Aim: Individual differences in chemosensitivity and clinical outcome in non-small cell lung cancer (NSCLC) patients treatment with platinum-based chemotherapy may be due to genetic factors. Our study aimed to investigate the prognostic role of GSTP1, XRCC1 and XRCC3 in NSCLC patients treated with chemotherapy. Methods: A total of 460 cases were consecutively selected from The Affiliated Hospital of Nantong University between Jan. 2003 to Nov. 2006, and all were followed-up until Nov. 2011. Genotyping of GSTP1 Ile105Val, XRCC1 Arg194Trp, XRCC1 Arg399Gln and XRCC3 Thr241Met was conducted by duplex polymerase-chain-reaction with confronting-two-pair primer methods. Results: Patients with GSTP Val/Val exhibited a shorter survival time, and had a 1.89 fold greater risk of death than did those with the IIe/IIe genotype. For XRCC1 Arg194Trp, the variant genotype Trp/Trp was significantly associated with a decreased risk of death from NSCLC when compared with the Arg/Arg. Individuals carrying XRCC1 399Gln/Gln genotype had a longer survival time, with a lowered risk of death from NSCLC. Conclusion: This study indicated that GSTP1 Ile105Val, XRCC1 Arg194Trp and XRCC1Arg399Gln genes have a role in modifying the effect of platinum-based chemotherapy for NSCLC patients in a Chinese population. Our findings provide information for therapeutic decisions for individualized therapy in NSCLC cases.
Background: Reactive oxygen and nitrogen are produced by rheumatoid arthritis (RA) synovial tissue and can induce mutations in key genes. Normally, this process is prevented by a DNA mismatch repair (MMR) system that maintains sequence fidelity. Key members of the MMR system include MutS${\alpha}$ (comprised of hMSH2 and hMSH6), which can sense and repair single base mismatches and 8-oxoguanine, and MutS${\beta}$ (comprised of hMSH2 and hMSH3), which repairs longer insertion/deletion loops. Methods: To provide further evidence of DNA damage, we analyzed synovial tissues for microsatellite instability (MSI). MSI was examined by PCR on genomic DNA of paired synovial tissue and peripheral blood cells (PBC) of RA patients using specific primer sequences for 5 key microsatellites. Results: Surprisingly, abundant MSI was observed in RA synovium compared with osteoarthritis (OA) tissue. Western blot analysis of the same tissues for the expression of MMR proteins demonstrated decreased hMSH6 and increased hMSH3 in RA synovium. To evaluate potential mechanisms of MMR regulation in arthritis, fibroblast-like synoviocytes (FLS) were isolated from synovial tissues and incubated with the nitric oxide donor S-nitroso-N-acetylpenicillamine (SNAP). Western blot analysis demonstrated constitutive expression of hMSH2, 3 and 6 in RA and OA FLS. When FLS were cultured with SNAP, the RA synovial pattern of MMR expression was reproduced (high hMSH3, low hMSH6). Conclusion: Therefore, oxidative stress can relax the DNA MMR system in RA by suppressing hMSH6. Decreased hMSH6 can subsequently interfere with repair of single base mutations, which is the type observed in RA. We propose that oxidative stress not only creates DNA adducts that are potentially mutagenic, but also suppresses the mechanisms that limit the DNA damage.
본 연구에서는 고경질 도막재로 PET 필름을 접합하는 시공방법에 있어서 PET 필름의 표면처리 조건 및 겹침이음 길이에 따른 접합특성 및 내구특성을 검토하였는데, 그 결과 무처리 보다 코로나 방전 처리에 의해 접촉각, 접합 인장강도, 벗김저항성이 개선되었다. 특히 시험체 E (코로나 방전+프라이머+강접접착제+폴리에스터 부직포)에서 가장 높은 것으로 나타났으며, 특히 겹침 이음길이 15mm 이상일 때 16주간 장기간 열화처리에도 안정적인 접합성능을 확보하는 것으로 나타나 방수재료로서 수밀성을 확보할 것으로 판단된다.
