Temporal Variation of Water Quality of the Western Chinhae Bay in Summer (진해만 서부해역의 하계 수질의 시간변동 특성)

  • Temporal changes of Chl-α, physical and chemical factors were investigated by diurnal observation at 2-hour interval at three fixed stations in the western Chinhae Bay from 12 Aug. to 13 Aug. 1999. Difference of dissolved oxygen between surface and bottom layer was maximum when the thermocline were strong. Organic distribution such as COD was affected by the growth of phytoplankton. Limitting factor was nitrogen, that is, inorganic nitrogen plays a significant role on regulating the algal growth. Surface distribution of dissolved inorganic nitrogen was very low compared to bottom layer by uptake of organisms. Maximum value of Chl-α at station C2 and C11 were observed from subsurface layer, ranges of which exceeded possibility concentration of red tide outbreak, 10 mg/㎥. On the other hand, that of C15 exist at surface layer. In this area, DIN and DIP concentrations increased by input sources such as rainfall and benthic flux before the bloom of phytoplankton. Accumulation of phytoplankton occurred at subsurface layer by the rapid uptake of DIN, especially nitrate ion, when strong thermocline existed as approach to the afternoon, which led to the increase of organics in water column and oxygen deficiency water mass at bottom layer until late at evening. Since then, DIN increases gradually as water temperature decrease to minimum. The quantitative understanding of nitrogen of fluxed to and from the various sources is necessary for environmental management.

Molecular Cloning and Characterization of the Gene Encoding Phytoene Desaturase from Kocuria gwangalliensis (Kocuria gwangalliensis 유래 phytoene desaturase 유전자의 cloning과 특성 연구)

  • Carotenoids such as phytoene, lycopene, and ${\beta}-carotene$ are used as food colorants, animal feed supplements, and for human nutrition and cosmetic purposes. Previously, we reported the isolation of a novel marine bacterium, Kocuria gwangalliensis, which produces a pink-orange pigment. Phytoene desaturase (CrtI), encoded by the gene crtI, catalyzes lycopene formation from phytoene and is an essential enzyme in the early steps of carotenoid biosynthesis. CrtI is one of the key enzymes regulating carotenoid biosynthesis and has been implicated as a rate-limiting enzyme of the pathway in various carotenoid synthesizing organisms. Here, we report the cloning of the crtI gene responsible for lycopene biosynthesis from K. gwangalliensis. The gene consisted of 1,584 bases encoding 527 amino acid residues. The nucleotide sequence of the crtI gene was compared with that of other species, including Kocuria rhizophila and Myxococcus xanthus, and was found to be well conserved during evolution. An expression plasmid containing the crtI gene was constructed (pCcrt1), and Escherichia coli cells were transformed with this plasmid to produce a recombinant protein of approximately 57 kDa, corresponding to the molecular weight of phytoene desaturase. Lycopene biosynthesis was confirmed when the plasmid pCcrtI was co-transformed into E. coli containing the plasmid pRScrtEB carrying the crtE and crtB genes required for lycopene biosynthesis. The results from this study will provide valuable information on the primary structure of K. gwangalliensis CrtI at the molecular level.

Development of Line Density Index for the Quantification of Oceanic Thermal Fronts (해양의 수온전선 정량화를 위한 선밀도 지수 개발)

  • Line density index(LDI) was developed to quantify a densely isothermal line rate as standard index in the ocean environment. Theoretical background on the LDI development process restricting index range 0 to 100 was described. And validation test was done for the LDI application condition that total line length is not greater than 1/10 of unit area. NOAA SST(Sea Surface Temperature) data were used for the experimental application of LDI in the South Sea of Korea. Using GIS, $0.1^{\circ}C$ isothermal lines were linearized as vector data form SST raster data, and unit area were built as polygon data. For the LDI calculation, spatial overlapping(line in polygon) was implemented. To analyze the effect of unit area size for the LDI distribution, two cases of unit area size were designed and descriptive statistics was calculated including performing normality test. The results showed no change of LDI's essential characteristics such as mean and normality except for the range of value, variance and standard deviation. Accordingly, it was found that complex structure of thermal front and even smaller scale of front width than unit area size could influence on the LDI distribution. Also, correlation analysis performed between LDI and difference of temperature(${\Delta}T^{\circ}C$), and horizontal thermal gradient(${\Delta}T^{\circ}C/km$) on the front was obtained from linear regression model. This obtained value was compared with the results from previous researches. Newly developed LDI can be used to compare the thermal front regions changing spatio-temporally in the ocean environment using absolute index value. It is considered to be significant to analyze the relationship between thermal front and marine environment or front and marine organisms in a quantitative approach described in this study.

