Authentication and quality control of Cordyceps sinensis, a traditional Chinese medicine known as winter-worm summer-grass

  • Cordyceps, one of the most valued traditional Chinese medicines, consists of the dried fungus Cordyceps sinensis growing on the larva of caterpillar. It is also known as 'winter-worm and summer-grass' because of its appearance during different seasons. The parasitic complex of the fungus and the caterpillar is found in soil of a prairie at an elevation of 3,500 to 5,000 meters in northwestern part of China. According to Chinese medicinal theory, Cordyceps is used to replenish the kidney and soothe the lung, and indeed many clinical applications have been reported. The natural Cordyceps is rare and expensive on the local market, and therefore, several mycelial strains have been isolated from natural Cordyceps and manufactured in large quantities by fermentation technology, and they are commonly sold as health food products in Orient. The adulterants of Cordyceps are commonly found on the market, and therefore the authentication of these products has to be defined. Having the urgent need from current market, different chemical markers such as nucleoside, ergosterol, mannitol and polysaccharide are being used for quality control of Cordyceps. Unfortunately, these markers are far from optimization, and therefore extensive works are needed to define the pharmacological efficiency of these markers.

Microbiological Purification of L-Arabitol from Xylitol Mother Liquor

  • As a rare sugar alcohol, L-arabitol can be used in food and can prevent extra fat deposits in the intestinal tract. Commercially, L-arabitol is prepared from pure L-arabinose by hydrogenation, which needs a high temperature and high pressure, leading to a high production cost for Larabitol. Therefore, this study describes a novel L-arabitol production method based on biological purification from the xylitol mother liquor, a cheap and readily available raw material that contains a high concentration of Larabitol. First, a novel Bacillus megaterium strain was screened that can utilize xylitol, sorbitol, and mannitol, yet not L-arabitol. The isolated strain was inoculated into a medium containing the xylitol mother liquor under formulated culture conditions, where a high L-arabitol yield (95%) and high purity (80%) were obtained when the medium was supplemented with 50 g/l of xylitol mother liquor. Upon further purification of the fermentation broth by ion exchange and decolorization, L-arabitol was crystallized with a purity of 98.5%.

  • Anaerobic bacteria are suggested to be potential source for new antibiotics. In order to search for antibiotics from domestic origin, we collected 800 soil samples across Korean locations and could isolate as many as 989 anaerobic strains. Among them 10, strains were found to have good producing capacity of antibiotics. An anaerobe was finally selected due to secreting antibiotics having high antimicrobial activity towards multiple resistant microorganism(E coli JM 83) transformed by genetic engineering technique. Its morphological, physiological and biochemical charateristics were investigated, together with antimicrobial spectrum therefrom. On antimicrobial spectrum study, substance secreted from this strain, had no activities to fungus and yeast. The selected strain showed G(+) and coccal shape, on Gram, staining and electron scanning microscopy, respectively. Biochemically this strain utilized glucose, fructose lactose, sucrose, but did not arabinose, cellulose, rhamnose, sorbitol, trehalose, mannitol. Catalase test showed negative property. Optimal growth temperature was $37^{\circ}C$. The results obtained above suggest this strain Streptococcus faecium subspp. and we named it Streptococcus sp. An-21-1.

해남식초에서의 Acetobacter sp. CS 균주의 분리 및 특성 (Isolation and Characterization of Acetobacter sp. CS Strains from Haenam Vinegar)

  • Two strains of the gram-negative acetic acid bacteria, Acetobacter sp. strain CS2- AND CS5, were isolated form the traditional raw rice wine vinegar of Haenam area. The strains oxidized ethanol to acetic acid and over-oxidized acetate and lactate to $CO^{2}$ and $H ^{2}$O. They produced 2-ketogluconic acid from glucose but did not produce .gamma.-pyrones from glucose and dihydroxyacetone from glycerol. The CS strains possessed ubiquinone-9 as a major isoprenoid quinone and contained straight-chain $C_{18 :1}$, $_C{16 : 0}$, and $C _{14 : 0}$ fatty acids. The DNA base composition of the CS2 and CS5 strains was 56.2 and 57.3 mole% G + C, respectively. The isolates were grown well on methanol, gluconate, erythritol, raffinose, dulcitol and xylitol as sole sources of carbon and energy which are different from those of other Acetobacter species and producedd acid from sucrose, glycerol, fructose, inositol, mannitol, and ribose.

  • Fowl typhoid (FT) is a septicemic disease caused by Salmonella gallinarum. The purpose of this study was to investigate the antimicrobial resistance and pulsed -field gel electrophoresis (PFGE) patterns of S gallinarum isolated from broiler. During 1999 to 2004, there was isolated a total of 26 strains in liver and spleen. The biochemical characteristics of S gallinarum isolates was nonmotile, no production of $H_2S$, glucose gas, non-fermented rhamnose, indole-negative, fermentation of dulcitol, mannitol, maltose, and ornithine decarboxylase. At antimicrobial susceptibility, all of isolates were susceptible to amoxicillin/clavulanic acid, amikacin, neomycin, kanamycin, and cephalothin. Twenty-six isolates were divided into 19 resistant patterns and 6 strains was 8-multi-drug resistance. PFGE of Xba I restriction fragments of S gallinarum isolates was 22 patterns.

