  • The purpose of this study was to evaluate the effect of concentration and pH of lactic acid on the formation of artificial root caries lesions formation in bovine teeth. The characterictics of artificially produced early root lesions were observed with polarized light microscope and the depth of lesions were mearsured with measuring microscope The results were as follows: 1. In the group of low pH and high concentration of lactic acid, the progress of lesion formation was faster than that of high pH and low concentration lactic acid group. 2. In the same group, initial lesion progress faster, but progression rate was reduced as time goes by. 3. In the development of initial root caries, cementum was act as a barrier to protect dentin from lesion progression.

A pH Measurement Study on Commercial Alcoholic Drinks (시판주류의 pH 측정 연구)

  • The purpose of this study was to determine pH value of some alcoholic drinks sold in Korea and to provide the basic information which can cause dental erosion. Alcoholic drinks commercially sold were purchased from various big markets in Korea. The sorts of drinks tested in this study consisted of 5 brands of beers, 24 brands of makgeollis, 9 brands of wines and 12 brands of sojus. The test groups were selected randomly and the pH of each beverage was determined using a pH meter he each pH was measured. For statistical data, Mann-Whitney test was used to analyze difference for red wine and white wine and Kruskal-Wallis test was used to compare the pH of each test group. The result of this study was as followings: the mean pH of 5 brands of beers was 4.21, that of 24 brands of makgeollis 3.88, of 9 brands of wines 3.34 and of 12 brans of sojus 7.86. Each test group was significantly different (p<0.05). Except for soju groups, the test groups had a low pH value which can cause dental erosion. In terms of comparing between pH value of red wine and white wine, the result of this study represented that the mean pH of red wine was 3.45 and that of white wine was 3.21. This result showed the pH of two kinds of wine had a low pH which can lead to dental erosion and the difference of two wine were significantly different (p<0.05). As a result, some drinks sold in Korea have a high erosive potential on teeth since they have a comparatively low pH expect soju. Hence, when we consume some kinds of alcoholic drinks, we make sure to remember that the alcoholics which had a low pH, can have an effect on dental erosion that mean we should avoid to drink some alcoholic drinks with low pH for long time.

The Effects of Different Feeding Levels on the Number of Fecal Lactic Acid-producing Bacteria and Fecal pH in Horses (말에서 사료 급여 수준이 분 젖산 생산 박테리아 및 pH 변화에 미치는 영향)

  • This study was conducted to determine the effects of different feeding levels on the number of fecal lactic acid-producing bacteria and fecal pH in horses. In Exp. 1, 6 female cross-bred horses ($332.4{\pm}50.4\;kg$, 5 year old) were fed 2% hay or 2% concentrates of BW for 30 d, and the number of fecal Lactobacilli and Streptococci was measured. For Exp. 2, 5 castrated Thoroughbred ($474.5{\pm}64.6kg$, 4 year old) and 5 female cross-bred horses ($343.6{\pm}56.3\;kg$, 5 year old) were allotted to high- or low-concentrates diets (5 levels) for 30 d in a $5{\times}5$ Latin square design, and the fecal pH was assessed. In Exp. 3, 3 castrated Thoroughbred ($482.6{\pm}53.3\;kg$, 4 year old) were fed high- or low-alfalfa hay diets (3 levels) for 30 d in a $3{\times}3$ Latin square design, and the fecal pH was measured. Feeding high-concentrates diets increased (P<0.05) the number of fecal lactic acid-producing bacteria (Lactobacilli and Streptococci) and decreased (P<0.01) the fecal pH. Feeding alfalfa hay with high-concentrates diet (2% alfalfa hay + 1% concentrates of BW) decreased (P<0.01) the fecal pH. These results showed that feeding high-concentrates diets increased lactic acid-producing bacteria in the large intestine and decreased the intestinal or fecal pH causing colic or laminitis, indicating that the proper fiber/concentrates (1:1) should be maintained in horses.

