Development of an Automatic Silkworm Breeding System

  • This paper is about the development results of an automatic silkworm breeding system to reduce labor and time by automatically performing the works for silkworm droppings changing and feed its food. It consists of an automatic guided vehicle and a processing unit. The automatic guided vehicle transports a silkworm dropping changing frame mounted on a silkworm tray stand, and the processing unit takes over the dropping changing frame on it, removes excrement contained the droppings changing frame and feeds silkworm food. In the case of the current silkworm farming, because the breeding period for large silkworms (4 to 5 stage) is short to 14 days and the supply of mulberry leaves takes 98% of the total amount of mulberry leaves needed for breeding silkworms at this time, labor concentration is intensive, and all breeding works depends on manpower. Therefore, it was difficult to breed large silkworms on a large scale. Moreover, silkworms are bred by adding Silkworm bed (Seop) and mulberry in the silkworm tray, and their droppings changing is to separate silkworms and excrement by moving silkworm trays one by one, and the production cost increases due to the high-cost manpower for silkworm breeding. To solve this problem, technology for automating silkworm breeding has also been developed. However, there is still a limitation that silkworm feeding and droppings changing works are not suitable for mass breeding because a lot of labor and time are spent depending on manual work. Therefore, a new silkworm breeding system for breeding silkworm automatically is needed and so we developed an Automatic Silkworm Breeding System applying the droppings change frame, the inverting unit, the feeding silkworm food device and automatic guided vehicle.

Comparative Proteomic Analysis of Yak Follicular Fluid during Estrus

  • The breeding of yaks is highly seasonal, there are many crucial proteins involved in the reproduction control program, especially in follicular development. In order to isolate differential proteins between mature and immature follicular fluid (FF) of yak, the FF from yak follicles with different sizes were sampled respectively, and two-dimensional gel electrophoresis (2-DE) of the proteins was carried out. After silver staining, the Image Master 2D platinum software was used for protein analysis and matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS) was performed for differential protein identification. The expression level of transferrin and enolase superfamily member 1 (ENOSF1) was determined by Western blotting for verification analysis. The results showed that 2-DE obtained an electrophoresis map of proteins from mature and immature yak FF with high resolution and repeatability. A comparison of protein profiles identified 12 differently expressed proteins, out of which 10 of them were upregulated while 2 were downregulated. Western blotting showed that the expression of transferrin and ENOSF1 was enhanced with follicular development. Both the obtained protein profiles and the differently expressed proteins identified in this study provided experimental data related to follicular development during yak breeding seasons. This study also laid the foundation for understanding the microenvironment during oocyte development.

Effects of Soil Textures on Infectivity of Root-Knot Nematodes on Carrot

  • This study was conducted to examine infectivity (penetration and gall and egg-mass formations) of the root-knot nematodes, Meloidogyne incognita and M. hapla, on carrots grown in soil conditions of 5 different soil textures consisting of bed-soil (b) and sand (s) mixtures (b-s mixtures) at the ratios of 10:0, 7:3, 5:5, 3:7, and 0:10. For M. incognita, the nematode penetration rates in b-s of 0:10 (100% sand) were significantly higher than in the other b-s mixtures, more greatly at 2 and 5 days after inoculation than at 10 DAI, while no significant differences in the penetration rates were mostly shown for M. hapla at the above DAI. However, for both nematodes, gall and egg-mass formations were remarkably increased in the b-s mixture of 0:10, compared to the other b-s mixtures, which is coincided with the general aspects of severe nematode infestations in sandy soils. This suggests the increased gall and egg-mass formations of M. incognita should be derived from the increased penetration rates in the sandy soil conditions, which provide a sufficient aeration due to coarse soil nature for the nematodes, leading to their mobility increased for the enhanced root penetration. For M. hapla, it is suggested that the sandy soil conditions affect positively on the healthy plant growth with little accumulation of the inhibitory materials and sufficient aeration, enhancing the nematode growth and feeding activities. All of these aspects provide information reliable for the development screening techniques efficient for the evaluation of the nematode resistance in the breeding programs.

Expression Analysis of the Caspase10 from Olive Flounder (Paralichthys olivaceus) against Viral Hemorrhagic Septicemia Virus (VHSV) Challenge

  • The caspase10 encodes an initiating caspase that plays an important role in the maintaining the cellular homeostasis by regulating the steps involved in the immune response and cell death. We investigated the expression of caspase10 during the different developmental stages and in olive flounder tissues. Caspase10 increased in the late stage of the formation of immune tissue, and high expression was observed in the gills, kidney, skin, and spleen. The current study analyzed the expressional changes of caspase10 in olive flounder infected with viral hemorrhagic septicemia virus (VHSV). One of the major causes of mass mortality, VHSV infection in olive flounder attributes to significant expression of caspase10 in the gills, spleen, skin, and kidneys. The results indicate a close association of caspase10 expression with the immune response to VHSV infection in olive flounder. The observations could form the basis data for exploration of other fish immune system.

Individual Recognition between Siblings of the Young Black-tailed Gull (Larus crassirostris)

  • We had 18 eggs artificially hatched in a mass breeding place of black-tailed gulls and examined the individual recognition between young siblings in a laboratory environment. The results of the experiment showed that the young gulls selectively responded to their siblings and non-siblings at an early stage after hatching. It was shown that they began to recognize the begging call among the voice signals of siblings and non-siblings 15-16 days after hatching, and the chirirah call 11-12 days after hatching. Also, more significant results were shown with the chirirah call than with the begging call. In an experiment of visual recognition between siblings and non-siblings, the young black-tailed gulls approached their siblings significantly 9-10 days after hatching. The recognition between young siblings in a mass breeding place provides an important evolutionary indicator in terms of their social behaviors.