Correlation Analysis of UA Using Wind Data of AWS/ASOS and SST in Summer in the East Sea (AWS/ASOS 바람자료를 이용한 여름철 동해 연안역의 용승지수와 수온과의 상관성)

  • In this study, we examined the UA (upwelling age) using wind data of AWS/ASOS in the East Sea coast and the correlation between UA and SST (sea surface temperature) from May to August in 1995 to 2016. The data used the 6 observations of the wind data of AWS/ASOS and the SST data of the COD/RISA provided by the National Institute and Fisheries Science near the East Sea coast. The UA was calculated quantitatively low but it rose when the actual cold water mass occurred. Correlation analysis between UA and SST showed the negative (-) r (correlation coefficient) predominately. At the time of cold-water mass in June to August 2013, the r had a very high negative value of -0.65 to -0.89 in the 6 observations. It proved that as the UA increases, the SST is lower. By knowing the UA, we were able to evaluate the trend of upwelling in the cold-water mass of the East Sea coast in the long term and it will contribute to minimizing the damage to aquatic organisms according to the size and intensity of the upwelling.

Inherent Optical Properties of Red Tide Algal for Ocean Color Remote Sensing Application (해색원격탐사 활용을 위한 적조생물종 고유 광특성 연구)

  • This research is about the inherent optical properties(IOPs) of algae which is collected from Nam-Hae for basic research of red tide remote sensing technique development. 21 kinds of red tide organisms were cultivated to investigate IOPs of them in the level of laboratory, and specific absorption coefficient of phytoplankton($a^*$) and backscattering coefficient of phytoplankton(${b_b}^*$) are estimated by using spectrophotometer. Absorption spectrums according to species appeared to range from 0.005 to 0.06 ($m^2{\cdot}mg^{-1}$), and the shapes of spectrums were also different. The range of ${b_b}^*$ appeared to be $10^{-2}{\sim}10^{-4}\;m^2{\cdot}mg^{-1}$, which had about 100 times differences between species, and the shape of spectrum have significant difference between species. These results will input as a remote sensing reflectance model input parameter from ocean color.

Toxic Effect of Micropollutants on Coastal Organisms -I. Toxicity on Some Marine Fishes- (Micropollutants가 연안 생물에 미치는 독성효과에 관한 연구 -1. 어류에 미치는 독성-)

  • The lethal concentration(LC50) of several micropollutants for three species of the fish Paralichthys olivaceus, Mugil cephalus and Sebastes schlegeli were determined by acute toxicity tests. For the determination of definitive test concentrations, the fish were exposed to three test material concentrations spaced at order-of-magnitude intervals based on a logarithmic ratio in range finding tests. LC50 was determined by five concentrations of test material in a geometric progression by means of range finding tests. The 96hr-LC50 values(mg/l) were estimated by the graphical interpolation of probability-logarithm transformations. These indicated that the order of sensitivities to three kind of micropollutants was Mugil cephalus > Paralichthys olivaceus > Sebastes schlegeli.

Fishing Characteristic of Beam Trawl Fishery in the Coastal Waters of Chungnam Province, Korea (충남 연안 조망어업의 어획특성)

  • To understand the fishery characteristics by a beam trawl off the coast of the Chungnam province, covering $125^{\circ}$ 50'~$126^{\circ}$ 05'E, $36^{\circ}$ 15'~$36^{\circ}$ 25'N, a beam trawl was deployed 2~4 times a month from March 2008 to February 2009. The catch was consisted of 96 species of 63 families belonging to 19 orders. Among these, organisms were identified as 54 species of fish, 27 species of crustaceans, 9 species of gastropoda, bivalves, and 6 species of cephalopods. Trachysalambria curvirostris which is the target species of the beam trawl fishery occupied only 2.1% of the total catch, showing a low ratio of catches. Most of T. curvirostris was harvested from July to November while small amount of T. curvirostris was captured from May to December. In this study, we concluded that the bycatch ratio except T. curvirostris in T. curvirostris fishery was relatively high, thus the fishery of T. curvirostris might affect the fishery resource management of bycaught species off the coast of Chungnam province.

