A Study on Marketing of Cultured Laver Products (양식해태의 유통에 관한 조사 연구)

  • Laver io one of the most necessary and seasonal items in Korean food from oldtimes. Laver is lagely eaten in dried form, and its supply depends entirely upon culture weeds. The history of laver culture in Korea about sixty or seventy years is older than in Japan. Significance of laver culture is divided into two aspects, one is food supply in the nation, and the other is export to other countries. Houses engaged in laver culture are about foully thousands, and laver production in 1972 is estimated as 1, 3 bitten sheets. (1 sheet is a dried laver of 20 cm sq, in the shape of paper) Especcially meaning of layer production is the concentration of labour input, and systematic management of labour. From around 1920, the method of laver culture was introduced by Japanese Imperialism for mono culture in shallow seas, and mass products of laver is provided to Japan market, DOMESTIC MARKET Fundamental consume function calculates at below, $D_{(68_71)}$=16354 $Y^{0.471}$ $P^{-1.0662}$ where D is total layer demand, Y income variable, P price variable. Futher new culture farms have been developed by means of break- water fences or by floating culture method. These improvements have been backed up with increased demand of laver products. Import quantity and price of Korean laver products are restrained by three organizations, that is producer, distributor and consumer. This relationship calculated by regression equation shows that import is influenced only producer organization, at the sacrifice of consumer profit. For increase to export of laver products, we urgently require to open foreign trade of laver products for Japanese consumer, .and Japan has political responsibility to solve Korean laver structure. But with long run timeseries, as regards Japanese production and import quantity, importing function shows increasing trend as below, 250 million sheets <3, 947.1674+0.005 $L_{g}$ >) 600 million sheets where $L_{q}$ is relative production quantity of laver in Japan. Comparison between Single and Co-culture of Adipocyte and Muscle Cell Lines in Cell Morphology and Cytosolic Substances (지방과 근육 세포주의 단독 및 공동배양을 통한 세포형태학 및 세포물질 비교 연구)

  • Present study was performed to investigate the effect of single and co-culture of adipocyte and muscle cell lines on cell differentiation. 3T3-L1 (adipocyte) and L6 (muscle) cell lines were single-cultured on the condition of 10% fetal bovine serum (FBS)/Dulbeco's modified eagle's medium (DMEM) for 48 h followed by culture within 5% FBS/DMEM as a growth media. Then, the growth media was replaced by differentiation media composed of 2% FBS/DMEM without additives in single- or co-culture of the 3T3-L1 and the L6 cells to induce differentiation of both cell types. In co-culture system, the 3T3-L1 and the L6 cells were grown in separated places by being seeded on a $0.4{\mu}m$ insert membrane and on the bottom of 6 well plate, respectively. Cell differentiation was measured using morphological investigation and cytosolic analysis of glycerol-3-phosphate dehydrogenase (GPDH; for 3T3-L1) and creatine kinase (CK; for L6). Based on the GPDH results, the presence of L6 cells did not stimulate 3T3-L1 differentiation showing more differentiation of 3T3-L1 cells in the single-culture compared to the co-culture condition. In contrast, 3T3-L1 cells in the co-culture promoted differentiation of L6 cells. Enzymatic analysis supported this result showing that 3T3-L1 cells showed statistically (P<0.05) higher GPDH activity in the single-culture than the co-culture, whereas CK results of L6 cells were vice versa (P<0.05). Overall, present results may indicate that co-culture system is more reliable and precise technique compared to single-culture. Further studies on several co-culture trials including different media conditions, supplementation of differentiating substances, molecular biological analysis, etc. should be required to obtain practical and fundamental mass data.

