmoss(1 : 1, v/v). Thus this media seems to be best sad production media for rehabilitation works of Close-to-Nature River and man-made wetlands. Phragmites japonica and Miscanthus sacchariflorus were not significantly different in shoot height and the number of tillers by different planting distance for the first one year of experiment. Pennisetum alopecuroides shows high possibility to be used for Close-to-Nature River rehabilitation works by seeding.

Proposal for Deep Learning based Character Recognition System by Virtual Data Generation (가상 데이터 생성을 통한 딥러닝 기반 문자인식 시스템 제안)

  • In this paper, we proposed a deep learning based character recognition system through virtual data generation. In order to secure the learning data that takes the largest weight in supervised learning, virtual data was created. Also, after creating virtual data, data generalization was performed to cope with various data by using augmentation parameter. Finally, the learning data composition generated data by assigning various values to augmentation parameter and font parameter. Test data for measuring the character recognition performance was constructed by cropping the text area from the actual image data. The test data was augmented considering the image distortion that may occur in real environment. Deep learning algorithm uses YOLO v3 which performs detection in real time. Inference result outputs the final detection result through post-processing.

Fermentation of a Potential Biocontrol Agent, Bacillus amyloliquefaciens SKU-78 Strain (풋마름병균의 길항세균 Bacillus amyloliquefaciens SKU-78의 대량 배양 조건 확립)

  • Mass production of biocontrol agent is an essential step for its commercial use. Media composition and culture conditions for production of Bacillus amyloliquefaciens SKU-78, a potential biocontrol agent against bacterial wilts, were optimized by a flask culture. Low cost media combining nitrogen and carbon sources were tested. Maximum cell growth (> $2{\times}10^9$ CFU/ml) was obtained in a medium of 5% soy flour combined with 3% corn starch after 24 h cultivation. The optimum initial pH, temperature and shaking speed was 5.5, $30^{\circ}C$ and 150-250 rpm, respectively. Fermentation of SKU-78 was scaled up in 30 L fermenter and the profiles of cell density, pH, dissolved oxygen and spore formation were recorded. After 8 h lag phase, exponential growth occurred and reached at maximum viable cell number ($1.2{\times}10^{11}$ CFU/ml) after 20 h. The SKU-78 strain grown in a low cost medium exhibited the high suppression of bacterial wilts. The results indicate that SKU-78 strain can be produced in a low cost medium and provide a basis for scaling up to industrial level.

Oak Mushroom Cultivation on Larch(Larix leptolepis Gorden) sawdust and its economic efficiency (낙엽송 톱밥을 이용(利用)한 표고버섯 재배(栽培)와 경제성)

  • To enhance the utilization of Larch(Larix leptolepis Gorden) wood, the sawdusts were tested as cultivating media for Lentinus edodes(Mori 290) by varying the composition of the media. The economic efficiency for producing mushrooms from larch sawdusts was evaluated. The results are as follows. 1. Larch sawdust was as much efficient as oak sawdust in the growth of the mycelia and production of good quality of oak mushrooms. 2. The mushrooms were able to be harvested starting six months after beginning the cultivation. No differences were found among the qualities of the mushrooms produced by various treatments. 3. The contents of lignin(28.6-28.7%) in the fresh sawdust of larchwood are higher than those in the residues of the used media(18.2%). But lignin content of 1% NaOH extracts(30.4-34.4%) in fresh sawdust of larch was lower than those in the residues(68.0%). 4. The production of mushroom was relatively high to be 29.7-31.5%(760-780g.)/2.5kg pack) of mushroom weight. The net profit estimated was 1,475Won per pack.

  • This study was carried out to determine the effects of media compositions on cell growth and casein hydrolysis for cell production in order to add Micrococcus sp. LL3 as a potential agent for industrial application for the shortening of ripening period. Monosaccharides like glucose, mannose and fructose were mare excellent as carbon source, but arabinose and xylose markedly inhibited cell growth and caseinolysis. Among the organic nutrients, yeast extract was more effective for cell growth and for caseolysis. However, inorganic nitrogen sources were less effective than organic sources. Urea inhibited cell growth severely. Cell growth and caseinolysis were rather increased a little in the broth containing 1% NaCl, and the organism tolerated and grew in relatively high concentrations of NaCl up to 9%. Addition of vitamin did not affect cell growth and caseolysis in level of $0.1\;{\mu}g/ml$ concentration. Cell growth and caseinolysis were stimulated by addition of glutamic acid and $MgSO_4$ with concentration of 0.2% and 0.05% respectively.
