Analysis of Environment and Production of Tricholoma matsutake in Matsutake-infected Pine Trees (송이 감염묘를 이용한 송이 발생 및 발생환경 분석)

  • Tricholoma matsutake (Pine mushroom) is expensive, and its artificial cultivation has been tried in several countries. Until date, the only successful cultivation of artificial pine mushroom in pine forests uses matsutake-infected pine trees. The National Institute of Forest Science in Korea has been restudying this method since 2000. Success in fruit production and reproduction was achieved in 2010 and 2017, respectively, in the same locale. The successes proved that pine mushrooms could be cultivated artificially in the field using matsutake-infected pine trees. The fruiting of pine mushroom in October 2010 occurred 6 years, 6 months after the transplantation of matsutake-infected pine trees. Five pine mushrooms reoccurred in September 2017, 13 years, 5 months and 15 years, 5 months after the transplantation of the respective matsutake-infected pine trees. The distance between the matsutake-infected pine tree and the pine mushrooms was 12 cm at 6.6 years, 90~115 cm at 13.5 years, and 95 cm at 15.5 years. Fruiting bodies occurred 13~16 days after the underground temperature declined to below $19^{\circ}C$. In conclusion, the use of matsutake-infected pine trees remains the only way to artificially cultivate pine mushrooms. More knowledge of the environmental factors affecting matsutake fruiting would be beneficial.

Spore Dispersion of Tricholoma matsutake at a Pinus densiflora Stand in Korea

  • The spore of Tricholoma matsutake is considered to be the starting point of the mushroom growth cycle, but the mechanism of mycelial development from the spore stage is not yet clarified. In this study, we tried to measure how far the spores of T. matsutake disperse from a fruiting body located at a Pinus densiflora stand in Korea. We established 16 slide glasses coated with glycerin near a fruiting body in four directions separated by four different distance intervals within a mushroom productive stand after removing all other fruiting bodies from three plots. The number of dispersed spores increased with time from the first day (475 $spores/cm^2$) to the fourth day (836 $spores/cm^2$) after the pileus opened. The number of spores dispersed downward was about 1.5 times greater than that dispersed toward the ride. The number of dispersed spores decreased exponentially as the distance from each fruiting body increased. More than 95% of the spores dropped within a meter from the fruiting body, with 75% dropping within 0.5 m. Even so, the number of spores dispersed over 5 m from the fruiting body was more than 50 million when considering the total number of spores produced by a fruiting body is about 5 billion.

Cloning and Organization of the Ribosomal RNA Genes of the Mushroom Trichloma matsutake

  • A portion (7.4 kb) of ribosomal DNA tandem repeat unit from a genome of the mushroom T. matsutake has been cloned. A 1.75 kb EcoRI fragment was cloned first using S. cerevisiae 255 rRNA gene as a probe, and this was then used for further cloning. A chromosomal walking experiment was carried out and the upstream region of the 1.75 kb fragment was cloned using SmaI/BamHI enzyme, the size was estimated to be 5.2 kb in length. Part of the downstream region of the 1.75 kb fragment was also cloned using XbaI/BamHI enzymes. Restriction enzyme maps of three cloned DNA fragments were constructed. Northern hybridization, using total RNA of T. matsutake, and the restriction fragments of three cloned DNAs as probes, revealed that all four ribosomal RNA genes (large subunit[LSU], small subunit [SSU], 5.85 and 5S rRNA genes) are present in the cloned region. The gene organization of the rDNA are regarded as an intergenic spacer [IGS]2 (partial) - SSU rRNA - internal transcribed spacer [ITS]1 - 5.8S rRNA - ITS2 - LSU rRNA - IGS1 -5S rRNA - IG52 (partial).

  • This study was attempted to investigate the effect of Tricholoma matsutake on experimentally induced hyperlipemia in rats. The levels of cholesterol, triglyceride and phospholipid were measured. Diagnostic attention has been paid to the cholesterol concentration associated with high density lipoprotein (HDL), which appears to be inversely related to the incidence of coronary artery disease. For these reasons, determination of HDL-cholesterol has significant meaning. Total cholesterol was determined by Abell-Kindall method, and to measure HDL-cholesterol, serum low density lipoproteins (LDL) were first selectively precipitated by HDL-precipitating reagent. The results obtained were as follows : 1) The level of total cholesterol in serum of hyperlipemic rats was decreased by oral administration of Tricholoma matsutake extract. 2) The level of HDL-cholesterol was increased by the mushroom extract. 3) The level of triglyceride was significantly decreased by the mushroom extract. 4) The level of phospholipid was slightly decreased by the mushroom extract.