체세포배의 생산, 보호 및 저장조건의 구명과 체세포배의 기내분화 및 체세포배 배양식물의 유전변이 여부를 탐색하기 위한 연구를 수행한 결과 MS기본배지 보다 1/2MS배지가 체세포배형성에 가장 효과적이었으며 BA, kinetin처리 모두 $0.1{\sim}1mg/L$에서 체세포배가 발이하여 식물체 형성이 양호하였고 농도가 높을수록 억제되는 경향을 보였으며 shoot의 발근과 신장을 위해서는 IAA처리가 효과적이었다. 1/2MS기본배지에 2.0%의 알진산 용액으로 알진산캡슐을 씌운 체세포배의 기내에서의 식물체형성(전환율)은 $AgNO_3$5mg/l처리가 86%로 가장 앙호하였다 . $5^{\circ}C$에서 3개월 동안 체세포배를 저온저장하여 65%의 발아가 이루어졌으며 저장기간이 길어질수록 발아율이 저하되었다. 체세포배 배앙식물은 RAPD(Randomly Amplified Polymorphic DNA)분석을 통하여 여러 major band 수준에서 DNA polymorphism이 관찰되었다.

Objectives: Nail has been a substitute DNA source for genotyping. To investigate the integrity and consistency of nail DNA amplification for biomarker study, nail clippings from 12 subjects were collected at monthly intervals. The possibility of longer amplification and existence of GAPDH RNA/protein, were also investigated with three nail samples. Methods: Three primer sets were designed for quantitative amplification of nuclear and mitochondrial genes and analysis of their consistency. The mean threshold cycles in amplification of the target genes were compared to test the consistency of polymerase chain reaction (PCR) performance among individual factors including age groups, sex, family, the nail source, and by the size of the amplification segments. Results: The amplification of the target genes from nail DNA showed similar integrity and consistency between the nail sources, and among the serial collections. However, nail DNA from those in their forties showed earlier threshold cycles in amplification than those in their teens or seventies. Mitochondrial DNA (mtDNA) showed better DNA integrity and consistency in amplification of all three targets than did nuclear DNA (nucDNA). Over 9 kb of mtDNA was successfully amplified, and nested quantitative PCR showed reliable copy numbers (%) between the two loci. Reverse transcription PCR for mRNA and immunoblotting for GAPDH protein successfully reflected their corresponding amounts. Regarding the existence of RNA and protein in nails, more effective extraction and detection methods need to be set up to validate the feasibility in biomarker study. Conclusions: Nail DNA might be a feasible intra-individual monitoring biomarker. Considering integrity and consistency in target amplification, mtDNA would be a better target for biomarker research than nucDNA.
Simple sequence repeats (SSRs) and interSSR (ISSR) marker systems were used in this study to reveal genetic changes induced by artificial selection for short/long larval duration in the tropical strain Nistari of the silkworm Bombyx mori. Artificial selection separated longer larval duration (LLD) ($29.428{\pm}0.723days$) and shorter larval duration (SLD) ($22.573{\pm}0.839days$) lines from a base, inbred population of Nistari (larval span of $23.143{\pm}0.35days$). SSR polymorphism was observed between the LLD and SLD lines at one microsatellite locus, Bmsat106 ($CA_7$) and at two loci of 1074 bp and 823 bp generated with the ISSR primer UBC873. Each of these loci was present only in the LLD line. The loci segregated in the third generation of selection and were fixed in opposite directions. In the $F_2$ generation of the $LLD{\times}SLD$ lines, the alleles of Bmsat106 and $UBC873_{1074bp}$ segregated in a 1:1 ratio and the loci were present only in the LLD individuals. $UBC873_{823bp}$ was homozygous. Single factor ANOVA showed a significant association between the segregating loci and longer larval duration. Together, the two alleles contributed to an 18% increase in larval duration. The nucleotide sequences of the $UBC873_{1074bp}$ and $UBC873_{823bp}$ loci had 67% A/T content and consisted of direct, reverse, complementary and palindromic repeats. The repeats appeared to be "nested" (59%) in larger repeats or as clustered elements adjacent to other repeats. Of 203 microsatellites identified, dinucleotides (67.8%) predominated and were rich in A/T and T/A motifs. The sequences of the $UBC873_{1074bp}$ and $UBC873_{823bp}$ loci showed similarity (E = 0.0) to contigs located in Scaffold 010774 and Scaffold 000139, respectively, of the B. mori genome. BLASTN analysis of the $UBC873_{1074bp}$ sequence showed significant homology of (nt.) 45-122 with upstream region of three exons from Bombyx. The complete sequence of this locus showed ~49% nucleotide conservation with transposon 412 of Drosophila melanogaster and the Ikirara insertions of Anopheles gambiae. The A + T richness and lack of coding potential of these small loci, and their absence in the SLD line, reflect the active process of genetic change associated with the switch to short larval duration as an adaptation to the tropics.
