Effects of Fermented Turmeric Extracts on the Obesity in Rats Fed a High-Fat Diet (강황 발효액이 고지방 섭취 흰쥐의 비만에 미치는 효과)

  • This study was designed to investigate the effects of fermented turmeric extract on body weight, abdominal fat mass and biochemical markers related to obesity in rats fed high fat diet. Turmeric and brown sugar was equally mixed and fermented for one year. Wistar rats (weighing about 125 g (15 rats)) were divided into three groups: high fat diet (20.9%) and brown sugar 7.2% (HFD), turmeric powder in replace of corn starch 12.8%, brown sugar 7.2% (TP), fermented turmeric powder 20% (FTP) for four weeks. The final body weight was about 425g. The retroperitoneal fat weights in TP group showed the tendency to decrease regardless of feed intake in the rats. Serum levels of total cholesterol and LDL-cholesterol in the FTP group showed the lowering tendency than those of the HFD group (p<0.05). Serum levels of leptin, adiponectin and aspartate aminotransferase (AST) in the FTP group was significantly (p<0.05) lower than that of the HFD group, and serum level of tumor necrosis factor-$\alpha$ (TNF-$\alpha$) in all the groups were similar. These results suggest that fermented turmeric extract might be effective to prevent obesity in rats fed high fat diet.

Effects of Zeolite Levels on the Performance of Broilers (Zeolite의 첨가수준이 육계의 생산성에 미치는 영향)

  • This experiment was carried out to investigate the effects of Zeolite levels on the performance of broilers. 360 starting chicken of Arbor Acre were selected for this study. Chickens were fed 4 levels (0.2.4 and 6%) of Zeolite in the basal ration for 7 weeks. The results obtained were as follows: 1. The body weight gain was highest in the 2% supplement of Zeolite, but more than 4% supplement resulted in lower body weight gain (P<0.05). 2. No significant differences were found in the feed intake among the treatments. 3. Feed conversion had no significant differences, among treatments but 6% Zeolite group was slightly higher than the others. 4. The viabilities were not significantly differences among the experimental groups. 5. Moisture in excreta was tend to be decreased significantly as Zeolite levels were increased (p<0.01) 6. Carcass yield and abdominal fat deposition were not different significantly over all experimental groups. 7. Highest income was attained by 2% supplement of Zeolite, but 6% group was reduced significantly (P<0.05).

Effects of Dietary Supplementation of Pine Needle Powder on Carcass Characteristics and Blood Cholesterol Contents of Broiler Chicken (사료내솔잎분말첨가급여가육계의도체특성및혈액콜레스테롤함량에미치는영향)

  • This study was conducted to investigate the effects of supplementation diets of pine needle powder on performance, proximate composition, carcass characteristics, blood cholesterol concentration, TBARS (thiobarbituric acid reactive substance), WHC (water holding capacity), and shear force of chicken meat. Broiler chicks were fed diets for five weeks containing 0% pine needle powder (Control), 0.3% pine needle powder (T1), 0.6% pine needle powder (T2), and 0.9% pine needle powder (T3). Performance was no significant difference among treatment groups, but mortality was decreased in diets by the supplementation of pine needle powder than that of T1 and control. The moisture, crude protein, crude fat, and crude ash were no significant difference. The liver weight and abdominal fat was significantly decreased by the supplementation of pine needle powder compare to the T1 and control (P<0.05). The total cholesterol and LDL-cholesterol concentration of control was higher than treatment groups, and the HDL-cholesterol concentration and glucose of control was lower than treatment groups (P<0.05). The TBARS value was significantly decreased by the supplementation of pine needle compared to the control (P<0.05). In conclusion, these data indicate that 0.6% pine needle powder supplementation (T2) was most effective in decreasing mortality, total cholesterol and LDL-cholesterol and increasing HDL-cholesterol concentration.

