Visible Surface-Assisted Laser Desorption/Ionization Mass Spectrometry of Small Macromolecules Deposited on the Graphite Plate

  • Visible surface-assisted desorption/ionization mass spectrometry (SALDI-MS) has been investigated for several small macromolecules deposited on the graphite plate using laser radiation at 532 nm where most of the macromolecules are transparent. The graphite surface functioned well as a photon absorbing material and an energy transfer mediator for visible light. The results show that visible SALDI is a much softer ionization technique than UV-MALDI and FAB-MS in our results with synthetic macromolecules, PPG, PPGMBE and cavitand molecules. For the SALDI of biomolecules, glycerol as a proton source was essential with the graphite plate. As in visible SALDI, the role division of the photon absorbing material and the cationization agent can provide a generality in mass spectrometric analysis of macromolecules compared with MALDI using the dual functional matrix.

  • Salivary research is at a critical crossroads regarding the clinical application of the basic knowledge. The purpose of this article is to introduce the current progress on salivary research to Korean dental scientists. The accumulated results based on advance technologies such as protein chemistry, molecular biology, and structural biology have showed that salivary macromolecules need structural requirements for proper function. Currently, several concepts or principles, which can be applied to salivary macromolecules, have been suggested. These include the role of molecules' conformation on biological activity, their multifunctional nature, their redundancy of function, their amphifunctional properties, and the potential importance of complexing between molecules. These concepts and the information available will help the development of saliva substitutes, the design of drug carriers and chimera molecules with enhanced function and the development of gene therapy protocols. These approaches will alleviate or restore lost salivary function and can be used to treat various kinds of oral and systemic diseases.

Synthesis and Characterization of Sulfonated Poly(arylene ether) Polyimide Multiblock Copolymers for Proton Exchange Membranes

  • Novel multiblock copolymers, based on segmented sulfonated hydrophilic-hydrophobic blocks, were synthesized and investigated for their application as proton exchange membranes. A series of segmented sulfonated poly(arylene ether sulfone)-b-polyimide multiblock copolymers, with various block lengths, were synthesized via the coupling reaction between the terminal amine moieties on the hydrophilic blocks and naphthalene anhydride functionalized hydrophobic blocks. Successful imidization reactions required a mixed solvent system, comprised of NMP and m-cresol, in the presence of catalysts. Proton conductivity measurements revealed that the proton conductivity improved with increasing hydrophilic and hydrophobic block lengths. The morphological structure of the multiblock copolymers was investigated using tapping mode atomic force microscopy (TM-AFM). The AFM images of the copolymers demonstrated well-defined nanophase separated morphologies, with the changes in the block length having a pronounced effect on the phase separated morphologies of the system. The self diffusion coefficient of water, as measured by $^1H$ NMR, provided a better understanding of the transport process. Thus, the block copolymers showed higher values than Nafion, and comparable proton conductivities in liquid water, as well as under partially hydrated conditions at $80^{\circ}C$. The new materials are strong candidates for use in PEM systems.

Movement and evolution of macromolecules in a grooved micro-channel

  • This paper presented an investigation of macromolecular suspension in a grooved channel by using the dissipative particle dynamics (DPD) with finitely extensible non-linear elastic (FENE) bead spring chains model. Before studying the movement and evolution of macromolecules, the DPD method was first validated by modeling the simple fluid flow in the grooved channel. For both simple fluid flow and macromolecular suspension, the flow fields were analyzed in detail. It is found that the structure of the grooved channel with sudden contraction and expansion strongly affects the velocity distribution. As the width of the channel reduces, the horizontal velocity increases simultaneously. Vortices can also be found at the top and bottom corners behind the contraction section. For macromolecular suspension, the macromolecular chains influence velocity and density distribution rather than the temperature and pressure. Macromolecules tend to drag simple fluid particles, reducing the velocity with density and velocity fluctuations. Particle trajectories and evolution of macromolecular conformation were investigated. The structure of the grooved channel with sudden contraction and expansion significantly influence the evolution of macromolecular conformation, while macromolecules display adaptivity to adjust their own conformation and angle to suit the structure so as to pass the channel smoothly.

Fouling Mechanism of Microfiltration/Ultrafiltration by Macromolecules and a Suppression Strategy from the Viewpoint of the Hydration Structure at the Membrane Surface

  • This short review focuses on fouling by proteins and macromolecules in microfiltration/ultrafiltration. First, an experimental system that enables investigation of how the extent of the adsorption of proteins and macromolecules on membrane surfaces contributes to a decrease in filtrate flux in microfiltration/ultrafiltration is described. Using this system, a causal relationship - not a correlation - indicating that adsorption results in a decrease in filtrate flux could be clearly demonstrated in some cases. Second, a hydration structure at the membrane surface that can suppress adsorption is discussed, inspired by biomaterial research. In their hydrated states, polymers with low-fouling properties have water molecules with a particular structure. Finally, some successful examples of the development of low-fouling membranes via surface modification using low-fouling polymers are discussed.

