  • Daphne pseudomezereum var. koreana is native to Korea and is distributedin Kangwon-do, Jeollabuk do, and Gyeongsang-do. This economically valuable species has experienced a dramatic decrease in natural habitat due to climate change and is difficult to cultivate. In this study, we investigate a mass propagation method for D. pseudomezereum through in vitro culture and genetic resource preservation.WPM medium was better than the MS medium for shoot growth. As a result, we compared the shoot number and length of apical (W/AP) and non-apical shoots (W0/AP) with BA and GA3 treatments in WPM medium. Their shoots and length grew well in both BA 8ìM + GA38ìM-treated apical shoot and without-apical shoot. NAA did not effectively induce rooting of the in vitro plantlet.

Establishment of propagation system for in vitro calla plants (Zantedeschia spp.) by treatment of taurine (타우린 처리를 통한 칼라 기내 식물체 대량증식체계 확립)

  • Zantedeschia spp. calla is very popular as a cut flower. It is very important to establish a micro propagation system through plant tissue culture with the problem that colored calla with various colors are low in natural reproduction rate and vulnerable to high temperature. In this study, we conducted the experiment by adding taurine to improve the growth of calla plant. When 20 mg/L of taurine was added with plant growth effect, 54.0 % of the cases of multiple shoots and 17.2 times of fresh weight were the most effective. Taurine 20 mg/L treatment showed 16.0 % and 39.2 %, respectively, than the untreated control. Taurine may contribute to mass propagation of elite breeding lines as well as an improvement of farm income by positively influencing the overall growth of calla plants, thereby positively affecting the establishment of the micro propagation system of calla shoot tips.

Comparative Bioreactor Studies in Terms of Oxygen Transfer between Suspended and Immobilized Fungal Systems for Cyclosporin A Fermentation (Cyclosporin A 생산을 위한 액체배양과 고정화배양의 생물반응기에서의 산소전달 비교 연구)

  • In fermentations with a 4-liter stirred tank bioreactor, a better than two-fold enhancement of the gas-liquid mass transfer coefficient$(k_La)$ in the celite-immobilized fungal cultures of Tolypocladium in flatum over the parallel conventional free-cell was observed at identical biomass concentrations, despite the higher specific oxygen uptake rate of the immobilized fungi during exponential growth. As a result oxygen sufficient conditions, i. e., dissolve oxygen(D.O.) concentrations exceeding 75% air saturation, could be maintained throughout exponential growth period of the immobilized culture, in contrast to the suspended fungal culture, whose D.O. levels fell below 50% air saturation. A linear monotonic dependence of $k_La$ upon impeller agitaion rate was found for both immobilized and conventional cultivation modes over a range of 250 to 550rpm, the slope being a function of biomass concentration for the free but not for the immobilized cell system In contrasts oxygen transfer rate was a much weaker function of aeration rate up to about 2.5 vvm for both culture configurations. Above this level, aeration rate had no further effect on the mass transfer. In addition, the immobilized cultures sustained good morphological and physiological states, leading to almost two times higher cyclosporln A (CyA) productivity overt the parallel free cell system. These experiments suggest that the celite-immobilized fungal system in a stirred tank reactor has considerable promise for scaling up cyclosporin A production in terms of high-density cultivation.

  • According as the fashion at the latter half part of the 20th century is diversified and made into high-class gradually, it is demanding the individuality of each one. Especially, since the beginning of 1980s, fashion came to represent the restoration mood in a modern style because of the popularization of post-modernism. And with this, it came to result in the reappearance of minimalism which was one of anti-culture in 1960s. In this study, this researcher grasped the appearance background, concept and structural concept of minimalism, and tried to study the influence that he minimalism had on fashion in the flow of fashion over the first half period of the 20th century. Minimalism is the fine art trend of American characteristic which quicked by the meeting of European culture and American culture, according as the new avant-grade art of Europe moved to America after World War II Minimalism which appeared, refusing the mechanisation, industrialization and mass production of America and the falsehood of consciousness and sensitivity under existing system remained obscure by the pop art which is the commercial fine art of same period, but it came to expand the domain of work possibility of modern fine art, and it became the basis of appearance of post-modernism. Minimalism has 4 kinds of structural features which are nonside nature, repetitiousness and environment nature with 3 dimensional feature which is issue nature. Minimalism is appearing in the exfcellent and future-oriented fashon of Andre Kureju t the time of 1960 well. Then science revolution and minimalism aroused the mini-revolution, and they made the compact radio in electric home appliances and the miniskirt in fashion appear. The features of minimal fashion as Retro '60' which appeared in the decedent mood with the 21st century just before on are as follows: First, the are the pursuit of concise plastic line. Second, it is the use of high tech material Third, the miniaturization in size amy be enumerated. Minimal fashion will appear s the positive confrontation from of mankind as the optimistic viewpoint about feature in the situation hat environment was destroyed and the resources were exhausted.

