  • Several types of scavenger receptors, including the Collectin-Placenta 1 (CL-P1) receptorthat is present in mammals, are molecules that are expressed on the surfaces of endothelial cells, macrophages and smooth muscle cells. These molecules are cell surface glycoproteins that can be conjugated to oxidized low density lipoprotein (oxLDL). Among these molecules, the effect of quercetin on CL-P1 activation has been confirmed. Quercetin is known as an antioxidant that stops oxidation because it acts to remove free radicals that are responsible for the oxidation reaction. In this study, fragments from the transcription start site of the mouse CL-P1 gene promoter to the -500th base were cloned using DNA polymerase. These fragments were then introduced into macrophage like RAW264.7 cells and fibroblast-like NIH3T3 cells to study the effect of quercetin on the CL-P1 gene expression. As a result, we found that bases ranging from -250 to -350 in the anterior part where gene expression starts are important for producing CL-P1 protein. Among them, the DNA mutation experiments we performed confirmed that the E2F binding sites are critical for producing the CL-P1 protein? In addition, when quercetin was added to the RAW264.7 culture medium, which was a culture of adherent cells, observedthe phenomenon of the cells falling off from the surface of the culture container.

Anti-Obesity Effects and the Regulation of Energy Metabolism in Skeletal Muscle Tissues of Allii Fistulosi Bulbus Extract in High Fat Diet-Induced Obesity Mice (총백추출물의 고지방식이 유도 비만 마우스에서의 항비만 효과 및 근육조직에서의 에너지대사 조절기전 연구)

  • Objectives: We investigated the effects of Allii Fistulosi Bulbus (AFB) on high fat diet (HFD)-induced obesity in mice and the regulation of energy metabolism in muscle tissues of mice. Methods: The C57BL/6 mice (6 weeks, male) were fed a HFD for 8 weeks and then administrated with AFB extract at 500 mg/kg (p.o.) once daily for 4 weeks. The body weight (BW), muscle weight, calorie intake, fasting blood glucose (FBG) and serum glucose, insulin, and low-density lipoprotein-cholesterol (LDL-C) levels were measured in mice. It was also observed the histological changes of pancreas, liver, and fat tissues with hematoxylin and eosin staining. It was investigated the phosphorylation of insulin receptor substrate 1 (IRS-1), Ser/Thr kinase (AKT), and adenosine monophosphate-activated protein kinase (AMPK), and the expression of phosphoinositide 3-kinase, glucose transporter type 4 (GLUT4), and sirtuin1 (Sirt1) in gastrocnemius tissues by western blot, respectively. Results: The increases of BWs, calorie intakes and FBG levels in obesity mice were decreased significantly by the administration of AFB extract. The AFB extract administration was reduced significantly serum levels of glucose, insulin, and LDL-C in obesity mice. The AFB extract inhibited lipid accumulation in liver tissues, hyperplasia of pancreatic islets, and enlargement of fat tissues in obesity mice. The phosphorylation of IRS-1 and AKT was increased significantly in muscle tissues and AMPK phosphorylation and the GLUT4 and Sirt1 expression were decreased significantly in muscle tissues after the AFB administration. Conclusions: Our study indicates that AFB extract improves symptoms of obesity through regulation of energy regulating proteins in muscle tissues.

Consistency of 1-day and 3-day average dietary intake and the relationship of dietary intake with blood glucose, hbA1c, BMI, and lipids in patients with type 2 diabetes (제2형 당뇨병 환자의 1일과 3일 평균 식이섭취량의 일관성과 혈당, 당화혈색소, 체질량지수, 지질과의 관련성)

  • Purpose: This study aimed to determine the consistency of 1-day and 3-day average dietary intake using the 24-hour diet recall method and to investigate the relationship of diet intake with physiological indicators potentially associated with diabetic complications in patients with diabetes. Methods: This study conducted a secondary data analysis using pretest data of a nursing intervention study entitled "Development of deep learning based AI coaching program for diabetic patients with high risk and examination of its effects." Data were analyzed through descriptive analysis, one-way repeated-measures analysis of variance, and Pearson correlation coefficients using SPSS 26.0. Results: The average total daily calorie intake over 3 days was 1,494.48 ± 436.47 kcal/day: 1,510.90 ± 547.76 kcal/day on the first day, 1,414.22 ± 527.58 kcal/day on the second day, 1,558.34 ± 645.83 kcal/ day on the third day, showing significant differences (F = 3.59, p = .031). The correlation coefficient between the 1-day and 3-day average dietary intake was 0.41-0.77 for each nutrient and 0.62-0.80 for each food group. Vegetable intake showed negative correlations with body mass index (BMI; r = -.19, p = .023) and triglycerides (r = -.18, p = .036), whereas dairy intake was positively associated with low-density lipoprotein-cholesterol (LDL; r = -0.18, p = .034) and triglycerides (r = .40, p<.001). Conclusion: This study demonstrated that 1-day dietary intake was highly correlated with 3-day average dietary intake using the 24-hour diet recall method. Food groups showed significant associations with physiological indicators of potential diabetic complications such as BMI, triglycerides, and LDL levels. Further studies are needed to improve the knowledge base on the relationships between physiological indicators and food groups.

