Vegetative and Male Reproductive Anatomy of Laurencia intercalaris sp. nov. (Rhodomelaceae, Rhodophyta) in Korea

  • The vegetative and male reproductive anatomy of a marine alga, Laurencia intercalaris sp. nov. (Rhodomelaceae, Rhodophyta), is described from subtidal habitats of eastern and southern Korea. This species has terete thalli with entangled fibrous holdfasts and regularly alternate branching of ultimate branchlets, and is inseparable from L. okamurae Yamada on the basis of habit. Vegetative axial cells produce a trichoblast and four pericentral cells in an alternating sequence. Spermatangia are produced intercalary or subterminally from one of two laterals on suprabasal cells of trichoblasts arising from axial cells in apical pits of branchlets. The other lateral remains sterile. In this sterile lateral, budding-like regeneration occurs on older segments that are oabscised. Comparison is made with other related Laurencia species, particularly those with terete thalli. The vegetative anatomy and the regeneration in sterile laterals of male trichoblasts, with the mode of spermatangial formation, distinguish the new species from previously described species of Laurencia including L. okamurae.

역병-풋마름병 복합저항성 세포질웅성불임계 고추 육성 (Breeding Cytoplasmic Male Sterile Lines Resistant to Phytophthora capsici and Ralstonia solanacearum in Capsicum Pepper)

  • 칠성초에 역병 저항성을 도입한 칠복1호에 베트남 도입 풋마름병 저항성 계통을 교배하여 육성한 $F_2$$BC_1F_1$에서부터 $F_4$$BC_1F_3$까지 역병-풋마름병 복합 저항성 선발을 2009년도와 2010년도에 걸쳐 수행하였다. 매 세대 역병을 접종하여 저항성을 평가하여 선발하고 선발개체에 풋마름병을 접종하여 감염되는 개체는 도태하였다. 역병에 대한 저항성은 선발과 함께 현저히 향상되었으며, 선발계통들은 역병 저항성으로 판매되고 있는 교배종 '무한질주'와 비슷한 수준의 저항성을 나타내었다. $BC_1F_2$ 선발개체를 칠복CMS-A라인에 교배를 하여 $F_1$의 임성을 보고 화분친의 CMS-Rf유전자형을 검정하였다. 대부분Nrfrf로 고정되고, 칠복 ${\times}$ KC995, 칠복 ${\times}$ KC1009 조합의 일부 개체가 이형(heterozygote) 상태인 것으로 확인되었다.

Determination of Cytoplasmic Male Sterile Factors in Onion Plants (Allium cepa L.) Using PCR-RFLP and SNP Markers

  • We have developed a polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP) marker that can distinguish male-fertile (N) and male-sterile (S) cytoplasm in onions. The PCR-RFLP marker was located in a chloroplast psbA gene amplicon. Digesting the amplicons from different cytoplasm-containing varieties with the restriction enzyme MspI revealed that N-cytoplasm plants have a functional MspI site (CCGG), whereas the S-cytoplasm plants has a substitution in that site (CTGG), and thus no MspI target. The results obtained using this PCR-RFLP marker to distinguish between cytoplasmic male sterile factors in 35 onion varieties corresponded with those using a CMS-specific sequence-characterized amplified region (SCAR) marker. Moreover, the PCR-RFLP marker can identify N- ot S-cytoplasms in DNA sample mixtures in which they are in up to a 10-fold minority, indicating that use of the marker has high diagnostic precision. We also demonstrated the usefulness of the SNP detected in the psbA gene for high-throughput discrimination of CMS factors using Real-time PCR and a TaqMan probe assay.