Effect of biocide addition on plantlet growth and contamination occurrence during the in vitro culture of blueberry

  • Interest and great demand for blueberry (Vaccinium corymbosum) have increased, as V. corymbosum is now one of the most economically important crops in Korea. It is expected that blueberry production and the area planted for cultivation will increase consistently in the years ahead because of high profitability and the consumer's demand for healthy ingredients. Effective mass production of blueberry is urgently needed for commercial cultivation establishment, but a main limitation is lack of a propagation system that produces a disease-free plant material for commercial plantation. A large amount of research has focused entirely on developing tissue culture techniques for blueberry propagation. However, controlling fungal and bacterial contamination of woody plant material is extremely difficult. Our study was conducted to investigate the effect of biocide addition during the in vitro culture of blueberry on plantlet growth and contamination occurrence. Four biocides, including Plant Preservative Mixture ($PPM^{TM}$), vancomycin, nystatin and penicillin G, were used in varying concentrations during the in vitro propagation of blueberry. When nystatin was added into the medium at low concentrations, the overall growth of blueberry plantlets was retarded. Addition of vancomycin and penicillin G in high concentrations decreased contamination but induced plantlet mortality. On the other hand, when 1ml/L $PPM^{TM}$ was added, the growth characteristics of blueberry plantlets did not significantly differ from non-treatment (control), and the contamination occurrence rate was very low. From these results, we found that the addition of the appropriate biocide could provide an effective method to reduce contamination in the culture process, thereby raising in vitro production efficiency.

Overproduction of Streptomyces griseus Protease A and B Induces Morphological Changes in Streptomyces lividans

  • The sprA and sprB gene encoding chymotrypsin-like proteases Streptomyces griseus protease A (SGPA) and Streptomyces griseus protease B (SGPB) and the sprT gene that encodes Streptomyces griseus trypsin (SGT) were cloned from Streptomyces griseus ATCC10137 and overexpressed in Streptomyces lividans TK24 as a heterologous host. The chymotrypsin activity of tole culture broth measured with the artificial chromogenic substrate , N-succinyl-ala-ala-pro-phe-p-nitroanilide, was 10, 14 and 14 units/mg in the transformants haboring the sprA, sprB and sprD genes, respectively. The growth of S. lividans reached the maximum cell mass after 4 days of culture, yet SGPA and SGPD production started in the stationary phase of cell growth and kept increasing for up to 10 days of culture in an R2YE medium. The trypsin activity of the culture broth measured with the artificial chromogenic substrate , N-${\alpha}$-benzoyl-DL- arginine-p-nitroanilide , was 16 units/mg and SGT production started in the stationary phase of cell growth and kept increasing for up to 10 days of culture in an R2YE medium. The introduction of the sprA gene into S, lividans TK24 triggered the biosynthesis of pigmented antibiotics, actinorhodin and undecylprodigiosin, and induced significant morphological changes in the colonies in Benedict, R2YE, and R1R2 media. In addition, the introduction of the sprT gene also induced morphological changes in the colony shape without affecting the antibiotic production, thereby implying that certain proteases would appear to play very important and specific roles in secondary-metabolites formation and morphological differentiation in Streptomyces.

  • This study was conducted to evaluate changes in food consumption during the 1950's on the basis of articles that reflected national policy and changes in society during that time period. Many factors are involved in the development and changes in food consumption culture, and these factors can affect each other. As a result, the process involved in the development of food consumption culture acts as a living system. This study evaluated the food consumption culture during the 1950's because this period was subjected to obvious influences that may explain the modern food consumption market logic and commercialism. Changes in the national food consumption are dependent on natural changes such as income enlargement or cultural exchange with a foreign country. Accordingly, food consumption during the 1950's was influenced by changes in economical, social, and political needs. In addition, the influx of surplus agricultural products from the United States had an adverse effect on local agriculture and resulted in an increased external dependence on food during the 1950s. Moreover, the import of raw materials and simple manufacturing techniques led to the development of an industrial food processing industry that enabled accelerated mass production of food at a low-price. Furthermore, the importation of surplus agricultural products from the United States that were used as the raw materials for foods that had traditionally been produced domestically led to an increased burden and qualitative decline in the local food-service industry. Taken together, the results of this study indicate that during the 1950's fresh food began to be replaced with processed foods in Korea.

