Cholesterol-induced inflammation and macrophage accumulation in adipose tissue is reduced by a low carbohydrate diet in guinea pigs

  • BACKGROUND/OBJECTIVES: The main objective of this study was to evaluate the effects of a high cholesterol (HC) dietary challenge on cholesterol tissue accumulation, inflammation, adipocyte differentiation, and macrophage infiltration in guinea pigs. A second objective was to assess whether macronutrient manipulation would reverse these metabolic alterations. MATERIALS/METHODS: Male Hartley guinea pigs (10/group) were assigned to either low cholesterol (LC) (0.04g/100g) or high cholesterol (HC) (0.25g/100g) diets for six weeks. For the second experiment, 20 guinea pigs were fed the HC diet for six weeks and then assigned to either a low carbohydrate (CHO) diet (L-CHO) (10% energy from CHO) or a high CHO diet (H-CHO) (54% CHO) for an additional six weeks. RESULTS: Higher concentrations of total (P < 0.005) and free (P < 0.05) cholesterol were observed in both adipose tissue and aortas of guinea pigs fed the HC compared to those in the LC group. In addition, higher concentrations of pro-inflammatory cytokines in the adipose tissue (P < 0.005) and lower concentrations of anti-inflammatory interleukin (IL)-10 were observed in the HC group (P < 0.05) compared to the LC group. Of particular interest, adipocytes in the HC group were smaller in size (P < 0.05) and showed increased macrophage infiltration compared to the LC group. When compared to the H-CHO group, lower concentrations of cholesterol in both adipose and aortas as well as lower concentrations of inflammatory cytokines in adipose tissue were observed in the L-CHO group (P < 0.05). In addition, guinea pigs fed the L-CHO exhibited larger adipose cells and lower macrophage infiltration compared to the H-CHO group. CONCLUSIONS: The results of this study strongly suggest that HC induces metabolic dysregulation associated with inflammation in adipose tissue and that L-CHO is more effective than H-CHO in attenuating these detrimental effects.

  • This experiment was conducted to investigate the factor of phytotoxicity for herbicides(bensulfuron methyl, pyrazosulfuron-ethyl, dimepiperate, molinate) on flood direct-seeded rice. The phytotoxicity of herbicides was evaluated under controlled environment condition. Bensulfuron methyl and pyrazosulfuron-ethyl reduced more rice growth, especially root growth on low temperature(20/$11^{\circ}C$) than high temperature(30/$22^{\circ}C$) cultivations. The phytotoxicity of bensulfuron methyl and pyrazosulfuron-ethyl were increased relatively by non-nutrient and nutrient solution. cultivation, respectively. The rice applied bensulfuron methyl and pyrazosulfuron-ethyl with pH 3.5, 5.5, 7.5 and 9.5 solution became low growth rate on low pH of pH 3.5 and 5.5 solution cultivation. Bensulfuron methyl application with pH 5.5 and pH 7.5 solution, and pyrazosulfuronethyl application with pH 7.5 and 9.5 solution reduced rice growth inhibition. The root growth of rice seeded in 6cm depth of water solution applied herbicides was suppressed by bensulftuon methyl and pyrazosulfuron-ethyl, the growth of shoot was suppressed heavily by dimepiperate and molinate, in particular dimepiperate suppressed about the growth of 90%. The phytotoxicity of pyrazosulfuron-ethyl became high on light clay soil of non-fertilizer condition and sand loam soil of fertilizer condition, bensulfuron methyl became high on sand loam soil.

NECESSITY OF READY ELECTRON DISPOSAL AND INTERSPECIES HYDROGEN TRANSFER FOR THE UTILIZATION OF ETHANOL BY RUMEN BACTERIA

  • Ethanol was utilized by mixed rumen microbes, but addition of pentachlorophenol (25 mg/l), a methanogen inhibitor, suppressed the utilization of ethanol. Carbon monoxide (50% of the gas phase), a hydrogenase inhibitor, more strongly suppressed the utilization of ethanol, propanol, and butanol. These results suggest that the major ethanol utilizers are $H_2$ producers. Ethanol utilization was depressed at low pH (below 6.0). Since methanogens were shown to be relatively resistant to low pH, it appears that ethanol utilizers are particularly sensitive to low pH. Ruminococcus albus and R. flavefaciens in mono-culture produced ethanol from carbohydrate (glucose and cellobiose), even when a high level (170 mM) of ethanol was present. Ethanol was not utilized even in the absence of carbohydrate, but the co-culture of these bacteria with methanogens resulted in the utilization of ethanol, i.e., when $H_2$ was rapidly converted to $CH_4$, R. albus and R. flavefaciens utilized ethanol. These results suggest that ethanol is utilized when the electrons liberated by the oxidation of ethanol are rapidly removed, and ready electron disposal in ethanol-utilizing, $H_2$-producing bacteria is accomplished by the interspecies transfer of $H_2$.