Occurrence of severe soybean-sprout rot caused by Pythium deliense in the recirculated production system

  • Severe soybean-sprout rot was found at the mass productive factory in 2000 and 2001 and it caused 10-20% loss of the production. Pythium sp. was isolated almost 90% by potato dextrose agar from rotted root and hypocotylsof the sprouts. And the pathogencity tests using test tubes with 2% water agar and small containers (30 ${\times}$ 30 ${\times}$ 50 cm, WxLxH) cultivation were shown a similar rot on roots and hypocotyls. The fungal mycelium grew rapidly on the water agar and it prevented the seed germination. Density of the Pythium sp. in the recycled water system at the factory was periodically measured using a selective medium, corn meal agar with Pimaricin 10 mg, Rifampicin 10 mg, Ampicillin 100 mg per 1 liter in order to check the contamination of recycled water. After fitering step using 5 and 1 ml in the recycled system was applied and it was effectively controlled Pythium rot. The daily yield of sprout was stable and the occurrenceof Pythium in the recycled water was much less after filtering. The fungal isolates were identified as Pythium deliense Meurs based on various mycological characteristics on corn meal agar and sucrose-asparagine bentgrass leaf culture medium. P. deliens oogonia were spherical, smooth, 19-23 urn in diameter, and their stalk bending toward antheridia. Antheridia were straw hat-shaped, curred club-shaped, therminal or intercalary, monoclinous, occasionally diclinous, 12∼15 ${\times}$ 8∼11 um, 1(∼2) per oogonium.

Performance Analysis of Ostermeier's Hamlet (공연분석: 오스터마이어의 <햄릿> (프랑스 2008, 한국 2010))

  • Ostermeier's Hamlet has no particular contemporary reinterpretation. Alternately devoting to show retrospect in illusion and existence through revenging actions. However, Shakespeare's metaphorical and implicative language is dissipated and the style of the play is not an illusive space-time, but a tragic theatrical production that uses rough language to express the depth of the story. The Perfomance of Hamlet is a sensuous jumble up of a diverse range of mass media. The double roles that the actors carry out give an affect of isolation between the audience and the play itself showing both empathy and liberty. Ostermeier's Hamlet distinctively shows a post-modern performance through the prominent elements of dirt, the use of mixed genre, theatric emphasis, making an image and the fulfillment of acting. Nonetheless, Ostermeier's performance stays off the point on the breakup strategy of the post-modern drama without suspending the narrative of Shakespeare's Hamlet. Besides aiming to show a performance centered by the imagery of physical expression, his performance shows New Realism in the 1960's, showing everyday life. Ostermeier thinks, that theatre helps give contemporary people an accurate reality check in the constant unstable periods of time. Therefore, Hamlet shows post-modern physical expression and outspoken dramaturgy using the effects of mass media in New Realism without breaking up realistic narration. With being the aberration of the Castle Helsingor, the main character Hamlet, expresses lunacy and can be considered as metaphor for young adults whom are broken down and isolated from the economic system. He is a substitute for those who experience agony, anger, torment, etc. and other suppressed emotions in everyday life. With the method of direction in the portrayal of Hamlet show signs of succession in the abundant popularization of the classics by communicating with the audience by following the trend of modern mass media and audio-visual perception; emphasizing the point of the philosophical topic 'life and death,' 'life and theatre,' and 'illusion and reality.'

주정증류 폐액을 이용한 Actinobacillus sp. EL-9로부터 Poly-$\beta$-Hydroxybutyrate의 생산 및 폐약의 처리

  • Alcoholic distillery wastes are utilized as dual purposes to produce PHB in lower production cost and to reduce the amount of waste to be treated. In this study, various attempts were made to increase PHB production under various conditions by Actinobacillus sp. EL-9 in a shaker culture. The addition of glucose, NH$_{4}$NO$_{3}$ to alcoholic distillery wastes slightly promoted cell mass and PHB production. Enzyme hydrolysis of alcoholic distillery wastes increased the production of PHB than that of untreated waste and acid hydrolysis treatment. The PHB weight in alcoholic distillery wastes was 1.91 g/l. Fermentation process of PHB production reduced the amount of COD value up to 54%, which reduced organic loading rate and capacity of activated sludge system.

  • The MRS medium is widely used as an optimized medium for the growth of Lactobacillus spp. and also used for the growth of Leuconostoc spp. Leuconostoc mesenteroides shows quite different physicochemical properties compared to Lactobacilli spp. and it is one of the major strain of kimchi fermenting microorganisms with its usefulness in our traditional foods and availability in biotechnology in the future, specifically tailor-made medium is necessary for the growth of Leuconostoc mesenteroides. Sequential experimental designs (Plackett-Burman, fractional factorial, steepest ascent, central composite design and response surface methodology) were introduced to optimize and improve the Leuconostoc medium. Fifteen medium ingredients were investigated and fructose, sodium acetate and ammonium citrate were determined to give a critical and positive effect for cell-growth. The yield of biomass using the optimal medium was improved more than that of the MRS medium and the result of fed-batch culture showed the capability of the improvement in cell mass similar to the E.coli system.

