Helicobacter pylori Infection and a P53 Codon 72 Single Nucleotide Polymorphism: a Reason for an Unexplained Asian Enigma

  • Aim: P53, the most commonly mutated tumor suppressor gene in all types of human cancer, is involved in cell cycle arrest and control of apoptosis. Although p53 contains several polymorphic sites, the codon 72 polymorphism is by far more common. There are divergent reports but many studies suggest p53 pro/pro SNP may be associated with susceptibility to developing various cancers in different regions of the world. The present study aimed to find any correlation between H. pylori infection and progression of carcinogenesis, by studying apoptosis and the p53 gene in gastric biopsies from north Indian population. Materials and Methods: A total of 921 biopsies were collected and tested for prevalence of H. pylori by rapid urease test (RUT), imprint cytology and histology. Apoptosis was studied by the TUNEL method. Analysis of p53 gene polymorphism at codon 72 was accomplished by PCR using restriction enzyme BstU1. Observation: Out of 921 samples tested 56.7% (543) were H. pylori positive by the three techniques. The mean apoptotic index (AI) in the normal group was 2.12, while gastritis had the maximum 4.24 followed by gastric ulcer 2.28, gastropathy 2.22 and duodenal ulcer 2.08. Mean AI in cases with gastric cancer (1.72) was less than the normal group. The analysis of p53 72 SNP revealed that p53 (Arg/Arg), (Pro /Arg) variant are higher (40.59% & 33.66%) as compared to p53 pro/pro variant (25.74%) inthe healthy population. Conclusions: The North Indian population harbors Arg or Pro/Arg SNP that is capable of withstanding stress conditions; this may be the reason of low incidence of gastric disease in spite of high infection with H. pylori. There was no significant association with H. pylori infection and AI. However, there is increased apoptosis in gastritis which may occur independent of H. pylori or p53 polymorphism.

Modeling the potential climate change-induced impacts on future genus Rhipicephalus (Acari: Ixodidae) tick distribution in semi-arid areas of Raya Azebo district, Northern Ethiopia

  • Background: Climate change is believed to be continuously affecting ticks by influencing their habitat suitability. However, we attempted to model the climate change-induced impacts on future genus Rhipicephalus distribution considering the major environmental factors that would influence the tick. Therefore, 50 tick occuance points were taken to model the potential distribution using maximum entropy (MaxEnt) software and 19 climatic variables, taking into account the ability for future climatic change under representative concentration pathways (RCPs) 4.5 and 8.5, were used. Results: MaxEnt model performance was tested and found with the AUC value of 0.99 which indicates excellent goodness-of-fit and predictive accuracy. Current models predict increased temperatures, both in the mid and end terms together with possible changes of other climatic factors like precipitation which may lead to higher tick-borne disease risks associated with expansion of the range of the targeted tick distribution. Distribution maps were constructed for the current, 2050, and 2070 for the two greenhouse gas scenarios and the most dramatic scenario; RCP 8.5 produced the highest increase probable distribution range. Conclusions: The future potential distribution of the genus Rhipicephalus show potential expansion to the new areas due to the future climatic suitability increase. These results indicate that the genus population of the targeted tick could emerge in areas in which they are currently lacking; increased incidence of tick-borne diseases poses further risk which can affect cattle production and productivity, thereby affecting the livelihood of smallholding farmers. Therefore, it is recommended to implement climate change adaptation practices to minimize the impacts.