Comparison of the proximate composition and amino acid content of domestic and imported Tricholoma matsutake and Sarcodon aspratus (국내산 및 수입산 송이버섯과 능이버섯의 일반성분 및 아미노산 함량 비교)

  • This study investigated the proximate composition and total amino acid contents in Tricholoma matsutake and Sarcodon aspratus to compare the food quality according to production area. The crude ash contents in Tricholoma matsutake from Yanji (China), Yangyang (Korea), Tibet (China), and Yunnan (China) were 6.95%, 6.40%, 5.52%, and 5.40%, respectively. Yangyang showed the lowest crude fat content (1.19%), whereas Tibet exhibited the lowest crude protein content (16.83%), showing a difference according to the production area. The total amino acid content of Tricholoma matsutake in Yanji, Yunnan, Tibet, and Yangyang was $11,490.14{\pm}892.07mg%$, $8,000.03{\pm}207.25mg%$, 6,815.48 mg%, and $6,074.74{\pm}814.86mg%$, respectively. The contents of proximate composition and total amino acids in Sarcodon aspratus showed no difference according to production area. The results of proximate composition in Tricholoma matsutake suggest that the crude ash content can be used as a distinguishing indicator between Tricholoma matsutake from Yangyang and Tibet. Further studies are needed to analyze the mineral contents for the establishment of distinguishing mineral indicators between Tricholoma matsutake from Yangyang and Tibet.

Determination of Differences in the Nonvolatile Metabolites of Pine-Mushrooms (Tricholoma matsutake Sing.) According to Different Parts and Heating Times Using $^1H$ NMR and Principal Component Analysis

  • The differences in the nonvolatile metabolites of pine-mushrooms (Tricholoma matsutake Sing.) according to different parts and heating times were analyzed by applying principal component analysis (PCA) to $^1H$ nuclear magnetic resonance (NMR) spectroscopy data. The $^1H$ NMR spectra and PCA enabled the differences of nonvolatile metabolites among mushroom samples to be clearly observed. The two parts of mushrooms could be easily discriminated based on PC 1, and could be separated according to different heattreated times based on PC 3. The major peaks in the $^1H$ NMR spectra that contributed to differences among mushroom samples were assigned to trehalose, succinic acid, choline, leucine/isoleucine, and alanine. The content of trehalose was higher in the pileus than in the stipe of all mushroom samples, whereas succinic acid, choline, and leucine/isoleucine were the main components in the stipe. Heating resulted in significant losses of alanine and leucine/isoleucine, whereas succinic acid, choline, and trehalose were the most abundant components in mushrooms heat-treated for 3 min and 5 min, respectively.

Ectomycorrhizal Mushroom Occurrence around the Fairy Ring of Tricholoma matsutake at a Pine-Mushroom Forest (송이 발생림의 송이 균환 주변에 출현하는 외생균근성 균류)

  • The occurrences of ectomycorrhizal fungi were investigated in a pine-mushroom forest at Hongcheon, Korea. The fairy rings of Tricholoma matsutake were figured by the sporocarp places of T. matsutake for three years $(1995{\sim}1997)$, and the occurrences of other ectomycorrhizal fungi were surveyed with x and y dimensions for two years $(1996{\sim}1997)$. The diameters of fairy rings of T. matsutake ranged from 2m to 10m, which indicated that the age of the fairy rings as $10{\sim}50$ years when we consider that the growth of the fairy ring used to show about 10 cm per year. Russula bella, R. sororia, R. delica and Cantharellus minor were the major species occurred on the site during the survey period, and each species occupied 16.0%, 12.8%, 12.4% and 7.0% of total mushroom occurrence, respectively. From the results, we could conclude that the surveyed stand was a productivity-declining forest from the view point of pine-mushroom production. In addition, Amanita pantherina, Suillus bovinus, Ramaria flaccida and Laccaria amethystea were considered to be the indicator species for declining of pine-mushroom productivity since some fruiting bodies of the species appeared around the fairy ring of T. matsutake.

