  • Korea is the urgent need to diversify the bio-energy raw materials because we have narrow area of land cultivated plants and lack the bio-mass resources. Using a resource-rich the marine environment enclosed on three sides by the sea. Through bio-energy production-technical and carbon dioxide reduction measures which will be a very effective alternative. Sustainable development about algae bio-energize industry through SSaM-GG(Smart, Shared and Mutual-Green, Growth) is expected. Algae has high carbon dioxide absorption factor, extracting oil from algae is thirty times much more than sunflower.

Biochemical Composition of a Korean Domestic Microalga Chlorella vulgaris KNUA027 (한국 토착 미세조류 클로렐라 불가리스 KNUA027 균주의 생화학적 조성)

  • A unicellular green alga, Chlorella vulgaris KNUA027, was isolated from the roots of Panax ginseng seedlings and its biotechnological potential was investigated. The results of GC/MS analysis showed that C. vulgaris KNUA027 was rich in nutritionally important polyunsaturated fatty acids (PUFAs) such as alpha-linolenic acid (C18:3 ω3, 45.8%, 50.8 mg/g) and hexadecatrienoic acid (C16:3 ω3, 11.8%, 13.1 mg/g). Therefore, this Korean indigenous microalga may have potential as a source of omega-3 PUFAs. It was also found that the saturated palmitic acid (C16:0, 37.1%, 41.2 mg/g), which is suitable for biodiesel production, was one of the major fatty acids produced by strain KNUA027. The proximate analysis showed that the volatile matter content was 88.5%, and the ultimate analysis indicated that the higher heating value was 19.8 MJ/kg. Therefore, the results from this research with C. vulgaris KNUA027 may provide the basis for the production of microalgae-based biofuels and biomass feedstock.

Marine-derived Ca-Mg complex influences lipid and glucose metabolism, serum metabolites, colostrum profile, and stress hormone in sows over four-parity periods

  • Minerals is required small amounts among various nutrients, but it has a significant impact on sow longevity and reproduction performance. This study was carried out to see the beneficial effects of marine-derived Ca-Mg complex on the reproductive performance of sows during four-parity periods. Seventy-two gilts ([Yorkshire × Landrace] × Duroc), with an average body weight of 181 kg, were randomly allocated to three groups; CON (basal diet), 0.3LC (CON - MgO - 0.3% limestone + 0.4% Ca-Mg complex), and 0.7LC (CON - MgO - 0.7% limestone + 0.4% Ca-Mg complex). During parity 3 and 4, the expression level of SCD gene was lower in the umbilical cord of piglets born to 0.3LC and 0.7LC sows compared with the CON sows. During parity 2, 3 and 4, SLC2A2 and FABP4 gene expressions were higher in the umbilical cord of piglets born to 0.7LC sows and the placenta of sows from 0.3LC groups, respectively. Ca-Mg complex increased (p < 0.05) Ca and Mg concentrations in sows and their piglets' serum as well as in colostrum regardless of parities. The serum vitamin D concentration was higher (p < 0.05) in their first parity, whereas serum prolactin and estrogen concentrations were higher (p < 0.05) during the fourth and third parity, respectively. The growth hormone concentrations were higher (p < 0.05) in the piglets born to sows during the first and second parity. The fat and immunoglobulin A (IgA) concentrations in colostrum were higher (p < 0.05) during the third and fourth parity, respectively. A reduction (p < 0.05) in salivary cortisol, epinephrine, and norepinephrine concentrations was observed in 0.3LC and 0.7LC sow groups compared with CON after farrowing regardless of parity, however before farrowing, a reduction in norepinephrine was observed. Before farrowing, the epinephrine and norepinephrine concentrations were higher (p < 0.05) during the first and second parity. After farrowing, the concentration of these hormones was higher during the second parity. Taken together, sows' parity and dietary Ca-Mg complex supplementation influenced serum metabolites, colostrum nutrients, stress hormones as well as the gene expressions related to lipid and glucose metabolism.

