Effects of alchohol and fat content in a balanced diet on chemical composition and morphology of liver were investigated in growing rats. Fourth eight male rats of Sprague-Dawley strain weighing about 160g were divided into 4 groups ; high fat diet group, alcohol-administered high fat diet group, low fat diet group and alcohol-administered high fat diet group, low fat diet group and alcohol-administered low fat diet group. High and low fat diets supplied 30% and 12%, respectively, of total calorie intake from fat, and alcohol was given by adding ethanol in drinking waster at 10%. Diets contained adequate amounts of all nutrients required for rats, including lipotrpoic agents(choline and methionine) to minimize effects of factors other than alcohol on liver damage. Ratios of liver weight to body weight were statistically different among groups. Liver/dody weight ratios alcohol-administered rats were significantly higher than those of non-alcohol groups after 6 weeks treatment. Although total lipid and triglyceride per gram liver were increased in alcohol-administered rats, especially low fat diet fed rats, the values were not significantly different. Opticmicroscopical observation revealed increase in cell size and no change in morphology of liver. Examination of hepatocytes by electron microscopy showed that fat droplets were observed in all groups but enlarged in the alcohol-administered low fat diet fed rat. Contents of protein, cholesterol and phospholipid were not affected by alcohol consumption. The level of lipid peroxide was significantly lower in the livers of alcohol-administered rats than in the livers of non-alcohol groups. The results of this study indicate that even moderate alcohol drinking and dietary fat content did not affect any significant change in composition and morphology of liver until 6 week treatment but that even moderate alcohol drinking caused some signs of steatosis of liver.

Effects of Sardine Oil Feeding and Vitamin E Supplementation on Histopathological Changes and $\alpha$-L-fucosidase activity in experimental hepatocarcinogenesis. Sprague-Dawley rats weighing 80~90 g were fed the diet containing either 15% corn oil (CO) or sardine oil (SO) with or without vitamin E supplements (dl-$\alpha$-tocopherol acetate 800 IU/kg diet) for 8 weeks. After 2 weeks of feeding, the rats were given a single intraperitoneal injectin of diethylnitrosamine (DEN, 200 mg/kg BW). From the fifth week, rats were given 0.02% acetylaminofluorene (AAF) in diet for 4 weeks. At the seventh week, 0.05% phenobarbital in liver and hepatic glutathione S-transferase palcental form positive (GST-P+) foci were examined by Hematoxylin& Eosin (H&E) staining and immunohistochemical method, respectively. Serum $\alpha$-L-fucosidase activity was determined. The livers fromt he carcinogen treated rats showed significantly increased formation of GST-P+ foci at sacrifice points while the livers fromthe non-carcinogen treated groups showed almost no foci. Although GST-P+ foci formation was not affected by dietary oil, it was increased unexpectedly by vitamin E supplementation. Histopathological changes were similar to patterns of GST-P+ foci formation in almost all groups. Serum $\alpha$-L-fucosidase activities were increased by carcinogen treatment in all dietary groups. $\alpha$-L-fucosidase activities were positively correlated with GST-P+ foci formation. There results suggest that excessive vitamin E supplementation can enhance hepatocarcinogenesis although the mechanisms involved are not clearly understood.
In the past few years, scintigraphy has become increasingly important in clinical practice, and the use of a color-printing technique has permited a more accurate interpretation of the scan image. Our liver color scintigrams consist of 51 hepatomas, 35 liver cirrhosis, 22 liver abscessis, 10 hepatitis and other 13 cases of the liver diseases which were clinically arid pathologically diagnosed at Sevarance Hospital, Yonsei Univ., since Feb. 1969 through Sept. 1969. These scintigrams have been analized in terms of various pathologic morphology, such as size, shape, margin of the liver, distribution of radioactivity, and shape of the space occupying lesions. The results are as follows: 1. Enlargement of the liver was the most common finding in the diseased livers. The Rt. lobe enlargement was particularly prominent in the liver abscess. 2. Irregular distribution of radioactivity in the liver (so called mottling) was present in 78% of hepatoma, while it was seen only in 31% of liver abscesses. 3. Liver cirrhosis tends to show perihilar accumulation of the isotope (57%). 4. The deformity of the lower most angle of the Rt. lobe, and the Lt. lateral margin of the Lt. lobe was also impressive throughout the cases ($74{\sim}95%$ of all diseased livers). 5. The frequency of visualization of the spleen was influenced by the size of space occupying lesions and the amount of functioning liver. 6. Differentiation between the liver abscess and hepatoma seems to be possible on scintigram, when shape and margin of defect and patterns of distribution of radioactivity in the remaining liver are clearly demonstrated.
