The Longevity Properties of 1,2,3,4,6-Penta-O-Galloyl-β-D-Glucose from Curcuma longa in Caenorhabditis elegans

  • Here in this study, we isolated 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucose (PGG) from Curcuma longa L. and elucidated the lifespan-extending effect of PGG using Caenorhabditis elegans model system. In the present study, PGG demonstrated potent lifespan extension of worms under normal culture condition. Then, we determined the protective effects of PGG on the stress conditions such as thermal and oxidative stress. In the case of heat stress, PGG-treated worms exhibited enhanced survival rate, compared to control worms. In addition, PGG-fed worms lived longer than control worms under oxidative stress induced by paraquat. To verify the possible mechanism of PGG-mediated increased lifespan and stress resistance of worms, we investigated whether PGG might alter superoxide dismutase (SOD) activities and intracellular ROS levels. Our results showed that PGG was able to elevate SOD activities of worms and reduce intracellular ROS accumulation in a dose-dependent manner.

  • Clonorchis sinensis is a biological carcinogen inducing human cholangiocarcinoma, and clonorchiasis is one of the important endemic infectious diseases in East Asia. The present study investigated survival longevity of C. sinensis adult worms in various in vitro conditions to find the best way of keeping the worms longer. The worms were maintained in 0.85% NaCl, 1${\times}$PBS, 1${\times}$Locke's solution, RPMI-1640, DMEM, and IMDM media, and in 1${\times}$Locke's solution with different supplements. All of the worms died within 3 and 7 days in 0.85% NaCl and 1${\times}$PBS, respectively, but survived up to 57 days in 1${\times}$Locke's solution. The worms lived for 106 days in DMEM, and 114 days in both RPMI-1640 and IMDM media. The survival rate in RPMI-1640 medium was the highest (50%) compared to that in DMEM ($20{\pm}10%$) and in IMDM ($33.3{\pm}25.2%$) after 3 months. The 1${\times}$Locke's solution with 0.005% bovine bile supplement showed increased duration of maximum survival from 42 days to 70 days. Higher concentration of bile supplements than 0.005% or addition of glucose were disadvantageous for the worm survival. The worms died rapidly in solutions containing L-aspartic acid, L-glutamic acid, and adenine compared to L-arginine, L-serine, and L-tryptophan. In conclusion, the 1${\times}$Locke's solution best supports the worms alive among inorganic solutions for 57 days, and the RPMI-1640 medium maintains living C. sinensis adults better and longer up to 114 days in vitro than other media.

곤달비의 수명 연장 효과 (Lifespan Extending Effects of Ligularia stenocephala)

  • Ligularia stenocephala has a wide range of types of constituents with various pharmacological properties. Here in this study, we examined the effect of methanolic extract of L. stenocephala (MLS) on the lifespan and stress tolerance using Caenorhabditis elegans model system. We found that lifespan of wild-type worms was significantly lengthened in the presence of MLS in a dose dependent manner. MLS also elevated the tolerance of worms against osmotic, heat shock, and oxidative stress. We also demonstrated in vivo antioxidant capacity of MLS by checking intracellular reactive oxygen species levels as well as antioxidant enzyme activities such as catalase and superoxide dismutase. We further investigated several aging-related factors, including pharyngeal pumping rate and body length. Here, we showed that MLS exerts longevity effect independent of both factors. In addition, body movement of aged worms was significantly elevated, suggesting MLS could enhance healthspan as well as lifespan.

  • 벼검은줄오갈병 매개충인 애멸구의 기주식물 선호성은 실내유균접종과 포장상태에서 공히 옥수수가 벼보다 낮으나 발병율은 아주 높았다. 바이러스가 매개충의 생태에 미치는 영향은 보독충이 무독충보다 성충의 생존일수, 부화약충수 및 성충율에서 심한 장애를 나타냈다. 벼에서는 감염초기부터 후기까지 건물중이 이병주가 건전주보다 낮았다. 그러나 옥수수에서는 감염초기에 이병주가 건전주보다 건물중이 높았다. 한천내확산법 및 미량심강법에 의한 감염바이러스의 정량결과, 항혈청과의 반응정도는 옥수수잎, 벼줄기, 벼잎, 옥수수줄기에서 부분순화한 항원바이러스의 순으로 나타났다. 이병기주식물의 phloem gall 형성에서 저온처리의 효과가 인정되었다.

