Quality Comparison of Sausage and Can Products in Korean Market (국내시장에 유통중인 소시지 및 캔류 제품의 품질 비교)

  • The wienner sausage(natural casing, N), wienner sausage(collagen casing, C), frankfurter sausage(F) and can products[spam(S), luncheon meat(L), jangjorim(J)] were obtained from different Korean meat processing companies and investigated for their salinity, saccharinity, pH, moisture and fat content, meat color and sensory evaluation. In sausage products, the saccharinity percent ranged 4.9∼5.0 in N, 6.6∼8.0 in C, and 5.2∼6.5 in F. The salinity percent of C and F were slightly higher than that of N. The pH values of all sausage product were above 6.0. The L* values of N were ranged 49.8∼56.7, which were slightly lower than those of C and R The sausage with high content of crude fat and high L*value earned the highest score in overall acceptability. In can products, saccharinity percentage was higher in J compared to the S and L. The salinity percentage of S was slightly higher than those of Land J. In meat color, L* and a* values were not different between S and L, although b* value of L was slightly higher than that of S and J. There were not significantly different among can products, however, the product containing low-salt had the highest score in overall acceptability.

Effect of Steaming Pressure and Time and Storage Period on Quality Characteristics of Baeksulgi (백설기 제조에서 찌는 압력과 시간 및 저장 기간에 따른 품질 특성)

  • The objective of the study was to investigate the effect of steaming pressure (0.5, 1.0, and 1.5 bar), steaming time (5, 10, and 15 min), and storage period on Hardness, hardness rate constant and sensory characteristics of Baeksulgi prepared with optimum formula (rice new 100%, water 60%, salt 1.5%, and sugar 15%). Although the hardness was slowly increased during storage period from 0 hr to 8 hr and sharply increased after 12 hr of storage in case of 0.5 bay steam pressure, it was slowly increased after 12 hr storage time in case of steaming pressure at 1.0 ban Hardness rate constant of Baeksulgi was $0.217{\sim}0.184hr^{-1}$ at range of steaming time for $8{\sim}10$ min and pressure at $0.8{\sim}1.0$ bar which was relatively low. Hardness and elasticity of sensory evaluation were increased with the increases in steaming pressure, steaming time, and storage period but cohesiveness was decreased The highest overall preference was steaming pressure at 10 bar, steaming time for 10 min, and storage period for 12 hr. In conclusion, steaming pressure, steaming tim, and storage period influenced Baeksulgi quality such as hardness and sensory characteristics.

Medium compositions reveal potential organogenesis in the diploid and tetrploid Codonopsis lanceolata

  • Medium composition plays a key role on influencing organogenesis in plant tissue culture. This study was carried out to examine the effects of medium composition on organogenesis in diploid and tetraploid Codonopsis lanceolata and obtain in-vitro mass propagation of superior species of C. lanceolata. Diploid C. lanceolata was found to be declined regarding MS medium composition for each concentration. However, shoot and adventitious root formation were suppressed with higher mineral salt concentration, and active growth of shoot and adventitious root was exhibited as 4.9 cm and 3.2 cm respectively in 1/2 MS medium. While in tetraploid C. lanceolata, it showed 2.9 cm and 3.2 cm respectively in 1/4 MS medium. In the case of sucrose concentration, no consistent decrease was observed for growth of shoot and the adventitious root of diploid both at high and low concentration. The growth of shoot (at 3% concentration) and adventitious root (at 7% concentration) was 2.3 cm and 2.0 cm respectively. Although there was no difference in shoot formation of tetraploid C. lanceolata in all concentrations with the range of 1.7~1.8, there was a slight decrease in shoot growth at high concentration. Results revealed that the adventitious root formation was suppressed at high concentration. The concentration of agar exhibited no significant difference in shoot formation of diploid C. lanceolata at all concentrations. The maximum result of adventitious growth (4.1 cm) was observed at 0.8% concentration. Slight inhibition of shoot formation and root formation of tetraploid C. lanceolata was observed at higher concentration. Shoot formation of diploid C. lanceolata also exhibited inhibition at higher concentration. Shoot formation of diploid C. lanceolata was increased at lower pH and shoot growth was the highest (2.3 cm) at pH 3.8. Adventitious root formation was higher at lower pH. However, both the adventitious root formation and growth exhibited comparatively higher result at pH 5.8. Taken together, the levels of pH had an effect on shoot and root formation in diploid and tetraploid of C. lanceolata

Characteristics of Geochemical Processes along the Salinity Gradient in the Han River Estuary (한강 기수역에서 염분구배에 따른 지화학적 특성 변화)

