Characteristic of Aromatic Amino Acid Substitution at α96 of Hemoglobin

  • Replacement of valine by tryptophan or tyrosine at position $\alpha$96 of the $\alpha$ chain ($\alpha$96Val), located in the ${\alpha}_1{\beta}_2$ subunit interface of hemoglobin leads to low oxygen affinity hemoglobin, and has been suggested to be due to the extra stability introduced by an aromatic amino acid at the $\alpha$96 position. The characteristic of aromatic amino acid substitution at the $\alpha$96 of hemoglobin has been further investigated by producing double mutant r Hb ($\alpha$42Tyr$\rightarrow$ Phe, $\alpha$96Val$\rightarrow$Trp). r Hb ($\alpha$42Tyr$\rightarrow$Phe) is known to exhibit almost no cooperativity in binding oxygen, and possesses high oxygen affinity due to the disruption of the hydrogen bond between $\alpha$42Tyr and $\beta$99Asp in the ${\alpha}_1{\beta}_2$ subunit interface of deoxy Hb A. The second mutation, $\alpha$96Val$\rightarrow$Trp, may compensate the functional defects of r Hb ($\alpha$42Tyr$\rightarrow$Phe), if the stability due to the introduction of trypophan at the $\alpha$96 position is strong enough to overcome the defect of r Hb ($\alpha$42Tyr$\rightarrow$Phe). Double mutant r Hb ($\alpha$42Tyr$\rightarrow$Phe, $\alpha$96Val$\rightarrow$Trp) exhibited almost no cooperativity in binding oxygen and possessed high oxygen affinity, similarly to that of r Hb ($\alpha$42Tyr$\rightarrow$Phe). $^1$H NMR spectroscopic data of r Hb ($\alpha$42Tyr$\rightarrow$Phe, $\alpha$96Val$\rightarrow$Trp) also showed a very unstable deoxy-quaternary structure. The present investigation has demonstrated that the presence of the crucible hydrogen bond between $\alpha$42Tyr and $\beta$99Asp is essential for the novel oxygen binding properties of deoxy Hb ($\alpha$96Val$\rightarrow$Trp).

초염기성 사문암 토양 중 세균군집의 계통학적 특성 (Phylogenetic Characteristics of Bacterial Populations Found in Serpentinite Soil)

  • 충남 홍성군 광천 사문암 토양지역의 석면폐광석(KS1)과 인근 토양(KG, KS2)은 pH8.5-9.2를 나타내어 초염기성 토양임이 확인되었다. KSI과 KS2 토양으로부터 통상농도의 alkaline 배지(AL)와 AL 배지를 $10^{-2}$로 희석한 DAL배지를 사용하여 평판법으로 세균수를 측정한 결과 AL 배지에서보다 DAL배지에서 10-100배 이상 높은 계수치를 나타내었다. 초염기성 사문암 토양으로부터 분리된 75균주에대해 통상농도의 AL 배지에서의 중식 유무를 확인한 결과, 통상농도의 AL배지에서 증식 가능한 [AL세균군]과 AL배지에서는 증식이 저해되고 DAL 배지에서만 증식 가능한 [DAL세균군으로 크게 나누었다. DAL세균(42균주)은 $10^{-3}$ AL 배지(약 6mg C/L)에서도 증식 가능한 저영양성세균(oligotrophic bacteria)으로 사문암 토양 중 50% 이상 분포해 있음이 확인되었다. 분리된 75 균주의 16S rDNA 염기서열을 결정하여 계통해석한 결과 proteobacteria $\alpha$-subdivision (3균주), $\beta$-subdivision (7균주), $\gamma$-subdivision (2균주), high G+C gram-positive bacteria (19균주)와 low G+C gram-positive bacteria (14 strains)의 계통군을 나타내었다. 이들 세균중 AL세균군(34균주)은 high G+C gram positive bacteria 에 속하는 streptomyces과 low G+C gram positive bacteria에 속하는 Bacillus로 구성되었다. 한편, DAL세균군(42균주)은 high G+C 및 low G+C gram positive 계통군 이외에도 proteobacteria -subdivision에 속하는 Afipia와 Ralstonia, proteobacteria -subdivision에 속하는 Variovorax, proteobacteria $\beta$-subdivision에 속하는Pseudomonas로 구성되어 계통학적으로 다양한 세균임이 확인되었다.