  • Che Guevara spearheading the Cuban Revolution was not only the icon as the revolutionary to the New-leftists of the 1960s advocating the ideology of Marxism but, as the cultural revolutionary, had a tremendous influence on the younger generation living in the digital age Che Guevara take on a figure of both the symbol of the Leftist but the romantic revolutionary because he had the external features such as black beret, red stars, military upper jackets and trousers, beard and pipe tobacco. In fact, the symbolic image of Che Guevara was made as the popular image by the avantgarde artists and political vanguard forces of the times under the necessity of Cuban government. Afterwards, the image of Che Guevara has been patronized in making people of aware of the resistant and revolutionary image to capital, power and the power of the media and symbolized as the resistant image to the American capital as well as the revolutionary guerrilla. And his image has continued to be reproduced and symbolized for the commercial and political purposes and as the enthusiastic image of youth culture. This can be seen as having been created as a new image by the popular culture formed by the development of the cyber culture and mass media in the cyberspace shaped by contemporary 'N' generations. The use of Che Guevara's symbolic image was made in the fashion field as well as in the cultural and artistic circles. The borrowing of the icon of Che Guevara represented in a fashion field is attributed to his free spirit, and it can be seen that fashion exists as the vehicle for representing both the symbol system and the sign system containing ideologies and texts as the communicator of resistance to the regime and to social issues. Therefore, this study attempted to investigate the commercial iconization of Che Guevara in the 1990s by comparing the ideology of the symbol in the 1960s and the 1990s and inquire into the borrowing of his image by the fashion domain as well as the fashion worn by him by reference to domestic books and materials on the fashion site. Thereby, this study attempted to make clear that the borrowing of Che Guevara in the realm of fashion since the 1990s not only contained the meaningful interpretation as the symbolic code in the culture of young people living in a digital era but fashion performs an intervening role in the cultural phenomenon.

Establishment of Mass Propagation System of Virus-Free Sweetpotato Plants and Conservation

  • Sweetpotato fields in Korea are highly infected with virus and virus like diseases that greatly diminish both yield and quality as indicated by field observations and laboratory tests. In order to solve this problem, there is an urgent need to produce and mass propagate virus-free planting materials for distribution to the farmers. These experiments were conducted, firstly, to determine the most appropriate culture media, nutrient solution, and cutting intervals to maintain growth and vigor of tissue cultured plantleta as mother plants for propagation in insect-proof greenhouse. And as a labor saving method, the production efficiency of plug trays for rapid propagation of stem cuttings as a source of planting materials was likewise evaluated. Results showed that plants grown in medium B supplied with 0.5 and 1.0 strength of MS nutrients had high growth rate, and 20-day cutting interval was the best. 72-plug tray was better than 128-plug. Secondly, it was to develop a technique for the production of first-generation seed roots using hydroponics cultivation system. The yield of virus-free plants propagated in the non-insect proof and open-field cultivation was 2,402 kg/10a, 6% higher than those in the insect-proof cultivation, and the rate of virus re-infection was 18% higher compared to 3.3% with insect-proof cultivation. Lastly, it was to investigate the growth performance of virus free plants in farmers' field. Differences were existed in the yield depending on the variety used, but virus free plants showed an increase of $6{\sim}24%$ over virus infected plants.

In Vitro Propagation of Medicinal Herbs in Korea

  • Mass production of forest medicinal plants is related to quality control of raw medicinal materials. Plant tissue culture is an important technology to produce high-quality plant materials. Numerous factors are reported to influence the success of in vitro regeneration of medicinal plants. Embryogenesis is known to be the most effective techniques and it has developed in some medicinal plant species. Various in vitro cultural condition for direct and/or indirect somatic embryogenesis systems have developed in Epimedium koreaum, Bupleurum falcatum, Paeonia lactiflora, Chrysanthemum zawadskii, Houttuynia cordata etc. In this study, we provide the present statue and information of in vitro propagation techniques that is able to apply as an efficient system for rootstock propagation system of forest medicinal plants.