  • To understand the geochemical processes in the Han River Estuary, distributions and behaviors of nutrients, dissolved organic matters, and uranium were investigated and analyzed during estuarine tidal mixing in June 2000 and February 2001. The distribution of inorganic nutrients showed very dynamic distributional patterns implying an apparent nitrification process and a concave non-conservative mixing along the salinity gradient. Dissolved organic carbon was high in the upstream region and decreased sharply in the low salinity region of around 5 psu. The 3-D fluorescence characteristic of dissolved organic matter showed two distinct fluorophores in the study area. Biomacromolecules originated mainly from the indigenous biochemical processes and geomacromolecules from terrestrial humic materials. In the study area, the distribution of geomacromolecule showed a concave non-conservative property along the salinity gradient presumably due to the flocculation and removal processes in the estuary. Meanwhile, distribution of the dissolved uranium, mainly in the form of stable uranium carbonate complex, also showed a concave non-conservative property along the salinity gradient in the Han River Estuary. From this study, the removal rate of dissolved uranium in the Han River Estuary was estimated to be about 7.1 ton per year.

Production of Novel Flavonoids in Cell Cultures of Cudrania tricuspidata (꾸지뽕나무 (Cudrania tricuspidata)세포배양에 의한 신규 Flavonoids 생산)

  • To produce novel bioactive flavonoids, Gericudranin A and Gericudranin B, a cell culture system of Cudrania tricuspidata including callus induction and optimization of culture conditions was established. Friable calli were efficiently induced from the hypocotyl segments of seedlings on B5 medium supplemented with 1.0 mg/L NAA, 0.1 mg/L kinetin and 3% sucrose. Several factors were optimized for the Gericudranin production and the cell growth in suspension cultures. Low level of basal salt medium (1/8 MS), 1.0 mg/L IAA and 0.1 mg/L zeatin, and high level of sucrose (5%) were effective for the production of Gericudranins, whereas WPM with 1.0 mg/L NAA, 0.1 mg/L zeatin, and 5% sucrose were more effective for the cell growth. When cells were cultured on MS liquid medium supplemented with 1.0 mg/L IBA, about 2200 $\mu\textrm{g}$/g dry wt of Gericudranin A could be produced. The level might be about 10 times of the native inner bark. About 2350 $\mu\textrm{g}$/g dry wt of Gericudranin B was also produced on MS liquid medium with 5% sucrose, 1.0 mg/L NAA, 0.1 mg/L kinetin. The content was estimated about 3 times of the level of native inner bark.

Effect of Temperature, pH and Salt Concnetration on formation of N-nitrosamines during Kimchi Fermentation (김치숙성중 숙성온도, pH 및 소금 농도가 니트로스아민의 생성에 미치는 영향)

  • The effect of ripening temperature, pH and salinity on the formation of Nnitrosamine (NA) during Kimchi fermentation and in vitro was studied, respectively. During Kimchi fermentation for six weeks at cold storage temperature ($4^{\circ}C$) and room temperature ($16{\pm}2^{\circ}C$), the contents of nitrite and dimethylamine (DMA) showed variation at room temperature but no variation at cold storage temperature. The maximum generation of nitrosodimethylamine (NDMA) resulted low content ($2.69\;\mu\textrm{g}/kg$) at cold storage temperature but started to increase after one week fermentation and reached to the 18-fold higher generation ($49.6\;\mu\textrm{g}/kg$) at room temperature. During Kimchi fermentation, no correlation was observed between the variation of nitrite and DMA content and the generation of NDMA. However, pH showed effective relation to NDMA generation such as the highest NDMA generation was obtained at lowest pH 4. During in vitro test, higher temperature and lower pH resulted more NDMA generation and generation amount was affected more by pH. Also, the salinity of Kimchi provided inhibitory effects on the formation of NDMA. NDMA was produced $5.86\;\mu\textrm{g}/kg$ at normal salinity (2.5%) but $90.9\;\mu\textrm{g}/kg$ at lower salinity (15%) after three week. The higher salinity showed lower formation of NDMA in vitro test, too.

Comparison of antioxidant, ${\alpha}$-glucosidase inhibition and anti-inflammatory activities of the leaf and root extracts of Smilax china L. (청미래덩굴 잎 및 뿌리 추출물의 항산화, ${\alpha}$-Glucosidase 억제 및 항염증 활성비교)

  • This study was conducted in order to compare the biological activities of leaf and root water extracts of Smilax china L. (SC) by measuring the total polyphenol and flavonoid contents, anti-oxidant activity, inhibitory effect on ${\alpha}$-glucosidase, and anti-inflammatory gene expression. The total polyphenol and flavonoid contents of SC leaf (SCLE) and root (SCRE) water extracts were 127.93 mg GAE/g and 39.50 mg GAE/g and 41.99 mg QE/g and 1.25 mg QE/g, respectively. The anti-oxidative activities of SCLE and SCRE were measured using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity assay and reducing power assay. Both SCLE and SCRE scavenged radicals in a concentration-dependent manner, and SCLE showed stronger radical scavenging activity and reducing power than SCRE; however, both SCLE and SCRE exhibited lower activities than ascorbic acid. Compared to the anti-diabetic drug acarbose, which was used as a positive control, SCLE and SCRE exhibited low ${\alpha}$-glucosidase inhibition activities; nevertheless, the activity of SCLE was 3.7 fold higher than that of SCRE. Finally, SCLE caused significantly decreased expression of the LPS-induced cytokines, iNOS, and COX-2 mRNA in RAW264.7 cells, indicating anti-inflammatory activity. These results indicate that SCLE might be a potential candidate as an anti-oxidant, anti-diabetic, and anti-inflammatory agent.

