Analysis of Low-level ${\alpha}$-D-glucose-1-phosphate in Thermophilic Enzyme Reaction Mixuture Using High pH Anion-exchange Chromatograph (고성능 액체 크로마토그래프를 이용한 내열성 효소반응 산물인 ${\alpha}$-D-glucose-1-phosphate의 저농도 분석)

  • We have used high pH anion-exchange chromatography to analyze low level (below $20{\mu}M$) $\alpha$-D-glucose-1-phosphate (G-1-P) that can be used as a cytostatic compound, an antibiotic, and immunosuppressive drug. Our chromatographic method afforded excellent peak resolution and seletivity for glucose-6-phosphate and various maltooligosaccharides as well as G-1-P. The pulsed amperometric detector yielded linear response on G-1-P ranging from 2 - $20{\mu}M$, giving slope of $4.8{\times}10^4$(peak area/${\mu}M$). The detection limit was $2{\mu}M$. This method was applied to the purification of thermophilic $\alpha$-glucan phosphorylase from Thermus caldophilus. The technique will be extremely useful in future studies concerning carbohydrate metabolism in living organisms.

Methods for Preparing Indigestible Dextrin with High Indigestible Fraction (난소화성 획분이 높은 난소화성 덱스트린의 제조 방법)

  • The indigestible dextrin with high indigestible fraction was prepared by treating the enzyme hydrolysate of pyrodextrin with ethanol or strongly acidic cation exchange resin(UBK 530). Optimum conditions of ethanol treatment for preparing the indigestible dextrin from $\alpha-amylase$ and amyloglucosidase treated hydrolysate were determined based on the indigestible fraction, dietary fiber content, and yield. Ethanol was added 5-fold by weight to 30%(w/w) enzyme hydrolysate, and the mixture was kept at room temperature for 3 hr. Low molecular weight saccharides containing glucose and high molecular weight saccharides were separated by strongly acidic cation exchange resin. While initial enzyme hydrolysate by $\alpha-amylase$ and amyloglucosidase showed 43.6% of DPI(glucose) and 51.1% of DP4+(maltotetraose and over), the indigestible dextrin collected to 50% of initial enzyme hydrolysate by treatment of cation exchange resin showed 7.1% of DPI(glucose) and 91.2% of DP4+(maltotetraose and over). In conclusion, 44.5% of indigestible fraction of initial enzyme hydrolysate increased to 78.9% after separation of low molecular weight saccharides.

Efficiency Improvement Using Two Balanced Subsets (두 개의 balanced subset을 이용한 효율성 개선)

  • Efficiency is one of the most important factors in cryptographic systems. Cheon et al. proposed a new exponent form for speeding up the exponentiation operation in discrete logarithm based cryptosystems. It is called split exponent with the form $e_1+{\alpha}e_2$ for a fixed element ${\alpha}$ and two elements $e_1$, $e_2$ with low Hamming weight representations. They chose $e_1$, $e_2$ in two unbalanced subsets $S_1$, $S_2$ of $Z_p$, respectively. We achieve efficiency improvement making $S_1$, $S_2$ balanced subsets of $Z_p$. As a result, speedup for exponentiations on binary fields is 9.1% and speedup for scalar multiplications on Koblitz Curves is 12.1%.

Evaluation of the Kit's Efficiency of Alpha Fetoprotein (AFP) Test (Alpha feto protein(AFP)검사 키트의 유효성 평가실험)

  • Purpose Alpha fetoprotein(AFP) is a fetal serum protein that increases in germ cell tumors derived from liver cancer or egg yolk. AFP test has been used for screening of liver cancer, determination of tumor stage, determination of therapeutic effect, and fetal congenital malformations. The purpose of this study was to compare the results of the four kits, identify the advantages and disadvantages of each kit, and select the appropriate kits for our laboratory. Materials and Methods Blood samples were obtained from 89 patients attending the Seoul national university hospital. Experiments were carried out in accordance with manufacturer's instructions of four companies(A, B, C, D). The precision, recovery, linearity, and sensitivity test were performed for each kit. Results In case of the precision within the measurement, the CV value of the C kit was less than 5% at the low, middle, and high concentrations. The A, B and D kit's the CV value was less than 5% at the concentrations except the low concentration. The recovery rates of the A, B, C, and D kits were $100{\pm}15%$, $100{\pm}30%$, $100{\pm}16%$ and $100{\pm}14%$, respectively. All kits showed good linearity. Sensitivity was measured as 0.5 IU/mL for A, 0.4 IU/mL for B, 0.98 IU/mL for C, and 0.3 IU/mL for D. Conclusion The CV values of the four kits were within 10%, and the correlation coefficients were close to 1 for $R^2=0.978$, $R^2=0.992$ and $R^2=0.8957$. As a result, they are clinically available. Therefore, each laboratory should select the appropriate kit for their experiment's environment.

