G009의 간 보호작용에 관한 연구 (Effects of G009 on Chemical-Induced Liver Damage in Rats)

  • The present study was performed to determine the protective effect of G009 on liver damage induced bv ethanol $CCl_4$ and thioacetamide in rats. In acute fatty liver animal model induced by ethanol, triglyceride accumulation was markedly decreased to the normal control level by 25 mg/kg G009 treatment. In addition, G009 significantly reduced serum ALT and AST levels in $CCl_4$-induced acute hepatitis animals. Treatment of G009 to the acute hepatitis rats induced by thioacetamide resulted in a dose dependent reduction of serum ALT level as well as AST level up to the normal control level. These protective effects of G009 were confirmed by histological examinations of the liver. These results suggested that G009 could be effective for the protection from the liver damage induced by ethanol, $CCl_4$and thioacetamide.

명간보(明肝補) 추출물의 Acetaminophen 유도 간 손상에 대한 보호효과 (Protective Effect of Myeongganbo Extract on Acetaminophen-Induced Liver Injury)

  • Objective : Myeongganbo (MGB) composited with Hovenia Semen, Puerariae Radix and Dioscoreae Rhizoma is the prescription for protection of liver function. The purpose of this study was to investigate the effects of MGB extract against acetaminophen (APAP)-induced liver injury in mice. Methods : MGB extract was prepared by extracting with hot distilled water. The extract was freeze-dried following filtration through vacuum distillation system. Mice fasted for overnight were orally administrated with or without MGB extract of different doses (25-200 mg/kg/day). After 30 min, APAP was orally applied with a single dose (400 mg/kg). The levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured in plasmas of mice. Glutathione (GSH), glutathione peroxidase GSH-px), cyclooxygenase-2 (COX-2) activity and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) level were investigated in liver homogenates. Liver sections were stained with haematoxylin & eosin, anti-TNF-${\alpha}$ and anti-mouse COX-2 antibodies. Results : APAP treatment remarkably increased AST and ALT activities in plasma but inhibited GSH and GSH-px levels in liver homogenates. Also, liver injury was significantly accelerated by APAP treatment. Furthermore, APAP remarkably elevated COX-2 activity and TNF-${\alpha}$ levels in liver homogenates. However, administration of MGB extract was able to counteract these effects. Histological studies provided supportive evidence for biochemical and molecular analysis Conclusions : These results suggest that MGB extract has potent hepatoprotective effect against APAP-induced liver injury, these properties may contribute to liver disease care.

SD Rat에서 어성초, 자소엽, 녹차 혼합 추출물의 방사선 방호 효과 연구 (Radiation Protection Effect of Mixed Extracts of Hottuynia, Perilla Frutescens, Camellia Sinnensis in the SD Rat)

  • 본 연구는 어성초, 자소엽, 녹차 혼합 추출물이 천연 방사선 방호제로 SD Rat의 전립선, 소장, 간에 방사선 방호 효과를 확인했다. 어성초, 자소엽, 녹차는 각각 전립선, 소장, 간에 항산화 효과가 있는 것으로 알려져 있다. 본 연구에서는 어성초, 자소엽, 녹차 혼합 추출물의 방사선 방호 효과를 확인하기 위해 SD Rat에 감마선 8 Gy를 조사하였다. 방사선 조사 후 전립선, 소장, 간의 조직학적 분석을 진행하였다. 전립선의 경우 투여 조사군 (HPC+IR Group)이 조사군 (IR Group)에 비해 방사선에 의한 전립선 손상이 적고 회복도 더 잘 됐다. 투여 조사군의 전립선 면적이 조사군보다 1일, 7일에 11.48 %p, 24.54 %p 높은 수치를 통해 확인하였다. 소장의 경우 투여 조사군이 조사군에 비해 방사선에 의한 소장 손상이 적고 회복도 더 잘 되었다. 투여 조사군의 소장 융모의 길이가 조사군 보다 1일, 7일에 23.73 %p, 24.27 %p 높은 수치를 통해 확인하였다. 간의 경우 투여 조사군이 조사군에 비해 방사선에 의한 간 손상이 적고 회복이 더 잘 됐다. 이는 간문맥과 주변 세포를 통해 확인하였다. 본 연구의 결과는 혼합 추출물을 이용한 천연 방사선 방호제의 연구에 기초 자료로 활용될 것으로 사료된다.

