Attenuated Neuropathic Pain in CaV3.1 Null Mice

  • To assess the role of $\alpha_{1G}$ T-type $Ca^{2+}$ channels in neuropathic pain after L5 spinal nerve ligation, we examined behavioral pain susceptibility in mice lacking $Ca_{V}3.1$ (${\alpha}_{1G}{^{-/-}}$), the gene encoding the pore-forming units of these channels. Reduced spontaneous pain responses and an increased threshold for paw withdrawal in response to mechanical stimulation were observed in these mice. The ${{\alpha}_{1G}}^{-/-}$ mice also showed attenuated thermal hyperalgesia in response to both low-(IR30) and high-intensity (IR60) infrared stimulation. Our results reveal the importance of ${\alpha}_{1G}$ T-type $Ca^{2+}$ channels in the development of neuropathic pain, and suggest that selective modulation of ${\alpha}_{1G}$ subtype channels may provide a novel approach to the treatment of allodynia and hyperalgesia.

Study of Antimicrobial Activity of New Zealand's Tea Tree Essential Oil, Grapefruit Seed Extract and its major Component.

  • Manuka oil sometime named New Zealand's tea tree oil is soluble in oil and come from nature. The $\alpha$-pinene extracted from Manuka oil and R-limonene which is one of the component of extracted Citrex from Grapefruit were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion and broth dilution methods were used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus is similar when the concentration of Manuka oil and $\alpha$-pinene is $10{\mu}l$. However, Antimicrobial activity of Manuka oil for Escherichia coli, Staphylococcus aureus is better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene is low. Antimicrobial activity of Citrex is superior to that of R-limonene. The proper ratio of Maunka oil and Citrex can improve the antimicrobial activity. The proper ratio obtained from studies was 75% of Maunka oil and 25% Citrex for Escherichia coli, 25% of Maunka oil and 75% Citrex for Staphylococcus aureus.

Effects of CoCl2 on multi-lineage differentiation of C3H/10T1/2 mesenchymal stem cells

  • Mesenchymal stem cells (MSCs) in the bone marrow and other somatic tissues reside in an environment with relative low oxygen tension. Cobalt chloride ($CoCl_2$) can mimic hypoxic conditions through transcriptional changes of some genes including hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$) and vascular endothelial growth factor (VEGF). This study evaluated the potential role of $CoCl_2$ preconditioning on multi-lineage differentiation of C3H/10T1/2, a murine MSC line to understand its possible molecular mechanisms in vitro. $CoCl_2$ treatment of MSCs markedly increased HIF-$1{\alpha}$ and VEGF mRNA, and protein expression of HIF-$1{\alpha}$. Temporary preconditioning of MSCs with $CoCl_2$ induced up-regulation of osteogenic markers including alkaline phosphatase, osteocalcin, and type I collagen during osteogenic differentiation, followed by enhanced mineralization. $CoCl_2$ also increased chondrogenic markers including aggrecan, sox9, and type II collagen, and promoted chondrocyte differentiation. $CoCl_2$ suppressed the expression of adipogenic markers including $PPAR{\gamma}$, aP2, and $C/EBP{\alpha}$, and inhibited adipogenesis. Temporary preconditioning with $CoCl_2$ could affect the multi-lineage differentiation of MSCs.

The Solubilization Behavior of DOPE-Immunoliposomes with Immunoglobulin G(IgG) by Added Bile Salts (Immunoglobulin G(IgG)를 함유한 DOPE 리포솜의 제조와 담즙산염에 의한 용해 특성)

  • The effects of bile salts (BS) on the stability of dioleoylphosphatidylethanolamine (DOPE) liposomes were investigated, observing apparent absorbance of vacant liposomes and calcein release from entrapped liposomes. Unilamellar liposomes were prepared by using a small quantity of palmitoly-immunoglobulin G(IgG) ($2.5{\times}10^{-4}$ mo1/lipid mol) to stabilize the bilayer phase of the unsaturated DOPE which by itself does not form stable liposomes. The destabilization of PE immunoliposomes by papain, clearly demonstrates that the IgG is essential for stabilization of PE bilayer. Approximately 4% of the entrapped calcein was released from the PE liposomes after 1 hr from liposome formation. Calcein release and absorbance of liposomes depended on the BS/lipid ratio because of the solubilization of lipid molecule in bilayer and the formation of mixed micelles. At very low BS concentrations, the incorporation of BS induced BS/lipid aggregates in the outer vesicles monolayer, while high BS concentrations, mixed micelles were formed. Chelate and its conjugates as $3{\alpha},\;7{\alpha},\;12{\alpha}-trihydroxy$ BS induce the concentration of the $3{\alpha}$, $12{\alpha}-dihydroxy$ BS at half-maximal solubilization of immunoliposomes to approximately 2.5-, or 5-fold. Conjugation of BS with glycine or taurine slightly enhanced their capacities to perturb membranes.