Induction of apoptosis by a hexane extract of aged black garlic in the human leukemic U937 cells

  • BACKGROUND/OBJECTIVES: In this study, the apoptogenic activity and mechanisms of cell death induced by hexane extract of aged black garlic (HEABG) were investigated in human leukemic U937 cells. MATERIALS/METHODS: Cytotoxicity was evaluated by MTT (3-(4, 5-dimethyl-thiazol-2-yl)-2, 5-diphenyl tetrazoliumbromide) assay. Apoptosis was detected using 4,6-diamidino-2-phenyllindile (DAPI) staining, agarose gel electrophoresis and flow cytometry. The protein levels were determined by Western blot analysis. Caspase activity was measured using a colorimetric assay. RESULTS: Exposure to HEABG was found to result in a concentration- and time-dependent growth inhibition by induction of apoptosis, which was associated with an up-regulation of death receptor 4 and Fas legend, and an increase in the ratio of Bax/Bcl-2 protein expression. Apoptosis-inducing concentrations of HEABG induced the activation of caspase-9, an initiator caspase of the mitochodrial mediated intrinsic pathway, and caspase-3, accompanied by proteolytic degradation of poly(ADP-ribose)-polymerase. HEABG also induced apoptosis via a death receptor mediated extrinsic pathway by caspase-8 activation, resulting in the truncation of Bid, and suggesting the existence of cross-talk between the extrinsic and intrinsic pathways. However, pre-treatment of U937 cells with the caspase-3 inhibitor, z-DEVD-fmk, significantly blocked the HEABG-induced apoptosis of these cells, and increased the survival rate of HEABG-treated cells, confirming that HEABG-induced apoptosis is mediated through activation of caspase cascade. CONCLUSIONS: Based on the overall results, we suggest that HEABG reduces leukemic cell growth by inducing caspase-dependent apoptosis through both intrinsic and extrinsic pathways, implying its potential therapeutic value in the treatment of leukemia.

  • We studied the maturation and spawning of female Spanish mackerel (Scomberomorus niphonius) based on 445 specimens collected monthly from January to December 2004 in the coastal waters off Busan. The fork length (FL) of S. niphonius ranged from 26.1 to 105.4 cm, the gonadosomatic index (GSI) of the females was highest in May 2004, and the spawning season take place from April to July. A significant difference was detected in the sex ratio between females and males ($X^2$ test, p<0.01). The percentage of sexually mature females exceeded 50% in the 50-60 cm (FL) size group and reached 100% in the over 70 cm (FL) size group. Fecundity (F) varied between 201,156 and 836,426 egg per female. The relationship between F and FL of the fish was expressed as $F=5.8756FL^{3.8465}$. The relationship F and body weight (BW) was expressed as F=581.421 n (BW)-4108.5. The first spawning length was 41.8 cm (FL.)

  • Here, we introduce a means of utilizing waste oyster shells which were obtained from temporary storage near coastal workplaces as $CO_2$ adsorbents. The calcined CaO can be easily dissociated to $Ca^{2+}$ cation and $CO_3{^{2-}}$ anion by hydrolysis and gas-liquid carbonation reaction and converted to precipitated calcium carbonate (PCC) in algae-containing water. The calcium hydroxide and carbonation combination in algae-containing water significantly contributed to improving water quality which is very dependent on the addition amount of calcined powders.

  • Morphology and histochemistry of gill of the abalone, Haliotis discus hannai were described using light microscopy and scanning electron microscopy (SEM). The abalone has bipectinate gill of protobranch. The cilia and microvilli were well-developed on the free surface of the gill filaments. The epithelial layer of gill filament was simple and composed columnar epithelia and mucous cells. Result of AB-PAS (pH 2.5 and 1.0) and AF-AB (pH 2.5) reaction showed that mucous cells contained mainly acidic carboxylated mucosubstances.

