The effect of liquid cultures of Cordyceps militaris (LCM) on weight gain, food intakes, food efficiency ratios, serum and hepatic lipids, fecal lipids excretion, serum protein and enzyme activities, were investigated in adult female rats (30 weeks old). Sprague-Dawley rats were assigned to one normal and four hyperlipidemic diet groups, Hyperlipidemic diet groups (20% fat, 1% cholesterol) were divided into high fat diet (LCM free water), 10%, 20% or 30% LCM diet groups (10%, 20% or 30% LCM in water) according to the levels of LCM supplementation. After 35 days of experimental diet consumption, the body weight gains, hepatic weights, and food efficiency ratios of the rats fed hyperlipidemic diets were significantly increased compared with those of the rats fed normal diet. The concentrations of serum and hepatic triglycerides, hepatic total lipid, and atherogenic index of the rats fed 20% or 30% LCM diets were significantly lower than those of the rats fed the high fat diet. The concentration of serum HDL-cholesterol of the rats fed all LCM diets was significantly higher than those of the rats fed the high fat diet. The fecal excretion of triglyceride in the rats fed 20% or 30% LCM diets was significantly higher than those of the rats fed high fat diet. The concentrations of serum and hepatic total cholesterol, serum LDL-cholesterol, and HDL-cholesterol/total cholesterol ratio, fecal excretion of cholesterol, and the activities of serum glutamic pyruvic transaminase and alkaline phosphatase of the rats fed all LCM diets were similar to those of the rats fed high fat diet. No differences were noted in the weights of kidney and femur, the serum concentration of glucose, total protein and albumin, and the activities of glutamic oxaloacetic transaminase and ${\gamma}$ -glutamyltranspeptidase, among the rats on all the experimental diets. These results showed that the 20% or 30% LCM diets feeding decreased the serum and hepatic triglycerides, and the atherogenic index, and increased the serum HDL-cholesterol of the adult female rats.
This study was conducted to evaluate effects of halogenated compounds on in vitro rumen fermentation characteristics and methane emissions. A fistulated Holstein cow of 650 kg body weight was used as a donor of rumen fluid. Five kinds of halogenated compounds (bromochloromethane (BCM), 2-bromoethane sulfonic acid (BES), 3-bromopropanesulfonic acid (BPS), chloroform (CLF), and pyromellitic diimide (PMDI) known to inhibit methyl-coenzyme M reductase activity were added to an in vitro fermentation incubated with rumen fluid. The microbial population including bacteria, protozoa, and fungi were enumerated, and gas production including methane and fermentation characteristics were observed in vitro. The pH values ranged from 6.25 to 6.72 in all the treatments, and these showed a similar level at 48 hr. The total gas production in the treatments showed a similar pattern with C at 48 hr, whereas methane production in the treatments was lower (p<0.05) than C. Concentrations of total volatile fatty acids (VFAs) and propionic acid were higher (p<0.05) in the treatments than in C at 12 hr. Therefore, halogenated compounds (BCM, BES, BPS, CLF, and PMDI) inhibited in vitro methane emissions by inhibiting methanogens in the rumen. Further studies on safety are needed.

The objective of this study was to evaluate the in vitro effects of organic acids on methane emission and ruminal fermentation characteristics. We expected our methodology to result in a decrease of methanogens attached to the surface of rumen ciliate protozoa by addition of organic acids and in particular a decrease in methane emission. A fistulated Holstein cow of 650 kg body weight was used as a donor of rumen fluid. Organic acids (aspartic acid, fumaric acid, lactic acid, malic acid, and succinic acid) known to be propionate enhancers were added to an in vitro fermentation system and incubated with rumen fluid. The microbial population, including bacteria, protozoa, and fungi, were enumerated, and gas production, including methane and fermentation characteristics, were observed in vitro. Organic acids appeared to affect the rumen protozoan community. The rumen protozoal popuation decreased with the addition of aspartic acid, fumaric acid, lactic acid, and malic acid. In particular, the methane emission was reduced by addition of lactic acid. The concentration of propionate with all organic acids that were added appeared to be higher than that of the control at 12 h incubation. Addition of organic acids significantly affected rumen bacteria and microbial growth. The bacteria in added fumaric acid and malic acid was significantly higher (p<0.05) and protozoa was significantly lower (p<0.05) than that of the control. Microbial growth with the addition of organic acids was greater than the control after 48 h incubation.