  • In this paper, simulation tests were conducted with similar materials to study the distribution of residual voids in longwall goaf. Short-time step loading was used to simulate the obvious deformation period in the later stage of arch breeding. Long-time constant loading was used to simulate the rheological stage of the arch forming. The results show that the irregular caving zone is the key area of old goaf for the subsidence control. The evolution process of the stress arch and fracture arch in stope can be divided into two stages: arch breeding stage and arch forming stage. In the arch breeding stage, broken rocks are initially caved and accumulated in the goaf, followed by the step deformation. Arch forming stage is the rheological deformation period of broken rocks. In addition, under the certain loads, the broken rock mass undergoes single sliding deformation and composite crushing deformation. The void of broken rock mass decreases gradually in short-time step loading stage. Under the water lubrication, a secondary sliding deformation occurs, leading to the acceleration of the broken rock mass deformation. Based on above research, the concept of equivalent height of residual voids was proposed, and whose calculation equations were developed. Finally, the conceptual model was verified by the field measurement data.

국내 육성 거베라의 우량묘 생산에 미치는 식물생장조절제의 영향 (Effect of Plant Crowth Regulators on Production of Good Quality Plant for Korean Gerbera Lines)

  • 본 연구는 국내에서 육성된 절화용 거베라 19품종의 우량묘 생산을 위하여 적정 식물생장조절물질의 종류 및 조합을 알아보고, 국내 육성품종의 우량묘 대량생산을 위한 기초 자료로 제공하고자 실시하였다. 우량묘 생산을 위한 생장조절물질의 종류 및 조합에서 NAA와 BA의 혼용보다는 BA와 Kinetin의 흔용조건에서 대량증식이 양호하였다. 대량증식이 양호한 BA와 Kinetin의 혼용조합에서 품종에 따라 대량증식의 조건이 달리 나타났다. 19개 품종 중 대부분은 BA 1.0mg/L + Kinetin 0.5, 1.0 mg/L의 혼용조건에서 우량묘의 대량증식이 양호하였다. 그러나 '선녀', '옥샘', '피니', '핑크라이트'등 4개 품종은 BA의 농도가 높을수록 투명화묘의 발생이 많이 나타났다. 이들 품종들은 BA 0.5 mg/L + Kinetin0.1 mg/L의 흔용처리구에서 우량묘의 대량증식이 가능하였다. 생체중과 건물중은 BA 0.5 mg/L + Kinetin 0.1, 0.5mg/L 의 혼용 처리구에서 양호한 경향이었다. 엽병장에서는 전 처리구 모두 큰 차이는 없었다. 투명화묘의 조직관찰결과, 투명화묘의 기공형태는 정상묘에 비하여 기공이 열려있는 상태로 돌출되어 있었으며, 둥근 모양을 하고 있었다.

  • 월동기간 동안 월동조류의 생리 및 영양학적 상태는 이후 번식지로의 이동성공과 번식성공에 영향을 줄 수 있음은 잘 알려져 있다. 그러나 환경적 요인들이 월동기간 동안 어떻게 몸 상태에 영향을 주어 장기적으로 이동과 번식에 영향을 주는지에 대해서는 아직까지 잘 알려져 있지 않다. 본 연구는 월동기간 동안 온도변화와 월동지에서의 인간 활동에 따른 방해가 개체수준에서의 월동하는 조류의 생활사에 미치는 영향과 번식지로의 이동 및 잠재적 번식 성공에 미치는 영향을 알아보기 위해 동적상태의존 월동 생활사 모델을 개발하였다. 모델에 사용된 지수는 월동개체군에 대한 연구가 잘 수행되어 있는 철원의 두루미 자료를 이용하였다. 모델은 온도 변화나 인간의 방해요인의 영향이 생존과 번식지로의 이동을 위한 에너지 축적 그리고 누적된 스트레스의 감소를 위한 자원 분배에 영향을 주는 것으로 예측하였다. 특히 월동지에 도래한 두루미 몸무게의 회복률은 기온변화가 적고 방해요인의 영향이 낮을수록 빨랐으며, 체내의 누적 스트레스는 기온변화가 크고 방해요인의 영향이 높을수록 회복속도가 낮을 것으로 예측되었다. 또한 월동지의 취식지를 공유하는 다른 종의 밀도가 높을수록 두루미의 몸무게 회복률이 낮은 것으로 예측되었다. 끝으로 모델의 예측된 결과를 통해 월동지에서의 월동조류 보전전략에 대해 고찰하였다.

Proteomic Comparison between Japanese Black and Holstein Cattle by Two-dimensional Gel Electrophoresis and Identification of Proteins

  • Differences of meat qualities between Japanese Black and Holstein have been known in Japan, however, the causative proteins and/or the genetic background have been unclear. The aim of this study was to identify candidate proteins causing differences of the meat qualities between the two breeds. Using technique of two-dimensional gel electrophoresis, protein profiling was compared from samples of the longissimus dorsi muscle and subcutaneous adipose tissue. Five protein spots were observed with different expression levels between breeds. By using LC-MS/MS analysis and Mascot program, three of them were identified as ankyrin repeat protein 2, phosphoylated myosin light chain 2 and mimecan protein. Subsequently, we compared the DNA coding sequences of three proteins between breeds to find any nucleotide substitution. However, there was no notable mutation which could affect pI or molecular mass of the proteins. The identified proteins may be responsible for different characteristics of the meat qualities between Japanese Black and Holstein cattle.
