In this study, the physicochemical and microbiological properties of market-available skate (Raja kenojei) kimchi were evaluated in comparison to long-term fermented Baechu kimchi. The comparative results for the skate (Raja kenojei) and Baechu kimchi products are as follows: salt concentrations = 3.40% and 3.95%, respectively; pH and acidity values = 4.01 and 3.90, and 0.65% and 0.82%, respectively; redox potential Eh values = 119.82 mV and 123.08 mV respectively; reducing sugars levels = 15.51 mg% and 13.23mg%, respectively; ascorbic acid levels = 24.21mg% and 22.18mg%, respectively; color L-values = 46.86 and 44.54, a-values = 15.46 and 12.28, and b-values = 29.94 and 28.36, respectively; hardness properties = 11.36 kgf and 10.23 kgf, respectively; hot water soluble pectin (HWSP) contents and sodium hexametaphosphate soluble pectin (NaSP) contents = 15.23% and 17.35%, and 32.51% and 29.64%, respectively; hydrochloric acid soluble pectin (HClSP) contents = 55.1% and 53.2%, respectively; total viable cell counts = $4.78{\times}10^8$ and $1.35{\times}10^8$, respectively; lactic acid bacteria counts = $5.18{\times}10^5$ and $1.32{\times}10^5$, respectively; and yeast levels = $8.52{\times}10^5$ and $5.23{\times}10^5$, respectively.
It is in its infancy to use bacteria as a novel biotechnology for leaching precious and heavy metals from raw materials. The objective of this study was to investigate biogeochemical processes of iron leaching from magnetite reduction by iron-reducing bacteria isolated from intertidal flat sediments, southwestern part of Korea. Microbial leaching experiments were performed using commercial magnetite, Aldrich magnetite, in well-defined mediums with and without bacteria. Water soluble Fe production was determined by ICP analysis of bioleached samples in comparison to uninoculated controls, and the resulting precipitated solids were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The extent of iron leaching from magnetite in the aerobic conditions (Fe = 107 ppm) was higher than that in the anaerobic environments (Fe = 94 ppm). In the anaerobic conditions, Fe(III) in commercial magnetite was also reduced to Fe(II), but no secondary mineral phases were observed. Amorphous iron oxides formed in the medium under aerobic conditions where there was sufficient supply of oxygen from the atmosphere. SEM observation suggests that the reduction process involves dissolution-precipitation mechanisms as opposed to solid state conversion of magnetite to amorphous iron oxides. The ability of bacteria to leach soluble iron and precipitate amorphous iron oxides from crystalline magnetite could have significant implications for biogeochemical processes in sediments where Fe(III) in magnetite plays an important role in the largest pool of electron acceptor as well as the tool as a novel biotechnology for leaching precious and heavy metals from raw materials.
To develop an effective use for poor-quality individually quick-frozen (IQF) oysters Crassostrea gigas stored for a long period, the extract conditions, quality characteristics, and optimum reaction flavoring (RF) conditions of a complex extract from these IQF oysters were investigated. The moisture, pH, and volatile basic nitrogen contents of IQF oysters stored for 18 months (18M-IQFO) were 77.9%, 6.32, and 17.9 mg/100 g, respectively. Three different kinds of extract were prepared from 18M-IQFO: a hot-water extract (HE), scrap enzymatic hydrolysate (EH), and complex extract (CE). The respective extracts contained 5.5, 8.6, and 6.6% crude protein and 281.7, 366.0, and 343.0 mg/100 g amino nitrogen, and had 811, 359, and 1,170 mL/kg extraction yields. The CE was superior to the traditional HE in terms of the extraction yield, amino-nitrogen content, and organoleptic qualities, except for the odor. To improve flavor via the Maillard reaction, the reaction system used to produce a desirable flavor comprised CE (Brix $30^{\circ}$), 0.4 M glucose, 0.4 M glycine, and 0.4 M cysteine solution (4:2:1:1, v/v). The reaction time and pH were the independent variables, and the sensory scores for baked potato odor, masking shellfish odor, and boiled meat odor were the dependent variables. The surface response methodology (RSM) analysis of the multiple responses optimization gave a reaction time of 120.6 minutes and pH 7.33 at $120^{\circ}C$. The reaction improved the flavor of CE considerably, as compared to that of the unreacted extract.