The concentration of cordycepin produced by crossbred Cordyceps militaris JLM 0636 (CM${\alpha}$) was 7.42 mg/g dry weight, which was 7-fold higher than that of C. militaris (CM). However, cordycepin was not detected in Paecilomyces japonica (PJ). The improvement effects of CM${\alpha}$, CM and PJ on orotic acid (OA)-induced fatty liver in male Sprague-Dawley rats was investigated. Rats were randomly divided into five groups (n=6) based on five dietary categories: normal (N), OA control (C), OA plus 3% (w/w) PJ (PJ), OA plus 3% CM (CM), and OA plus 3% CM${\alpha}$ (CM${\alpha}$). OA treatment induced the retardation of body weight gain and enlargement of the liver. The concentration of hepatic triglyceride was markedly increased by OA-feeding, whereas this rise was significantly reduced by simultaneous feeding of OA, PJ, CM, and CM${\alpha}$, and this phenomenon was more pronounced by cordycepin-enriched CM${\alpha}$. The contents of total lipid, triglyceride, and free fatty acid in the serum were significantly or slightly lower in the OA control group than those of the N group, but there were no significant differences among the OA treatment groups. The hepatocytes in the OA-induced fatty liver contained numerous large lipid droplets, but PJ, CM, and CM${\alpha}$ feeding prevented the OA-induced lipid droplet accumulation in the hepatocytes. This effect was more pronounced in cordycepin-enriched CM${\alpha}$ than in PJ or CM in OA treatment rats. Accordingly, cordycepin-enriched CM${\alpha}$ could be an ideal candidate material as a dietary supplement in healthy functional foods to improve the effects of fatty liver.

The effect of high stocking density (HSD) on the expression of stress and lipid metabolism associated genes in the liver of broiler chickens was examined by chicken genome array analysis. The chickens in a control group were randomly assigned to a $495cm^2/bird$ stocking density, whereas the chickens in a HSD group were arranged in a $245cm^2/bird$ stocking density with feeding ad libitum for 35 days. The chickens assigned to the HSD group had a significantly lower body weight, weight gain, and feed intake compared with those of the control group (p<0.05). The mortality of chickens was higher in the HSD group than in the control group. The microarray analysis indicated up-regulation of stress associated genes such as HMGCR, $HSP90{\alpha}$, HSPA5 (GRP78/Bip), DNAJC3 and ATF4, and down-regulation of interferon-${\gamma}$ and PDCD4 genes. The endoplasmic reticulum stress associated genes, HSPA5 (GRP78/Bip), DNAJC3 and ATF4, were highly expressed in the HSD group. The genes, ACSL5, TMEM195 and ELOVL6, involved in fatty acid synthesis, were elevated in the HSD group. The genes, ACAA1, ACOX1, EHHADH, LOC423347 and CPT1A, related to fatty acid oxidation, were also activated in the HSD group. These results suggest that a HSD rearing system stimulates the genes associated with fatty acid synthesis as well as fatty acid oxidation in the liver of broiler chickens.

This study investigated the effects of diet of different forages on growth performance and carcass characteristics of Hanwoo steers. Twenty-one Hanwoo steers were randomly allocated to three groups (fed hay, reed, and reed with rice straw) of seven steers each. Initial and final body weights of control, T1, and T2 groups were 125.5, 128.3, 128.3 kg and 697.4, 614.6, 706.7 kg, respectively. Average daily gain tended to increase in controls (0.70 kg/d) and the T2 group (0.71 kg/d) but not as much in the T1 group (0.60 kg/d); however, there was no significant difference. DMI was not significantly different among the treatment groups, but T1 was relatively lower than the other groups. For the yield traits, carcass weight was not significantly different between controls and the T2 group but was greater in the T2 group compared to the T1 group (p<0.05). Back fat thickness and rib eye area were higher in controls and T2 compared to T1; yield grade (A:B:C, %) was greater in T1 (43:57:0) compared to the other groups (control 0:71:29; T2 29:42:29). For the quality traits, fat color and texture were not significantly different among groups. However, meat color and maturity were significantly greater in T1 compared to T2 (p<0.05). Marbling score and appearance rate of over 1st meat quality grade were greater in the control and T2 groups compared to the T1 group. Based on the results, growth performance, feed utilization, and carcass traits appeared to improve when roughage containing rice straw plus reed was offered. Therefore, reed is worth considering as a roughage source for fattening Hanwoo steers.
Polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and dioxin-like polychlorinated biphenyls (dl-PCBs) are bioaccumulative chemicals that are considered to be toxic contaminants based on several epidemiological studies. These chemicals in colostrum were investigated and estimated for their residual consistencies by maternal characteristics like parity and maternal ages. Test subjects were healthy primipara and multipara mothers with a mean age of 31.5 (S.D=3.6) in 2007. Seven isomers of PCDDs, 10 of PCDFs, 4 of non-orthopolychlorinated biphenyls(non-ortho PCBs) and 8 of mono-orthochlorinated polychlorinated biphenyls (mono-ortho PCBs) were analyzed by HRGC/HRMS. From the analyzed data, the mean level of total WHO-TEQs was 9.41 pg TEQ/g lipid, which is significantly lower than the level found in individuals from other countries. The main contributors to the total WHO-TEQs with increasing percentages were 2,3,4,7,8-PeCDF, 1,2,3,7,8-PeCDD and 3,3',4,4',5-PCB (#126), and they accounted for more than 60% of the total WHO-TEQs. PCDFs concentrations and total WHO-TEQs were negatively associated with parity (p<0.05), and maternal age was positively associated with total WHO-TEQs (p<0.01). However, the associations with body mass index (BMI) and fish intake during pregnancy were not significant. These results were suggested that parity and maternal age are an important factor affecting the concentrations of PCDD/DFs and dl-PCBs in these specimens.