Property of Nano-thickness Nickel Silicides with Low Temperature Catalytic CVD (Catalytic CVD 저온공정으로 제조된 나노급 니켈실리사이드의 물성)

  • 10 nm thick Ni layers were deposited on 200 nm $SiO_2/Si$ substrates using an e-beam evaporator. Then, 60 nm or 20 nm thick ${\alpha}$-Si:H layers were grown at low temperature (<$200^{\circ}C$) by a Catalytic-CVD. NiSi layers were already formed instantaneously during Cat-CVD process regardless of the thickness of the $\alpha$-Si. The resulting changes in sheet resistance, microstructure, phase, chemical composition, and surface roughness with the additional rapid thermal annealing up to $500^{\circ}C$ were examined using a four point probe, HRXRD, FE-SEM, TEM, AES, and SPM, respectively. The sheet resistance of the NiSi layer was 12${\Omega}$/□ regardless of the thickness of the ${\alpha}$-Si and kept stable even after the additional annealing process. The thickness of the NiSi layer was 30 nm with excellent uniformity and the surface roughness was maintained under 2 nm after the annealing. Accordingly, our result implies that the low temperature Cat-CVD process with proposed films stack sequence may have more advantages than the conventional CVD process for nano scale NiSi applications.

Effects of an Agro-healing Program on Promoting Mental Health of the Middle-aged

  • Background and objective: This study was conducted with 10 men and women in their 50s-60s to investigate the effect of agro-healing activities on the improvement of mental health. Methods: The experimental group participated in total 8 sessions of agro-healing activities, once a week for 2 hours each, at a care farm in Wanju-gun. Physiological measurements were taken with an electroencephalogram (EEG), Salivettes samples, and blood pressure before and after the activities. Results: As a result of analyzing the changes in brainwaves of the experimental group before and after agro-healing activities, relative slow alpha (RSA), relative fast alpha (RFA), and ratio of alpha to high beta (RAHB), the indices of stability and relaxation, increased after the program with statistical significance. Also, the ratio of SMR to theta (RST) of the attention index increased on the right frontal lobes, temporal lobes, and left occipital lobes, and relative low beta (RLB) increasd on the frontal lobes, temporal lobes, and occipital lobes after the program with statistical significance. The sympathetic nervous system activity, which is a stress index, decreased after the program, whereas the parasympathetic nervous system activity, which is a relaxation index, increased, showing statistical significance (p < .05). As a result of analyzing the changes in blood pressure after the program, systolic blood pressure and diastolic blood pressure decreased from prehypertension to normal blood pressure, showing statistical significance. SThese results indicate that participating in agro-healing activities at a care farm for the 50-60s helps reduce stress and improve stability and relaxation as well as attention. Conclusion: Thus, developing and applying customized agro-healing programs for participants will have a positive effect on brain activity and psychophysiological improvement by relieving tension and stress. However, there are limitations in generalizing the results of this study since most of agro-healing farms have low accessibility that leads to a low level of participants.

Effect of Dietary Protein on the Changes of Lipoprotein Fractions in Carbon Tetrachloride-Treated Rats (식이성 단백질 함량에 따른 흰쥐에 사염화탄소 투여시 Lipoprotein 분획비의 변동)

  • To evaluate an effect of dietary protein on lipoprotein profile serum of carbon tetrachloride-treated rats, carbon tetrachloride (50% in olive oil) was twice given at 0.1ml/100g body weight at intervals of 24hours to the male rats and then the degree of liver damage in carbon tetrachloride-treated animals fed a low protein diet was compared with that fed a high protein diet. The increasing rate of liver weight/body weight and the serum levels of alanine aminotransferase in carbon tetrachloride-treated rats to the control group were higher in rats fed high protein diet than those fed low protein diet. In the serum levels of lipid (total lipid, total cholesterol and triglyceride) remarkable differences were not found between low protein diet group and high protein diet group. But these serum lipids in carbon tetrachloride-treated rats were decreased and the decreasing rate of serum lipids to control group were higher in carbon tetrachloride-treated rats fed high protein diet than those fed low protein diet. Under the animal model as identified by the present data herein, serum pre $\beta$-lipoprotein and $\alpha$-lipoprotein fractions were decreased in carbon tetrachloride-treated rats, but the serum levels of $\beta$-lipoprotein were rather increased in the both group by the injection of carbon tetrachloride. Especially, the decreasing rate of $\alpha$-lipoprotein fraction was higher in $CCl_4$-treated rats fed a high protein diet than those fed a low protein diet to its control group and the increasing rate of serum $\beta$-lipoprotein fraction was also higher in $CCl_4$-treated rats fed high protein diet than those fed low protein diet.