Effect of Fermented Water Extracts from Ligularia fischeri on Hepatotoxicity Induced by D-Galactosamine in Rats (D-Galactosamine 투여 랫트에서 곰취 열수 추출 발효물이 간 독성 저하에 미치는 영향)

  • This study was conducted to determine the effect of fermented water extracts from Ligularia fischeri (LAF) on reduction of hepatotoxicity induced by D-galactosamine (D-GalN) in rats. In this experiment, male Sprague-Dawley rats were used as experimental animals, which were divided into eight groups: normal group, D-GalN-treated group (control), D-GalN and non-fermented water extracts from Ligularia fischeri (LA)-treated groups [100, 200, and 400 mg/kg BW (body weight)], and D-GalN and LAF-treated groups (100, 200, and 400 mg/kg BW). ${\gamma}$-Glutamyl transferase, aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase activities in serum of the D-GalN and LAF-treated groups decreased significantly compared to those of the control group (P<0.05). The high density lipoprotein-cholesterol levels of the D-GalN and LAF-treated groups increased significantly compared to those of the control group (P<0.05). The low density lipoprotein-cholesterol and triglyceride levels of the D-GalN and LAF-treated groups decreased significantly compared to those of the control group (P<0.05). The atherogenic index values of the D-GalN and LAF-treated groups decreased significantly compared to those of the control group (P<0.05), and their high density lipoprotein cholesterol by total cholesterol ratio increased significantly in these groups (P<0.05). Superoxide dismutase activity of liver tissues were enhanced significantly (P<0.05) in the D-GalN and LAF-treated groups compared to that of the control group (P<0.05), whereas their malondialdehyde content decreased significantly in these groups (P<0.05). The histopathological observations revealed apoptotic cells and mild portal inflammation in liver tissues of the D-GalN and LAF-treated groups. Taken together, these results demonstrate that LAF may improve plasma lipid profile and alleviate hepatic damage.

Serum Insulin, Insulin-like Growth Factor-I and Insulin-like Growth Factor Binding Protein-3 Levels in Obese Adolescents (비만 청소년에서 Insulin, Insulin-like Growth Factor-I, Insulin-like Growth Factor Binding Protein-3에 대한 연구)

  • Purpose : Childhood Obesity is increasing throughout the world, and it is known to incur many diseases especially in later life such as diabetes and cardiovascular disorders. The purpose of this study was to investigate the association between obesity and insulin, insulin-like growth factor-I (IGF-I), insulin-like growth factor binding protein-3(IGFBP-3) and know if these factors are useful in predicting cardiovascular diseases. Methods : The study group consisted of 64 moderate and severe obese adolescents and the controls were normal adolescents of the same age. body mass index(BMI) was calculated by height and weight; systolic and diastolic blood pressure was measured at resting state. After 10-hour fasting period, blood cholesterol, triglyceride, high density lipoprotein(HDL) cholesterol, low density lipoprotein(LDL) cholesterol, glucose, insulin, free fatty acid, IGF-I and IGFBP-3 were measured. Results : Insulin was significantly higher in the obese adolescent group than the control group(obese group $15.6{\pm}7.0{\mu}IU/mL$, P<0.01). IGF-I was also significantly higher in the obese adolescent group than the control group(obese group $498.1{\pm}122.2ng/mL$, P<0.05). In addition, IGFBP-3 was significantly higher in the obese adolescent group than the control group(obese group $3,777{\pm}4,721ng/mL$, P<0.05). Insulin showed significantly positive correlation with BMI(r=0.3944) and obesity index(r=0.34). IGFBP-3 were significantly correlated with obesity index(r=0.419), diastolic blood pressure (r=0.264) and BMI(r=0.247). Insulin resistance index significantly positive correlation with BMI(r=0.595), blood triglycerid level(r=0.515) and obesity index(r=0.469). Conclusion : Serum insulin, insulin resistance index, IGF-I and IGFBP-3 levels may be useful to predict cardiovascular diseases in adolescent obesity.

