Construction of a Biofilter Immobilized with Rhodococcus sp. B261 for Removal of H2S Gas Generated by Livestock

  • To explore the optimal conditions for the removal of $H_{2}S$ gas by biofiltration, various conditions, including inlet $H_{2}S$ concentration, flow rate, moisture, and cell number, were examined. Heterotrophic bacteria were isolated from the compost of the animal excreta. A strain that effectively removed $H_{2}S$ was selected and identified as Rhodococcus rhodochrous B261 by analysis of its 16S rDNA sequence. A cell number of $10^{7}\;cfu/g^{-}compost$ was sufficient to dominate the microbiota, and an effective removal was observed at $H_{2}S$ gas concentrations below 220 mg/L. The moisture content of 33-38% was suitable for activation of the microbial activity and delaying the desiccation. Higher flow rates resulted in lower removal rates of the $H_{2}S$ gas. Under the conditions of $10^7\;cfu/g^{-}compost$, $H_{2}S$ gas concentrations of 220 mg/L, and moisture content of 33-38%, the inlet $H_{2}S$ gas concentrations of 120 and 400 mg/L were completely removed for 34 and 12 days, respectively. The amount of sulfur removed was $2.99{\times}10^{-9}H_{2}S-S/cell$, which was suggested as the amount of sulfur removed by a single cell. The biofilter consisting of the compost and R. rhodochrous B261 could be suitable for a long-term biofilteration for the removal of $H_{2}S$ and other malodorous compounds.

Studies of gas chromatographic analysis of malodorous S compounds in air (대기 중 악취황 성분들에 대한 GC 분석의 특성)

  • In this study, analytical characteristics of S gas detection technique were investigated against four major reduced S compounds (including hydrogen sulfide; methyl mercaptan; dimethyl sulfide (DMS); and dimethyl disulfide (DMDS)). To analyze such properties, an analytical system was constructed by combining the GC/PFPD system with the loop injection method. The results of our analysis indicated that response behavior of S gases differs greatly between compounds; H2S exhibited the weakest sensitivity of all compounds, while DMDS with two S-atom compounds the strongest sensitivity. To learn more about their response behavior on GC/PFPD method, their calibration patterns were compared using the three arbitrarily set concentration ranges of low, intermediate, and high. The results showed that calibration patterns of each compound are distinguished because of different factors. There was a line of evidence that calibration of $H_2S$ was affected noticeably by adsorptive loss within the system, whereas those of DMS and DMDS were influenced most sensitively by such factor as the linearity response at a given PMT voltage setting. The overall results of our study suggest that quantification of malordorous S compounds require a better knowledge of compound-specific response behavior against GC detection.

Deodorization of H2S, CH3SH in Soil Filter Reactors Packed with Montmorillonites, Rice Hulls and Thickening-activated Sludge (Montmorillonites, 왕겨 및 농축활성슬러지를 충진한 토양상에서의 H2S, CH3SH의 제거)

  • Deodorization characteristics and removal rate of sulfur-containing odor have been investigated in the soil filter reactors packed with montmorillonites (Mont.), rice hulls(Rh.), and thickening-activated sludge(Ts.). And variation of pH and $SO_4{^{2-}}$ with the removal of malodorous sulfur compounds have been investigated together. As compared removal rate of montmorillonites between wet and dry condition for sulfur compounds through batch test, it showed that wet condition was better than dry one; removal ratio, as wet/dry, was $H_2S$ of 1.2 and $CH_3SH$ of 1.9, and decrease of pH and increase of $SO_4{^{2-}}$ concentration in the wet condition also showed to be larger than in dry condition. In continuous test for biological deodorization experiment, removal rate of sulfur compounds in reactor packed with Mont., Rh. and Ts, was more than 98 %, and the variation of static pressure was maintained stably under condition of SV $150h^{-1}$, LV 4.2 mm/sec and SV $200h^{-1}$, LV 5.6 mm/sec, and in reactor packed with Mont. and Rh., $H_2S$ was 76.4 % to 87.2 % and $CH_3SH$ was 87.8 % to 93.3 % under the same condition. From above results, it ascertained that it can obtain the high deodorization efficiency by inoculating thickening-activated sludge in soil filter using montmorillonites.

Evaluation of Odors and Odorous Compounds from Liquid Animal Manure Treated with Different Methods and Their Application to Soils (액상 가축분뇨의 처리 및 토양환원에 따른 악취 및 악취물질의 평가)

  • To comply with stricter regulations provoked by increasing odor nuisance, it is imperative to practice effective odor control for sustainable livestock production. This study was conducted to assess odor and odorous compounds emitted from liquid animal manure with different treatment methods such as Fresh Manure(without treatment, FM), Anaerobic Digestion(AD) and Thermophilic Aerobic Digestion(TAD) and their application to soil. Air samples were collected at the headspace of liquid manure, upland and paddy soil, and analyzed for odor intensity and offensiveness using an olfactometry; odor concentration index using odor analyser; nitrogen-containing compound such as ammonia(NH3) using fluorescence method; and sulfur containing compounds such as hydrogen sulfide(H2S), methyl mercaptan(MeSH), dimethyl sulfide(DMS) and dimethyl disulfide(DMDS) using gas chromatography-pulsed flame photometric detector, respectively. Odor intensity, offensiveness and concentration index from TAD liquid manure was statistically lower than those from FM and AD(p<0.01). Mean concentrations of H2S, MeSH, DMS, DMDS and NH3 were 65.93ppb, 18.55ppb, 5.26ppb, 0.33ppb and 10.57ppm for liquid manure with AD; and 5.15ppb, 0.97ppb, 0.80ppb, 0.56ppb and 1.34ppm for liquid manure with TAD, respectively. More than 60% of malodorous compounds related to nitrogen and sulfur were removed by heterotrophic microorganisms during TAD treatment. When liquid manure was applied onto upland and paddy soil, NH3 removal efficiencies ranged from 51 to 94% and 22 to 91% for AD and TAD liquid manure, respectively. The above results show that liquid manure with TAD is superior to AD and FM with respect to the odor reduction and odor problem caused by land applied liquid manure is directly related to the degree of odor generated by the manure treatment method.

  • Recently there is much interest in the antibacterial activity of nano-sized silver particle (nanosilver) since silver is known to be safe and effective as disinfectant for a long time. Oral malodor is considered to originate in the oral cavity primarily as a result of production of malodorous compounds by oral bacteria. Major compounds responsible for oral malodor are volatile sulfur compounds, which is thought to be generated by the G(-) anaerobic bacteria found normally in the oral cavity, especially on the dorsum of the tongue. The purposes of this study were to investigate the effect of nanosilver on growth of oral malodor generating microorganisms, including Fusobacterium nucleatum, Prevotella melaninogenica, Klebsiella pneumonia, and to determine the optimal culture condition of them. The results were as follows: 1. The optimal culture condition for P. melaninogenica was vacuum culture using desiccator after evacuation of air by vacuum pump in chopped beef meat media. 2. The growth of K. pneumonia was temporarily inhibited by nanosilver (5 ppm and 10 ppm). 3. The morphological alteration and cell damage caused by nanosilver were observed in K. pneumonia.
