The corazonin (Crz) gene showed two subtypes of different length at laval and pupal stage. The long subtype fade out in adult central nerve system (CNS) but the short one survive through all the life cycle from larva to adult. The short subtype has the same base sequences with mature Crz mRNA and detected in both brain and ventral nerve cord (VNC). The long one, on the contrary, was detected only in the brain tissue. As observed in above results, Crz neurons develop in different pattern in the CNS of scuttle fly and the Crz gene expresses two different subtypes. These results suggest that this neurotransmitter may perform differential neurophysiological functions in the scuttle fly. Variation in the amino acid composition of the final active undecapeptide supports in strong those possibilities. We expect further studies on the relationship between neurophysiological functions of Crz and behavioral characteristics of the scuttle fly.
ACP (annealing control primer)를 사용하여 DDRT (differential display reverse transcription)-PCR 방법으로 볼락의 성장단계에 따라 발현량 차이를 나타내는 DEG (differentially expressed gene)를 확보하였다. ACP 120개를 분석하여 18개월령 근육조직에서보다 6개월령 근육조직에서 발현량이 많은 DEG 16개와 6개월령 근육조직에서보다 18개월령 근육조직에서 발현량이 더 많은 DEG22개의 염기서열을 분석하였다. DEG 염기서열을 BLAST 검색한 결과, parvalbumin (PVALB) 등 18개의 유전자(PVALB, NDKB, TPM, TnI, GAPDH, CKM2, factor 2 SERF2, AMPD, TRICA, ARHGAP15, ESD, hsp70, COL1A2, GST, Midllip1, MYL1, SERCA1B, FTH1)와 69~95%의 상동성을 나타냈다. Real time PCR 분석법으로 6개월령 근육조직에서 발현량이 많은 DEG14와 PVALB 유전자의 성장단계별 발현양상을 조사한 결과, 볼락이 성장함에 따라 발현량이 감소하였으며, 특히 PVALB 유전자는 6개월령 이후에는 발현량이 극히 적었다. 6개월령 근육조직에서보다 18 개월령 근육조직에서 발현량에서 많았던 CKM2 유전자는 성장함에 따라 발현량이 계속 증가하였고, 4세 이후에는 발현량이 감소하였다. DEG의 조직특이적 발현양상을 분석한 결과, DEG14는 근육, 간, 신장, 및 비장조직에서 발현되었으며, PVALB 유전자는 근육과 신장조직에서 발현되었고, 간과 비장조직에서는 발현되지 않았다. CKM2 유전자는 근육, 신장 및 비장조직에서 발현되었고, 간 조직에서는 발현되지 않았다. PVALB 유전자의 mRNA 크기는 659 bp 이며, 110개의 아미노산으로 구성되어 있다. Parvalbumin과 CKM2 유전자는 성장속도가 빠른 어류 선발에 이용할 수 있는 분자마커 개발에 활용하고자한다.
We cloned and sequenced the open reading frame (ORF) cDNA encoding myostatin from the muscle of abalone (Haliotis discus hannai). The ORF cDNA of the abalone myostatin is 1134 bp and encoded 377 amino acid residues that were 60-96% homologous with the amino acids of other organism myostatins. In addition, the ORF contained a conserved proteolytic cleavage site (RXRR) and nine conserved cysteine residues in the C-terminus. Semi-quantitative RT-PCR revealed the presence of myostatin mRNA in various tissues. The strongest expression was observed in the mantle of female abalone, and the gills and heart of male abalone.

The ectomycorrhizal fungus Tricholoma matsutake grows symbiotically with Pinus densiflora. Phenylalanine ammonia-lyase (E.C. 4.3.1.24) catalyzes the conversion of L-phenylalanine to trans-cinnamic acid. The role of fungal phenylalanine ammonia-lyase, however, has not been clear until now. In this study, the gene encoding phenylalanine ammonia-lyase (PAL), which was isolated from T. matsutake, was cloned and characterized. The PAL gene (tmpal) consists of 2,160 nucleotides, coding for a polypeptide containing 719 amino acid residues. The deduced amino acid sequence of tmpal from T. matsutake shows high identity (70%) with that from Laccaria bicolor. Comparative analysis of the PAL genes among T. matsutake and other species of the class Agaricomycetes showed that both active sites and binding sites were significantly conserved among these genes. The transcriptional analysis of the PAL gene revealed a differential gene expression pattern depending on the developmental stages (mycelium, primordium, stipe, pileus, and gills) of T. matsutake. These results suggest that the PAL gene in T. matsutake plays an important role in multiple physiological functions.