A new sudden death in rabbits appeared in China and Korea in 1984 and 1985, respectively, and was recognized to be an acute infectious disease caused by a virus. The disease was reported as a "new viral disease," and thereafter, a tentative name of "viral hemorrhagic disease", "hemorrhagic pneumonia" or "viral hemorrhagic pneumonia" has been described in the case reports. But authors had called the viral disease "rabbit viral hepatitis" due to picornavirus infection, because the principal lesion of the disease was an acute hepatitis. The purpose of this report is to describe the electron microscopic findings on the livers in experimentally infected rabbits. All the livers of the affected rabbits were shown to have degenerative changes of a type that is characteristic of acute hepatitis. In the liver cells, there were dilation of rER and mitochondria, vacuole formation of various sizes, and appearances of many virus-like particles in the vicinity of rER, granular bodies and crystalline arrays of viral particles in the cytoplasm with necrotic changes of the nucleus. Clusters of virus-like particles and viral crystals appeared in the cytoplasm of sinusoid endothelial cells and Kupffer's cells with morphological changes of organelles. Also viral crystals were demonstrated in the cytoplasm of macrophages among the liver cells. On the whole, the liver cells had many virus-like particles and a few crystalline arrays of viral particles. Therefore, this implies that the liver cells are the main site of the viral replication in inducing the viremia. It was concluded that the liver was the primary target organ of this viral disease, and the pathological and the ultrastructural evidence suggest that the virus may be belong to genus enterovirus.
Total lipid were extracted from the eggs, livers, intestines, muscles, and testis of Fugu xanthopterus. The fatty acid composition of total lipid was analyzed by gas liquid chromatography. Proximate percentage of total lipid from the samples was shown to be ; 81.72% in the livers, 14.53% in the intestines, 12.63% in the eggs, 0.85% in the muscles, and 1.74% in the testis. Toxins(tetrodotoxins) were completely removed from the liver of Fugu xanthopterus with 1% acetic acid/methanol solution before total lipid was extracted with CHCI3:methanol solution(2:1, v/v). The toxicity left in the total lipid was checked using mice. The content of DHA in each tissues of Fugu xanthopterus was the most abundant in muscle(32.4%), followed by liver(23.0%), intestine(26.7%) and testis(15.4%). The level of total polyenoic acids comprising DHA was more abundant in muscle(57.4%) than in the liver(50.9%).
The study was carried out to observe the effects of Clonorchis sinensis Infection on induction of cholangiocarcinoma in Syrian golden hamsters to which 15 ppm dimethylnitrosamine (DMN) solution was administered for 8 weeks. The histopathological changes of the bile duct and liver cells were observed at the 11th week. In six of 8 hamsters (75%) which were treated with DMN and then infected with C. sinenis, the livers developed cholangiocarcinoma at 10 weeks after the infestation of C. sinenis. The features of cholangiocarcinoma lesions were adenomatous or papillary hyperplasia of the bile duct epithelia showing distinct anaplastic changes with mucinous cell metaplasia and necrotic area. In the hamsters which received either DMN or C. sinenis alone, the livers showed only hyperplastic changes of the bile duct epithelial cells. It was suggested that C. sinensis infection and DMN administration could be a synergism on the development of cholangiocarcinoma in Syrian golden hamsters.
Casting specimens of portal system and hepatic vein system in livers were made by injection of vinylite into the portal vein and postcava in 20 adult dogs. The author classificated the ramification of portal system and hepatic vein system. The results obtained were summerized as follows: 1. Portal system in livers were divided into left and right trunks. The left trunk subdivided into papillary process (caudate lobe,) left medial lobe, left lateral lobe, quadrate lobe and right medial lobe rami. The right trunk were subdivided into right lateral lobe and caudate proccss(caudate lobe.) 2. The lateral superior and medial inferior rami of portal system in left lateral lobe were subdivided 1 or 2 branches from left trunk. 3. The lateral superior ramus of portal system in left medial lobe did not appeared in 40% of the cases examined. 4. Portal system in quadrate lobe were subdivided 1-3 branches from left trunk, 5. Portol system in right medial lobe rami were relatively simple in ramification. 6. The lateral superior and medial inferior rami of portal system in right lateral lobe were subdivided 1 or 2 branches from right trunk. 7. Hepatic vein system of left lateral, left medial, quadrate and right medial lobe rami were originated from same ramus divided from the postcava in all cases. 8. Hepatic vein system of left and right rami in right medial lobe were divided from postcava.