웅담이 CCl4로 유발된 mouse의 간 손상에 미치는 영향 (Protective Effects of near nile Juice Against Carbon Tetrachloride-Induced Acute Hepatotoxicity in ICR Mice)

  • Objectives : This study was aimed at investigating liver protection mechanism of bear bile juice (Fel Ursi)by inducing liver toxicity through $CCl_4$ in mice and evaluated histological and serological findings. Methods : Experiment groups was categorized into untreated normal group, $CCl_4$ treated control group, and orally administered bear bile juice experiment group. At the termination of experiment, gross examination of the liver as well as histological findings, and Total protein, Albumin, Total bilirubin, Direct bilirubin SGOT, SGPT, and ALP contents in the serum were evaluated. Results : 1. For gross examination and histological findings, $CCI_4$ treated control group showed destroyed lobular structure, increased fibrosis, as well as hepatic cirrhosis. For the group treated with bear bile juice, the lobular structure suffered less damage, and showed lower level of fibrosis and liver cirrhosis compared to the control group. 2. For serum analysis, Total protein and Albumin were significantly increased in the bear bile juice experiment group than the control group. Total bilirubin and Direct bilirubin didn't show significant differences between the two groups. SOOT, SGPT, and ALP were significantly decreased in the normal and bear bile juice experiment groups compared to the control group. Conclusion : Taken together, bear bile juice can be effectively used for recovering the liver functions and further researches must be conducted to verify the efficacies of bear bile juice.

Protective Effect of Soybean against Hepatocarcinogenesis Induced by DL-Ethionine

  • There has been increasing interest in the value of using soybean to delay or reduce the tumor incidence. This study was undertaken to investigate the possible protective effects of soybean against hepatocarcinogenesis induced by DL-ethionine. Accordingly, we measured biochemical changes occurring in serum and liver of rats treated with DL-ethionine in the presence or absence of soybean. Male albino rats were fed a control diet containing the hepatocarcinogen, DL-ethionine, or the control diet plus soybean 30%, or the control diet plus soybean plus DL-ethionine 0.25% for three months and then returned to a control diet for up to nine months. Rats fed a control diet plus DL-ethionine showed a gradual decrease in liver DNA, RNA, total protein, and liver weight and enzyme activites of liver transaminases (GOT and GPT) and alkaline phosphatase over the 7-month study period. This was followed by a large increase in the liver parameters at the end of the $9^{th}$ month, except for 5'-nucleotidase and glucose-6-phosphatase that showed a large decrease. On the other hand, a gradual increase in the serum enzyme activities of GOT, GPT, 5-nucleotidase, alkaline phosphatase, and in the albumin/globulin (A/G) ratio is observed in the group of rats fed a control diet plus DL-ethionine compared to the control group over 8 months, and this was followed by a large increase in all serum parameters studied at nine-months. The administration of 30% soybean to the rat diet in addition to DL-ethionine maintained all parameters studied at near control values until the end of the $9^{th}$ month. This study suggests that soybean has a protective effect against the hepatocarcinogenesis induced by DL-ethionine.

Lipid-lowering and hepatoprotective effects of Vitis vinifera dried seeds on paracetamol-induced hepatotoxicity in rats

  • BACKGROUND/OBJECTIVES: Red grape seeds as functional food are a good source of important bioactive components such as phenolics and antioxidants, which decrease oxidative stress that contributes to the pathogenesis of hepatotoxicity. The current study was conducted in order to evaluate the protective effect of red grape dried seeds (RGDS) on antioxidant properties, lipid metabolism, and liver and kidney functions of rats with paracetamol (750 mg/kg) induced hepatotoxicity. MATERIALS/METHODS: RGDS was added to the basal diet at 5, 10, and 20%. Thirty five adult male rats were assigned to five groups (n = 7) for a six-week feeding period; group (1) normal control, group (2) induced control, groups (3, 4, and 5) fed a diet with RGPS at different levels, 5, 10, and 20%, respectively. At the end of the feeding period, animals' blood and tissues were collected for estimation of serum lipid profile, serum liver, and kidney biomarkers. The protection was measured by detecting lipid peroxidation (LPO), glutathione (GSH), superoxide dismutase (SOD), Catalase (CAT) (in liver tissues), and liver histological examination. RESULTS: The results showed a significant (P < 0.05) decrease in levels of serum cholesterol, triglycerides, low density lipoprotein (LDL-C), and very low density lipoprotein (VLDL-C), with a significant increase in level of high density lipoprotein (HDL-C) for RGDS groups compared to induced control. Rats administered a diet containing RGDS levels produced significant (P < 0.05) hepatoprotection by decreasing the activities of liver enzymes, kidney parameters, and lipid peroxidation, while levels of GSH, SOD, and CAT were increased significantly to near the normal levels. CONCLUSION: The RGDS 20% group was more effective than others against hepatotoxicity of paracetamol, which may be attributed to RGDS total phenols and antioxidant contents, which were 1.438 mg and 1.231 mg, respectively.