To substitute fromzed and freeze-dried food for live food in the development of the artificial seedling production of bivalve larvae, the dietary value of live T. suecica was compared with those of freeze-dried T. suecica, frozed T. suecica, live T. suecica (50%)+frozen T. suecica (50%), and live T. suecica (50%)+freeze-dried T. suecica (50%) in the rearing of oyster (Crassotrea gigas) and hen clam (Mactra chinensis) larvae, and manila clam (Tapes philipninarum) spats. Oyster larvae fed live T. suecica showed the highest growth (shell hight $231.9^{\mu}$m) and survival rate (72.6%) and those fed freeze-dried T. suecica showed the lowest growth (shell height $168.9^{\mu}$m) and survival rate (35.3%). However, in the hen clam larvae, there were not significantly different among diet group in growth and survival rate. The small spats of manila clam fed live t. suecica or live T. suecica (50%)+freeze-dried T. suecica (50%) showed higher growth and survival rate than those fed other diet group. In the case of large spats of manila clam, live T. suecica and live T. suecica (50%)+frozen T. suecica (50%) showed better growht. But, survival rates were not different among diet groups. Dietary valuse of frozen and dried T. suecica were different on species and growth stage, and frozen and freeze-dried T. suecica can be partially used as substitute food for T. suecica live T. succica in shellfish hatchery.
Fully cholesteryloxycarbonated and (8-cholesteryloxycarbonyl) heptanoated disaccharide derivatives were synthesized by reacting cellobiose, maltose, and lactose with cholesteryl chloroformate or 8- cholesteryloxycarbonylheptanoyl chloride, and their thermotropic liquid crystalline properties were investigated. All the cholesteryloxycarbonated derivatives (CH1DSs) formed enantiotropic cholesteric phases, whereas all the (8-cholesteryloxycarbonyl) heptanoated derivatives (CH8DSs) exhibited monotropic cholesteric phases with left-handed helicoidal structures whose optical pitches (${\lambda}m's$) decrease with increasing temperature. All the CH1DSs, contrast with the CH8DSs, did not display reflection colors over the full cholesteric range, suggesting that the helicoidal twisting power of the cholesteryl group highly depends on the length of the spacer joining the cholesteryl group to the disaccharide chain. The thermal stability and degree of order in the mesophase and the temperature dependence of the ${\lambda}m$ observed for EH8DSs were entirely different from those reported for the cholesterol-bearing dimers and triplet and the (8-cholesteryloxycarbonyl) heptanoated polysaccharide derivatives. The results were discussed in terms of the difference in the number of the mesogenic units per mole of repeating unit and the flexibility of the main chain.
All the radionuclides in high-level nuclear waste will decay to harmless levels eventually but for some radionuclides decay is so slow that their radiation remains dangerous for times on the order of tens or hundreds of thousands of years. At the present time, the most favorite disposal plan for high-level radioactive waste is a mined geological disposal in which canister enclosing stable solid form of radioactive waste is placed in mined cavities locating hundred meters below the surface. The chief hazard in such disposal is dissolution of radionuclides from the waste in the groundwater that will eventually carry the dissolved radionuclides to surface environments. The hazard from possible escape of the radionuclides through groundwater can be delayed by engineered and geologic barriers. The engineered barriers can become useless by unexpected geologic catastrophe such as volcanism, earthquake, and tectonic movement and by fraudulent work such as careless construction, improperly welded canisters within the first few decades or centuries. As a result, dangerously radioactive waste which is still intensively radioactive is directly exposed to attack by moving groundwater. All the more, it is almost impossible to control repositories for times more than 10,000 years. Therefore, naturally controlled geologic, barriers whose properties will not be changed within 10,000 years are important to guarantee the safety of repositories of high-level radioactive waste. In Sweden and France, the suitability of granite for the mined geological disposal of high-level waste has been studied intensively. According to the research in Sweden and France, granites has the following physio-chemical characteristics which can delay the transportation of radionuclide by groundwater. First, the permeabilities of granites decreases as the depth increases and is $10^{-8}{\sim}10^{-12}m/s$ at depth below 300 m. Second, groundwater at depth below 300 m has pH=7-9 and reducing condition (Eh=-0.1~0.4). This geochemical condition is desirable to prevent both canister and solid waste from corrosion. Third most radionuclides are not transported by low solubilities and some radionuclide with high solubility such as Cs and Sr are retarded by absorption of geologic media through which ground water flows. Therefore, if high-level waste is disposed at depth below 300 m in the granite body which has a low permeability and is geologically stable more than 10,000 years, the safety of repositories from the hazard due to radionuclide escape can guaranteed for more than 10,000 years.
Major ions and nitrogen isotopic analyses were performed to determine the geochemical characters and to identify the source of nitrate of the shallow groundwater around agricultural field in the Youngdong area. The pH value of groundwater ranges from 60. to 8.2 (pH 7.2, mean). The average of EC, Eh and DO is 369 ${\mu}S/cm$ (70~729 ${\mu}S/cm$), 165.6 mV (29~383.2 mV), 4.3 mg/L (1.8~8.0 mg/L) respectively. The ion concentraion of groundwater was in the order of $Ca^{2+}$>$Na^{2+}$>$Mg^{2+}$>$K^{2+}$ and ${HCO_3}^-$>${NO_3}^-$>${SO_4}^{2-}$>$Cl^-$>$F^-$. Most of groundwater is Ca-$HCO_3$ type. The groundwater was affected by water-rock interaction in the shallow depth. Some groundwater is Ca-Cl or Na-$HCO_3$ (2.5%) type that was due to agricultural activities. The $NO_3$_N concetration of grondwater range from 10.2 mg/l to 26.9 mg/l, which show that this area is under nitrate pollution. ${\delta}^{15}N-NO_3$ value of the groundwater is the origins of are a combination of animal wastes and man-made fertilizers.