The combined effects of iron and selenium status on glutathione peroxidase (GSHPx) activity, cytochrome P-450 activity, and lipid peroxidation in the liver and intestinal mucosa of rats were investigated. In experiment one, four experimental groups (+Se+Fe, -Se+Fe, +Se++Fe, -Se++Fe) were manipulated for 3 weeks with intramuscular administration of irondextran (++Fe) and/or normal diet (+Fe) and deionized water (-Se) and/or selenium-supplemented deionized water (+Se). In experiment two, 2% dietary carbonyl iron (instead of the parenteral administration) was fed for 3 weeks to rats. Body weight of rats was significantly decreased in both parenterally and orally iron-overloaded groups (p<0.01), regardless of Se supplement. Serum iron was significantly increased in parenterally iron-overloaded groups but it was marginally increased in orally iron-overloaded groups. There was no significant difference in hemoglobin content among experimental groups in either experiment one or two. Total iron in the small intestine, intestinal mucosa, and livers was significantly high in both parenterally and orally iron-overloaded rats, regardless of selenium status. In the liver and intestine, GSHPx activity was significantly higher in all selenium-supplemented groups, compared to Se-deficient groups (p<0.01) and lipid peroxidation was significantly enhanced in both parenterally and orally iron-overloaded groups, compared to iron-adequate groups. There was no significant difference in cytochrome P-450 activity in the livers between groups in both experiment one and two. These results indicated that GSHPx activity in liver and intestinal mucosa was depended on selenium status, regardless of iron status, and iron-overload enhances lipid peroxidation in liver and intestinal mucosa by increasing the tissue iron content.
This study investigated whether trinitroglycerine (TNG) as nitric oxide (NO) releasing agent had anti-leishmanial effects and mediated pathology in BALB/c mice infected with Leishmania major. Cutaneous leishmaniasis (CL), a zoonotic infection caused by leishmania protozoa is still one of the health problems in the world and in Iran. NO is involved in host immune responses against intracellular L. major, and leishmania killing by macrophages is mediated by this substance. Moreover, application of CL treatment with NO-donors has been recently indicated. In our study, TNG was used for its ability to increase NO and to modify CL infection in mice, in order to evaluate NO effects on lesion size and formation, parasite proliferation inside macrophages, amastigote visceralization in target organs, and NO induction in plasma and organ suspensions. Data obtained in this study indicated that TNG increased plasma and liver-NO, reduced lesion sizes, removed amastigotes from lesions, livers, spleens, and lymph nodes, declined proliferation of amastigotes, hepatomegaly, and increased survival rate. However, TNG reduced spleen-NO and had no significant effects on spelenomegaly. The results show that TNG therapy reduced leishmaniasis and pathology in association with raised NO levels. TNG had some antiparasitic activity by reduction of positive smears from lesions, livers, spleens, and lymph nodes, which could emphasize the role of TNG to inhibit visceralization of L. major in target organs.
The effects of estrogen and androgens on Vg gene expression were examined in primary hepatocyte culture and livers of the immature male trout. Specific primers of Vg cDNA were designed with already reported Vg gene nucleotide sequences. PCR product was sequenced and verified with Vg cDNA of rainbow trout. Total RNA was extracted from the cultured hepatocytes and livers of steroid-treated rainbow trout and then it was analyzed by reverse transcriptase- polymerase chain reaction (RT-PCR) analysis. The Vg mRNA and Vg protein synthesis were increased in rainbow trout in vivo and in vitro with E$_2$ and methyltestosterone (MT) There were dose and time-related effects of E$_2$ and MT on vitellogesis. Androgens such as progesterone androsterone and testosterone also stimulated Vg mRNA expression in vitro. The results show that androgens as well as E$_2$ can induce expression of Vg mRNA in trout in vivo and in vitro.