Enhanced Macrophage Antitumor Effects of Protein A in Combination with $IFN-{\Upsilon}$

  • In this study we examined the potential for the synergistic augmentation of the antitumor activity of inflammatory mouse peritoneal macrophages by stimulation with protein A combined with $IFN-\gamma$. The moderate augmentative effect induced by preincubation with protein A was demonstrated to be concentration-dependent, whereas IFN-, had a very low activating effect. Following preincubation with both protein A and $IFN-\gamma$, a marked enhancement of macrophage activity was noted. In addition, based on the utilization of neutralizing antibody to TNF-$\alpha$ or the inhibition of NO Production, TNF-$\alpha$ and NO were proven to be involved as mediators during the activation of tumoricidal macrophages by protein A in combination with $IFN-\gamma$. We also demonstrated that supernatants from macrophages treated with protein A plus $IFN-\gamma$ contained both TNF-$\alpha$ and NO at markedly increased levels. Thus, tumor cell lysis in the combined system was mediated via TNF-$\alpha$ or NO. These results demonstrate the synergistic effects on mouse pertioneal macrophage function of protein A in combination with $IFN-\gamma$ and suggest that combinations of such agents may serve as the basis for future in vivo immunotherapy.

  • This paper examines the crack growth behavior of 7075-T651 aluminum alloy under high-low block loading condition. The cantilever beam type specimen with a chevron notch is used in this study. The crack growth and closure are investgated by compliance method. The applied initial stress ratios are R=-0.5, R=0.0 and R=0.25 Crack length($\alpha$), effective stress intensity factor range(ΔKeff), ratio of effective stress intensity factor range(U) and crack growth rate(d$\alpha$/dN) etc. are inspected fracture mechanics estimate.

  • This study was performed to find the atmospheric concentrations of organochlorine pesticides (OCPs) and polychlorinated biphenyls (PCBs) at four different heights (l0, 50, 150 and 1,500 cm) using low volume polyurethane foam (PUF) plug air sampler in semi-rural area. ${\alpha}-/{\beta}-/{\gamma}-HCH$ with low molecular weights and Koa (octanol-air partitioning coefficient) in OCPs were similarly high concentrations at all heights, but the other OCPs (p,p'-DDE, p,p'-DDD, p,p'-DDT, Heptachlor epoxide, ${\gamma}$-CHL, ${\alpha}$-CHL, Trans-nonachlor) with high molecular weights and Koa decreased with increasing heights. However, the concentrations of PCBs increased with increasing height.

Comparison of evaluation methods for measuring pressure of compressionwear (컴프레션웨어의 압박감 측정 방법 비교 연구)

  • The aim of this study was comparing measuring tools for detecting physical comfortness with variation of garment pressure. The measuring tools for detecting physical comfortness were EEG and survey questionnaire. Two low-pressure compressionwears and a commercial compressionwear (girdle) were tested. Results showed that the questionnaire survey well detected suffocation or motion comfortness. But it did not discrete the appropriate tightness of the compressionwears. The results of EEG analysis show that the absolute power of ${\alpha}$-wave value was elevated with the low-pressure compressionwears. It also showed lower stress value. The high-pressure compressionwear presented decreased absolute power of ${\alpha}$-wave value. It showed higher stress value. These results implicate that EEG can appropriatly indicate the change of physical comfortness of compressionwear. The appropriate tightness of compressionwear can be measured with EEG analysis rather than survey questionnaire.

Synthesis and Characterization of Tin Nitride Thin Films Deposited by Low Nitrogen Gas Ratio

  • Thin nitride thin films were synthesized by reactive radio-frequency magnetron sputtering in the ultra high vacuum (UHV) chamber. To control the characteristics of thin films, tin nitride thin films were obtained various argon and nitrogen gas mixtures, especially low nitrogen gas ratios. Tin nitride thin films were analyzed with alpha step, scanning electron microscopy (SEM), X-ray diffraction (XRD), X-ray photoelectron spectroscopy (XPS), and 4 point probe measurement. The result of alpha step and SEM showed that the thickness of thin nitride thin films were decreased with increasing nitrogen gas ratios. The metallic tin structure was decreased and the amorphous tin nitride structure were observed by XRD with higher nitrogen gas ratios. The oxidation state of tin and nitride were studied with high resolution Sn 3d and N 1s XP spectra.