  • Alkaline bacterium producing a high pro-tease activity at low temperature was isolated by using enrichment culture from soil samples and identified as Bacil-lus sp. WRD-1 Cell growth was maximal at 10 hours and the optimal initial pH and culture time of culture condition for enzyme production was pH 7 and 10 hours, respectively. Temperature range of high enzyme activity were $10~40^{\circ}C$. The optimal pH and temperature for the enzyme activity were pH9 and $30^{\circ}C$.

Flavor Compounds and Physicochemical Properties of Low-fat Functional Sausages Manufactured with Chitosans during Refrigerated Storage (키토산을 첨가한 저지방 기능성 소시지의 향미 성분 분석과 냉장 저장 중의 이화학적인 특성)

  • The objective of this study was to evaluate the physico-chemical properties and flavor compounds of sausages with various levels and molecular weight (MWs) of chitosans, during storage at $4^{\circ}C$. Various MWs (Low: 1.5 kDa; Medium: $30{\sim}50$ kDa; High: 200 kDa) and two levels (0.3 and $0.6\%$) of chiosans were dissolved and measured the viscosity at $4^{\circ}C$, pH values were not affected (p>0.05) by either MWs or levels of chitosans. The addition or high MWs or chitosan into the pork salt soluble protein (SSP) increased the viscosity, whereas no differences were observed in low and medium MWs of chitosan. Textural profile analysis (TPA) was affected by the addition of medium or high MWs of chitosan. As a result, the addition of medium of chitosan increased the hardness, gumminess, chewiness, cohesiveness and springiness values, whereas increased level of chitosan didn't affect TPA values, except few cases. Approximately twenty-nine flavor compounds were identified in the low-fat and regular-fat sausages, however the addition of chitosans didn't impair the flavor composition of the sausages, These results indicated that the addition of chitosans didn't affect the flavor profiles, but affected the textural properties in the sausages, especially MWs higher than 30 kDa.

Effect of High Hydrostatic Pressure on the Quality of Chinese Cabbage Kimchi (초고압 처리가 배추김치의 품질특성에 미치는 영향)

  • Effects of high hydrostatic pressure on pH, titratable acidity, color, hardness and microorganisms of Chinese Cabbage Kimchi were investigated. Kimchi was pressurized at $200{\sim}600$ MPa for 5 min. There were no significant differences in color and hardness between control and pressurized Kimchi (p>0.05). Total aerobes and lactic acid bacteria were effectively inactivated by high hydrostatic pressure above 400 MPa. Changes in pH, titratable acidity, color, hardness and microbial counts for 4 weeks storage of Kimchi were investigated Kimchi was pressurized at 400 MPa for 5 min and stored at $4^{\circ}C$. The pH of control decreased to 3.94 but pressurized Kimchi maintained its initial pH value throughout the storage. The color of control showed significantly low values compared with pressureized Kimchi (p<0.05), but hardness was not significantly changed (p>0.05). Total aerobes and lactic acid bacteria in the control were reduced from the initial value of $10^8{\sim}10^9$ CFU/mL to $10^6$CFU/mL after 4 weeks storage. Whereas microbial counts in pressurized Kimchi was maintained about $10^3{\sim}10^4$ CFU/mL during storage.

Development of pH-Responsive Core-Shell Microcapsule Reactor

  • A novel type of intelligent microcapsule reactor system was prepared. The reactor can recognize pH change in the medea and control reaction rate by itself. For the reactor system, acrylic acid (AA), N-isopropylacrylamide (NIPAM), and glucose oxidase (GOD) were selected as a pH-responsive device, a gating device according and a reaction device, respectively. Poly(NIPAM-co-AA) (P-NIPAM-co-AA) are known to change its hydrophilicity-hydrophobicity due to pH change. They were integrated in a core-shell microcapsule space. GOD was loaded inside the core space and the pores in the outside shell layer were filled with P-NIPAM-co-AA linear grafted chains as pH-responsive gates by plasma graft filling polymerization method. When P-NIPAM-co-AA gates are hydrophilic at high pH value, this microcapsule permits glucose penetration into the core space and GOD reaction proceeds. However, when P-NIPAM-co-AA gates are hydrophobic at low pH value, this microcapsule forbids glucose penetration and GOD reaction will not occur. The accuracy of this concept was examined.