Proteome Analysis of the Young Spikelets of Photoperiod-Sensitive Rice Mutant Treated in Different Photoperiods

  • Photoperiod sensitive genetic male sterile (PGMS) rice is sterile mutant controlled by photoperiod. A PGMS mutant 920S was sterile grown under long-day (LD) photoperiod (14 h light/10 h dark) but fertile grown under short-day (SD) photoperiod (10 h light/14 h dark). Proteome analysis revealed that 12 protein spots were differentially expressed in the spikelets of 920S plants either treated with LD or SD photoperiod. Among these proteins, three proteins including chlorophyll a/b binding protein, vacuolar ATPase ${\beta}-subunit,\;{\alpha}-tubulin$ and an unknown protein were more than three-fold abundant in the spikelet of the SD-treated plants than those of the LD-treated plants. On the other hand, eight proteins including acetyl transferase, 2, 3- biphosphoglycerate, aminopeptidase N, pyruvate decarboxylase, 60S acidic ribosomal protein and three unknown protein spots were more abundant in the spikelets of the LD-treated plants than those of the SD-treated plants. The results suggest that the observed proteins may be involved in sterile or fertile pollen development under LD or SD photoperiod respectively in the PGMS mutant rice.

양질 다수성 일대잡종 벼 육성 연구 I. 우리나라 품종 배경의 세포질-유전자적 웅성불임 및 임성회복 계통 육성 (Breeding Hybrid Rice with Good Quality and High Yield I. Breeding of Cytoplasmic-Genetic Male Sterile and Restorer Lines with Backgrounds of Korean Rice Varieties)

  • 우리나라 japonica 벼의 일대잡종을 다양하게 육성할 목적으로 세포질 유전자적 웅성불임인 BTCMS에 대한 임성회복력이 있는 계통 AR-3을 교잡하여 그 F1을 모본으로 하고 조ㆍ중ㆍ만생의 한국 japonica형 벼 4품종씩을 교본 반복친으로 5회 backcross하여 우리나라 벼 품종 배경의 우엉불임 계통과 임성회복계통을 육성하고 이들의 주요 작물학적 특성을 각각의 반복친과 비교한 결과를 요약하면 다음과 같다. 1. 우리나라 japonica형 품종의 배경을 갖는 세포질 유전자적 웅성불임계통을 조ㆍ중ㆍ만생군으로 각각 4개씩 도합 12계통 육성하였으며, 동일한 품종의 배경을 갖는 임성회복계통친도 조ㆍ중ㆍ만생군으로 각각 4개씩 12계통 육성하였다. 2. 육성된 웅성불임계통은 조중군에서 소백벼 A, 오대벼,A, 관악벼A, 대성벼A, 중생군에서 화진벼A, 팔달A, 수원 224A, 이리 386A, 만생군에서 낙동벼A, 팔굉A, 화청벼A 및 밀양 97A 등으로 명명하였다. 3. 육성된 임성회복계통은 조생군에서 소백벼R, 오대벼R, 팔달R, 수원 224R, 이리 386R, 만생군에서 낙동벼R, 팔굉R, 화청벼R 및 밀양 97R 등으로 명명하였다. 4. 육성된 웅성불임계통들은 반복친(불임유지친)에 비하여 간장이 단축되는 경향이었으나 출수기, 수장 및 수량구성요소 등은 대체로 비슷하였다. 5. 육성된 임성회복계통들은 반복친과 비교하면 대체로 간장, 수장, 수량구성요소 및 수량의 차이가 없었다.

  • 연초(N. tabacum L.)의 세포질 웅성불임 F$_1$ 식물체와 N. africana의 종간교배에 의한 웅성불임 반수체식물의 육성 및 특성을 조사한 결과는 다음과 같다. 1. (CMS NC82$\times$burley 21) F$_1$과 N. africana의 종간교배에 의한 세포질 웅성불임 반수체 식물의 출현빈도는 삭당 0.5주로 나타났다. 2. 종간교배으로 얻은 웅성불임 반수체식물 중 잎과 줄기의 색이 green(황색종)과 yellow(버어리종)의 분리비는 3 : 1로 나타났다. 3. 반수체 식물 중 잎과 줄기의 색이 yellow(버어리종 type)인 개체에 TMV저항성을 검정한 결과 저항성과 이병성의 비는 1 : 1로 나타났다. 4. (CMS MC82 Burley 21)F$_1$과 N. africana의 종간교배으로 세포질 웅성불임 isogenic Burley 21 반수체 식물의 육성이 가능하였고 이 반수체 식물의 염색체를 배가함으로써 여교배육종법에 비하여 조기에 세포질 웅성불임 Burley 21을 육성할 수 있을 것으로 생각된다.