The extracellular endoinulase from Streptomyces sp. 556 was purified and characterized, The culture broth was fractionated by ammonium sulfate saturation followed by DEAE-cellulose column chromatography and 5ephadex G-200 gel filtration, The ultimately purified fraction revealed a single band in 7.5% polyacrylamide gel electropherogram. The purified enzyme showed the maximal activity at pH 5.5-6.0 and $50^{\circ}C$, but lost 93% of inulase activity after 30 min incubation at $55^{\circ}C$ . The essen.tial amino acid residue for catalytic activity appeared to be tryptophan. This endo inulase was activated by $Mn^{2+}$, whereas inactivated by $Ag^{+}$, $Hg^{+}$, $Cu^{2+}$, $Zn^{2+}$, $Fe^{3+}$ and $Mo^{6+}$ EDTA and 8-hydroxyquinoline inhibited the enzyme so that the enzyme was considered to be a metalloenzyme. The Km value for inulin was 0.287 mM, and no invertase or $\alpha$-glucosidase activity was found in the enzyme.

The formation of brown-colored precipitates is one of the serious problems frequently encountered in the development and supply of groundwater in Korea, because by it the water exceeds the drinking water standard in terms of color. taste. turbidity and dissolved iron concentration and of often results in scaling problem within the water supplying system. In groundwaters from the Pajoo area, brown precipitates are typically formed in a few hours after pumping-out. In this paper we examine the process of the brown precipitates' formation using the equilibrium thermodynamic and kinetic approaches, in order to understand the origin and geochemical pathway of the generation of turbidity in groundwater. The results of this study are used to suggest not only the proper pumping technique to minimize the formation of precipitates but also the optimal design of water treatment methods to improve the water quality. The bed-rock groundwater in the Pajoo area belongs to the Ca-$HCO_3$type that was evolved through water/rock (gneiss) interaction. Based on SEM-EDS and XRD analyses, the precipitates are identified as an amorphous, Fe-bearing oxides or hydroxides. By the use of multi-step filtration with pore sizes of 6, 4, 1, 0.45 and 0.2 $\mu\textrm{m}$, the precipitates mostly fall in the colloidal size (1 to 0.45 $\mu\textrm{m}$) but are concentrated (about 81%) in the range of 1 to 6 $\mu\textrm{m}$in teams of mass (weight) distribution. Large amounts of dissolved iron were possibly originated from dissolution of clinochlore in cataclasite which contains high amounts of Fe (up to 3 wt.%). The calculation of saturation index (using a computer code PHREEQC), as well as the examination of pH-Eh stability relations, also indicate that the final precipitates are Fe-oxy-hydroxide that is formed by the change of water chemistry (mainly, oxidation) due to the exposure to oxygen during the pumping-out of Fe(II)-bearing, reduced groundwater. After pumping-out, the groundwater shows the progressive decreases of pH, DO and alkalinity with elapsed time. However, turbidity increases and then decreases with time. The decrease of dissolved Fe concentration as a function of elapsed time after pumping-out is expressed as a regression equation Fe(II)=10.l exp(-0.0009t). The oxidation reaction due to the influx of free oxygen during the pumping and storage of groundwater results in the formation of brown precipitates, which is dependent on time, $Po_2$and pH. In order to obtain drinkable water quality, therefore, the precipitates should be removed by filtering after the stepwise storage and aeration in tanks with sufficient volume for sufficient time. Particle size distribution data also suggest that step-wise filtration would be cost-effective. To minimize the scaling within wells, the continued (if possible) pumping within the optimum pumping rate is recommended because this technique will be most effective for minimizing the mixing between deep Fe(II)-rich water and shallow $O_2$-rich water. The simultaneous pumping of shallow $O_2$-rich water in different wells is also recommended.
High-ester pectin was demethylated by the treatments of HCl alone and a combination of HCl and $NH_4OH$. The low-ester pectin prepared were analyzed for chemical composition and the pectin gels were evaluated for firmness by sag values and strength by puncture stress. Gels made from HCl demethylated sample showed brittle, weak and poor elastic characteristics while the $HCl-NH_4OH$ treated samples generally resulted in a smooth and elastic gels except those samples having very low content of ester group or acid amide group. Statistical analysis showed that significant correlations were found between sag values and ester content or molecular weight, and puncture stress and ester content, acid amide groups or molecular weight. The equations derived for sag, puncture stress and sag/puncture stress from chemical data could be useful for prediction of some of the physical properties of low-ester pectin gel.