Effect of pH values and inoculation amounts for α-glucosidase inhibitory activity in mulberry leaf fermentation

  • Mulberry leaves containing 1-deoxynojirimycin (DNJ) have been recognized as a potentially important source for prevent or treat hyperglycemia. However, DNJ content of natural mulberry leaf are as low as 0.1%. Thus, the most effective method for increasing ${\alpha}$-glucosidase inhibitory activity with the DNJ high-production is needed. In this study, we investigated the influence of ${\alpha}$-glucosidase inhibitory activity according to different pH values (6-9) and inoculation amounts (0.1-0.5%) when Bacillus subtilis cultured on mulberry leaf powder media. We confirmed that ${\alpha}$-glucosidase inhibitory activity was difference according to culture conditions of different pH values, inoculation amounts, and fermentation times. The results of mulberry leaf fermentation according to pH values and inoculation amounts were shown that the optimal conditions for ${\alpha}$-glucosidase inhibitory activity were defined as pH 7 and 9, inoculation amount 0.4%, and incubation until 2 to 4 days. These results can be provided a basic data for the optimal culture conditions increasing ${\alpha}$-glucosidase inhibitory activity from mulberry leaf fermentation.

  • 비정질 WNx 박막이 반응성 스퍼터링법으로 제조되었다. 비정질 형성을 위한 질소의 농도범위는 10~40at%이었다. 비정질 W(sub)67N(sub)33 박막은 1273K에서 1분 동안 급속 열처리되어 저항이 낮은 등축정의 $\alpha$-텅스텐 상과 과잉의 질소로 변태되었다. 이러한 박막의 저항은 순수한 텅스텐 박막과 유사하였다. $\alpha$-텅스텐 상으로부터 방출된 과잉의 질소는 $\alpha$-텅스텐/다결정 실리콘의 계면에 편석되었다. 편석된 질소는 Si$_3$N$_4$나노 결정으로 균일한 확산 장벽층을 형성시켰고, 저항이 높은 텅스텐 실리사이드의 반응을 억제하였다.

$\alpha$-Amylase 저해제 생성균 Streptomyces actuosus DMCJ-49와 Streptomyces minoensis DMCJ-144의 종간 융합에 의한 균주 개량 (Strain Improvement by Interspecific Fusion of Streptomyces actuosus DMCJ-49 and Streptomyces minoensis DMCJ-144 producing $\alpha$-Amylase Inhibitor)

  • Streptomyces actuosus DMCJ-49 and Streptomyces minoensis DMCJ-144 produce the .alpha.-amylase inhibitor. Inerspecific protoplast fusion technique was used to increase the productivity of .alpha.-amylase inhibitor. Four auxotrophic mutants were obtained respectively from two strains by N-methyl-N'-nitor-N-nitrosoguanidine(3mg/ml) treatment. The optimum conditions for the protoplast formation of Streptomyces actuosus DMCJ-49 ade was as follows; 1.2% w/v of glycine, 3mg/ml of lysozyme, and 30 min of lysozyme treatment followed by 36 hr. incubation in the protop-last formation medium. In case of DMCJ-144-his those were 1.2%w/v, 3 mg/ml, 30 minutes and 60 hours, respectively. Regeneration was accomplished with hypertonic soft agar medium that contained 0.4M sucrose, 20mM CaCl$_2$, 50 mM MgCl$_2$ and low levels of phosphate. Fusion of protoplasts carrying different auxotrophic markers was achieved by treatment with polyethylene glycol. The optimum concentration of polyethylene glycol 1450 for the production of recombinants was 40%w/v. When the protoplasts was treated with 40% polyethylene glycol for 30 minutes, the frequency of recombinants was 6.5$\times$$10^{-3}$ and the $\alpha$-amylase inhibition activity of $ade^-his^-$ No. 4, which is the fusant with the most improved activity increased from 33 to 125 I.U./ml.

Isolation and identification of Bacillus megaterium producing Alkaline ${\alpha}$-amylase

  • A bacterial strain, Bacillus megaterium L-49 has been isolated and identified that produces alkaline ${\alpha}$-amylase. The cell is ellipsoidal, about $1.0-1.2{\times}3.0-3.6{\mu}m$ in diameter, Gram-positive, motile, and central partial central. Growth occurs in media containing 7% of NaCl. This strain could utilize D-glucose, lactose, xylose, sucrose, mannose, and maltose, and but it does not utilize D-fructose, and glycogen. Among the various concentrations of saturated ammonium sulfate, the retractation ratio in range of 70 to 100% was about 93%. However, in the case of acetone, it was about 98.7%. EDTA has activating effect and Ca2+ has no effect on alkaline ${\alpha}$-amylase activity. The alkaline ${\alpha}$-amylase has low thermal stability. The optimal temperature for reaction is $50^{\circ}C$. The alkaline ${\alpha}$-amylase activity maintained stabilizing at pH 6-11 and the optimal pH for reaction was 9-10.