Characteristics and Pathogenicity for Japnaes Eel Anguilla japonica of Vibrio vulnificus Isolated from Oyster, Sediment and Seawater in the Korea Coast

  • Biotyping of Vibrio vulnificus strains isolated from marine environments along the south coast of Korea showed that the majority of the isolates (94.7%) belonged to biotype 1 and the remaining isolates (5.3%) belonged to biotype 2. Analysis of 16S rRNA V. vulnificus strains isolated from marine environments using a multiplex polymerase chain reaction (PCR) revealed that 78.7% were type A and 21.3% were type B. Random amplified polymorphic DNA (RAPD) was used to analyze the genomic differences in V. vulnificus among the biotype 2 strains isolated from marine environments (newly isolated strains group) and reference strains obtained from infected eels (reference strains group). The two groups had distinctly different profiles of the amplicons produced from RAPD. Additionally, biochemical comparison of these strains revealed that all four strains isolated from marine environments differed from the strains isolated from eels in their ability to promote D-mannitol fermentation. Two (NH 1 and NH 2) out of four isolates of biotype 2 from marine environments showed pathogenicity in eels Anguilla japonica in a challenge test. These isolates did not agglutinate with antisera against V. vulnificus NCIMB 2137 (serovar E), ATCC 27562 (non-serovar E), and ATCC 33816 (atypical serovar E).

At antimicrobial susceptibility, all of isolates were susceptible to amoxicillin/clavulanic acid, amikacin, neomycin, kanamycin, norfloxacin and enrofloxacin. One hundred isolates were divided into 54 resistant patterns and 37 strains was 6-multi drug resistance. PFGE of Xba I restriction fragments of S. Gallinarum isolates was 20 patterns.

  • Salmonella Typhimurium is a virulent pathogen for human and animal. We studied serotypes, biochemical characteristics, and antimicrobial resistance of S. Typhimurium isolated from diseased pigs in Gyeongbuk province over 1998 to 2008. One hundred sixteen isolates were identified as S. Typhimurium by biochemical characteristics and serotypes from 90 farms. The biochemical characteristics of S. Typhimurium isolates was production of $H_2S$, indole-negative, fermentation of mannitol, dulcitol, sorbitol, inositol, rhamnose, and maltose, and ornithine decarboxylase. At antimicrobial susceptibility test, the majority of isolates were highly susceptible to amoxicillin/clavulanic acid, cefepime, ciprofloxacin, while were highly resistant streptomycin, cephalothin, enrofloxacin, nalidixic acid, apramycin, chloramphenicol, tetracycline. The isolates were divided into 65 resistant patterns and 47 of the isolates were shown as a DT104 ASSSuT resistant phenotype.

Staphylococcus aureus의 Coagulase 생성능과 R-플라스미드 분리에 관한 연구 (Studies of Coagulase Production and Isolation of R-plasmid from Staphylococcus aureus)

  • A total of 129 clinical isolates of Staphylococcus species was characterized by the tests of coagulase production, haemagglutination, mannitol fermentation, DNase production and hemolysis. Ninety-nine out of them showed positive reactions to the tests, therefore they were identified as Staphylococcus aureus. The isolates showing positive reaction in haemagglutination test also showed 100% of tube coagulase positive reaction. The haemagglutination test was a reliable method for identifying Staphylococcus aureus in the clinical laboratory. S. aureus produced stronger hemolysis with human blood agar than with sheep blood agar. Antibiotic resistant S. aureus isolates(S-46, S-112, S-126) had 4 to 6 p]asmid DNA elements. The S-112 strain had 6 plasmid DNA elements(1.8, 2.2, 3.7, $26.3{\sim}50$, and 70 Mdaltons), the S-126 had 4 elements(2.6, 4.2, $4.6{\sim}60Md$), and the S-46 had 1 element(${\sim}100Md$). PPSA strain had 4 plasmid DNA elements(2.5, 4.2, $4.6{\sim}60Md$) and S. aureurs(ATCC) strain contained 9.4, 26.3 and ${\sim}50Md$ plasmid DNA elements.

함경도 지방의 전통가자미식해의 소금 첨가수준에 따른 숙성중 맛 성분의 변화에 관한 연구 (Effect of Salting Levels on the Changes of Taste Constituents of Domestic Fermented Flounder Sikhae of Hamkyeng-Do)

  • 함경도 지방의 전통 향토식품인 가자미식해를 소금절임농도를 달리하여 맛 성분의 변화를 관찰하였다. 당은 원료(대조구)에서 fructose, ${\beta}$-D-(+)-glucose, sucrose, maltose, mannose가 주로 검출되었고 그중 fructose가 가장 많이 검출되었으나 숙성된 식해에서는 fructose가 급격히 감소하는 반면 원료에서 검출되지 않았던 mannitol이 급격히 증가되어 15% 소금절임 식해에서 8.97%로 가장 높았다. 수용성 질소에 대한 아미노태 질소의 비가 가장 높은 것은 15% 소금절임 식해로 53.4%를 나타내었다. 식해에 많이 함유된 유리아미노산으로는 leucine, alanine, arginine, glutamine, isoleucine, valine, glutamic acid, lysine등으로 이들이 주로 식해의 맛 성분에 관련하는 것 같았다. 5'-Nucleotide는 CMP, UMP, CTP, AMP, ADP, ATP등이 검출되었고 처리에 관계없이 UMP가 가장 많이 검출되었으나 처리에 따른 큰 변화는 없었다. 원료에 비하여 15% 소금절임 식해에서 CMP가 2배, CTP가 1.25배 증가하였다. ATP, ADP 및 AMP는 원료에 비하여 식해에서 감소하였다.