GenBank database로부터 돼지 genomic 서열과 사람의 CFB 유전자의 CDS를 정렬하여 돼지 CFB 유전자의 CDS를 추정하였다. 이를 바탕으로 제작된 primer를 이용하여 RT-PCR을 실시하여 CDS 내부서열을 결정하였으며, 결정된 서열을 바탕으로 primer 제작 및 RACE-PCR을 실시하였다. 돼지 CFB 유전자의 전체 CDS 길이는 2298 bp였으며, 사람 및 마우스와의 비교결과 염기삽입/결실이 확인되었다. CDS 및 아미노산 서열을 사람 및 마우스와 비교한 결과 CDS는 84% 및 80%, 아미노산 서열은 79%, 77%의 상동성을 보였다. 포유류의 CFB에서 일반적으로 나타나는 보체조절단백질(complement control protein, CCP) 영역, Von willebrand factor A(VWFA) 영역, 그리고 serine protease 영역이 확인되었으며, 단백질 기능에 중요하게 작용하는 아미노산 잔기들은 돼지를 포함한 사람, 마우스, 소, 말에서 동일하게 나타났다. 사람, 마우스, 소, 말, 돼지 CFB 유전자의 아미노산 서열에 의한 유전적 거리지수 및 neighbor-joining tree 작성 결과 돼지는 같은 우제목에 속하는 소와 가장 가까운 계통유전학적 유연관계를 나타내었다. 결정된 CDS를 바탕으로 exon 영역을 증폭하기 위한 primer를 제작하였고, cSNP 분석을 위해서 돼지 6품종을 대상으로 direct sequencing을 실시하였다. 그 결과 아미노산 치환을 일으키는 3개(C13T, A1696G, A2015C)의 cSNP가 동정되었다. 동일한 DNA를 사용하여 동정된 3개의 cSNP를 대상으로 Multiplex- ARMS 방법으로 유전자형 분석 결과 direct sequencing 결과와 일치하였다. Multiplex-ARMS 방법의 재현성 확인을 위해 무작위로 2개의 DNA 시료를 선발한 후 direct sequencing과 Multiplex-ARMS 분석을 각각 실시하였으며, 3개의 cSNP에 대한 유전자형이 일치함을 재확인하였다. 따라서 본 연구에서 확인된 3개의 cSNP는 SLA class III 영역의 haplotype 분석을 위한 기초 자료로 사용될 수 있으며, Multiplex- ARMS 기법은 이종장기 개발에 필수적인 SLA 전체 영역 내 유전자들의 유전자형 분석을 위한 효율적인 분석방법이라고 사료된다.
본 연구는 차세대 염기서열 분석법(NGS)을 이용하여 방어의 microsatellite 마커를 개발하고, 개발된 마커를 이용하여 방어 집단의 유전적 특성을 분석하기 위해 수행되었다. 차세대 염기서열 분석 장비인 Illumina Hiseq2500를 이용하여 총 28,873,374개의 read들을 얻어 assembly를 수행한 결과, 전체의 약 1.6%에 해당하는 466,359개의 read들이 assembly 되었으며, 이 read들의 총 길이는 7,247,216,874 bp로 확인되었다. 크기가 518 bp 이상이 되는 contig는 30.729개로 나타났으며, 이 중 microsatellite 영역을 포함하는 contig 132개(0.43%)를 1차로 선별하고, PCR 증폭 여부 및 유전자형 분석을 통해 microsatellite 후보 60개를 2차로 선별하였다. 그 중 방어집단의 마커로서 유용한 15개의 microsatellite 마커를 선택하였다. 방어집단을 대상으로 개발된 15개의 microsatellite 마커로 분석한 결과, 관찰된 유효 대립유전자수(NA)는 평균 18.5(11~30)로 나타났다. 평균 관측치 이형접합도(HO)와 평균기대치 이형접합도(HE)는 각각 0.812(0.431~0.972)와 0.896(0.782~0.949)으로 나타났다. 다형성이 관찰된 모든 microsatellite 마커 간의 연관불평형은 나타나지 않았으며, 해산어의 평균 HE 값인 0.79 이상의 수치를 나타내었다. 따라서 본 연구에서 개발된 15개의 microsatellite 마커는 방어 집단의 유전적 다양성 분석에 유용할 것으로 사료된다.
