Biphenyldimethyl dicarboxylate(DDB)가 염화 제2수은-유발 간독성 흰쥐에서의 지질 과산화와 Oxygen Free Radical 제거효소 활성도에 미치는 영향 (Effects of Biphenyldimethyl Dicarboxylate(DDB) on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in Mercuric Chloride-induced Hepntotoxic Rats)

  • In an attempt to define the effects of biphenyldimethyl dicarboxylate (DDB) on the lipid peroxidation and oxygen free radical scavenging enzymes activities in mercuric chloride-induced hepatotoxic rats, we studied malondialdehyde (MDA) level and the activities of catalase and superoxide dismutase (SOD) in liver of the rats at 24 hr after the injection of mercuric chloride. Sprague-Dalwey albino rats were injected subcutaneously with mercuric chloride (5 mg/kg) only and mercuric chloride (5 mg/kg) plus. DDB (200 mg/kg/day, p.o) is administered for 4 days prior to 3 days from the injection of mercuric chloride. The group treated with mercuric chloride showed significantly higher MDA level and lower catalase and SOD activities as compared with that of control group. The group treated with mercuric chloride plus DDB showed significantly lower MDA level and catalase activity and higher SOD activity as compared with that of mercuric chloride-treated group. These results suggest that the excessive oxygen free radicals resulting from the depression of superoxide dismutase activity is an important determinant in the pathogenesis of mercuric chloride-induced hepatotoxicity and DDB has antioxidant effects.

  • In an attempt to dofine the early biochemical determinants that participate in the pathogenesis of glycerol-induced nephrotoxicity, especially focusing on oxygen free radicals and $N-acetyl-{\beta}-D-glucosaminidase$ (NAG) activity, we studied 24-hours urine outflow, 24-hours urinary protein excretion and urinary NAG activity after the injection of glycerol and also we studied malondialdehyde(MDA) level and superoxide dismutase(SOD) activity in the kidney of rats at 24hr after the injection of glycerol. Sprague-Dawley albino rats weighing 240 to 260 gm were injected intramuscularly with a 50% solution of glycerol(2ml/kg, 4ml/kg and 8ml/kg). The group treated with glycerol showed significantly lower urine outflow level and urinary protein excretion level and higher urinary NAG activity after the injection as compared to those of control group. Also the group treated with glycerol showed significantly higher MDA level and lower SOD activity at 24hr after the injection as compared to those of control group. These results suggest that the excessive oxygen free radicals resulting from the depression of SOD activity is an important determinant in the pathogenesis of glycerol-induced nephrotoxicity and higher urinary NAG activity is an index of renal tubular cell damage in the glycerol-induced nephrotoxicity.

Malondialdehyde Level by Ethanol Exposure in Mouse According to the ALDH2 Enzyme Activity

  • Excessive alcohol consumption is associated with increased risks of many diseases including cancer. Individuals who regularly consume excessive quantities of alcohol have a greater risk of developing head and neck cancers such as esophageal, pharyngeal and oral cavity cancers if they are deficient in ALDH2 expression compared to normal populations. We evaluated lipid peroxidation in Aldh2 +/+ and Aldh2 -/- mice after they had been subjected to acute ethanol exposure. Malondialdehyde(MDA) level in liver tissue was evaluated as a biomarker of oxidative lipid peroxidation. Although the ethanol treatment did not increase the hepatic MDA level both in Aldh2 +/+ mice and in Aldh2 -/- mice, the MDA level was significant higher in the Aldh2 -/- mice than in the Aldh2 +/+ group. The MDA level was also significantly correlated with olive tail moment in blood and the level of 8-OHdG in liver tissue. This is a strong evidence to support our hypothesis that oxidative stress is more intense in Aldh2 -/- mice than in Aldh2 +/+ mice. Our results suggest that ALDH2-deficient individuals may be more susceptible than wild-type ALDH2 individuals to ethanol-mediated liver disease, including cancer.

Prooxidant-antioxidant balance and malondialdehyde over time in adult rats after tubal sterilization and vasectomy

  • Objective: Sterilization (tubal sterilization and vasectomy) is a widely applied contraceptive method worldwide. Although most studies have described sterilization as a safe method, there are reports of tubal ligation (TL) and vasectomy complications. The aim of this study was to evaluate the effects of TL and vasectomy on the serum oxidative stress, specifically prooxidant-antioxidant balance (PAB) and malondialdehyde (MDA) levels, over time. Methods: Male and female rats were classified into vasectomy, sham-vasectomy, TL, and sham-TL groups, respectively. The PAB and MDA levels were measured on days 15 and 45 and months 3 and 6 after the intervention. For female rats, blood sampling was performed during the diestrous phase and estradiol and progesterone were also measured. Results: Serum PAB and MDA increased after TL (p<0.05). Vasectomy increased serum MDA remarkably after 45 days, 3 months, and 6 months (p<0.05). After vasectomy, serum PAB also increased although not significantly. Serum estradiol and progesterone decreased remarkably in the TL group compared to the sham group (p<0.05). Conclusion: Bilateral TL and vasectomy both increase the serum oxidative stress; however the imbalance after TL was very noticeable. As for the TL, the reduction of serum estrogen levels can be involved in this imbalance. Complications followed by TL or vasectomy could be due to increased levels of oxidants. Thus, prescribing antioxidants during and or after surgery may be a solution.