Ribgrass Mosaic Tobamovirus Occurred on Chinese Cabbage in Korea

  • A tobamovirus, Ribgrass mosaic virus (RMV), was identified newly from chinese cabbage (Brassica campestris L. pekinensis) in Korea. Virus disease incidence of RMV on chinese cabbage was 37.9% in alpine area on August in 1993. RMV induced the symptoms of necrotic ring spots, necrotic streak on midrib and malformation. RMV, Ca1 and Ca3 isolate, could infect 35 species out of 45 plants including Chenopodium amaranticolor. Physical properties of RMV Ca1 isolate were very stable as 10.8 over for dilution end point, $95^{\circ}C$ for temperature inactivation point and 18 weeks for longevity in vitro. RMV had the soil transmission rate of 75.0% for the chinese cabbages, 'Chunhawang' and 'Seoul' cultivars. The purified virions of RMV had the typical ultraviolet absorption spectrum of maximum at 260 nm and minimum at 247 nm. RMV of Ca1 isolate was related serologically with antisera of Tobacco mosaic virus (TMV)-Cym, TMV-O and Pepper mottle virus, but not related with antiserum of Odontoglossum ring spot virus. coat protein gene of RMV-Ca1, sized 473 nucleotides, encoded 158 amino acid residues. Nucleotide identity of RMV-Ca1 CP gene was 96.4% with RMV-Shanghai (GenBank accession No. of AF185272) from China and 96.0% with RMV-Impatiens (GenBank accession No. of AM040974) from Germany. Identity of amino acids between RMV-Ca1 and the two RMV isolates was 96.8%. Specific three primers were selected for rapid and easy genetic detection of RMV using Virion Captured (VC)/RT-PCR method.

Evaluation of Control Efficacy of Biocontrol Agent, Epicoccosorus nematosporus on Eleocharis kuroguwai in the Field

  • This study was conducted to determine the efficacy of Epicoccosorus nematosporus for the control of Eleocharis kuroguwai and to evaluate the meteorological factors which affect weeding efficacy in field conditions for three years (1996-1998). The best time to control E. kuroguwai with E. nematosporus as a biological control agent in the field was in July, when temperature ranged from 20.4 to $23.4^{\circ}C$; the surface wetness duration was 12.6-16.1 hours, and application time of 6:00 p.m. and 8:00 p.m.; and weeding efficacy was 81-90%. On 10 June 1996 in Milyang area, where the field experiments were performed, mean temperature was $16.5^{\circ}C$ with 11.3 hours of dew duration. Meanwhile, on 20 Aug. 1996 the temperature was $21.3^{\circ}C$ with 15.4 hours of dew duration. During these periods, the weeding efficacy was recorded at 61.8 and 60.8%, respectively. Time required for complete plant death was 25.8 and 25.6 days at application times 10 June and 20 Aug., respectively. At the time of application on 7,18, and 27 July 1996, mean temperature was 20.4-$23.4^{\circ}C$ with 12.6-16.5 hours of dew duration. The weeding efficacies of these periods were very hi일 with 81.4-90.8%. Three years of field observations from 1996 to 1998 showed that infection in the field can occur at any time through the summer season, although total infection rates vary between months and between years. In 1996, plant infection rapidly increased from 56% on 30 June, 82.4% on 15 July, 94.6% on 15 August, and 92.8% on 15 September under favorable meteorological conditions such as minimum temperature of $17.6^{\circ}C$ and maximum temperature of $27.1^{\circ}C$, with 86% relative humidity and 977.5 mm of rainfall during E. kuroguwai growing season. However, in 1997, the disease incidence was very low because of unfavorable weather conditions brought about by the hot temperature and the low amount of rainfall at 321.5 mm. Disease progress was slow from 24.4% on 30 June to 49.2% at the end of the growing season.

A Prospect for Growth and Economic Size of Foods-for-Elderly Industry -Focused on Health Functional Foods and Foods for Special Dietary Uses- (고령친화식품산업의 성장과 규모 전망 -건강기능식품과 특수용도식품을 중심으로-)

  • The purpose of this study is to predict the economic size of foods-for-elderly market, which will be valuable information for establishing related policy and backup system. After setting the scope of related industry, detailed information for current market situation was investigated and a systematic forecast for market changes in the future was performed. Economic growth, changes in consumer expenditure and economic status of the elderly, current subscription of medical insurance and saving for pension were reflected. In addition, a survey toward related firms was completed and changes in aged population and incidence of chronic disease in the elderly were taken into account. Results show that the annual growth rate of the market was predicted to be the minimum 4.54% through the maximum 8.32% from 2010 to 2025 and its market size was forecasted to be the minimum 7,073 ten million won through the maximum 10,976 ten million won. It is expected that the market of foods-for-elderly will grow rapidly with development of foods technology and fast increase of aged population. Especially, growth of health functional foods and foods for special dietary uses for elderly will be distinguished. However, it seems that related firms are on the hedge, watching current trend of the related industry. This may results in insufficient supply against the demand. Therefore, policy for foods-for-elderly should be introduced and systematically administered, including R&D support, standardization and authentication for foods-for-elderly, construction of related database system.

