Protection of Radiation induced Somatic Damage by the Reduction of Oxidative Stress at Critical Organs of Rat with Naringenin Administration

  • Free radicals originate due to the radiolysis of cytoplasmic water with low "Linear Energy Transfer" (LET) radiations. Naringenin (Ng) is a natural antioxidative compound found in citrus fruits. This study revealed that Naringenin (Ng) reduced the radiation damage of critical organs by scavenging oxidative free radicals. In the study, Ng was orally administrated to rats daily for 7 consecutive days, prior to whole body exposure to gamma-rays. The scavenging efficacy was evaluated biochemically by measuring the concentration of cytotoxic byproducts and the activity of enzymes relevant to oxidative free radicals, after extracting the organs from the exposed rat. We observed increased levels of malondialdehyde (MDA) concentration, and decrease in the activities of superoxide dismutase (SOD) and catalase (CAT) in the exposed control group. However, pretreatment with Ng significantly reduced the MDA concentration, and increased the activities of SOD and CAT, as compared to the control group, due to the free radical scavenging by Ng. The results indicate that Ng administration prior to irradiation could protect critical organs from radiation damage.

  • Cervical cancer is the second most common gynecological malignancy worldwide. According to HPV Information Centre, Spain (Aug'2014), in India approximately 1,22,844 women are diagnosed with the disease every year and of them 67,477 die due to the disease. CaCx is said to be mediated by HPV but recent data published reveal the role of Oxidative Stress in different Cancers. Arsenic is also one of the agents for causing Oxidative Stress. Arsenic has been linked with different types of cancer. Arsenic is considered responsible for generation of free radicals and eventually for apoptosis. Early diagnosis of CaCx is presently a matter of concern and clinical presentation in advanced stages become difficult for complete clinical response. For determination of oxidative stress, Malondialdehyde (MDA) was taken as an identifier and arsenic estimation was performed with the help of Atomic Absorption Spectrometer (AAS). RBC count and Haemoglobin levels were performed according to standard protocol. MDA was in direct proportion with arsenic concentration and inversely proportional to RBC and Haemoglobin in CaCx patients. Arsenic is one of the major causative agents for oxidative stress and hence may be a risk factor leading to cancer including CaCx.

Interactions between Oxidative Stress, Lipid Profile and Antioxidants in Breast Cancer: A Case Control Study

  • Oxidant/antioxidant balance has been suggested as an important factor for initiation and progression of cancer. The objective of this study was to determine changes in the levels of malondialdehyde (MDA), nitric oxide (NO), total cholesterol, triglycerides, LDL-cholesterol, HDL-cholesterol, total antioxidant capacity (TAC), glutathione peroxidase (GSH-Px), and superoxide dismutase (SOD) activities in serum samples of breast cancer patients (n=30) and healthy subjects (n=100). MDA and NO levels were found to be increased in breast cancer patients compared to the healthy subject group (p<0.05). Total cholesterol and triglycerides were elevated; and HDL-cholesterol level was found to be decreased in the cancer patients as compared to the healthy subjects (p<0.05). Compared to the healthy group, both serum TAC levels (p<0.001) and activity of SOD and GSH-Px (p=0.05) were found to be decreased in the breast cancer patients as compared to the healthy controls. Considering the data presented in this study, we suggest that free radicals induce lipid eroxidation and peroxidation of unsaturated fatty acid with decreased activity of enzymatic antioxidants in breast cancer.

EFFECTS OF GINSENOSIDE $Rb_2$ ON THE ANTIOXIDANTS IN SENESCENCE - ACCELERATED MICE(SAM - R/1)

  • In biological system, there are enzymes such as superoxide dismutase(SOD), catalase and glutathione(GSH) peroxidase which scavenge reactive oxygen species as well as antioxidants such as ceruloplasmin, albumin and nonprotein-bound SH including GSH related to defense mechanism. In the present study, the protective effects of ginsenoside $Rb_2$ against oxidative stress were investigated in the SAM-R/1 mice. Treatment with ginsenoside $Rb_2$ significantly increased Cu, Zn-SOD and Mn-SOD in the liver. Ginsenoside $Rb_2$ tended to increase hepatic catalase activity and significantly increased serum albumin and nonprotein-bound SH levels in the liver. But treatment with ginsenoside $Rb_2$ showed a significant decrease in hepatic malondialdehyde(MDA) levels compared to control group. Furthermore, we compared the effects in the hepatic SOD, MDA and serum albumin. These findings suggest that the increase of antioxidants by ginsenoside $Rb_2$ results in the protective effects against reactive oxygen species.

