The protective effect of jujube methanol extract on benzo(a)pyrene(B(a)P)-induced liver injury was studied in rats in vitro and in vivo. Jujube methanol extract significantly recovered the enzyme activities(GOT, GPT, LDH and ALP) and lipid contents(total cholesterol, triglyceride and HDL-cholesterol) changed by B(a)P to normal levels in vivo. in viro experiment jujube methanol extract didn't stimulate hepatocyte proliferation but significantly recovered the enzyme activities(GOT, GPT and LDH) in comparison to group Ⅱ administered B(a)P only. It was suggested that jujube methanol extract have a protective effect on liver injury by B(a)P.

The protective effect of SAPP, an extract from a novel herbal complex, on acute liver injury was investigated using mouse animal model in this study. The content of total phenol in SAPP was increased at dose dependent manner. Consistent with the content of total phenol, SAPP showed the significant anti-oxidative effects on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) method. Acute liver injury was induced by D-galactosamine (D-GalN) in mouse. Treatment with SAPP significantly reduced the level of alanine transaminase (ALT) and aspartate transaminase (AST) in serum. Histological observation revealed that whereas D-GalN treated mouse showed vacuolization of hepatocytes, sinusoidal dilation and congestion, loss of cell boundaries and ballooning degeneration, loss of architecture and cell necrosis, treatment with SAPP improved D-GalN-induced liver injury. These results suggest that SAPP shows protective effects against D-GalN-induced hepatotoxicity in vivo acute mouse model.
The protective effect of omija methanol extract on benzo(a)pyrene induce liver injury was studied in rats in vitro and in vivo. In vitro experiment, primary cultured hepatocytes(5${\times}$105cells/$m\ell$) were cultured for 20∼24 hours after adding omija methanol extract(5.1$\mu\textrm{g}$/$m\ell$) and B(a)P(50$\mu\textrm{m}$) in culture medium. In vivo experiment, omija methanol extract(0.1g/kg/day, per os) was administered for 7days and B(a)P(0.1mg/kg body weight, intraperitoneally) was given to the rats after the last administration of extract. Omija methanol extract significantly recovered serum enzyme activities(AST, ALT and LDH) and lipid contents(total cholesterol, triglyceride and HDL-cholesterol) changed by benzo(a)pyrene (B(a)P) to normal levels in vivo. In vitro experiment, as a result of 3-(4, 5-dimethlythiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) assay, omija methanol extract showed a little hepatotoxicity compared with group I (normal) but significantly recovered enzyme activities(AST, ALT and LDH) changed by B(a)P in comparison to group IIadministered B(a)P only. It was suggested that omija methanol extract has a protective effect on liver injury induced by B(a)P.
This study aimed to find out the effect of freeze-drying leek against cadmium poisoning on the cholesterol and enzyme activities in serum and superoxide radical, SOD and catalase in liver and kidney of the male rats during the administrered period. In this experiment, male rats of Sprague-Dawley strain were used. The rats were divided into 3 groups which were fed differently either for 5 weeks or for 10 weeks:basal diet, basal diet and cadmium in water and 3 % leek added diet and cadmium in water. Cadmium was administered ad libiturn 100 ppm CdCl$_{2}$ in water. The followings are the results of this experiment. 1. Leek reduced the cholesterol and the activities of GPT increase resulted from cadmium treatment. 2. Leek reduced the rate of cadmium in liver and kidney. 3. Leek reduced the activities of SOR and catalase in liver and kidney, while it enhanced the activities of SOD. 4. Leek reduced the necrosis and swelling in liver and kidney casused by cadmium treatment. This experiment showed that leek-addition group had protective effect against cadmium poisoning and increased ALPase activities in serum. Leek alleviated GPT activities in serum and cadmium concentration, necrosis, and swelling in liver and kidney. Therefore, this experiment concluded that leek has defensive power against cadmium poisoning.