버섯생산성 향상을 위해서는 주년재배체계가 확립되어야 하는데 여름철 재배가 가능한 중고온성 봉지재배용 고품질버섯 '곤지5호'의 주요특성은 다음과 같다. '곤지5호'는 균사생장적온은 $26{\sim}29^{\circ}C$이고 버섯발생 및 생육온도는 $18{\sim}20^{\circ}C$로 중고온성을 나타내었다. 갓색은 $20^{\circ}C$재배시 회색(색차값(L)=56.0)이고 얕은 깔대기형이며 다발형으로 발생하였다. 또한, 대는 회백색이고 굵고 긴형태를 나타내었으며 '수한1호'에 비해 탄력성이 우수하였다. 봉지재배시 $20^{\circ}C$에서 초발이 소요일수는 3일, 자실체 생육 일수는 4일이었다. 수량은 생산력검정시 1kg 봉지에서 221.4g을 나타내어 '수한1호' 201.0g에 비해 10% 증수효과가 있었다. 또한, 농가실증재배시 Farm 1과 Farm 3 지역에서는 대조구인 '수한1호' 217g/1kg봉지 대비 13% 증수효과가 있었으며, Farm 2지역에서는 '부평33호'의 230.8g/1kg봉지와 비슷하였다. DNA다형성을 비교 분석한 결과 URP-2F, URP-2R, URP-25F등의 모든 primer에서 모본과 다른 밴드양상을 나타내어 품종간의 구분이 확실하였고, 교잡모본과 혼합된 밴드 양상을 나타내었다.
본 연구는 자가부식 접착제의 치질로의 침투와 레진 tag 형성을 알아보고 부가적인 산부식이 레진 tag 형성에 미치는 효과를 관찰하기 위하여 계획되었다. 3종의 자가부식 접착제(SE bond, AQ bond and L Pop)와 one bottle adhesive(Single bond)를 사용하였고 발거한 구치로 교합면 법랑질과 상아질 시편을 제작하였으며 각 군당 5개씩 네 개의 군으로 나눈 총 20개의 시편을 양분하여 각각 접착제를 적용하기 전 35% 인산으로 산부식하거나 산부식처리하지 않았으며 이후 레진을 적용시키고 중합하였다. 시편은 Silverstone microtome으로 절단한 후 HCl 용액과 NaOCl 용액으로 처리하고 건조시킨 후 이온으로 피복하였으며 주사전자현미경으로 관찰하여 다음과 같은 결과를 얻었다. 1. 부가적 산부식 처리한 상아질은 모든 자가부식 접착제군에서 산부식하지 않은 상아질보다 resin tag의 길이와 두께가 증가하는 것이 관찰되었다. 2. 법랑질에서는 L Pop을 제외하고 부가적 산부식한 군에서 레진 tag의 두께가 산부식하지 않은 군보다 컸고 보다 명확한 부식양상을 관찰할 수 있었다. L Pop에서는 부식법랑질과 비부식법랑질간의 차이가 없었다. 본 연구의 결과는 자가부식형 접착제가 법랑질을 부식시키지 않고 상아세관으로 침투하는 정도가 산부식군에 미치지 못하다는 것을 의미한다. 이와 관련하여 접착강도와 미세누출 및 레진 tag 형태에 대한 부가적 연구가 필요할 것이다.