  • A major concern regarding the collection and storage of biodiversity information is the inefficiency of conventional taxonomic approaches in dealing with a large number of species. This inefficiency has increased the demand for automated, rapid, and reliable molecular identification systems and large-scale biological databases. DNA-based taxonomic approaches are now arguably a necessity in biodiversity studies. In particular, DNA barcoding using short DNA sequences provides an effective molecular tool for species identification. We constructed a large-scale database system that holds a collection of 5531 barcode sequences from 2429 Korean species. The Korea Barcode of Life database (KBOL, http://koreabarcode.org) is a web-based database system that is used for compiling a high volume of DNA barcode data and identifying unknown biological specimens. With the KBOL system, users can not only link DNA barcodes and biological information but can also undertake conservation activities, including environmental management, monitoring, and detecting significant organisms.

  • Schisandrae fructus (SF) and Mori folium (MF) have been used as traditional medicines for thousands of years in parts of Asia, including Korea, China, and Japan. Recent researches on SF and MF have documented a wide spectrum of therapeutic properties, including anti-microbial, anti-inflammatory, anti-oxidative, immunomodulatory and anti-angiogenesis effects. However, the toxicity and safety of SF and MF, and their mixture (medicinal herber mixture, MHMIX) were not confirmed. Therefore, this study was performed to evaluate the acute toxicity and safety of SF, MF and MHMIX. SF, MF and MHMIX were orally administered at a dose of 5,000 mg/kg in ICR mice. Animals were monitored for the mortality and changes in the body weight, clinical signs and gross observation during the 14 days after dosing, upon necropsy. We also measured parameters of organ weight, clinical chemistry, and hematology. No dead and no clinical signs were found during the experiment period after administration of a single oral dose of SF, MF and MHMIX. There were no adverse effects on clinical signs, body weight, or organ weight and no gross pathological findings in any treatment group. Therefore, LD50 value of SF, MF and MHMIX may be over 5,000 mg/kg and it may have no side toxic effect to ICR mice. The results on the single-dose toxicity of SF, MF and MHMIX indicate that it is not possible to reach oral dose levels related to death or dose levels with any harmful side effects.

Isolation and Characterization of an Agar-hydrolyzing Marine Bacterium, Pseudoalteromonas sp. H9, from the Coastal Seawater of the West Sea, South Korea (서해안 해수로부터 분리한 한천분해 해양미생물 Pseudoalteromonas sp. H9의 동정 및 특성 연구)

  • An agarolytic marine bacterium (H9) was isolated from the coastal seawater of the West Sea, South Korea. The isolate, H9, was gram-negative and rod-shaped with a smooth surface and polar flagellum. Cells grew at 20-30℃, between pH 5.0 and 9.0, and in ASW-YP (Artificial Sea Water-Yeast extract, Peptone) media containing 1-5% (w/v) NaCl. The G+C content was 41.56 mol%. The predominant isoprenoid quinone in strain H9 was ubiquinone-8. The major fatty acids (>10%) were C16:1ω7c (34.3%), C16:0 (23.72%), and C18:1ω7c (13.64%). Based on 16S rRNA gene sequencing, and biochemical and chemotaxonomic characterization, the strain was designated as Pseudoalteromonas sp. H9 (=KCTC23887). In liquid culture supplemented with 0.2% agar, the cell density and agarase activity reached a maximum level of OD = 4.32 (48 h) and OD = 3.87 (24 h), respectively. The optimum pH and temperature for the extracellular crude agarases of H9 were 7.0 and 40℃, respectively. Thin-layer chromatography analysis of the agarase hydrolysis products revealed that the crude agarases hydrolyze agarose into neoagarotetraose and neoagarohexaose. Therefore, the new agar-degrading strain, H9, can be applicable for the production of valuable neoagarooligosaccharides and for the complete degradation of agar in bio-industries.