Metabolic syndrome in the overweight and obese adolescents and the impact of obesity on the cardiovascular system (과체중과 비만 청소년에서 대사증후군과 비만이 심혈관에 미치는 영향)

  • Purpose : Metabolic syndrome (MS), characterized by obesity and insulin resistance, elicits risk factors such as hyperlipidemia, hypertension, and glucose intolerance with additive effects on atherosclerosis, leading to cardiovascular diseases. The purposes of this study were to evaluate the prevalence of MS among overweight and obese adolescents and to investigate the impact of obesity on the cardiovascular system. Methods : In all, 684 adolescents were included in the study. Blood pressure, body mass index (BMI), fasting blood glucose, total cholesterol, triglyceride, low-density-lipoprotein (LDL)-cholesterol, high-density-lipoprotein (HDL)-cholesterol, aspartate aminotransferase (AST), alanine aminotransferase (ALT), and high-sensitive C-reactive protein (hs-CRP) were measured in the patients with a BMI of >85 percentile. Brachial-ankle pulse wave velocity (BaPWV) and ankle brachial index were measured using Vascular Profiler (VP)-1000. Results : MS was confirmed in 19.5% of the overweight and obese adolescents and 50.8% of the obese adolescents. The systolic and diastolic blood pressure, height, weight, fat mass, %fat, BMI, obesity index, and waist circumference were higher in the overweight and obese adolescents with MS. Moreover, the triglyceride, AST, ALT, and hs-CRP levels were higher, whereas HDL-cholesterol level was significantly lower in this group. The overweight and obese adolescents with MS showed shorter diastolic and systolic times, higher heart rate and BaPWV, and longer E-wave deceleration time by echocardiography. Conclusion : Overweight and obese adolescents showed characteristic MS features such as hypertension and hyperlipidemia. Thus, obese adolescents predisposed to MS should be provided early treatment for obesity.

Dietary Effects of Fermented Soybean Curd Residue (Biji) on Body Weight, Serum Lipid Profiles, and Antioxidation-Related Enzymes Activity of Mice Fed a High Fat Diet (고지방식이 마우스의 체중과 혈청지질 및 항산화계 효소활성에 미치는 발효 비지의 식이효과)

  • We investigated the ability of soybean curd residue (SCR) and its fermented products to inhibit obesity and improve the blood lipid profiles of obese mice fed a high-fat diet. Samples were prepared by fermenting SCR with Aspergillus oryzae var effuses KACC 44990 (ASCR), a microbe used for the fermentation of traditional Korean Meju, and with Monascus pilosus IFO 4480 (MSCR), a microbe used for the production of red rice. In addition, AMSCR, a mixture composed of equal amounts of ASCR and MSCR, was also prepared. Male mice were divided into six groups and fed with either a normal diet, a high-fat diet, or a high-fat diet supplemented with SCR, ASCR, MSCR, or AMSCR. After 8 weeks, body weight gain, serum and hepatic lipid profiles, and the activities of enzymes that generate or scavenge reactive oxygen species (ROS) were evaluated. Compared with the high-fat diet group, all the test groups showed a significant reduction in body, organ, and epididymal fat weight gain. These effects were observed with supplements in the order AMSCR>ASCR>MSCR>SCR. Similarly, supplements of test samples reduced high levels of serum and hepatic triglycerides (TG), total cholesterol, and low-density lipoprotein (LDL) cholesterol caused by hight-fat diet, while high-density lipoprotein (HDL) cholesterol was increased. Interestingly, the ability of ASCR to lower serum TG was stronger than that of MSCR, while MSCR showed a stronger hypocholesterolemic effect than ASCR. Meanwhile, AMSCR returned comprehensively serum lipid levels to normal. In addition, hepatic damage was prevented with effects in the order AMSCR>ASCR>MSCR>SCR. Hepatic ROS generating system including xanthine oxidase (XO) and ROS scavenging system including superoxide dismutase (SOD), glutathione peroxidase (GPX) and glutathione S-transferase (GST) were recovered to normal level by all test diets. In conclusion, this study suggests that SCR and its fermented products can inhibit obesity and improve lipid profiles.