식품 거대분자의 열역학적 비혼합성 (Thermodynamic Incompatibility of Food Macromolecules)

  • Proteins and polysaccharides are major food macromolecules. Generally, the mixture of these macromolecules can be separated into two phases because of their thermodynamic incompatibility. Phase separ-ation is explained by equilibrium phase diagram, which comprises binodal curve, critical point, phase separation threshold, tie-line and rectilinear diameter. Phase separation of protein-polysacc-haride solution is affected by pH, temperature, ionic strength, molecular weight, molecular structure, etc. Membraneless osmosis has been developed to concentrate protein solutions, using the phase diagram constituted by proteins and polysaccharides. Protein-polysaccharide mixtures are very promising fat mimetics because solution of mixtures forms water-continuous system with two phase-separated gels, which give plastic texture and a fatty mouthfeel.

Major ginsenosides from Panax ginseng promote aerobic cellular respiration and SIRT1-mediated mitochondrial biosynthesis in cardiomyocytes and neurons

  • Background: Aerobic cellular respiration provides chemical energy, adenosine triphosphate (ATP), to maintain multiple cellular functions. Sirtuin 1 (SIRT1) can deacetylate peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α) to promote mitochondrial biosynthesis. Targeting energy metabolism is a potential strategy for the prevention and treatment of various diseases, such as cardiac and neurological disorders. Ginsenosides, one of the major bioactive constituents of Panax ginseng, have been extensively used due to their diverse beneficial effects on healthy subjects and patients with different diseases. However, the underlying molecular mechanisms of total ginsenosides (GS) on energy metabolism remain unclear. Methods: In this study, oxygen consumption rate, ATP production, mitochondrial biosynthesis, glucose metabolism, and SIRT1-PGC-1α pathways in untreated and GS-treated different cells, fly, and mouse models were investigated. Results: GS pretreatment enhanced mitochondrial respiration capacity and ATP production in aerobic respiration-dominated cardiomyocytes and neurons, and promoted tricarboxylic acid metabolism in cardiomyocytes. Moreover, GS clearly enhanced NAD+-dependent SIRT1 activation to increase mitochondrial biosynthesis in cardiomyocytes and neurons, which was completely abrogated by nicotinamide. Importantly, ginsenoside monomers, such as Rg1, Re, Rf, Rb1, Rc, Rh1, Rb2, and Rb3, were found to activate SIRT1 and promote energy metabolism. Conclusion: This study may provide new insights into the extensive application of ginseng for cardiac and neurological protection in healthy subjects and patients.

  • 소난자 체외성숙에 있어 소 태아혈청 (fetal bovine serum: FBS) 첨가시 높은 성숙율을 보이며 핵 이식 후 배 발달율에 있어서도 높다. 하지만 배양액 내에 포함된 소 태아혈청은 수정란 이식 후 태아의 발육 중 및 태어난 직후에 발생되는 송아지에서 나타나는 몇몇 비정상적인 현상들의 원인인 것으로 보고되고 있다 (Walker 등, 1998). 본 실험은 성숙배양시 소 태아혈청과 이의 대체물질로서 BSA 또는 PVA 와 같은 macromolecules의 첨가가 소 난자의 성숙과 핵 이식 후 배 발달에 미치는 효과를 알아보기 위하여 실시하였다.   • 면역학적인 방법을 사용하여 가돈 여포액내의 특이단백질의 존재여부와 이들을 추출하여 난자성숙에 미치는 영향을 조사한 결과 다음과 같았다. 1. 가돈 혈청 및 혈장에는 존재하지 않는 면역학적으로 특이한 두 단백질이 가돈 여포액내에서 검출되었다. 2. 이들 두 단백질은 전기영동적으로 각각 fast alpha-I globulin 과 beta-globulin 의 이동성을 나타냈다. 3. 이들 두 단백질은 생쥐 여포난자의 성숙을 억제시켰다. 본 실험의 결과로 미루어 가돈 여포액내에는 난자성숙을 억제시키는 특이단백질이 존재하는 것으로 사료된다.

  • This study was performed to examine the effects of various macromolecules in in vitro growth (IVG) media on the growth, maturation, and parthenogenesis (PA) of pig oocytes derived from small antral follicles (SAF). Immature oocytes were cultured for two days in IVG medium supplemented with 10% (v/v) fetal bovine serum (FBS), 10% (v/v) pig follicular fluid (PFF), 0.4% (w/v) bovine serum albumin (BSA), or 0.1% (w/v) polyvinyl alcohol (PVA) and then maintained for 44 h for maturation. After IVG, the mean diameters of the SAF treated with FBS, PVA, and no IVG-MAF ($113.0-114.8{\mu}m$) were significantly larger than that of no IVG-SAF ($111.8{\mu}m$). The proportion of metaphase II oocytes was higher in PFF (73.6%) than in BSA (43.5%) and PVA (53.7%) but similar to that in the FBS treatment (61.5%). FBS and PFF increased cumulus expansion significantly compared to PVA and BSA while the intraoocyte glutathione content was not influenced by the macromolecules. Blastocyst formation of PA oocytes treated with FBS (51.8%), PFF (50.4%), and PVA (45.2%) was significantly higher than that of the BSA-treated oocytes (20.6%). These results show that the PFF and FBS treatments during IVG improved the growth, maturation, and embryonic development of SAF.