Comparison of Macro and Micro Mineral Contents in Domestic and Imported Tricholoma matsutake (국내산과 수입산 송이의 다량 및 미량 미네랄 함량 비교)

  • The mineral content of Tricholoma matsutake was evaluated for comparison of mineral contents according to the area of cultivation. Ten domestic and thirty Chinese (10 Yanji, 10 Yunnan and 10 Tibet) T. matsutake specimens were assessed using an atomic absorption spectrophotometer (AAS) and inductively coupled plasma mass spectrometer (ICP-MS). The Na, Mg, K, and Ca contents of domestic T. matsutake were 128.12±85.25 mg/kg, 218.52±105.35 mg/kg, 7,534.58±2,691.52 mg/kg, and 17.69±7.14 mg/kg, respectively, while those of Yanji T. matsutake were 124.89±57.24 mg/kg, 64.07±27.52 mg/kg, 1,439.18±311.04 mg/kg, and 10.88±4.52 mg/kg, respectively. The Na, Mg, K, and Ca contents of Yunnan T. matsutake were 90.78±23.23 mg/kg, 77.40±28.36 mg/kg, 1,446.29±126.33 mg/kg, and 28.42±5.18 mg/kg respectively, while those of Tibet T. matsutake were 143.50±41.54 mg/kg, 124.64±50.18 mg/kg, 3,530.95±2,714.99 mg/kg, and 21.05±8.71 mg/kg, respectively. The Cu contents of domestic, Yanji, Yunnan, and Tibet T. matsutake were 105.43±32.97 mg/kg, 19.92±8.95 mg/kg, 54.51±16.91 mg/kg, and 64.80±23.01 mg/kg, respectively. Both domestic and Chinese T. matsutake samples showed significantly different K, Mg, and Cu levels in this study. Therefore, a comparative evaluation of the K, Mg, and Cu contents of multiple domestic and Chinese T. matsutake varieties is needed to determine the appropriate area of cultivation in the future.

Studies on Analyzing Meteorological Elements Related with Yield of Tricholoma matsutake (S. Ito et Imai) Singer. (송이(Tricholoma matsutake)생산(生産)과 관련되는 기후특성분석(氣候特性分析))

  • This study was carried out to obtain basic information elucidating relationship meteorological elements and yield of Tricholoma matsutake. The yields of pine mushroom in seven areas including Yangyang-gun more than 30,000 kg. Pine mushrooms were generally distributed in the red pine (Pinus densiflora) forests of East-Coast Region, Sobaek and Taebaek-Mountainous Regions, Yeongnam-Central Region and North-Central Region of Korea. Yields of pine mushroom were significant correlation with precipitation during April-October in last years, average minima temperature during July-September, rainy days during Aug. 21-Sep. 10 and precipitation during Sep. 11-30, etc. Air and soil temperature during pine mushroom yield period were 15-25 and $15-20^{\circ}C$, respectively. The first collection date of pine mushroom could be predicted by measuring the soil temperature at 5 cm depth.

Ergosterol and Water Changes in Tricholoma matsutake Soil Colony during the Mushroom Fruiting Season

  • The purpose of this study is to understand spatio-temporal changes of active fungal biomass and water in Tricholoma matsutake soil colonies during the mushroom fruiting season. The active fungal biomass was estimated by analyzing ergosterol content at four different points within four replicated locations in a single circular T. matsutake colony at Ssanggok valley in the Sogri Mt. National Park in Korea during 2003 to 2005. The four points were the ahead of the colony, the front edge of the colony and 20 cm and 40 cm back from the front edge of the colony. Ergosterol content was 0.0 to 0.7 ${\mu}g$ per gram dried soil at the ahead, 2.5 to 4.8 ${\mu}g$ at the front edge, 0.5 to 1.8 ${\mu}g$ at the 20 cm back and 0.3 to 0.8 ${\mu}g$ at the 40 cm back. The ergosterol content was very high at the front edge where the T. matsutake hyphae were most active. However, ergosterol content did not significantly change during the fruiting season, September to October. Soil water contents were lower at the front edge and 20 cm back from the front edge of the colony than at the ahead and 40 cm back during the fruiting season. Soil water content ranged from 12 to 19% at the ahead, 10 to 11% at the edge, 9 to 11% at the 20 cm back and 11 to 15% at the 40 cm back. Our results suggest that the active front edge of the T. matsutake soil colony could be managed in terms of water relation and T. matsutake ectomycorrhizal root development.