Effects of Peucedanum japonicum Thunb. on Lipid Metabolism and Antioxidative Activities in Rats Fed a High-Fat/High-Cholesterol Diet (갯기름나물이 고지방-고콜레스테롤 식이를 급여한 흰쥐의 지질대사 및 항산화 활성에 미치는 영향)

  • This study was carried out to investigate the effects of Peucedanum japonicum Thunb. (PJT) powder on lipid metabolism and antioxidative activity in rats fed a high-fat/high-cholesterol diet for 4 weeks. Male Sprague-Dawley rats were assigned to four groups: normal diet group (N), high-fat/high-cholesterol group (HFC), high-fat/highcholesterol with 5% PJT powder (HFC-PBL), and high-fat/high-cholesterol with 10% PJT powder (HFC-PBH). Body weight gain and food efficiency ratio (FER) increased in the HFC group, whereas they gradually decreased in the PJT powder-fed groups. Food intake was not significantly different between the experimental groups. Liver and adipose tissue weights of the HFC group were heavier than that of the N group, whereas the groups fed PJT powder showed gradual reduction of tissue weights. Serum alkaline phosphatase (ALP) activity significantly decreased after PJT powder administration. Serum triglyceride level significantly decreased in groups fed PJT powder compared to the HFC group. The serum low density lipoprotein cholesterol (LDL-cholesterol) level of the HFC group increased by 73.70% than that of the N group, whereas serum high density lipoprotein (HDL-cholesterol) tended to decrease in groups fed PJT powder compared to the HFC group. Levels of triglycerides in epididymal and mesenteric adipose tissues, as well as cholesterol levels in liver and adipose tissues were lower in the groups fed 10% PJT powder compared to the HFC group. The liver glutathione (GSH) level increased in the groups fed PJT powder compare to the HFC group. The liver thiobarbituric acid reactive substances (TBARS) content significantly decreased in the groups fed PJT powder compared to the HFC group. These results suggest that PJT powder may improve lipid metabolism in the serum, liver, and adipose tissue and prevent oxidative stress by stimulating antioxidative systems in rats fed a high fat-high cholesterol diet.

Effect of Vinegar Made with Hydroponic-cultured Panax ginseng C. A. Meyer on Body Weight and Lipid Metabolism in High-Fat Diet-Fed Mice (수경재배 인삼으로 제조한 식초가 고지방 식이를 섭취한 마우스의 체중 및 지질대사에 미치는 영향)

  • The aim of this study was to develop hydroponic-cultured ginseng vinegar (HGV) containing ginsenoside Rg2 in order to its anti-obesity and anti-hyperlipidemic effects in C57BL/6J mice. HGV was prepared by two-stage fermentation. The ginsenoside Rg2 contained in acetic acid-fermented HGV increased by 4.0 times compared to that in pre-fermented HGV. To measure the anti-obesity effect of HGV, thirty two mice were randomly divided into four groups: normal diet group (ND), high-fat diet group (HFD), high-fat diet-supplemented with HGV group (HGV), and high-fat diet-supplemented with green tea extract group (GT). Body weight, fat weight, and liver weight decreased in the HGV group. The HGV group also showed lower plasma levels of low-density lipoprotein (LDL)-cholesterol and triglycerides, and higher levels of high-density lipoprotein (HDL)-cholesterol compared to the corresponding levels in the HFD group. Furthermore, there were significant decreases in plasma aspartase aminotransferase (AST) and alanine aminotransferase (ALT) levels in the HGV group compared to the corresponding levels in the HFD group. These results suggest that HGV can be used as an anti-obesity therapeutic agent or functional ingredient.

Changes in Serum Lipid Components and Blood Glucose by Krill (Euphausia superba) Meal and NaF in Rats (크릴 분말 및 NaF 첨가 식이가 흰쥐의 혈청 지질성분 및 혈당 변동에 미치는 영향)

  • This study was conducted to examine the changes in serum lipid components and blood glucose by krill (Euphausia superba) meal and sodium fluoride (NaF) in rats fed on experimental diets and administered orally to NaF 10 mg for 5 weeks. Body weight of rats decreased as the amount of krill meal diet increased, it was observed the basal diet plus NaF group (BF group) compared to the lower basal diet group (BD group). The serum concentrations of total cholesterol, LDL (low-density lipoprotein) cholesterol, free cholesterol, triglyceride (TG), phospholipid (PL) and blood glucose in serum were higher in the BF group than the 10% krill meal group plus NaF 10 mg (KF10 group) or BD group, the 20% krill meal plus NaF 10 mg group (KF20 group), the 30% krill meal plus NaF 10 mg group (KF30 group). Conversely depending on the content of krill meal for the HDL (high-density lipoprotein) cholesterol level, it showed higher results. The concentration of total protein was no significant difference among the groups (p<0.05). The results indicate that a krill meal diet effectively inhibited increases in lipid elevation and blood glucose level in the sera of rats.