Effects of Morphological Characteristics of Muscle Fibers on Porcine Growth Performance and Pork Quality

  • The aim of this study was to investigate the effects of morphological characteristics of porcine muscle fibers on growth performance, muscle fiber characteristics, and pork quality taken from the longissimus dorsi muscle. A total of 239 crossbred pigs (164 castrated males and 75 females) were used in this study. Experimental pigs were categorized by the total number of muscle fiber (TNF: High and Low) and cross sectional area of muscle fiber (CSAF: Large, Middle, and Small). Their combinations were classified into six groups (High-Large, HL; High-Middle, HM; High-Small, HS; Low-Large, LL; Low-Middle, LM; Low-Small, LS). The TNF and CSAF were significantly (p<0.05) correlated with growth rate and carcass productivity, while the only of the type I number had no meaningful relationships excluding the correlation with loin area (p<0.001). The proportion of type I area was positively correlated with pH45 min while the proportion of type IIB area was negatively correlated with pH45 min and pH24 h (p<0.05). Drip loss and protein denaturation had strong relationships with the proportion of type IIB number or area. The HL group exhibited the greatest growth performance. In addition, the HL group had significantly greater values in protein solubility than the other groups. In conclusion, this study suggest that high TNF combined to large CSAF improve the ultimate lean meat productivity and assure normal meat quality simultaneously with increased both proportion of number and area of type I, type IIA muscle fibers and lowered proportion of number and area of type IIB.

Protective Effect of Astragalus polysaccharides on Liver Injury Induced by Several Different Chemotherapeutics in Mice

  • Side effects are an unavoidable consequence of chemotherapy drugs, during which liver injury often takes place. The current study was designed to investigate the protective effect of Astragalus polysaccharides (APS) against the hepatotoxicity induced by frequently-used chemical therapy agents, cyclophosphamide (CTX), docetaxel (DTX) and epirubicin (EPI)) in mice. Mice were divided into five groups, controls, low or high dose groups ($DTX_L$, $CTX_L$, $EPI_L$ or $DTX_H$, $CTX_H$, $EPI_H$), and low or high dose chemotherapeutics+APS groups ($DTX_L$+APS, $CTX_L$+APS, $EPI_L$+APS or $DTX_H$+APS, $CTX_H$+APS, $EPI_H$+APS). Controls were treated with equivalent normal saline for 28 days every other day; low or high dose group were intraperitoneal (i.p) injected with low or high doses of CTX, DTX and EPI for 28 days every other day; low or high dose chemotherapeutics+APS group were separately intraperitoneal (i.p) injected with chemotherapeutics for 28 days every other day and i.p with APS (100 mg/kg) for 7 days continually from the 22th to the 28th days. The body weight, serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST), histopathological features, and ultrastructure morphological change of liver tissues, protein expression level of caspase-3 were estimated at different time points. With high dose treatment of DTX, CTX and EPI, weight gain was inhibited and serum levels of ALT and AST were significantly increased. Sections of liver tissue showed massive hepatotoxicity in $CTX_H$ group compared to the control group, including hepatic lobule disorder, granular and vacuolar degeneration and necrosis in hepatic cells. These changes were confirmed at ultrastructural level, including obvious pyknosis, heterochromatin aggregation, nuclear membrane resolution, and chondrosome crystal decrease. Western blotting revealed that the protein levels of caspase-3 increased in $CTX_H$ group. The low dose groups exhibited trivial hepatotoxicity. More interestingly, after 100 mg/kg APS, liver injury was redecued not only regarding serum transaminase activities (low or high dose chemotherapeutics+APS group), but also from pathological and ultrastructural changes and the protein levels of caspase-3 ($CTX_H$+APS group). In conclusion, DTX, CTX and EPI induce liver damage in a dose dependent manner, whereas APS exerted protective effects.

  • The present study compared the proteomic characteristics of a low passage number (L-33) and high passage number (H-81) LNCaP cell clone. Marked differences in protein expression were noted in the response of L-33 and H-81 cells to androgens. To investigate if regulation of these proteins was androgen-dependent, expression of the androgen receptor was silenced via small interfering RNA. Consistent with the proteomic data, abrogation of androgen receptor production in H-81 cells resulted in the reversed expression level into L-33 cells compared with non-treated H-81 LNCaP cells. The results clarify the progression into an androgen-independent phenotype.