연무 종류별 강수 발생시간 관측 특성 및 에어로졸-강수 연관성 분석 (Observed Characteristics of Precipitation Timing during the Severe Hazes: Implication to Aerosol-Precipitation Interactions)

  • Characteristics of precipitation response to enhanced aerosols have been investigated during the severe haze events observed in Korea for 2011 to 2016. All 6-years haze events are classified into long-range transported haze (LH: 31%), urban haze (UH: 28%), and yellow sand (YS: 18%) in order. Long-range transported one is mainly discussed in this study. Interestingly, both LH (68%) and YS (87%) appear to be more frequently accompanied with precipitation than UH (48%). We also found out the different timing of precipitation for LH and YS, respectively. The variations of precipitation frequency for the LH event tend to coincide with aerosol variations specifically in terms of temporal covariation, which is in contrast with YS. Increased aerosol loadings following precipitation for the YS event seems to be primarily controlled by large scale synoptic forcing. Meanwhile, aerosols for the LH event may be closely associated with precipitation longevity through changes in cloud microphysics such that enhanced aerosols can increase smaller cloud droplets and further extend light precipitation at weaker rate. Notably, precipitation persisted longer than operational weather forecast not considering detailed aerosol-cloud interactions, but the timescale was limited within a day. This result demonstrates active interactions between aerosols and meteorology such as probable modifications of cloud microphysics and precipitation, synoptic-induced dust transport, and precipitation-scavenging in Korea. Understanding of aerosol potential effect on precipitation will contribute to improving the performance of numerical weather model especially in terms of precipitation timing and location.

  • 현대인들은 편리한 생활방식, 영양과잉, 운동부족 등으로 비만문제에 심각하게 노출되어 있다. 장수를 원하는 인간의 욕망이 클수록 비만문제는 중요한 사회문제가 되고 있다. 사람들은 비만을 개선하기 위해 동네 공원이나 산책로, 등산로 등을 걸으며 운동을 한다. 지자체는 주민들을 위해 산책로나 등산로에 운동표지판을 세워 각종 운동정보를 제공하며 운동을 권장한다. 그러나 기존의 운동 표지판은 붙박이 고정식이라 다양한 운동정보나 건강정보를 제공하지 못한다. 그래서 본 연구는 헬스케어 IT시대에 맞춰 스마트 헬스 스크린을 통한 과학적 운동관리를 제안한다. IT를 기반으로 한 스마트 헬스 스크린은 실시간으로 걸음수, 칼로리소모량 심박수 등의 운동정보를 제공한다. 스마트 헬스 스크린이 제공하는 체계적이고 과학적인 운동관리는 지역주민들에게 개개인에게 맞는 운동량과 강도를 조절할 수 있도록 도와준다. 더불어 특별한 비용 없이 걷기만으로도 건강관리와 비만관리가 가능하게 해준다. 우리의 연구가 비만해결에 작은 도움이 되길 바란다.

Ribgrass Mosaic Tobamovirus Occurred on Chinese Cabbage in Korea

  • A tobamovirus, Ribgrass mosaic virus (RMV), was identified newly from chinese cabbage (Brassica campestris L. pekinensis) in Korea. Virus disease incidence of RMV on chinese cabbage was 37.9% in alpine area on August in 1993. RMV induced the symptoms of necrotic ring spots, necrotic streak on midrib and malformation. RMV, Ca1 and Ca3 isolate, could infect 35 species out of 45 plants including Chenopodium amaranticolor. Physical properties of RMV Ca1 isolate were very stable as 10.8 over for dilution end point, $95^{\circ}C$ for temperature inactivation point and 18 weeks for longevity in vitro. RMV had the soil transmission rate of 75.0% for the chinese cabbages, 'Chunhawang' and 'Seoul' cultivars. The purified virions of RMV had the typical ultraviolet absorption spectrum of maximum at 260 nm and minimum at 247 nm. RMV of Ca1 isolate was related serologically with antisera of Tobacco mosaic virus (TMV)-Cym, TMV-O and Pepper mottle virus, but not related with antiserum of Odontoglossum ring spot virus. coat protein gene of RMV-Ca1, sized 473 nucleotides, encoded 158 amino acid residues. Nucleotide identity of RMV-Ca1 CP gene was 96.4% with RMV-Shanghai (GenBank accession No. of AF185272) from China and 96.0% with RMV-Impatiens (GenBank accession No. of AM040974) from Germany. Identity of amino acids between RMV-Ca1 and the two RMV isolates was 96.8%. Specific three primers were selected for rapid and easy genetic detection of RMV using Virion Captured (VC)/RT-PCR method.

The influence of nanofillers on the properties of ethanol-solvated and non-solvated dental adhesives

  • Objectives: The aim of this study was to evaluate the influence of different concentrations of nanofillers on the chemical and physical properties of ethanol-solvated and non-solvated dental adhesives. Materials and Methods: Eight experimental adhesives were prepared with different nanofiller concentrations (0, 1, 2, and 4 wt%) and 2 solvent concentrations (0% and 10% ethanol). Several properties of the experimental adhesives were evaluated, such as water sorption and solubility (n = 5, 20 seconds light activation), real-time degree of conversion (DC; n = 3, 20 and 40 seconds light activation), and stability of cohesive strength at 6 months (CS; n = 20, 20 seconds light activation) using the microtensile test. A light-emitting diode (Bluephase 20i, Ivoclar Vivadent) with an average light emittance of $1,200mW/cm^2$ was used. Results: The presence of solvent reduced the DC after 20 seconds of curing, but increased the final DC, water sorption, and solubility of the adhesives. Storage in water reduced the strength of the adhesives. The addition of 1 wt% and 2 wt% nanofillers increased the polymerization rate of the adhesives. Conclusions: The presence of nanofillers and ethanol improved the final DC, although the DC of the solvated adhesives at 20 seconds was lower than that of the non-solvated adhesives. The presence of ethanol reduced the strength of the adhesives and increased their water sorption and solubility. However, nanofillers did not affect the water sorption and strength of the tested adhesives.