The Role of Ascorbic Acid on the Redox Status and the Concentration of Malondialdehyde in Streptozotocin-Induced Diabetic Rats

  • We investigated the role of ascorbic acid on the redox status in streptozotocin-induced diabetic rats. In the plasma of diabetic rats, the ratio of reduced/total ascorbic acid was significantly decreased as compared with normal control. Ascorbic acid supplementation increased the reduced and total ascorbic acid contents as compared with diabetic control. In the rutintreatment group, reduced and total contents of ascorbic acid were significantly decreased, however, the ratio of reduced/total contents of ascorbic acid had no difference as compared with diabetic rats. In the insulin-treatment group, this ratio is not significantly different as compared with diabetic control. However, in the insulin plus ascorbic acid treatment group, reduced form and the ratio of reduced/total ascorbic acid were significantly increased as compared with diabetic control. In addition, we measured the contents of malondialdehyde (MDA) in the plasma of diabetic rats. The contents of MDA was increased as compared with normal control, however, in insulin-treatment group, the contents of MDA was decreased as compared with diabetic rats. Ascorbic acid had no effects on the increases of MDA in diabetic rats. In conclusion, plasma ascorbic acid level and its reduced/total ratio reflects the status of the oxidative stress in the diabetic rats. Supplement of ascorbic acid did not correct the ratio of the reduced/total ascorbic acid. However, supplement of insulin and ascorbic acid corrected the ratio of reduced/total ascorbic acid.

Questionable Reliability of Malondialdehyde to Measure Oxidative Stress in Sjögren's Syndrome: Preliminary Study

  • Purpose: To investigate the expression of malondialdehyde (MDA), lipid peroxidation marker for oxidative stress (OS), in autoimmune Sjögren's syndrome (SjS) by utilizing the SjS-prone C57BL/6.NOD-Aec1Aec2 (B6DC) mouse and the SjS patient plasma samples. Methods: The MDA concentrations in the lysates of the submandibular gland, liver, and serum samples from the SjS-prone B6DC mouse model were compared with those from the C57BL/6J as a control. A thiobarbituric acid reactive substance (TBARS) assay kit was used to measure MDA. Plasma samples from five SjS patients and five control subjects were also evaluated. Results: The MDA concentrations in experimental animals and controls were not significantly different. There were no significant differences between the plasma of SjS patients and of controls. Conclusions: The expression of MDA was investigated in the organs from the SjS-prone B6DC mouse for the first time and in the plasma samples of SjS patients. No significant differences were observed between SjS and control samples when MDA was the target molecule with the TBARS assay. MDA may not be a reliable marker to measure OS contrary to the published studies involving OS of SjS.

레드비트를 함유하는 화장품의 담배 연기에 의한 피부 지질 산화 방지 효과 (Protective Effect of Cosmetics Containing Red Beet against Cigarette Smoke-induced Oxidative Damage in Human Skin)

  • 최근 화장품 시장에서 안티폴루션 효능이 있는 화장품은 새로운 피부 건강을 위한 해결책으로 나타나고 있다. 외부(대기) 오염물질에 의해 피부의 산화 기전을 매개하는 주요 원인 인자들로 오존, UV, 미세먼지 및 담배연기 등을 꼽을 수 있다. 담배 연기의 노출은 직 간접적으로 피부 표피 내 지질의 산화를 야기한다. 피부의 지질 산화에 의해 squalene과 squalene monohydroperoxide의 비율 변화가 발생하고, malondialdehyde (MDA)가 지질산화 산물로 생성된다. 따라서, 본 연구에서는 담배연기 노출 시 발생하는 피부 산화에 대하여 레드비트를 함유하는 화장품이 방지할 수 있는 효능이 있는지에 대하여 MDA의 생성량으로 관찰하였다. 시험대상자 전박부를 세 영역(음성대조, 양성대조, 시험제품)으로 나누어 각 지름 3.3 cm의 원으로 구획하고, 15분간 담배연기에 노출 후 피부 표면으로부터 지질막을 걷어낸 후 TBARS assay를 통하여 MDA를 정량 하였다. 음성대조(무도포, 미노출)에 비해 양성대조(무도포, 담배연기 노출)의 MDA 생성량이 3.7배 증가된 결과로, 오염물질인 담배연기에 의한 피부의 산화적 손상을 확인하였다. 반면에 레드비트를 함유하는 화장품을 미리 도포한 영역은 양성대조에 비해 MDA 생성량이 25% 감소하는 결과를 나타내었다. 결론적으로, 담배연기 노출은 피부표피 내 지질 산화를 야기하며, 레드비트를 함유하는 화장품이 이러한 대기환경오염으로부터 방어적 효과(안티폴루션 효과)를 보이는 것을 확인하였다.

