Potential for measuring mobile cadmium concentration in sandy soil using polymer magnetic beads with carboxyl groups was investigated. Experiments for extracting cadmium were performed with contaminated soils, de-ionized water and magnetic beads. In this neutral experimental condition, reacting cadmium with magnetic beads indicate total amount of cadmium that can be moved in soil. The results showed that the mobile fraction of cadmium in soil could be combined with magnetic beads in short time. After binding between cadmium and magnetic beads, the beads were separated from soil suspension by outer magnetic force. The bound cadmium was dissolved from magnetic beads by acid solutions, which were then analyzed by atomic absorption spectroscopy (AAS). This method can determine mobile heavy metals in sandy soil effectively than existing method which use pollutant chemicals to environments such as EDTA.
1-Nitropyrene (1-NP), 2-NP and 2-nitrofluoranthene (2-NFR) are useful markers for studying the atmospheric behaviors of polycyclic aromatic hydrocarbons (PAHs) and nitropolycyclic aromatic hydrocarbons (NPAHs). However, present methods for measuring trace levels of these compounds are lesssensitive and laborious. Here we describe several improvements to a previously reported high-performance liquid chromatography-chemiluminescence detection system that allows it to determine trace levels of 1-, 2-NPs and 2-NFR. The proposed system was equipped with a reducer column packed with Pt/Rh instead of zinc whose life-time was limited. The combination of Cosmosil MS-II (monomeric ODS) and AR-II (polymeric ODS) columns was used instead of polymeric ODS columns as the separator column to improve the separation. An ethanol mixture with acetate buffer (pH 5.5) was used in place of an acetonitrile mixture with the same buffer to activate the reducer column. The same ethanol mixture was used as the mobile phase for the clean-up column. The switching time of the column switching valve was optimized to concentrate the amino-derivatives of above NPAHs quantitatively on the concentrator column. The concentrations of bis(2,4,6-trichlorophenly) oxalate and hydrogen peroxide in the chemiluminescence reagent solution were optimized to 0.4 mM and 30 mM, respectively, to increase the sensitivity. Under the above conditions, the detection limits (S/N=3) of 1-, 2-NPs and 2-NFR were 1 fmol (0.25 pg), 10 fmol (2.5 pg) and 4 fmol (1 pg), respectively. The proposed system was effectively used to determine trace levels of 1-, 2-NPs and 2-NFR in airborne particulates collected at Noto Peninsula. The atmospheric concentrations of 1-, 2-NPs and 2-NFR were not more than sub pg $m^{-3}$ levels. They were higher in winter (January) than in summer (July). In both seasons, the concentrations were in decreasing order, [2-NFR]>[1-NP]>[2-NP].
This study was conducted to investigate the effect of Lactobacillus sp. and Saccharomyces sp. on turf quality, shoot and root growth of creeping betgrass in golf course by measuring turf color index, chlorophyll content, dry weight of shoot and root, T/R ratio and root length. Fertilizer treatment was designed as follows; nonfertilizer (NF), control (CF; compound fertilizer), microorganism medium(MO; CF+MO)), microorganism medium contained Fe(MO-Fe; CF+MO-Fe) and microorganisum medium contained S (MO-S; CF+MO-S). Soil properties investigated after experiment was scarcely affected by applied fertilizers in root zone of creeping bentgrass. The turf color index and chlorophyll index of MO, MO-Fe, MO-S treatment were higher than those of NF, and similar to those of CF. The turfgrass root in MO and MO-Fe treatment was longer than others. The dry weight of shoot in MO and MO-S was higher than CF and that of root in MO and MO-Fe, and dry weight of MO was increased than that of NF and CF, by 26% and 6%, respectively. AS compared with NF, T/R ratio of CF, MO, MO-Fe and MO-S was increased, and MO and MO-Fe was similar to CF, MO-S higher. Nutrient content in CF, MO, MO-Fe and MO-S was contained more than in NF, and it was higher in shoot. These was suggested that application of MO induced the development of quality and growth of creeping bentgrass by assisting root growth and nutrients uptake.
Boiling is an efficient removal method to reduce radon in groundwater when ventilating indoor air. 13 groundwater samples with various radon concentrations were used to evaluate the reduction rate of radon with heating temperature and time. The groundwater samples were obtained by Bladder pump and on-situ measurements such as dissolved oxygen (DO) and hydrogen concentration (pH) and so on were carried out by a flow cell system isolated from the ambient atmosphere environment. All samples for measuring radon in groundwater were analyzed by liquid scintillation counter (LSC). The experiment result showed that increasing groundwater temperature enhanced radon removal rate but the initial radon concentration with high level lowered the removal rate. This means that radon reduction in groundwater by heating needs more heating energy and longer heating time with radon concentrations. Radon removal rate in groundwater, therefore, mainly depends on the initial radon concentration, heating temperature, and heating time.