Effects of Dietary Supplementation of Rare Earth and Chlortetracycline on the Performance and Intestinal Microflora in Broiler Chicks (희토와 클로르테트라시이클인이 육계의 생산성 및 장내 미생물에 미치는 영향)

  • Rare earth(RE) is a common name indicating 17 of specific elements including 15 of lanthanides, scandium (Sc), and yttrium(Y). This study was conducted to compare the effect of RE to growth stimulating antibiotic products in broiler diet. One of the products commonly used in broiler feeding, Chlorotetracycline(CTC) was used at a level of 200ppm for the antibiotic treatment and replaced by 100ppm RE for the RE treatment. Dietary addition of RE and CTC showed similar effects in improving broiler growth and feed conversion each other, and those were significantly better when compared to those of the control group(P<0.05). Abdominal fats of chicks fed RE were significantly lower than others(P<0.05). Intestinal E. coli and total microbial numbers were reduced by RE addition(P<0.05), but the number of Lactobacillus was not affected. The number of Salmonella was also decreased but it was not significant.

Inflammation Scan Using $^{99m}Tc-HMPAO$ Labelled Leukocytes ($^{99m}Tc-HMPAO$를 이용한 자가백혈구표지 및 그를 이용한 염증병소의 스캔)

  • Inflammation scan using radiolabelled leukocytes has high sensitivity and specificity. Several methods for labelling leukocytes have been evaluated using P-32 diisopropyl fluorophosphate (DFP-32), H-3 thymidine, Cr-51 chromate, Ga-67 citrate and Tc-99m-sulfur colloid. In-111-oxine has proved so far to be the most reliable agent for labelling leukocytes. In-111-oxine is, however, expensive, not easily available when needed, and its radiation dose to leukocytes is relatively high. Moreover, resolution of the resultant image is relatively poor. Tc-99m is still the agent of choice because of, as compared with the indium, its favorable physical characteristics, lower cost and availability. Now the technique for labelling the leukocytes with technetium is successfully obtained using the lipophilic HAPAO with higher efficiency for granulocytes than for other cells. With this technique it is possible to label leukocytes in plasma to improve the viability of the leukocytes. Inflammation scan using Tc-99m-HMPAO has been evaluated in several laboratories, and difference in methods for separation and labelling accounts for difference in efficiency, viability and biodistribution of the labelled leukocytes. We performed inflammation scan using leukocytes labelled with Tc-99m-HMPAO in three dogs 24 hours after inoculation of live E. Coli and A. Aureus in their right abdominal wall. We separated mixed leukocytes by simple sedimentation using 6% hetastarch (HES) and labelled the leukocytes with Tc-99m-HMPAO in 20% cell free plama diluted with phosphate buffer solution(Fig. 1). Uptake was high in the liver and spleen but is was minimal in the lungs on whole body scan. Kidneys and intestine showed minimal activity although it was high in the urinary bladder(Fig. 2). Uptake of labelled leukocytes in the inflammation site was do(mite on 2 hour-postinjection scan and abscess was clearly delineated on 24 hour-delayed scan with high target-to-nontarget ratio(Fig. 3, 4). Inflammation scan using mixed leukocytes labelled with Tc-99m-HMPAO is very sensitive and specific in early detection of inflammation.

Effects of Dietary Supplementation of Mulberry Leaves Powder on Carcass Characteristics and Meat Color of Broiler Chicken (사료 내 뽕잎 분말 첨가 급여가 육계의 도체특성 및 계육의 육색에 미치는 영향)

  • This study was conducted in order to investigate the effects of supplementation diets of mulberry leaves powder on performance, carcass characteristics, blood characteristics, and meat color of chicken meat. Broiler chicks were fed diets containing 0% mulberry leaves powder (Control), 1% mulberry leaves powder (T1), 2% mulberry leaves powder (T2), and 3% mulberry leaves powder (T3) for five weeks. The weight gain, feed intake, and feed conversion were no significant difference among treatment groups. The carcass weight, carcass ratio, gizzard, cecum, heart, and abdominal fat weight were not significantly different. The liver weight significantly decreased by the supplementation of mulberry leaves powder compared to the control (p<0.05). The total cholesterol and LDL-cholesterol concentration of the control were higher than T3, but the HDL-cholesterol concentration, glucose, and albumin of the control were lower than those of the treatment groups (p<0.05). However, there were no significant difference in GPT and triglyceride concentration among treatments. In meat color, the CIE $L^*$ and $a^*$ value of treatment groups (T3) showed significantly higher value compared to the control, however, no difference in the CIE $b^*$ values was observed among treatment groups. In conclusion, these data indicate that 3% mulberry leaves powder supplementation was most effective in decreasing the total cholesterol and LDL-cholesterol as well as increasing the HDL-cholesterol, glucose, and albumin concentration.