  • 역병(Phytophthora capsici)과 세균성점무늬병(Xanthomonas euvesicatoria)에 복합저항성인 핵유전형 웅성불임계(Genic male sterile line, GMS)를 육성하기 위하여 역병 저항성 핵유전형 웅성불임계에 역병-세균성점무늬병 복합저항성 계통을 교배하여 작성한 조합의 $F_3$$F_4$ 세대에 대하여 두 가지 병에 대한 저항성 선발을 실시하였다. 역병 저항성은 잘 알려져 있는 KC294(CM334)와 KC263(AC2258)에서 도입하고, 세균성점무늬병 저항성은 KC47(PI24467)에서 도입되었다. 역병에 고도의 저항성을 지닌 GMS 계통이 얻어졌으며, 이들은 세균성점무늬병에도 양적으로 저항성을 나타낼 것으로 기대된다.

Sources of Resistance to Bacterial Wilt Found in Vietnam Collections of Pepper (Capsicum annuum) and Their Nuclear Fertility Restorer Genotypes for Cytoplasmic Male Sterility

  • Eleven hot pepper accessions collected in Vietnam showed stable resistance to bacterial wilt as well-known resistance sources, MC4 and MC5, in repeated inoculation tests with different Ralstonia solanacearum isolates conducted from 2004 to 2010. Seven of these accessions (specifically KC981, KC1006, KC1021, KC1027, KC1045, KC1050, and KC1055) resulted in stable male sterile F1 plants in the crosses with a cytoplasmically male sterile (CMS) Chilseong (CMS-A, Srfrf ), and therefore, they were maintainers (CMS-B) with a genotype of Nrfrf. The rest (KC980, KC995, KC999, and KC1009) produced stable male fertile F1 plants in the crosses, and therefore, were restorers (CMS-C) with a genotype of N(S)RfRf. Therefore, the maintainer and restorer sources of resistance may be used in preference in breeding maternal (CMS and their maintainers) and paternal parents (restorers) for resistance to bacterial wilt, respectively, in the hybrid breeding system utilizing cytoplasmic male sterility.

Hybridization Using a New Male-sterile Germplasm as the Female Parent in Chinese Jujube

  • This study identified a new male-sterile germplasm of Chinese jujube, named male-sterile No. 2 (JMS2), and achieved controlled hybridization using that germplasm as the female parent. The anthers of JMS2 before flower bud opening became shrunken, dingy yellow and much smaller than normal ones, and they changed to brown after anthesis. No pollen was observed in anthers of JMS2 and its male-sterile trait remained stable over different years. A total of 1,642 fruits were obtained from ten intra- and interspecific cross combinations via controlled hybridization from 2008 to 2012 using JMS2 as the female parent. Of those, 27.3% produced seeds, on average (0-72.6%). The rate of fruit with seed (RFS) was significantly different between cross combinations or years. Compared to other cross combinations, the RFS in the combination of JMS2 ${\times}$ 'Xingguang' (a Chinese jujube cultivar with high resistance to jujube witches' broom disease) and JMS2 ${\times}$ 'Xing16' (a sour jujube genotype) remained high in different years and reached means of 48.7 and 58.1%, respectively. Finally, 150 plantlets were regenerated from immature embryos, and 51 of them were randomly selected and identified to be authentic hybrids using amplified fragment length polymorphism (AFLP) markers. This is the first report of hybrids obtained from a cross between Chinese jujube and sour jujube.