Evaluation of Heavy Metal Pollution in the Dumping Site of the Dredged Sediment, Masan Bay (마산만 오염퇴적물 준설토 투기해역의 중금속 오염평가)

  • A large amount 2.1×106 ㎥ of the polluted sediment was dredged from the Masan Bay and deposited in Gapo confined area, Masan. The dissolved metal concentrations of seawater in the dumping site (Gapo area) were observed during one tidal cycle and compared with those of seawater obtained from Jinhae Bay. The sediment was evaluated as from Non polluted to Moderately polluted by USEPA standards. It was judged that toxicological effects of sediment analyzed ranged from ERL to ERM with copper and zinc, and ERL with cadmium, chrome, lead, and nickel by the Adverse Biological Effects. The pollutant concentration was low in surface sediment compared to deeper sediment since the sediments with relatively low concentrations of pollutant were dumped to the surface. The benthic organisms in Gapo area had higher concentrations of trace metals (Oyster: Zn 238.96, Cu 5.29 ㎍/g wet wt., Clam: Zn 17.71, Cu 1.00 ㎍/g wet wt., Mussel. Zn 187.98, Pb 0.28, Cr 0.15, Mn 4.23, Sr 1.45 and Fe 100.33 ㎍/g wet wt.) compared to outside of dumping site. However, the trace metal level in the bivalves was less than the NFPQIS (National Fisheries Products Quality Inspection Service) standard.

Auranofin Enhances Sulforaphane-Mediated Apoptosis in Hepatocellular Carcinoma Hep3B Cells through Inactivation of the PI3K/Akt Signaling Pathway

  • The thioredoxin (Trx) system plays critical roles in regulating intracellular redox levels and defending organisms against oxidative stress. Recent studies indicated that Trx reductase (TrxR) was overexpressed in various types of human cancer cells indicating that the Trx-TrxR system may be a potential target for anti-cancer drug development. This study investigated the synergistic effect of auranofin, a TrxR-specific inhibitor, on sulforaphane-mediated apoptotic cell death using Hep3B cells. The results showed that sulforaphane significantly enhanced auranofin-induced apoptosis by inhibiting TrxR activity and cell proliferation compared to either single treatment. The synergistic effect of sulforaphane and auranofin on apoptosis was evidenced by an increased annexin-V-positive cells and Sub-G1 cells. The induction of apoptosis by the combined treatment caused the loss of mitochondrial membrane potential (ΔΨm) and upregulation of Bax. In addition, the proteolytic activities of caspases (-3, -8, and -9) and the degradation of poly (ADP-ribose) polymerase, a substrate protein of activated caspase-3, were also higher in the combined treatment. Moreover, combined treatment induced excessive generation of reactive oxygen species (ROS). However, treatment with N-acetyl-L-cysteine, a ROS scavenger, reduced combined treatment-induced ROS production and apoptosis. Thereby, these results deduce that ROS played a pivotal role in apoptosis induced by auranofin and sulforaphane. Furthermore, apoptosis induced by auranofin and sulforaphane was significantly increased through inhibition of the phosphoinositide 3-kinase (PI3K)/Akt pathway. Taken together, the present study demonstrated that down-regulation of TrxR activity contributed to the synergistic effect of auranofin and sulforaphane on apoptosis through ROS production and inhibition of PI3K/Akt signaling pathway.

Faunistic Investigation for Marine Mollusks In Jindo Island (진도의 해산 연체동물상)

  • As a commemorative joint survey for the 20th anniversary of the Korean Society of Systematic Zoology, molluscan fauna of Jindo Island was investigated from the materials collected from 3 localities of Jindo Island from 29th to 30th of June, 2004. A total of 77 species of 37 families were identified. Among them, 26 species of 18 families are reported for the first time in this area, including Onchidella kurodai (Taki, 1935) that is new to Korean fauna. For O. kurodai, external morphologies were described in detail with illustrations. Combining the present data with those of the previous works revealed that a total of 115 species from 43 families have been recorded in Jindo Island and its adjacent areas thus far. From a biogeographic point of view, some gastropod species (Bittium craticulatum, Mocroschisma dilatatum, Cantharidus japonicus, C. callichroa, Homalopoma sangarense, and Crepidula onyx) encountered from Jindo Island were the species previously recorded in the southern and/or eastern offshores of Korean peninsula, including Jeju Island (Choe, 1992). Along with the fact that Jindo Island contains a large number of molluscan species ranging over the Yellow Sea coast, this finding is another line of evidence supporting the Kim and Kwon (1983)'s idea that this area is an overtopping zone of marine organisms which dwell in both the Yellow sea and southern part of Korean sea waters. No comprehensive faunistic survey was not made in the present study. We expect that further supplementary study would generate more biogeographic and faunistic knowledges than what we understand through the present study.