  • Hydrogen induced cracking (HIC) is one of the hydrogen degradation phenomena of linepipe steels caused by $H_2S$ gas in the crude oil or natural gas. However, NACE TM0284-96 standard HIC test method is hard to satisfy the steel requirements for sour service application since it uses more severe environmental conditions than actual conditions. Therefore, in order to use steels effectively, it is required to evaluate HIC resistance of steels in the practical range of environmental severity. In this study, HIC resistance of two high strength low alloy (HSLA) steels being used as line pipe steels was evaluated in various test solutions with different $H_2S$ pressures and pH values. The results showed that the key parameter affecting crack area ratio (CAR) is $H_2S$ partial pressure of test solution when the pH value of test solution is not over 4. Hydrogen diffusivity was not a constant value, but it was rather affected by the hydrogen ion concentration (pH value) in the solution.

  • In this paper, for polymerization of poly(vinyl acetate-co-ethylene) (VAE) by redox system using poly(vinyl alcohol) (PVOH) as emulsifier on the properties of the final emulsion, and pH changes affect the physical properties of the final emulsion was investigated. The results of the molecular weight of PVOH had a dramatic impact on the emulsion properties. The used a low molecular weight of PVOH products was obtained low viscosity and using the high molecular weight of PVOH were obtained high viscosity product. However, changing the pH of the final polymerized product properties for the PVOH obtained different results. Generally, a poly(vinyl acetate) emulsion by a high degree of polymerization and high molecular weight of PVOH was obtained high viscosity of the final emulsion. But, in VAE was lower emulsion viscosity in high pH. This is the molecular weight of the emulsion during the synthesis of PVOH is considered to be affected by degradation. The final viscosity was decreased by grafting ratio and molecular weight were decreased with increasing of pH.

Effect of Dissolved Oxygen Concentration and pH on the Mass Production of High Molecular Weight Pullulan by Aureobasidium pullulans

  • The effects of DO and pH on the mass production of pullulan with high molecular weight and the morphology of A. pullulans ATCC 42023 were evaluated. A. pullulans showed a maximum production of pullulan (11.98 g/l) when the initial pH of the culture broth was 6.5 in a shake-flask culture. In a batch culture, the mixture of a yeast-like and mycelial cell forms was found at a pH of 4.5, and the maximum production of pullulan (13.31 g/l) was obtained. However, a high proportion of high molecular weight pullulan (M.W.>2,000,000) was produced at a pH of 6.5, with a yeast-like morphology. The maximum pullulan production yield ($51\%$) was obtained at a pH noncontrol (initial pH 6.5) and DO control (above $50\%$) condition. Pullulan degrading enzyme was activated when the pH of the broth was lower than 5.0 and the portion of low molecular weight pullulan was increased. The formation of a black pigment was observed at an initial stationary phase, at 40 h of fermentation. Therefore, the fermentation should be carried out in a pH noncontrol (initial pH of 6.5) and DO control (above $50\%$) condition, and should be harvested before reaching the stationary phase (around 40 h) for the production of high molecular weight pullulan.

Growth Inhibition of Cucumber by Absorbing Excess Al at Low Soil pH (강한 산성토양에서 Al의 과잉 흡수에 의한 오이 생육장해 양상)

  • On-site diagnosis applied to soil having a symptom of yellowing and whitening in cucumber leaf and stem. Soil pH determined 4.2 by methods of on-site analysis and 4.5 by soil test analysis. High aluminum in soil solution extracted with water saturation was detected. Leaf and stem tissue were abundant in Al content but not in Ca. Also, N content of leaf and stem was low compared to normal N ranges. This symptom of cucumber assumed to be from the Al and nitrous acid gas toxicity by low soil pH and Eh. Conclusionally, symptom in leaf and stem of cucumber was alleviated and cucumber normally recovered during cultivation period by applying calcium hydroxide solution to correct soil pH up to 6.5. These results showed that low soil pH resulted in aluminum toxicity and N deficiency to plant growth in on-site farming.