Effect of pH on the Vascular Tone and $^{45}Ca$ Uptake in the Aorta of Spontaneously Hypertensive Rats

  • The effect of extracellular and intracellular pH on vascular tone and $^{45}Ca$ uptake were investigated in aortic strips and dispersed single aortic smooth muscle cells of spontaneously hypertensive rats (SHR) and aged-matched Wistar-Kyoto rats (WKY). The contraction produced by a change of extracellular pH (pHo) in the range of $6.5{\sim}8.3$ was estimated by comparison with the level of vascular tone at pH 7.4. Contraction was induced below pHo 6.5 in WKY, pHo 7.1 in SHR, and over pHo 8.0 on both strains. The amplitude of contraction induced by high pHo (over pHo 7.7) was similar in SHR and WKY, but that induced by low pHo (below pHo 7.1) in SHR was greater than that in WKY. Either high pHo- or low pHo-induced contractions in WKY and SHR were not induced in the Ca-free Tyrode's solution and were induced by the addition of Ca. $^{45}Ca$ uptake increased progressively as pHo was increased from 6.8 to 8.1 in the single aortic smooth muscle cells of WKY and SHR. $NH_4Cl$ induced a gradually developing contraction in a dose-dependent manner $(5\;mM{\sim}30\;mM)$ and the removal of $NH_4Cl$ induced transient contraction was followed by profound relaxation in the aortic rings of both strains. The contractions induced by $NH_4Cl$ or by the removal of $NH_4Cl$ in SHR were significantly greater than that in WKY. These contractions were not induced in Ca-free Tyrode's solution. $^{45}Ca$ uptake was increased by $NH_4Cl$ (20 mM) and was not changed by the removal of $NH_4Cl$ (20 mM) in the aortic strips of WKY and SHR. As a summary of above results, the vascular tone of SHR was more sensitive to the change pHi and pHo than that of WKY. The contractions induced by change of extracellular or intracellular pH depended on extracellular Ca in the aorta of SHR nnd WKY. However, the Ca uptake was in accord with the changes of contraction but increase in contraction by low pH was not accompanied by an increase in Ca uptake in both strains.

Effect of Fermentation Temperature on Quality of Doenjang (숙성온도가 된장의 품질에 미치는 영향)

  • There were four types of Doenjang fermentation as following conditions for investigation ; 1) low temperature fermentation at $13^{\circ}C$ for 180 days, 2) low temperature at $13^{\circ}C$ for 7 days to room temperature at $30^{\circ}C$ for 10 days, to low temperature at $13^{\circ}C$ for 163 days, and for 173 days, 3) low temperature at $13^{\circ}C$ 7 days to room temperature at $30^{\circ}C$, 4) room temperature at $30^{\circ}C$ for 180 days. There were no changes of moisture, NaCl and total nitrogen content during fermentation period of four types conditions, but pH and amino type nitrogen decreased in room temperature at $30^{\circ}C$ for 180 days. It required 3 times more fermentation period until same quantity of the amino type nitrogen. The low temperature fermentation sample was lower than room temperature fermentation sample in pH and amino type nitrogen. The yeast decreased in low temperature fermentation sample taken 15 to 30 days longer than room temperature sample. The yeast is increased up to 30 days, and decreased little by little. After 60 days, it remained a few without effectiveness on the Doenjang quality. The low temperature fermentation sample showed brighter than room temperature fermentation sample. Different fermentation condition affected Doenjnag quality, especially, low temperature fermentation sample showed bright color in Doenjnag. So low temperature fermentation must be expected as good method for getting high quality Doenjnag.

MINERAL CONCENTRATION IN RICE STRAW AND SOIL IN KYONGBUK PROVINCE, KOREA

  • A field survey was carried out in Korea to assess the mineral composition of rice straw since it is a cheap and available cattle feedstuff. Forage and soil samples were collected in 4 localities in the Kyongbuk province. Soil analysis were also carried out in order to establish the relationships between soil composition and mineral content in the rice straw. Based on NRC tables, the rice straw samples provided adequate amounts of Mg, Ca, K, S, Mo, Mn and Zn. Percentages of samples deficient in P, Na, Cu and Se were 83, 50, 67 and 83 respectively. Soil samples, whose mean pH was 5.8 and mean organic matter content was 3.99%, were comparatively acidic and had high organic matter content. They also had high amounts of extractable Fe and Mn. Correlation coefficients between mineral content in soil and rice straw were low, i.e., 0.42 for Me (p < 0.05), and 0.37 for Mo (p < 0.05). The low or nonexistent correlation between soil and forage composition indicates the difficulty of establishing appropriate methods of mineral availability to the plants.