애반딧불이(딱정벌레목: 반딧불이과)의 생육 특성 (Bionomical Characteristics of Luciola lateralis (Coleoptera: Lampyridae) in Mass Breeding)

  • 우리나라에서 서식하는 애반딧불이의 생육 특성을 조사한 결과, $23^{\circ}C$, R.H. 80%에서 사육에서 성충 수명은 암컷이 $17.5{\pm}0.7$일, 수컷이 $20.6{\pm}1.0$일으로 수컷이 다소 높았으나, 사육밀도별 유의차는 없었다. 성충 출현 후 최초 산란일은 1쌍 $2.8{\pm}1.5$일, 2쌍 $2.5{\pm}1.5$일, 3쌍 $1.7{\pm}0.7$일, 4쌍 $1.3{\pm}0.5$일로 사육 밀도가 증가함에 따라 산란전기가 짧아졌다. 산란수는 평균 $248.9{\pm}80.7$개를 산란하며, 사육밀도와는 유의성이 없었다. 일별 산란은 산란 시작 1일째가 20.8%, 2일째 17.3%, 3일째 14.3%, 4일째 12.7%, 5일째 7.0%의 순으로 산란 시작 1일째가 가장 높고 시간이 경과함에 따라 점차 낮아졌다. 난기간은 $21.8{\pm}0.7$일이고, 부화율은 $95.5{\pm}4.8%$이었다. 유충 령기 구분은 탈피각과 유충반문으로 조사한 바 9령으로 확인되었고 1령 $1.0{\pm}0.1$일, 2령 $3.3{\pm}0.2$일, 3령 $10.2{\pm}1.0$일, 4령 $6.8{\pm}0.5$일, 5령 $6.9{\pm}2.0$일, 6령 $15.4{\pm}4.1$일, 7령 $18.3{\pm}5.9$일, 8령 $25.8{\pm}8.7$일, 9령 $31.2{\pm}13.2$일이었다. 성숙한 9령 유충은 $2.8{\pm}0.8$일 동안 용실을 만들고, 전용기간과 용기간은 $4.6{\pm}0.9$일과 $5.6{\pm}0.7$일이었으며, 성충이 된 후 약 $3.8{\pm}0.4$일 동안 뒷날개를 굳힌 후 용실을 뚫고 탈출하였다

  • 본 연구는 갈색날개매미충을 생물적 방제인자로 활용하기 위해 알 기생 천적인 날개매미충알벌의 발육 특성을 조사하였다. 날개매미충알벌의 온도별 발육기간은 25℃에서 알은 3.8일, 유충 11.1일 및 번데기 16.3일, 30℃에서는 알 3.4일, 유충 7.8일 및 번데기 15.3일이었다. 유충 기간을 제외하고 온도에 따라 유의미한 발육 기간의 차이를 보이지 않았다. 다만 알~번데기까지 전 발육기간은 25℃에 비해 30℃에서 발육기간이 짧아 온도에 따라 유의미한 차이를 보였다. 하루 중 날개매미충알벌의 우화는 불이 켜져 있는 명 기간에 모든 개체가 성충으로 우화하였다. 우화율이 가장 높은 시간대는 10:00~12:00 사이였고(34.5%), 그 다음으로 12:00~14:00 사이였다(22.99%). 암 기간(00:00~08:00)에서는 성충으로 우화하는 개체는 관찰되지 않았다. 날개매미충알벌 성충의 성비는 암컷이 75.8%, 수컷이 24.2%로 암수 비율은 약 3 : 1이었다. 성충의 수명은 20℃에서 암컷과 수컷이 각각 49.0일, 44.0일, 25℃에서 27.6일, 28.4일 및 30℃에서 18.0일, 14.0일로 조사한 온도 중 암수 모두 온도가 낮은 20℃에서 수명이 길었고, 온도가 높을수록 수명은 짧아지는 경향을 보였다. 암컷 성충은 00:00~02:00 사이를 제외하고 모든 시간대에 걸쳐 산란하였고, 산란수가 가장 많은 시간대는 20:00~22:00 사이로 26.1%를 차지하였고, 그 다음으로는 10:00~12:00 사이에 15.7%로 조사되었다. 날개매미충알벌은 수컷을 생산하는 단위생식을 하는 것으로 조사되었다. 따라서 이상의 결과로 날개매미충알벌은 갈색날개매미충을 방제하는 중요한 생물적 방제 인자로 활용할 수 있을 것으로 생각된다.