Baicalein (BA), a plant-derived active flavonoid present in the root of Scutellaria baicalensis, has been widely used for the treatment of stress-related neuropsychiatric disorders including depression. Previous studies have demonstrated that repeated restraint stress disrupts the activity of the hypothalamic-pituitary-adrenal (HPA) axis, resulting in depression. The behavioral and neurochemical basis of the BA effect on depression remain unclear. The present study used the forced swimming test (FST) and changes in brain neurotransmitter levels to confirm the impact of BA on repeated restraint stress-induced behavioral and neurochemical changes in rats. Male rats received 10, 20, or 40 mg/kg BA (i.p.) 30 min prior to daily exposure to repeated restraint stress (2 h/day) for 14 days. Activation of the HPA axis in response to repeated restraint stress was confirmed by measuring serum corticosterone levels and the expression of corticotrophin-releasing factor in the hypothalamus. Daily BA administration significantly decreased the duration of immobility in the FST, increased sucrose consumption, and restored the stress-related decreases in dopamine concentrations in the hippocampus to near normal levels. BA significantly inhibited the stress-induced decrease in neuronal tyrosine hydroxylase immunoreactivity in the ventral tegmental area and the expression of brain-derived neurotrophic factor (BDNF) mRNA in the hippocampus. Taken together, these findings indicate that administration of BA prior to the repeated restraint stress significantly improves helpless behaviors and depressive symptoms, possibly by preventing the decrease in dopamine and BDNF expression. Thus, BA may be a useful agent for the treatment or alleviation of the complex symptoms associated with depression.

This study was conducted to investigate the effect of Actinomyces sp. and Bacillus sp., United States granular microorganisms and Japan granular microorganisms on turfgrass growth and thatch decomposing of creeping bentgrass in golf course by measuring turf color index, chlorophyll index, thatch content of soil, root length, turf density and chemical properties and thatch content of soil. Fertilizer treatment was designed as follows; control(CF; compound fertilizer), microorganism medium(M; CF+M), microorganism medium and livestock manure fertilizer(M-L; CF+M+LMF), microorganism medium, livestock manure fertilizer and amino acid liquid fertilizer(M-L-A; MM+LMF+ALF), United States granular microorganisms(USGM; CF+USGM), Japan granular microorganisms(CF+JGM). Soil properties investigated after experiment was scarcely affected by applied fertilizers in root zone of creeping bentgrass. The turf color index and chlorophyll index of M, M-L, M-L-A, USGM, JGM treatment were higher than those of CF. The turfgrass root in M-L treatment was longer than others. The thatch content of soil in M treatment was longer than others. The thatch content of M was decreased than that of CF by 6.8%. These was suggested that application of M induced the development of quality and growth of creeping bentgrass by assisting turfgrass growth and thatch decomposing.
This study was designed to determine the most effective concentration of fluoride and levels of laser irradiation for the remineralization of decayed teeth. After irradiation with a pulsed Nd:YAG laser and the topical application of fluoride, phosphate and fluoride concentration in enamel were measured. And then the changes on surface enamel using an scanning electron microscope were observed. Samples by extraction healthy, permanent, mandibular bovine teeth with no caries were obtained. Among them 371 healthy samples were selected and artificial carious lesions were made. 20 samples were assigned to each experimental group. After irradiation with a pulsed Nd:YAG laser with total energy densities of 10J/$\textrm{cm}^2$, 20J/$\textrm{cm}^2$ for each group. On the teeth, 2% NaF, 1.9% NH4F, 1.6% TiF4 Elmex gel(amine fluoride) and 1.23% APF gel were applied. After pH circulatory procedures, concentrations of fluoride with and Ionalyzer (Orion Research, Model 901, USA) and phosphates with an Uv/V is spectrophotometer (Uvikon 860, Kontrom Co, Switzerland) were measured. By etching the teeth in layers and measuring fluoride concentrations, a profile of fluoride penetration according to depth could be developed. And also the changes on the surface of the enamel using an electron scanning microscope were observed. The comparative analysis yielded the following results : 1. Phosphate concentration was low in all groups compared with the control group except for teeth treated Elmex gel, irradiated with 10J/$\textrm{cm}^2$ and 30J/$\textrm{cm}^2$ energy densities. Teeth treated with APF gel and 30J/$\textrm{cm}^2$ irradiation gad the lowest phosphate concentration. 2. Among all groups, fluoride concentrations in tooth enamel were highest in APF gel and NaF groups irradiated at 30J/$\textrm{cm}^2$. The APF gel group had the highest fluoride concentrations across all energy densities. 3. In the APF gel group, and the NaF group, the greater the energy density of the laser, the higher the fluoride concentrations in the enamel. 4. In all groups, the concentration of fluoride in the enamel by depth tended to decrease with depth. 5. Under the scanning electron microscope, under the condition of irradiation with 20J/$\textrm{cm}^2$, enamel crack was detected for the first time. In the NH4F group, spherical deposits were found on the surface of the enamel, and in the TiF4 group the surface of enamel was covered with an irregular, thin membranous mass in places. In the APF gel and NaF groups irradiated with 10J/$\textrm{cm}^2$, spherical and irregular particles covered the teeth. When these groups were irradiated at 20J/cm2, they were covered with amorphous crystals. These results suggest that one could obtain more effective anticariogenic effects without damage to teeth when less than 20J/$\textrm{cm}^2$ energy densities and APF gel are used.