Dehydrating and bacterial elimination effects of fecal dehydrating system for reducing bovine mastitis derived from environmental contamination (환경유래 젖소유방염 저감을 위한 우분뇨 탈수 시스템의 탈수 및 유방염 원인체 제균 효과 규명)

  • Bovine mastitis is an important disease causing serious economic loss in dairy production and food poison in public health. Escherichia coli and Staphylococcus aureus are the major causative agents of bovine mastitis. These bacteria were found in milk and environmental condition such as feces, water, soil and so on. Bovine mastitis causative micro-organisms can survive in 1-2 weeks in feces and bed complexes. Low level of percentage of water content (PWC) of feces and bed complexes can reduce the spreading of bovine mastitis incidence from environmental contamination. In this study, we developed the fecal dehydrating system and determined the elimination rates of bovine mastitis causative agent from feces and bed complexes. To develop the fecal dehydrating system, the screw pressurized dehydrating system was used and the maximum rate of dehydrating was reached to 52% PWC using 90% PWC (wet base) of fecal and bed complexes. The elimination rates of the dehydrating system for E. coli and S. aureus were reached at 41.19 $\pm$ 7.84% to 62.55 $\pm$ 8.71% in various percentages of PWC of feces and bed complexes (80, 85 and 90%). These results suggested that the application of fecal dehydrating system would reduce the exposure of dairy cattle to bovine mastitis causing agents contaminated feces and bed complexes, and can be used for environmental bovine mastitis control avoiding misuse or abuse of chemical disinfectants and antibiotics in dairy farm.

Characteristics of Cucumber mosaic virus-VCH Causing Vein Chlorosis on Red Pepper in Korea (고추에 엽맥퇴록병을 일으키는 오이 모자이크 바이러스(CMV-VCH)의 특징)

  • Cucumber mosaic virus(CMV) was occurred on red pepper showing vein chlorosis or vein necrosis with the incidence rate of 52% from 62 specimens collected in natural fields. Among 32 samples infected with CMV, the specimens of 22 red pepper leaves showing vein chlorosis were infected singly with CMV-VCH. CMV-VCH induced vein chlorosis on the inoculated leaves, and vein banding and vein necrosis on the upper leaves of Nicotiana glutinosa, and then killed after showing bud necrosis. The typical symptoms of vein banding, malformation and blister were produced on the upper leaves of Nicotiana benthamiana and N. tabacum 'Ky-57' without symptoms on the inoculated leaves. The commercial cultivars of 'Bugang', 'Manitta' and 'Gwariput' were shown the typical symptom of vein chlorosis by the mechanical inoculation of CMV-VCH. CMV-VCH was detected specifically by RT-PCR. Virus particles of CMV-VCH were isometric shape having 30 nm diameter. Ultraviolet absorption of purified CMV-VCH was maximum at 260 nm and minimum at 242 nm. The ratio of A260/A280 was 1.71. CMV-VCH had the single nucleo-protein having the molecular weight of 24.5 kDa.

Preoperative Risk Factors for Pathologic N2 Metastasis in Positron Emission Tomography-Computed Tomography-Diagnosed N0-1 Non-Small Cell Lung Cancer

  • Background: Accurate mediastinal lymph node staging is vital for the optimal therapy and prognostication of patients with lung cancer. This study aimed to determine the preoperative risk factors for pN2 disease, as well as its incidence and long-term outcomes, in patients with clinical N0-1 non-small cell lung cancer. Methods: We retrospectively analyzed patients who were treated surgically for primary non-small cell lung cancer from November 2005 to December 2014. Patients staged as clinical N0-1 via chest computed tomography (CT) and positron emission tomography (PET)-CT were divided into two groups (pN0-1 and pN2) and compared. Results: In a univariate analysis, the significant preoperative risk factors for pN2 included a large tumor size (p=0.083), high maximum standard uptake value on PET (p<0.001), and central location of the tumor (p<0.001). In a multivariate analysis, central location of the tumor (p<0.001) remained a significant preoperative risk factor for pN2 status. The 5-year overall survival rates were 75% and 22.9% in the pN0-1 and pN2 groups, respectively, and 50% and 78.2% in the patients with centrally located and peripherally located tumors, respectively. In a Cox proportional hazard model, central location of the tumor increased the risk of death by 3.4-fold (p<0.001). Conclusion: More invasive procedures should be considered when preoperative risk factors are identified in order to improve the efficacy of diagnostic and therapeutic plans and, consequently, the patient's prognosis.