  • Aspergillus phoenicis에 의한 progesterone의 전환반응에 대하여 고찰하였다. 본 실험조건에서의 물질대사 결과 $11{\alpha}-hydroxyprogesterone$이 주생성물로 생성 되었다. $11{\alpha}-hydroxyprogesterone$의 농도는 반응시간에 따라 서서히 증가하였으나 40시간 이후부터는 점근값에 도달하였다. 반응액에의 glucose 첨가는 progesterone의 전환반응 속도를 증가시켜 점근값의 농도에 도달하는 시간이 괄목할 만큼 앞당겨졌다. Progesterone의 초기 농도를 증가시키는 것은 전환율이나 반응완결에 소요되는 시간에 큰 변화를 주지 않았으나 전환반응은 progesterone 농도에 대하여 1차반응 형태로 요약될 수는 없었다. 유기용매에 의한 효소 활성의 저하정도는 유기용매의 종류 및 그 농도에 의존하였으며, 낮은 유기용매의 농도에서는 acetone이 가장 높은 전환율을 나타내었다.

Determination of Trace Amounts of Nickel(II) with ${\alpha}-(2-Benzimidazolyl))-{\alpha}'{\alpha}$

  • A method is described for the fluorimetric determination of nickel, based on the formation of $Ni(II)-\alpha-(2-Benzimidazolyl)-\alpha'$, $\alpha''$ -(N-5-Nitro-2-Pyridylhydrazone)-toluene complex in the presence of a non-ionic surfactant. The complex has practically no fluorescence in the absence of surfactant, but the addition of Triton X-100 makes possible the fluorimetric determination of low concentrations of Ni(II) as it enhances the fluorescenceintensity of the complex by up to about 5-fold. This method is very sensitive and selectrive for the direct determination of nickel ion. The optimum conditions are a Triton X-100 concentration of 2.0 mL(5.0%, v/v) and pH $9.0\pm0.2(ammonium$ chloride-ammonia buffer). The fluorescence is measured at 337 nm of emission wavelength under 300 nm of excitation wavelength. The fluorescence intensity is a linear function of the concentration of Ni(II) in the range 5-70 ng/mL, and the detection limit is 2.0 nm/mL. The proposed method has been successfully applied to the determination of trace amounts of Ni(II) in food and human hair samples.

  • 핵연료 피복관 제조 및 사용 시에 필요한 자료를 얻기 위하여 지르칼로이-4재료에서 가공과 열처리의 영향을 조사하였다. 지르칼로이-4 재료는 저가공도에서는 경도가 급격히 증가하지만 10% 이상 가공도 에서는 점진적으로 증가하였다. 냉간가공된재료의 재결정은 가공도가 30%, 60%, 80%로 증가함에 따라서 64$0^{\circ}C$, 59$0^{\circ}C$, 555$^{\circ}C$에서 각각 완료되었다. $\beta$구역에서 열처리한후에 노냉, 공냉, 수냉을하였을 때 냉각속도가 증가함에 따라서 경도는 증가하고, 조직은 coarse widmanstatten($\alpha$) $\longrightarrow$ fine parallel plate($\alpha$) $\longrightarrow$ martensite($\alpha$$^{'}$)순으로 변화한다. 변화한다.

  • The purpose of this study is to prove how magnetic field (MF) acts on sympathetic neuro-transmissions using thermal response. Mice were divided into two groups and each one was exposed to MF (20 G, 24 hrs) or sham. Every vehicle or drugs were treated a half hour before the thermal response test. The pain threshold was lowered by MF (20 G, 24 hrs) alone. This reduction of pain threshold by MF was not blocked by a single treatment of $\alpha$-receptor antagonist (prazosin), $\alpha$$_2$-receptor agonist (clonidine, guanabenz), $\beta$$_1$-receptor antagonist (atenolol) or $\beta$$_1$,$\beta$$_2$-receptor antagonist (propranolol). But administration of $\alpha$$_2$-receptor antagonist (yohimbine) completely inhibited the decrease in pain threshold by MF. Moreover, it increased by high dose of yohimbine over normal condition. These results suggest that MF acts on sympathetic nerve terminal to induce hyperalgesia, in which pre-synaptic az receptor might be involved.