Development of a Numerical Model for Prediction of the Cooling Load of Nutrient Solution in Hydroponic Greenhouse (수경온실의 양액 냉각부하 예측모델 개발)

  • Cooling of nutrient solution is essential to improve the growth environment of crops in hydroponic culture during summer season in Korea. This study was carried out to provide fundamental data for development of the cooling system satisfying the required cooling load of nutrient solution in hydroponic greenhouse. A numerical model for prediction of the cooling load of nutrient solution in hydroponic greenhouse was developed, and the results by the model showed good agreements with those by experiments. Main factors effecting on cooling load were solar radiation and air temperature in weather data, and conductivity of planting board and area ratio of bed to floor in greenhouse parameters. Using the model developed, the design cooling load of nutrient solution in hydroponic greenhouse of 1,000$m^2$(300pyong) was predicted to be 95,000 kJ/hr in Suwon and the vicinity.

Effects of Auxins end Cytokinins on Callus Induction from Leaf Blade, Petiole, and Stem Segments of in Vitro-grown 'Sheridan' Grape Shoots

  • To establish an the mass production system of grape anthocyanin pigments through callus and cell suspension culture, the effects of various combinations of auxins and cytokinins on friable callus production were studied. for friable callus production, 2,4-D was superior to other regulators. IAA at 2 mg/L induced callus from stem and petiole while NAA resulted in rooting. Callus induction rate increased with the 2,4-D level, and stem segments were superior to leaf blade or petiole, showing nearly 100% with 1 and 2 mg/L 2,4-D from petiole and stem. Combined treatments of 2,4-D + kinetin and NAA + BA also yielded friable callus from stem segments. In treatments with 1 mg/L 2,4-D + 1 mg/L kinetin and 1 mg/L NAA + 1 mg/L BA, callus induction rate was nearly 100%. The combination effect of 2,4-D and BA on anthocyanin production was not significant.

Pharmacological Activities of the Mycelial Extract of Cultured Cordyceps sinensis

  • For defining the possibilities of the commercial mass liquid culture of Cordyceps sinensis, the pharmacological activities of mycelia were analyzed. The mycelium of C. sinensis consists of carbohydrate (5.1%) and fat (1.3%), and contains a low content of protein (0.7%) and ash (0.5%), and 92.4% moisture. The molecular sugar ratio of carbohydrate was composed mainly of glucose, mannose (1.0 : 0.9), in addition a small amount of galactose and arabinose (0.2 : 0.1). The cellular materials of mycelia were fractionated into ethylacetate (EA), MeOH (M) and hot-water extract fraction (HW). HW fraction showed the most potent intestinal immune system modulating activity, anti-coagulant activity, and anti-complementary activity, and M fraction had the inhibition activity of radical generation as effective as genistine. These results reveal that the mycelium of liquid cultured C. sinensis showed pharmacological activities and could be used for commercial purpose.

The Emotional Dimensions of North Korean Politics through the Lens of Historical Institutionalism

  • This study explores the following research question to address issues linked to emotions, identity, and institutions: how has the cult (institution) of the three Kims affected North Koreans' strong sense of nationalism (emotion), which is based on their Juche ideology (identity)? This paper investigates four fundamental elements of historical institutionalism: time boundedness, path dependency, institutional changes, and the shadow of the past. First, time boundedness illustrates how culture and education have been used to build trust and loyalty in the general public to construct individual and family cults. Second, path dependence reveals how the Songbun system has resulted in strong nationalism throughout Rodongdang's institutionalization. Third, institutional changes highlight the significance of age divides, as different age groups do not always support the three Kims' cult. Finally, the shadow of the past helps us understand the primary processes for generating mass ardent nationalism in the form of powerful impulses for self-sacrifice.