Characteristics of proteolytic microorganisms and their effects on proteolysis in total mixed ration silages of soybean curd residue

  • Objective: The objective of this study was to isolate proteolytic microorganisms and evaluate their effects on proteolysis in total mixed ration (TMR) silages of soybean curd residue. Methods: TMRs were formulated with soybean curd residue, alfalfa or Leymus chinensis hay, corn meal, soybean meal, a vitamin-mineral supplement, and salt in a ratio of 25.0: 40.0:30.0:4.0:0.5:0.5, respectively, on a basis of dry matter. The microbial proteinases during ensiling were characterized, the dominate strains associated with proteolysis were identified, and their enzymatic characterization were evaluated in alfalfa (A-TMR) and Leymus chinensis (L-TMR) TMR silages containing soybean curd residue. Results: Both A-TMR and L-TMR silages were well preserved, with low pH and high lactic acid concentrations. The aerobic bacteria and yeast counts in both TMR silages decreased to about 105 cfu/g fresh matter (FM) and below the detection limit, respectively. The lactic acid bacteria count increased to 109 cfu/g FM. The total microbial proteinases activities reached their maximums during the early ensiling stage and then reduced in both TMR silages with fermentation prolonged. Metalloproteinase was the main proteinase when the total proteinases activities reached their maximums, and when ensiling terminated, metallo and serine proteinases played equally important parts in proteolysis in both TMR silages. Strains in the genera Curtobacterium and Paenibacillus were identified as the most dominant proteolytic bacteria in A-TMR and L-TMR, respectively, and both their proteinases were mainly with metalloproteinase characteristics. In the latter ensiling phase, Enterococcus faecium strains became the major sources of proteolytic enzymes in both TMR silages. Their proteinases were mainly of metallo and serine proteinases classes in this experiment. Conclusion: Proteolytic aerobic bacteria were substituted by proteolytic lactic acid bacteria during ensiling, and the microbial serine and metallo proteinases in these strains played leading roles in proteolysis in TMR silages.

Improvement of floating ability and storage stability for jumbo granules (수면부상성 점보입제의 수면부상성 및 경시분해 안정성 개선)

  • This study was performed to develop jumbo granules floated and spreaded on the water surface after application, having low production and formulation cost, and safe to environment for paddy herbicides and to establish the formulation recipe. The jumbo granules of azimsulfuron with molinate(0.075+7.5%) was formulated by KCl as water soluble carrier and paraffin oil as solvent to impose the floating and spreading force to granules. That showed 100% of total granules to be floated on and spreaded upto the water surface within 25 minutes after application. Change in carriers, surfactants and pH did not affected to improve the time-course degradation of azimsulfuron in jumbo granules, but salt formation of azimsulfuron added by 1.15M solution of NaOH a little. Addition of N-methyl acrylate and modification of formulation process affected decrease in degradation of azimsulfuron upto 1.2, 2.1, and 7.2% after 2, 6 and 12 weeks under storage at $40{\pm}2^{\circ}C$, respectively, which showed the establishment of formulation recipe of the jumbo granules.

Enhanced drought and oxidative stress tolerance in transgenic sweetpotato expressing a codA gene (CodA 고발현 형질전환 고구마의 산화 및 건조 스트레스 내성 증가)

  • Glycine betaine (GB) is one of the compatible solutes that accumulate in the chloroplasts of certain halotolerant plants under salt or cold stress. The codA gene for choline oxidase, the enzyme that converts choline into GB, has been cloned from a soil bacterium Arthrobacter globiformis. We generated transgenic sweetpotato plants [Ipomoea batatas (L.) Lam] expressing codA gene in chloroplasts under the control of the SWPA2 promoter (referred to as SC plants) and evaluated SC plants under oxidative and drought stresses. SC plants showed enhanced tolerance to methyl viologen (MV)-mediated oxidative stress and drought stress due to induced expression of codA. At $5{\mu}M$ of MV treatment, all SC plants showed enhanced tolerance to MV-mediated oxidative stress through maintaining low ion leakage and increased GB levels compared to wild type plants. When plants were subjected to drought conditions, SC plants showed enhanced tolerance to drought stress through maintaining high relative water contents and increased codA expression compared to wild type plants. These results suggest that the SC plants generated in this study will be useful for enhanced biomass production on global marginal lands.