사물탕(四物湯)이 노화백서(老化白鼠) 뇌조직(腦組織)의 생화학적(生化學的) 변화(變化)에 미치는 영향(影響) (Effects of Samultang on the Biochemical Changes in Brain Tissue of Senile Rat)

  • The present experiment was desined to examine catecholamines, 5-hydroxytryptamine, amino acids, malondialdehyde(MDA) and free radical scavening activity, by administering Samultang extract of a variety of concentraction to senile brain The results were summarized as followings: 1. Samultang significantly increased noradrenaline in the cortex, striatum, hippocampus and hypothalamus of the brain tissue of senile rats, and even though Samultang increased noradrenaline also in other brain tissue, there was no significance. 2. Samultang had effects on dopamine changes in hypothalamus of the brain tissue of senile rats. 3. Samultang significantly increased 5-hydrotryptamine in pons-medulla oblongota and cerebellum, but decreased in hypothalamus. 4. Samultang increased amino acid in the brain tissue of senile rats. 5. Samultang significantly decresed lipid peroxide production in the brain tissue of senile rats. 6. Samultang significantly decresed MDA and free radical in the brain tissue of senile rats. According to the above results, Samultang is assumed to improve brain function by reacting on biochemical of the senile brain, and that Samultang can be used to treat regressive brain disease carrying symptoms of psychoactive disorders.

The Beneficial Effect of Melatonin for Toluene Hepatotoxicity in Rats

  • Toluene is mainly metabolized in liver by oxidative pathway. Oxigen free radicals occur through the process of toluene metabolism Therefore it causes tissue and cell min by the oxygen free radicals from the metabolism of toluene. Melatonin acts as a highly efficient free radical scavenger that protects cells from damage by oxygen free radicals. To test this hypothesis, toluene hepatotoxicity was induced by an abdominal injection of toluene. To see if the melatonin protects the rat's liver, melatonin was administrated orally, at the time of each toluene injection. Aspartate aminotransferase(AST), alanin aminotransferase(ALT), latic dehydrogenase(LDH) and alkaline phosphatase(ALP) levels in serum were measured to estimate hepatic function. Malondialdehyde(MDA), which gives an indirect index of oxidative injury was also measured. Hippuric acid is the last metabolic Production of toluene was measured by HPLC. There were significantly higher in AST, ALT, LDH, MDA and hippuric acid in toluene group, but there were no significant difference in melatonin group except ALT and hippuric acid. There was significantly lower in ALP level in toluene group, but there was no significant difference melatonin group, suggesting a significant hepatotoxicity due to oxygen free radicals through the process of toluene metabolism Melatonin treatment significantly protected hepatic function and free radical-mediated injury in the liver against toluene-induced changes. Accordingly, this study shows that melatonin is helpful in protecting liver injury by acute toluene intoxication.

실리마린의 간보호 작용에 미치는 쿠쿠마 추출물의 영향 (Curcuma Extract Enhances Hepatoprotective Action of Silymarin in Experimental Liver Damage Models)

  • Silymarin and curcumin have been used for supportive treatment of liver disease of difffrent etiology due to their hepatoprotective activities. The present study was carried out to investigate the hepatoprotective efffcts of silymarin and/or curcuma extract against hepatotoxins induced liver injury. To investigate hepatoprotective effects, the silymarin and/or curcuma extract were pre-treated orally to experimental animals. And thereafter a single dose of hepatotoxin, carbon tetrachloride ($CCl_4$) and acetaminophen were administered through oral or intraperitoneal route, respectively. Chronic liver damage was induced by subcutaneous injection of $CCl_4$ for 3 weeks (2 times/week). Hepatoprotective and therapeutic effects were monitored by estimating serurn ALT and AST levels and by measuring hepatic glutathione (GSH) and malondialdehyde (MDA)levels. Collagen type 1 was detected with irnrnunostaining to assess fibrosis. The results showed that the mix-ture of silymarin and curcuma extract significantly reduced serum biochemistry levels and MDA levels com-pared with those of control group in both acute and chronic animal models. In antifibrotic effect, the relative hepatic collagen content was significantly decreased by silymarin and/or curcuma extract treatment. It was concluded that the complex of silymarin and curcuma extract have a both hepatoprotective and therapeutic effect synergically in rat liver injury induced by heptotoxins.