OsTAT encodes a twin-arginine translocator (TAT) pathway signal protein. It contains a TRANS membrane domain and a chloroplast transit peptide. mRNA transcription profiling of OsTAT1 revealed that it is highly overexpressed in the leaves corroborating reports on its role in chloroplast. Moreover, its level of expression is more pronounced during earlier stages (germination, 3-leaf stage, and maximum tillering) of growth in rice. A lower disease progress curve of bacterial blight is evident in transgenic lines compared with the wild type, Dongjin indicating its involvement in immunity to Xoo. Expression pattern following infection of Xoo strain K2 depicts highest levels at 4 and 8 hour post-inoculation which implies crucial induction of resistance during early response. This study initially reports a new overview on the biological functions of plant's TAT pathway. Further molecular and genetic analyses are underway to provide detailed involvement of OsTAT in disease resistance.
본 연구에서는 아토피 피부염 동물 모델인 NC/Nga mice에 실크단백질 sericin과 fibroin을 식이 공급 후 피부의 세라마이드 함량 및 관련인자 발현 변화를 정상대조군인 BALB/c mice 및 아토피 피부염 대조군과 비교 분석하였다. 세라마이드 함량은 아토피 대조군인 CA군이 정상대조군 C군보다 현저히 낮았으나, 정제된 건조 분말을 별다른 처리 없이 그대로 실크단백질 sericin을 식이공급한 S군은 정상대조군인 C군 이상의 수준으로 높였으며, 실크단백질 fibroin(F군)은 세라마이드 함량을 정상대조군 수준이상으로 올려주지는 못하였다. 세라마이드 합성효소인 SPT의 mRNA 및 protein 발현은 아토피 대조군인 CA군은 정상대조군인 C군보다 높았으나, 실크단백질 공급군인 S군 및 F군은 모두 정상대조군 C군보다 현저히 낮았다. 반면, 세라마이드 분해효소인 ceramidase의 mRNA 및 protein 발현은 아토피 대조군인 CA군은 정상대조군인 C군보다 높았으나, 실크단백질을 식이섭취한 S군 및 F군에서는 정상대조군인 C군과 유사한 수준으로 CA군에 비해서는 유의적으로 낮았다. 결론적으로 실크단백질 sericin의 10주간 식이섭취는 아토피 피부염 동물모델 NC/Nga mice 표피의 세라마이드 함량을 정상대조군 수준 이상으로 증가시켰으며, 궁극적으로 표피의 장벽기능을 정상대조군의 수준으로 변화시켰다. 이는 serine이 다량 함유되어있는 실크단백질 sericin을 공급에 의해 증가되었는데, serine은 그 자체로서 보습기능을 하여 표피의 장벽기능을 보완하였기 때문에 SPT에 의한 생합성은 불필요하였으며 또한, ceramidase에 의한 세라마이드 분해를 정상수준이하로 억제하여 세라마이드의 생성보다는 분해억제에 의한 것으로 여겨진다.

Dendritic cells (DC) or macrophages possesses pattern recognition molecule (PRM) for binding $\beta$-glucan as pathogen-associated molecular pattern (PAMP). Recently $\beta$-glucan receptor was cloned from DC and named as dectin-l which belongs to type II C-type lectin family. Human dectin-1 is consisted of 7 exons and 6 introns. The polypeptide of dectin-1 has 247 amino acids and has cytoplasmic, transmembrane, stalk and carbohydrate recognition domains. Dectin-1 could recognize variety of beta-1,3 and/or beta-1,6 glucan linkages, but not alpha-glucans. In our macrophage cell line culture system, dectin-1 mRNA was detected in RA W264.7 cells by reverse transcription-polymerase chain reaction (RT-PCR). Dectin-1 was also detected in the murine organs of spleen, thymus, lung and intestines. Treatment of RA W264.7 cells with $\beta$-glucans of Ganoderma lucidum (GLG) resulted in increased expression of IL-6 and TNF-$\alpha$ in the presence of LPS. However, GLG alone did not increase IL-6 nor TNF-$\alpha$. These results suggest that receptor dectin-1 cooperate with CD14 to activate signal transduction that is very critical in immunoresponse.

IFIT1 (also known as ISG56) is a member of the interferon-inducible protein with tetratricopeptide repeats (IFITs) family. IFITs are strongly induced by type I interferon (IFN), double-stranded RNA and virus infection. Here, we investigated IFIT1 expression in human umbilical vein endothelial cells (HUVECs) and in human bronchus epithelial cells (BEAS-2Bs) induced by the H9N2 virus and inactivated viral particle at different time points. We also investigated the effect of H9N2 virus and viral particle infection on $IFN-{\alpha}/{\beta}$ production, and assessed whether hemagglutinin or neuraminidase protein induced IFIT1 expression. Results showed that both H9N2 virus infection and viral particle inoculation induced the expression of IFIT1 at mRNA and protein levels in the two cell lines. Hemagglutinin or neuraminidase protein binding alone is not sufficient to induce IFIT1 expression. Surprisingly, the expression patterns of IFIT1 in response to H9N2 virus and viral particles in the two cell lines were opposite, and production kinetics of $IFN-{\alpha}/{\beta}$ also differed. An additional finding was that induction of IFIT1 in response to H9N2 virus infection or viral particle inoculation was more sensitive in HUVECs than in BEAS-2Bs. Our data offers new insight into the innate immune response of endothelial cells to H9N2 virus infection.

Ski protein is implicated in proliferation/differentiation in a variety of cells. We had previously reported that Ski protein is present in granulosa cells of atretic follicles, but not in preovulatory follicles, suggesting that Ski has a role in apoptosis of granulosa cells. The alternative fate of granulosa cells other than apoptosis is to differentiate to luteal cells, however, it is unknown whether Ski is expressed and has a role in granulosa cells undergoing luteinization. Thus, the aim of the present study was to locate Ski protein in the rat ovary during luteinization to predict the possible role of Ski. In order to examine the expression pattern of Ski protein along with the progress of luteinization, follicular growth was induced by administration of equine chorionic gonadotropin to immature female rat, and luteinization was induced by human chorionic gonadotropin treatment to mimic luteinizing hormone (LH) surge. While no Ski-positive granulosa cells were present in preovulatory follicle, Ski protein expression was induced in response to LH surge, and was maintained after the formation of corpus luteum (CL). Though Ski protein is absent in granulosa cells of preovulatory follicle, its mRNA (c-ski) was expressed and the level was unchanged even after LH surge. Taken together, these results demonstrated that Ski protein expression is induced in granulosa cells upon luteinization, and suggested that its expression is regulated post-transcriptionally.