Role of Reactive Oxygen Species in the Adipogenesis of Adipose-derived Stem Cells (지방유래줄기세포의 지방분화과정에서 활성산소가 미치는 영향)

  • Purpose: Stem cells continue to receive research attention in the clinical fields, and adipose-derived stem cells (ADSCs) have been shown to be a good source raw material. Many plastic surgeons are researching the ADSC adipogenesis with a view of conducting clinical trials, and many attempts have been made to identify the factors that promote the adipogenesis of ADSCs, but comparatively few correlation studies have been undertaken to explore the relation between reactive oxygen species (ROS) and the ADSC adipogenesis. We undertook this study is to investigate the effects of ROS on ADSC adipogenesis. Methods: ADSCs were isolated and cultured from abdominal adipose tissue, and cultured in different media; 1) DMEM(control), 2) adipogenesis induction culture medium, 3) adipogenesis induction culture medium with ROS ($20{\mu}M/50{\mu}M\;H_2O_2$), 4) adipogenesis induction culture medium containing ROS ($20{\mu}M/50{\mu}M\;H_2O_2$) and antioxidant ($10{\mu}M/20{\mu}M$ Deferoxamine). We compared adipogenesis in these different media by taking absorbance measurements after Oil-Red O staining every 5 days. Results: After culturing for 20 days, significant differences were observed between these various culture groups. Absorbance results showed significantly more adipogenesis had occurred in media containing adipogenesis induction culture medium and $H_2O_2$ (in a $H_2O_2$ dose-dependently manner) than in media containing adipogenesis induction culture medium and no $H_2O_2$ (p<0.001). Furthermore, in media containing adipogenesis induction culture medium, $H_2O_2$, and antioxidant, absorbance results were significantly lower than in adipogenesis induction culture medium and $H_2O_2$ (p<0.001). Conclusion: These findings suggest that ROS promote the adipogenesis of ADSCs. We suggests that ROS could be used in the adipose tissue engineering to improve fat cell differentiation and implantable fat tissue organization.

Surgical Experience of the Remnant Thoracoabdominal Aortic Replacement after Aortic Surgery (대동맥수술 후 잔존 흉복부대동맥치환술에 대한 임상 경험)

  • Background: Aortic diseases tend to involve the entire aorta. Hence, there is the constant possibility of the need for a secondary operation at the remnant aorta. This study analyzed our cases of secondary aortic surgery in order to determine its characteristics and problems. Material and Method: Between April 2003 and June 2007, 12 patients (6 male and 6 female) underwent thoracoabdominal aortic replacement as a secondary aortic operation. Their clinical courses were analyzed. Four of the patients underwent lower thoracobadominal aortic replacement under the normothermic femorofemoral bypass, and the others underwent an entire thoracobdominal aortic replacement under deep hypothermic circulatory arrest. Result: There was no death or paraplegia. As local complications, there were 3 cases of wound infection and 2 cases of an immediate reoperation caused by bleeding and one case of delayed wound. revision for a contaminated perigraft hematoma. As a systemic complication, there was one case of renal insufficiency, which required hemodialysis and one case of respiratory insufficiency that needed prolonged ventilator care. The mean admission period was $30{\pm}21$ days. All the patients were followed up for $626{\pm}542$ days without reoperation or other problems. Conclusion: Using properly selected patients and a careful approach, thoracoabdominal aortic replacement can be performed safely as a secondary aortic surgery.

Effects of DMTU, SOD and Ischemic Preconditioning on the Ultrastructural Changes of the Rectus Femoris Muscles in Rats after Ischemia and Reperfusion (SOD, DMTU및 허혈양상화 처치가 허혈 및 재관류에 의한 흰쥐 넙다리곧은근의 미세구조 변화에 미치는 영향)