흰쥐 뇌의 측중격핵에서 망간에 의한 산화적 손상에 관한 연구 (Manganese-induced Oxidative Stress in Nucleus Accumbens of Rat Brain)

  • This study was carried out to identify the effect of oxidative stress on the pathogenesis of manganese intoxication. Five rats in experimental group were given with $MnCl_2$intraperitoneally for 4 weeks(4 mg/kg once daily 5 days per week) and another five rats for control group were given with normal saline. In experimental group, manganese concentrations increased significantly in nucleus accumbens by 142% (p<0.05), SOD activities increased significantly by 124%(p<0.01), and MDA concentrations increased significantly 148%(p<0.05) compared with control group. Among fatty acids, total n-6 polyunsaturated fatty acids(PU) increased significantly by 231%(p<0.05) compared with control group. Arachidonic acids(AA) increased by 224%(p<0.05), and these increase were composed mostly of n-6 polyunsaturated fatty acids(PUFA). Among n-3 PUFAs except linolenic acids, eicosapentanolc acid(EPA) decreased significantly by 38%(p 0.01) and docosahexanoic acids(DHA) decreased by 30% p<0.05) compared with control group. Our results suggest that the oxygen free radicals produced by manganese may cause compositional changes of fBtty acids in nucleus accumbens of the rat. Characteristics of fatty acids compositional changes by manganese were the decrease of EPAs and DHAs(n-3 PUFAs), and increase of AAs(n-6 PUFAs). These changes with the increase of MDA, suggest that manganese neurotokxcity is caused by lipid perokidation mediated with oxygen free radicals, especially superoxide radicals.

흰쥐 골격근의 허혈-재관류 손상후 생화학적 변화에 미치는 Melatonin의 효과 (The Effect of Melatonin on Biochemical Changes after Ischemia-Reperfusion Injury of Rat Skeletal Muscle)

  • The ischemia-reperfusion injury of the skeletal muscles is caused by generation of reactive oxygen during ischemia and reperfusion. Melatonin or N-Acetyl-5-methoxy- tryptamine is suggested to have antioxidant effects in several tissues. In present study, we examined the protective effect of melatonin in a rat hind limb ischemia-reperfusion injury. Dimethyl-sulfoxide(DMSO) was also tested for comparison. Ischemia was induced for 4 hours by vascular clamping and followed by 1 hour or 24 hours of reperfusion. Muscle injury was evaluated in 4 groups such as single laparotomy group(control), ischemia-reperfusion group, DMSO group, melatonin group. Eedema ratio and malondialdehyde(MDA) of muscle tissue and serum level of creatine kinase(CK), were measeured at the end of reperfusion. DMSO and melatonin group showed significant amelioration of edema and serum CK compared with ischemia-reperfusion group. The decreasing effect was more prominent in melatonin group. The muscle tissue MDA concentration is significantly lower in melatonin group than in ischemia-reperfusion group. The results show that melatonin prevents and improves ischemia-reperfusion injury more effectively in a rat hind limb than DMSO dose. Thus, clinically the melatonin may be used for a beneficial treatment of such injuries

Physiological and Biochemical Responses of Fifteen Rice Cultivars to UV-B Radiation

  • This study was conducted to examine the physiological and biochemical responses against UV-B radiation in the seedling of 15 different rice cultivars, having the different physiological sensitivities. Out of 15 rice cultivars tested, moderate and susceptible groups showed significant decreases in biomass and RGR (relative growth rate). Contents of total chlorophyll were reduced remarkedly by irradiation of UV-B. In all rice cultivars tested, the content of chlorophyll a was strongly decreased, while the contents of chlorophyll b were slightly reduced without showing clear different among three groups and 15 cultivars. Carotenoid content was largely reduced by UV-B radiation, whereas polyamine content was moderately increased. The contents of MDA (malondialdehyde) that reflect the level of lipid peroxidation of cell membranes were clearly increased by UV-B stress, showing higher content in susceptible cultivars than moderate and torelant cultivars. The physiological important parameters highly related to visible injury were leaf color, chlorophyll, carotenoid, and lipid peroxidation, whereas biomass and polyamines were not closely correlated. Based on this results, it was concluded that changes of visible injury and the contents of chlorophyll and MDA could be adequately applied and utilized as physiological indicators to UV-B radiation.