Diabetes mellitus is the fifth leading cause of death among Koreans. Control of hyperglycemia and dyslipidemia is strongly correlated with decrease in risks for cardiovascular diseases, the most common and fatal diabetic complication. The effects of chronic feeding of a mixture of Chinese herbs on blood lipid profile were measured in an animal model of type 2 diabetes mellitus, db/db mice (C57BL/Ks). The Chinese herb mixture was composed of Panax ginseng C. A. Meyer,Astragalus membranaceus, Glycyrrhiza uralensis, Lycium chinense, Morus, Pueraria thunbergiana, Prunella vulgaris var. lilacina, Acanthopanax sessiliflorus, Schizandra chinensis, Scutellaria baicalensis, Dioscorea batatas, Polygonatum doratumvar. pluriflorum, Paeonia lactiflora, and Rehmannia glutinosa in a ratio of 1 : 0.7 : 0.4 : 0.7 :0.4 : 0.7 : 1.1 : 0.9 : 0.4 : 0.4 : 0.7 :0.7 : 0.9 : 0.9. Methanol extract of the Chinese herb mixture was tested for the inhibitory activity against yeast ${\alpha}$-glucosidase in vitro. The Chinese herb mixture extract inhibited ${\alpha}$-glucosidase by 25.2% at the concentration of 0.5mg/mL. Four weekold male db/db mice (n = 14) were fed AIN-93G semipurified diet or diet containing 10% powder of the Chinese herb mixture for 6 weeks after 1 week of adaptation period. Body weight (39.5 ${\pm}$ 1.6 g) and food intake (4.3 ${\pm}$ 0.6 g/day) of the Chinese herb group were not significantly different from those of the control group (40.4 ${\pm}$ 2.6 g and 4.5 ${\pm}$ 0.6 g/day). Consumption of Chinese herb mixture significantly decreased plasma glucose level (442.5 ${\pm}$ 36.0mg/dL) compared with the control group (489.8 ${\pm}$ 34.6 mg/dL, p < 0.05). Plasma cholesterol level (159.2 ${\pm}$ 18.4 mg/dL) of the Chinese herb group was significantly lower than that of the control group (185.4 ${\pm}$ 13.7 mg/dL, p < 0.05). Blood glycated hemoglobin (6.3 ${\pm}$ 0.8%) and plasma triglyceride levels (99.4 ${\pm}$ 15.0mg/dL) of the Chinese herb group were not significantly different from those of the control group (6.7 ${\pm}$ 0.7% and 108.8 ${\pm}$ 11.0mg/dL). Thus, the Chinese herb mixture could be useful in the treatment of diabetes and cardiovascular complications of diabetes.

We previously demonstrated that zinc plus arachidonic acid (ZA) treatment lowered blood glucose levels in streptozotocin-induced diabetic rats, genetically diabetic obese (ob/ob) mice, and genetically diabetic, non-obese Goto-Kakizaki rats. However, plasma insulin levels did not increase with ZA treatment, suggesting that ZA lowers blood glucose levels not by stimulating pancreatic insulin secretion. However, it is unclear whether these agents lower blood glucose levels by decreasing hepatic glucose output (HGO) or by increasing glucose utilization in peripheral tissues, or both. In order to determine ZA target organ of insulin action, we divided 18 Sprague-Dawley rats weighing ${\sim}130g$ into 3 groups (6 rats per group) and treated them for four weeks with: (1) Control diet (regular rat chow), (2) High fructose (60.0%) diet only, and (3) the same fructose diet plus zinc (10 mg/L) and arachidonic acid (50 mg/L) containing drinking water. After 4 weeks, insulin action was assessed using the hyperinsulinemic euglycemic clamp technique. Food intake and body weights were comparable in all three groups of rats throughout the study period. Plasma glucose and insulin concentrations, glucose uptake, and HGO in the basal state were all the same in these three rat groups. During the clamp study, fructose-treated and fructose+ZA treated rat groups did not exhibit any detectable change on insulin-mediated glucose uptake compared to controls. High fructose feeding impaired insulin mediated suppression of HGO, compared to controls during clamp (4.39 vs. 2.35 mg/kg/min; p<0.05). However, ZA treatment in high fructose-fed rats showed a remarkable increase in hepatic insulin sensitivity compared to high fructose-fed rats, reflected by a complete recovery in suppression of HGO during the clamp (4.39 vs. 2.18 mg/kg/min; p<0.05). This data suggests that ZA increases insulin sensitivity in liver but not glucose utilization of peripheral tissues in high fructose-fed rats.