Effect of Soluble-silicate or Chitosan Foliar Spray on Ginseng Cultivated in Blue-white Plastic Film House

  • The experiments were performed in the Jinan (elevation: 300 meters above sea level), Jeollabuk-do. Seedlings (n = 63 per $3.3m^2$) of ginseng cultivar (Cheonpung, Yeonpung) were planted on April 10, 2015. Shading material of plastic film house was blue-white film. Before the Planting seedling, silicate (3 kg/10 a) or chitosan (40 kg/10 a) was fertilized and foliar sprayed on the leaves 1000 times dilution solution once a month from May to September every year. The growth results of 5-year old ginseng surveyed in 2018 are as follows. The average air temperature in the plastic film house was the highest at $26.6^{\circ}C$ and $26.5^{\circ}C$ in July and August, respectively, and the highest temperature was $40.5^{\circ}C$ in July. The maximum daily temperature of $35^{\circ}C$ or more was 30 days, with the average soil temperature being $24.9^{\circ}C$ in August. The chemical properties of the test soil are as follows. pH was 6.4~6.9 level and EC was 0.35~0.46 dS/m. The organic matter content was 33.5~41.4 g/kg, and available-P content was 251.9~306.8 mg/kg. Exchangeable cations contents, such as K, Ca and Mg were all the appropriate ranges. The soil microbial density surveyed by the dilution plate method was 10~50 times higher than that of control (Non-treatment) and actinomycete density was 3~6 times higher. Pathogens of the genus Fusarium by Metagenome analysis decreased 91.3% and 68.2% respectively in the foliar sprayed of chitosan and soluble-silicate. The light intensity (PAR) in the blue-white film plastic film house gradually increased until July and then decereased, with the average of light intensity in March-October was $120.3umol/m^2/s$. The growth of aerial parts such as plant height and stem length was better than non-sprayed group in silicate or chitosan treatments and Yeonpung cultivar was superior to the Cheonpung cultivar. The SPAD value was higher in Yeonpung cultivar foliar sprayed with soluble-silicate. The growth of underground parts such as root length and taproot length were better in chitosan and soluble-silicate treatment than control, especially in Yeonpung cultivar foliar sprayed with chitosan was good in taproot length and taproot diameter, and fresh weight of root was 60.1 g. Ginsenoside contents were 24.9 mg/g and 22.4 mg/g, in the Cheonpung cultivar foliar sprayed with soluble-silicate or chitosan respectively, 28% and 15% higher than control (19.5 mg/g). The incidence of disease by Alteraria panax and Botrytis cinerea was 3~9% and 4~9%, respectively. High temperature damage rate was 3~5%.

Emergence Characteristics of Fire Blight from 2019 to 2023 in Korea (2019-2023년 국내 과수 화상병의 발생 특성)

  • Erwinia amylovora is a gram-negative plant pathogen that causes fire blight in apple and pear trees, resulting in significant damage worldwide. In this study, we monitored the emergence of fire blight from 2019 to 2023 to determine the emergence patterns and the factors affecting the outbreak of the disease. As a result of the 5-year survey on the emergence of fire blight, a total of 2,029 cases have emerged, mostly in apple trees of 1,378 cases (67.9%) followed by 645 cases (31.8%) in pear trees, and from quince, hawthorn, and mountain ash trees. Fire blight appeared in specific areas of Gyeonggi, Chungnam, Gangwon, and Chungbuk provinces in 2019, but spread to Andong and Yesan in 2021, Muju and Bonghwa in 2023. In 2020 and 2021, there were 744 and 618 cases of fire blight outbreaks, respectively, compared to other years (188-245 cases/year). Notably, 914 of these cases were observed in apple trees from May to July, with 667 cases reported in Chungju and Jecheon. The incidence of fire blight was positively correlated with the daily maximum temperatures and rainy days in January and February, as well as the rainy days in May and June. The average age of the diseased pear trees was 25 years, higher than the 10-year average age of the apple trees. This study provides fundamental information to understand the status and factors affecting the fire blight emergence in Korea. Prevention measures should be established through continuous analysis of the status of fire blight.