MALDI Mass Spectrometric Analysis of Nonderivatized Steroids Using Cyclodextrin-supported 2,5-Dihydroxybenzoic Acid as Matrix

  • Sex hormones are important metabolites in vertebrates' development and reproduction. For rapid screening sex hormones, matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) is one of the promising analytical platforms, but MALDI MS faces many challenges in detecting steroids such as low ionization efficiency and matrix background interference. One potential strategy to overcome matrix interference in the low m/z region is using a cyclodextrin (CD)-supported matrix for steroid analysis since CD-supported matrixes are known to effectively suppress matrix-related ion signals. In this study, we aimed to find the optimal CD-supported matrix for the analysis of the nonderivatized sex steroids. Our results showed that the ${\alpha}CD$-supported 2,5-dihydroxybenzoic acid (DHB) matrix efficiently ionized all three major classes of sex hormones, estrogens, androgens, and progestagens, with low or no matrix background and also with high sensitivity. In addition, the ${\alpha}CD$-supported DHB matrix mainly generated molecular ions or protonated ions of sex hormones, and this enabled us to obtain information-rich tandem mass spectra which potentially lead to unambiguous identification of steroid species from complex metabolite mixtures.

Lipid Peroxidation and Vitamins E and A Levels in Tissues of Rats Fed Fish Oil or Soybean Oil Supplemented with Vitamin E (비타민 E 수준을 달리한 어유 또는 대두유를 먹인 흰쥐 조직의 지질과산화와 비타임 E 및 A 상태)

  • To investigate effects of dietary fish oil and vitamin E level on the tissue levels of vitamin E and vitamin A and to see which tissue is sensitive to lipid peroxidizability, male Sprague-Dawley rats were fed experimental diets composed of either menhaden oil or soybean oil nad either low(equivalent to 17 mg $\alpha$-tocopherol) or high (equivalent to 140mg $\alpha$-tocopherol) vitamin E level for 4 weeks. Palsma TBARS per mg lipid was significantly elevated in rats fed fish oil with low vitamin E level compared to soybean oil-fed rats. TBARS levels of liver, heart, kidney and liver microsomes were also increased by feeding fish oil with low vitamin E level. Plasma TBARS level was significantly correlated with TBARS levels of liver, heart, kidney and liver microsome. Plasma vitamin E level of groups with vitamin E supplementation was elevated significantly as compared to the those without vitamin E supplementation, whereas vitamin E levels of liver, heart and kidney were not changed significantly. Plasma TBARS was negatively correlated with plasma vitamin E(r=0.5763, P<0.001) and A(r=-0.4523, P<0.01) and seems to be a good indicator of in vivo lipid peroxidative stress.

Prediction of negative peak wind pressures on roofs of low-rise building

  • In this paper, a probability distribution which is consistent with the observed phenomenon at the roof corner and, also on other portions of the roof, of a low-rise building is proposed. The model is consistent with the choice of probability density function suggested by the statistical thermodynamics of open systems and turbulence modelling in fluid mechanics. After presenting the justification based on physical phenomenon and based on statistical arguments, the fit of alpha-stable distribution for prediction of extreme negative wind pressure coefficients is explored. The predictions are compared with those actually observed during wind tunnel experiments (using wind tunnel experimental data obtained from the aerodynamic database of Tokyo Polytechnic University), and those predicted by using Gumbel minimum and Hermite polynomial model. The predictions are also compared with those estimated using a recently proposed non-parametric model in regions where stability criterion (in skewness-kurtosis space) is satisfied. From the comparisons, it is noted that the proposed model can be used to estimate the extreme peak negative wind pressure coefficients. The model has an advantage that it is consistent with the physical processes proposed in the literature for explaining large fluctuations at the roof corners.