단회(單回) 및 분획(分劃) X선(線) 조사(照射)가 가토(家兎)의 간(肝) 및 폐조직(肺組織)의 NP-SH 및 NP-SS에 미치는 영향(影響) (Effect of Single or Fractionated X-Irradiation on the NP-SH and NP-SS of Rabbit Liver and Lung Tissues)

  • In view of the importance of sulfhydryl groups in producing a chemical protection against ionizing radiation, an attempt was made to evaluate the changes of intrinsic non-protein sulfhydryl (NP-SH) and non-Protein disulfide(NP-SS) of the lung and liver tissues of rabbits following the whole body X-irradiation with 900 r either in single or fractionated $(300\;r{\times}3)$ dose. NP-SH was measured by Ellman's method, and NP-SS was measured by the electrolytic reduction method described by Dohan. Experiment was performed at 1,3,5,24 and 48 hours post·irradiation, and the results were compared with the control. The results thus obtained are summarized as follows; 1) Intrinsic levels of NP-SH and NP-SS of normal rabbits were $0.77{\pm}0.10$ and $0.61{\pm}0.07\;{\mu}mol/gm$ wet weight in liver, and $0.28{\pm}0.03$ and $0.54{\pm}0.03$ in lung tissues respectively. 2) NP-SH of liver after single X-irradiation showed no significant change in general, but at 48 hours post·irradiation, it was elevated comparing with the normal value. 3) Levels of NP-SS in liver was decreased than the normal value in the irradiated groups, and the lowest level was observed at 3 hours after single X-irradiation and at 5 hours after fractionated irradiation. 4) In lung tissues, levels of NP-SS showed no significant change from the control at earlier experimental hours, but a great decrease was observed at later Part of the experiment.

Folic acid inhibits necrosis and apoptosis in ischemic and reperfusion induced injury in rat liver

  • Temporary clamping of the portal triad is a common strategy to minimize bleeding during liver transplantation. Increasing evidences suggests that oxygen derived free radicals and reintroduction of oxygen in ischemic tissue lead to ischemic and reperfusion injury (I/R) and lead to apoptosis and necrosis. Adult Wistar rat subjected to 60 min of partial liver ischemia followed by three hour reperfusion. Eighteen Wister rats were divided into sham-operated control group (I) (n = 6), ischemia and reperfusion group (II) (n = 6), folic acid treated group (1 mg/kg body weight/daily by oral route for 7 days before induced ischemia reperfusion maneuver) (III) (n = 6). Apoptotic and necrotic hepatocytes, mitochondrial antioxidant enzymes were measured. Liver injury was assessed by alanine transaminases (ALT), aspartate transaminases (AST), liver histopathology and electron microscopy. An ischemic and reperfusion hepatocellular injury was indicated by increased serum-ALT, AST, histopathology and electron microscopy studies. Apoptotic and necrotic cells were increased which was revealed by flow cytometry in I/R group. Pre- treatment with folic acid significantly decreased serum -ALT, AST levels, apoptotic and necrotic cells after 1 h ischemia followed by 3 h of reperfusion. Histopathology and TEM studies showed markedly diminished hepatocellular injury in folic acid pretreated rats during the hepatic I/R, which reached a level comparable to saline-treated rat of sham operated group. On the basis of our findings it may be concluded that folic acid afforded significant protection from necrosis and apoptosis in I/R injury.

사염화탄소에 의한 간손상에 대한 황백의 간보호 효과 연구 (Hepatoprotective Effect of Bark of Phellodendron amurense RUPR. on Liver Damage Induced by Carbon Tetrachloride)

  • We evaluated the hepatoprotective activity of crude hot-water extracts of the traditional Korean medicinal herb, Phellodendri Cortex (PC; Bark of Phellodendron amurense RUPR.), in an experimental model of hepatic damage induced by carbon tetrachloride ($CCl_4$). The serum marker of liver damage, sGOT, sGPT and sALP, were significantly decreased in the liver of the PC treated rats, compared with that of $CCl_4$ treated group. The histological observation of liver section of rats, showed the same protective effect of PC treatment. And the protective activity of PC was more significant in the post-treated group than pre-treated group. The significant decrease of malodialdehyde and increase of glutathion, catalase activity were observed in the liver homogenate of PC treated rats. Based on these findings, it is suggested that PC has potent hepatoprotective effects and the mechanism of the protection may be related to antioxidation pathways.