  • The ischemia and reperfusion injury of the skeletal muscles is caused by generation of reactive oxygen during ischemia and reperfusion. It is well known that over 4 hours of ischemia injures the skeletal muscles irreversibly. The author has demonstrated the effects of SOD (superoxide dismutase), DMTU (dimethyl thiourea) and ischemic preconditioning on ultrastructural changes of the muscle fibers in the rectus femoris muscles after 4 hours of ischemia and 1 day and 3 days of reperfusion. A total of 72 healthy Sprague-Dawley rats weighing from 200 gm to 250 gm were used as experimental animals. Under urethane(1.15 g/kg, IP, 2 times) anesthesia, lower abdominal incision was done and the left common iliac artery was occluded by using vascular clamp for 4 hours. The left rectus femoris muscles were obtained at 1 and 3 days after the removal of vascular clamp. The SOD (15,000 unit/kg) or DMTU (500 mg/kg) were administered intraperitoneally at 1 hour before induction of ischemia. The ischemic preconditioned group underwent three episodes of 5 minutes occlusion and 5 minutes reperfusion followed by 4 hours of ischemia and 1 day and 3 days of reperfusion. The specimens were sliced into $1mm^3$ and prepared by routine methods for electron microscopic observation. All specimens were stained with uranyl acetate and lead citrate and then observed with Hitachi-600 transmission electron microscope. The results were as follows: 1. SOD or DMTU alone did not affect the ultrastructure of muscle fibers in the rectus femoris muscles. The electron density of mitochondrial matrix was decreased by ischemic preconditioning. 2. Dilated cisternae of sarcoplasmic reticulum, triad, mitochondria and the loss of myofilament in the sarcomere were observed in the 4 hours ischemia and 1 day reperfused rectus femoris muscles. Markedly changed sarcoplasmic reticulum, triad, disordered or loss of myofilament, indistinct A-band and I-band, and irregular electron lucent M -line and Z-line are seen in the 4 hours ischemia and 3 days reperfused rectus femoris muscles. 3. SOD reduced the changes of organelles in the muscle fibers of the 4 hours ischemia and 1 day reperfused rectus femoris muscles of the rats, but SOD did not affect the changes of muscle fibers in the 4 hours ischemia and 3 days reperfused muscles. On the other hand, DMTU markedly attenuated considerably the ultrastructural change of the 4 hours ischemia and 1 day or 3 days reperfused rectus femoris muscles. 4. By the ischemic preconditioning, the change was attenuated remarkably in the 4 hours ischemia and 1 day reperfused rectus femoris muscles. As the ischemic reperfused changes of muscle fibers were regenerated or recovered by ischemic preconditioning, the ultrastructures of them were similar to those of normal control in the 4 hours ischemia and 3 days reperfused rectus formoris muscles. Consequently, it is suggested that DMTU is stronger inhibitor to ischemic reperfused change than SOD. The ischemia and reperfusion-induced muscular damage is remarkably inhibited by ischemic preconditioning.

Association between the Components of Metabolic Syndrome and Total Bilirubin at the Health Care Check Up in Korean Men and Women: Based on Adults Except for Diabetes and Hypertension (당뇨와 고혈압 환자를 제외한 건강검진을 실시한 한국 성인남녀의 대사증후군 구성요소와 총 빌리루빈간의 관련성)

  • Bilirubin has an anti-inflammatory effect as an endogenous antioxidant and has been reported to be inversely correlated with hypertension, diabetes, metabolic syndrome, and insulin resistance. On the other hand, there is a lack of research evaluating the association between bilirubin and metabolic syndrome compared to liver-specific indicators. This study examined the association between the total bilirubin and metabolic syndrome prevalence in Korean adults who underwent health screening compared to ALT and GGT. This cross sectional study included 22,568 adult males and females aged 20 years or older who underwent a health examination at the General Hospital of Gyeonggi Province from March 2015 to February 2018. Metabolic syndrome was diagnosed based on the American Heart Association/National Heart, Lung, and Blood Institute (AHA/NHLBI) guidelines. In both genders, the metabolic syndrome diagnosed group had higher liver enzymes, and the total bilirubin and direct bilirubin were lower than those without the metabolic syndrome risk factors. The serum total bilirubin showed a weak inverse correlation with abdominal obesity (r=-0.066 vs r=-0.055) and triglyceride (r=-0.127 vs r=-0.136) in both males and females, which is weaker than ALT and GGT The data were statistically significant. In addition, total bilirubin as well as ALT and GGT in both men and women were not predictors of the likelihood of developing metabolic syndrome.