This study was carried out to investigate the liver protective effects of extracts from Persicae Semen (WT-003, WT-005, WT-006). The acute hepatic injury was induced by intraperitoneal injection of alpha-naphtylisothiocyanate (75 mg/kg, p.o.) and treated with WT-006 (100 mg/kg/day, 200 mg/kg/day, 400 mg/kg/day). The experimental hepatic fibrosis was induced by bile duct ligation/scission(BDL/S), duration of 4 weeks and treated with WT-003, WT-005 or WT-006 (200 mg/kg/days for 4 week). In acute liver injury, WT-006 (200 or 400 mg/kg) lowered serum alanine transferase(ALT) and aspartate transferase(AST) significantly. In fibrotic rats, WT-006 treatment inhibited the hydroxyproline deposition in liver and lowered serum AST, ALT and ALP, significantly. These results suggest WT-006 extract, which does not contain amygdalin, from Persicae Semen have liver protective and antifibrotic effects in rats.
Sulfasalzine is a widely administered drug against inflammatory-based disorders in human. However several cases of liver injury are associated with its administration. There is no stabilized safe protective agent against sulfasalazine-induced liver injury. Current investigation was designed to evaluate if N-acetylcysteine (NAC) and dithioteritol (DTT) as thiol reducing agents and/or vitamins C and E as antioxidants have any protective effects against sulfasalazine-induced hepatic injury in an ex vivo model of isolated rat liver. Rat liver was canulated and perfused via portal vein in a closed recirculating system. Different concentrations of sulfasalazine and/or thiol reductants and antioxidants were administered and markers of organ injury were monitored at different time intervals. It was found that 5 mM of sulfasalazine caused marked liver injury as judged by rise in liver perfusate level of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH) (p < 0.05). A significant amount of lipid peroxidation and hepatic glutathione depletion were detected in drug-treated livers, accompanied with significant histopathological changes of the organ. Administration of NAC ($500{\mu}M$), DTT (${400\mu}M$), Vitamin C ($200{\mu}M$), or vitamin E ($200{\mu}M$) significantly alleviated sulfasalazine-induced hepatic injury in isolated perfused rat liver. The data obtained from current investigation indicate potential therapeutic properties of thiol reductants and antioxidants against sulfasalazine-induced liver injury.
Background: Bisphenol A (BPA), known as an endocrine disruptor, is widely used in the world. BPA is reported to cause inflammation-related diseases. Korean Red Ginseng (KRG) has been used safely in human for a long time for the treatment of diverse diseases. KRG has been reported of its mitigating effect on menopausal symptoms and suppress adipose inflammation. Here, we investigate the protective effect of orally administered KRG on the impacts of BPA in the liver and uterus of menopausal mice model. Methods: The transcriptome analysis for the effects of BPA on mice liver was evaluated by Gene Expression Omnibus (GEO) database-based data (GSE26728). In vivo assay to evaluate the protective effect of KRG on BPA impact in ovariectomized (OVX) mice were designed and analyzed by RNA sequencing. Results: We first demonstrated that BPA induced 12 kinds of gene set in the liver of normal mice. The administration of BPA and KRG did not change body, liver, and uterine weight in OVX mice. KRG downregulated BPA-induced inflammatory response and chemotaxis-related gene expression. Several gene set enrichment analysis (GSEA)-derived inflammatory response genes increased by BPA were inhibited by KRG in OVX mice. Conclusion: Our data suggest that BPA has commonly influenced inflammatory response effects on both normal and OVX mice. KRG protects against BPA impact of inflammatory response and chemotaxis in OVX mouse models. Our comparative analysis will provide new insight into the efficacy of KRG on endocrine disrupting chemicals and OVX mouse.
There has been increasing interest in the value of using soybean to delay or reduce the tumor incidence. This study was undertaken to investigate the possible protective effects of soybean against hepatocarcinogenesis induced by DL-ethionine. Accordingly, we measured biochemical changes occurring in serum and liver of rats treated with DL-ethionine in the presence or absence of soybean. Male albino rats were fed a control diet containing the hepatocarcinogen, DL-ethionine, or the control diet plus soybean 30%, or the control diet plus soybean plus DL-ethionine 0.25% for three months and then returned to a control diet for up to nine months. Rats fed a control diet plus DL-ethionine showed a gradual decrease in liver DNA, RNA, total protein, and liver weight and enzyme activites of liver transaminases (GOT and GPT) and alkaline phosphatase over the 7-month study period. This was followed by a large increase in the liver parameters at the end of the $9^{th}$ month, except for 5'-nucleotidase and glucose-6-phosphatase that showed a large decrease. On the other hand, a gradual increase in the serum enzyme activities of GOT, GPT, 5-nucleotidase, alkaline phosphatase, and in the albumin/globulin (A/G) ratio is observed in the group of rats fed a control diet plus DL-ethionine compared to the control group over 8 months, and this was followed by a large increase in all serum parameters studied at nine-months. The administration of 30% soybean to the rat diet in addition to DL-ethionine maintained all parameters studied at near control values until the end of the $9^{th}$ month. This study suggests that soybean has a protective effect against the hepatocarcinogenesis induced by DL-ethionine.

Objective : This study was undertaken to examine whether Carthami Semen aquacupuncture (CSA) exerts protective effect against Hg-induced cell injury in rabbit liver. Methods : The cell injury was evaluated by ALT activity and lipid peroxidation was estimated by measuring malondialdehyde (MDA). Results : Hg caused an increase of ALT activity and lipid peroxidation in a dose-dependent-manner over concentrations of 0.1-1 mM, which were prevented by addition of 0.005% CSA. The protective effect of CSA was dose-dependent in concentration range of 0.001 to 0.01%. The increase of ALT activity and lipid peroxidation induced by 0.5 mM Hg were almost completely decreased by addition of 0.01% CSA. When the liver tissues were exposed to 0.5 mM Hg, GSH content was decreased, which was significantly restored by 0.01% CSA. 0.5 mM Hg caused decrease in the amount of total and nonprotein sulfhydryl groups, and 0.01% CSA prevented Hg-induced reduction of nonprotein sulfhydryl group but not protein sulfhydryl group. Conclusions : These results suggest that CSA exerts protective effect against Hg-induced cell injury by antioxidant action resulting from enhancement of nonprotein sulfhydryl group content including GSH in liver.
A ginseng preparation consisting of ginseng ext., lycii fructus ext., four vitamins and caffeine was chosen and its efficacy was evaluated with respect to nutritional supplement, antifatigue activity and liver protective action. Animals administered orally in both one-third and three fold doses of the preparation showed no significant increments of their body weights when compared with those of the normal animals, suggesting no supplemental activity. However, the preparation in the above two doses significantly prolonged swimming time to 53 and 63%, respectively. Ginseng and lycii fructus ext. were found to be responsible for the antifatigue activity. And also the preparation significantly inhibited lipid peroxidation of mouse liver after ethanol-induced acute intoxication.