Effects of ethanol and/or some organic acid on the growth of some microorganism grown in Kimchi, Lactobacillus plantarum, Leuconostoc mesenteroides. Kluyveromyces marxianus, were identified and the cell injury was observed by measuring optical density at 260nm. When 0.0∼5.0% ethanol was added in the growth medium, the cell growth was inhibited depending on the concentration. Organic acids involving acetic, adipic and citric acid inhibited the growth of L. plantarum and Leu. mesenteroides, but K. marxianus, the yeast, was not at 0.1% of organic acid. When 2.0% of ethanol and 0.1% of organic acid were used, adipic acid was more effective on the growth inhibition. This inhibition of microbial growth seemed to be caused by the leakage of internal contents from microbes which were observed by the optical density at 260nm in the buffer supernatant. 5.0% of ethanol accelerated the optical density increase at 260nm in L. plantarum, Leu. mesenteroides and K. marxianus, but 2.0% of ethanol did not. 0.1% organic acid increased the absorbance of the supernatant in lactic bacteria, but not in yeast, K. marxianus. The measurement of absorbance at 260nm revealed that the cell injury increased when 2.0% of ethanol and/or 0.1% of organic acid. especially adipic acid were added.
This study, collaborated Gifu University, Japan, was performed to analyze chemical pollutants and microorganism and to clarify the distribution of sulfate-reducing bacteria and their insolubilization of heavy metal ions in leachates sampled seasonally between 1994 and 1996 from Nanjido waste landfill site, sampled 4 times between 1995 and 1996 from Pusan and Daejeon waste landfill site, and sampled 1 time between 1992 and 1994 from Hokkaido, Nagoya, Osaka and Hukuoka waste landfill site in Japan. The results were as follows: 1. The temperatures of internal leachate and leachate effluent were 40$\circ$C and 30$\circ$C, respectively, and the pH values of both leachates were about 8.0 at Nanjido waste landfill site. The concentration of SO$_4^{-2}$ gradually increased with the degree of stabilization and that of NO$_3$-N was detected in a part of sampling sites at one and half years, and in all sampling sites at 3 years after completion of landfill. 2. The organic substances in leachate of Nanjido waste landfill site decreased with the degree of stabilization and they were very fluctuated with measuring point and time. The concentration of organic substance and heavy metals in internal leachate were higher than in leachate effluent and those of Cd, Hg, and Pb were lower than detection limit except a part of samples in 1996. 3. APCs in internal leachate and leachate effluent were not much different and the minimum of APCs in internal leachate and leachate effluent were $1.0\times 10^4$/ml and $4.0\times 10^1$/ml, respectively. 4. The maximums of SRBs in Nanjido, Pusan, and Daejeon waste landfill site were 9180 MPN/ml, 24000 MPN/ml, and 348 MPN/ml, respectively and the maximum of SRBs in Japan waste landfill site was 9300 MPN/ml. 5. During 2-week-SRB culture, the values of MPN were high at 50$\circ$C for initial culture period and at 30$\circ$C for last culture period. MPN started to appear at first day and rapidly increased between 7th day and 9th day. 6. Cadmium and copper were insolubilized by SRB within 6 hr and iron and zinc were done within 48 hr. The rates of insolubilization of Cd, Cu, Fe, Zn, T-Cr were 100%, 99.5%, 95.0%, 99.8%, 16.1% after 48 hr treatment with SRB, respectively.
In the present paper, the effect of sodium sulfite treatment on tile inhibition of browning reactions occurring in the storage of dried oyster was tested and the supplementary effect of antioxidantsaddedwasalsomentioned. Dried oysters treated with sodium sulfite solutions as described in the previous paper(Lee and Choi, 1977) were stored in the bottles with silica gel bags at room temperature with or without the application of antioxidants. The ethanol solution of an antioxidant mixture(BHA, BHT, plus, synergists) was sprayed on the surface of cooked oyster before drying. The density of brown pigment was determined spectrophotometrically by measuring the absorbance at 420 and 440 nm of both fractions of pigment extract, namely chloroform-methanol and water soluble fractions, which represent the brown color developed by fat oxidation and Maillard reactions respectively. TBA value was also measured for the oxidative rancidity in oysters during the storage. It appeared from the results that the 0.5 M sodium sulfite-60minute treated samples showed better effect after 150 day storage at room temperature. Controlling tile pH of treating solutions, did not reveal so much different in inhibitory effect in the aspect of color but a more reduction of tyrosine and reducing sugar was resulted with acidic solution than with alkaline solution. The development of brown color in dried oyster seemed to be leaded rather by the oxidative rancidity of lipids than sugar-amino reactions particularly in a long-term storage since the browning of chloroform-methanol fraction progressed more rapidly than of water. soluble fraction. The application of antioxidant, therefore, could largely retard the browning of the product as appeared in the results that sodium sulfite treated oyster with addition of antioxidant kept the best color during the storage.