염화비닐 노출 근로자의 요중 Malondialdehyde 농도 수준 (The level of urinary malondialdehyde concentrations in vinyl chloride monomer exposure workers)

  • The aim of this study was to investigate whether the exposure to vinyl chloride monomer(VCM) induces lipid peroxidation in workers by evaluating the concentration of malondialdehyde(MDA) in the urine in order to assess worker's oxidative stress due to exposure of vinyl chloride monomer. The subjects investigated in the study were divided into the experimental group; 18 workers exposed to VCM, and the control group; 19 workers unexposed to VCM. A gas chromatography/pulsed flame photometric detector(GC/PFPD) was utilized to analyze thiodiglycolic acid(TDGA), which was methylated with trimethylsilyldiazomethane (2.0M in diethyl ether) in urine and the urinary MDA, the product of lipid peroxidation, was determined by high-performance liquid chromatography/ultraviolet-visible detector after derivatized with 2,4-dinitrophenylhydrazine(DNPH). The concentrations of urinary TDGA in controls and VCM exposure workers were 0.13(2.01)mg/g Cr. GM(GSD) and 0.35(1.96)mg/gCr. GM(GSD), respectively. The concentrations of urinary MDA were $0.12(2.21){\mu}mol/gCr$. GM(GSD) in controls and $1.35(1.79){\mu}mol/gCr$. GM(GSD) in VCM exposure workers. As a result of simple regressions analysis between urinary concentration of TDGA and MDA in VCM exposure workers, it was found that the $R^2$ value was 0.261 (p=0.03) and the drinking and smoking did not affect their level. In conclusion, the workers exposed to VCM have a potentially to suffered by oxidative stress due to VCM exposure and the urinary MDA can be applicable to the marker of effect to assess the level of worker's VCM exposure.

우리나라 일부 농업 종사자의 개인보호구 착용, 작업위생행위에 따른 소변 중 MDA, 8-OHdG 농도 변화 (Changes in Urinary MDA and 8-OHdG Concentrations due to Wearing Personal Protective Equipment and Performing Protective Behaviors among Agricultural Workers in Korea)

  • Objectives: Oxidative stress and DNA damage have been proposed as mechanisms linking pesticide exposure to health effects such as cancer and neurological diseases. We investigated whether protective measures could significantly reduce the levels of biomarkers for oxidative stress and DNA damage in agricultural workers. Methods: In the present study, the levels of malondialdehyde (MDA) and 8-hydroxy-2'-deoxyguanosine (8-OHdG), biomarkers related to oxidative stress and DNA damage, respectively, were analyzed in urine samples collected from agricultural workers in two provinces of Korea (n=60). The influence of wearing personal protective equipment (PPE) and performing protective behaviors on the levels of these two biomarkers was also evaluated. Results: The median urinary levels of MDA and 8-OHdG were 10.45 nmol/mg creatinine and 14.42 ng/mg creatinine in subjects living in region A, while they were 6.25 nmol/mg creatinine and 24.77 ng/mg creatinine in subjects living in region B, respectively. The levels of MDA and 8-OHdG were higher in male farmers. Farmers wearing greater numbers of PPE and performing more protective behaviors had significantly lower levels of MDA. Greater numbers of protective behaviors was significantly associated with lower levels of 8-OHdG. Conclusion: The results of the present study indicate that pesticide exposure could induce oxidative stress and DNA damage in agricultural workers, and that protective measures are important for mitigating pesticide exposure.

홍삼복용이 노인들의 유산소 운동시 발생된 활성산소에 미치는 영향 (The Effects of Red-ginseng Intaking on Free Radical Produced During Aerobic Exercise in the Elderly)

  • 노인들의 고강도 유산소운동시 많은 산소요구량에 의하여 산화적인 스트레스가 증가하고, 결과적으로 함께 발생되는 활성산소는 MDA수준을 증가시킨다. 이렇게 노인들의 강도 높은 운동 중에 발생되는 활성산소에 홍삼복용이 어떠한 영향을 미치는 지를 알아보기 위하여 젊은이들과 비교하여 연구하였다. 노인들은 젊은이들과 함께 홍삼복용 전 후에 걸쳐 안정시, 운동직 후, 그리고 회복기 30분의 MDA수준이 유의하게 낮아진 것으로 나타났다. 따라서 홍삼복용은 젊은이들뿐만 아니라 노인들의 축적된 과산화지질을 제거하거나 과산화지질의 생성을 억제하는 항산화 효과가 있음을 시사한다.
