만성적으로 알코올을 섭취한 쥐의 조직 내 Fatty Acid Ethyl Esters (FAEEs)와 지질과산화물 형성에 미치는 영향 (Effect of Chronical Ethanol Ingestion on the Levels of Fatty Acid Ethyl Esters (FAEEs) and Lipid Peroxidation in Rat Tissues)

  • The present study was designed to observe the effect of chronically ingested ethanol on the level of fatty acid ethyl esters (FAEEs), which is a non-oxidative metabolite of ethanol metabolism in tissues, and its correlation to the status of oxidative stress in rats. Forty male Sprague Dawley rats weighing 145 - 155 g were divided into 2 groups, Control and EtOH. All rats were fed Lieber-DeCarli liquid diet for 4 weeks by pair-feeding. An isocaloric maltose dextrin was added in replace of 50 g ethanol (36%kcal) in the control diet. Chronically ingested ethanol significantly increased the content of FAEEs in pancreas and liver, but not in brain. The level of 2-thiobarbituric acid reactive substances (TBARS) was significantly increased, but ${\alpha}-tocopherol$ level was significantly decreased in pancreas and liver. However, the levels of TBARS and ${\alpha}-tocopherol$ in brain were not significantly affected by ethanol ingestion. Therefore, chronically ingested ethanol might cause tissue damage by increasing the levels of FAEEs and TBARS and dissipating more ${\alpha}-tocopherol$ in tissues.

Lutein decreases oxidative stress and inflammation in liver and eyes of guinea pigs fed a hypercholesterolemic diet

  • Guinea pigs were fed a hypercholesterolemic diet (0.25 g/100 g cholesterol) and randomly allocated either to a Control group (n = 9) or to a Lutein (0.1 g/100 g) group (n = 10) for 12 weeks to evaluate oxidative stress and inflammation in both liver and eyes. Malondialdehyde (MDA) concentrations and inflammatory cytokines were measured as well as hepatic nuclear factor-kappaB (NF-${\kappa}B$) binding. Lutein concentrations were greater in eyes ($P$ < 0.01) and liver ($P$ < 0.001) in the Lutein group. All guinea pigs had high concentrations of hepatic cholesterol as well as high plasma ALT and AST levels indicative of liver injury. However, the Lutein group had 43% lower hepatic free cholesterol than the Controls ($P$ < 0.05). Hepatic MDA and MDA in the eye were lower in the Lutein compared to the Control group ($P$ < 0.05). Hepatic tumor necrosis factor-${\alpha}$ was 32% lower in the Lutein group ($P$ < 0.05). Lastly, the Lutein group presented lower NF-${\kappa}B$ DNA binding activity than the Control group ($P$ < 0.001). These results suggest that in the presence of high cholesterol, lutein exerts both antioxidant and anti-inflammatory effects, which can be explained by attenuated NF-${\kappa}B$ DNA binding activity. Furthermore, results also suggest that lutein accumulates in the eyes of guinea pigs to protect against oxidative stress.

Protective Effect of Administrated Glutathione-enriched Saccharomyces cerevisiae FF-8 Against Carbon Tetrachloride ($CCl_4$)-induced Hepatotoxicity and Oxidative Stress in Rats

  • The present work is aimed to evaluate the protective effect of glutathione-enriched Saccharomyces cerevisiae FF-8 strain on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity and oxidative stress in rats. The activities of liver markers (alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, lactate dehydrogenase), lipid peroxidative index (thiobarbituric acid-reactive substances), and the antioxidant status (reduced glutathione) were used to monitor those protective roles of FF-8 strain. The liver marker enzymes in plasma and the lipid peroxidation in the liver were increased when $CCl_4$ was treated but these were significantly decreased by FF-8 strain treatment. The hepatic concentration of glutathione in the current glutathione-enriched FF-8 strain fed animal was approximately twice as high as the normal, but this was slightly increased in response to $CCl_4$ plus glutathione-enriched FF-8 strain. The increased liver triglyceride concentration due to the $CCl_4$ treatment was significantly decreased by FF-8 strain and the reduced level reached to that of normal group. Administration of FF-8 strain in normal rat did not show any signs of harmful effects. Therefore, the current findings suggest that FF-8 strain could be an effective antioxidant with no or negligible side-effects and it might be useful for the purpose of protection treatment of hepatotoxicity and oxidative stress in $CCl_4$-treatment in rat.

Ginseng essence, a medicinal and edible herbal formulation, ameliorates carbon tetrachloride-induced oxidative stress and liver injury in rats

  • Background: Ginseng essence (GE) is a formulation comprising four medicinal and edible herbs including ginseng (Panax ginseng), American ginseng (Panax quinquefolius), lotus seed (Nelumbo nucifera), and lily bulb (Lilium longiflorum). This study was aimed at investigating the hepatoprotective effect of GE against carbon tetrachloride ($CCl_4$)-induced liver injury in rats. Methods: We treated Wistar rats daily with low, medium, and high [0.625 g/kg body weight (bw), 1.25 g/kg bw, and 3.125 g/kg bw, respectively] doses of GE for 9 wk. After the 1st wk of treatment, rats were administered 20% $CCl_4$ (1.5 mL/kg bw) two times a week to induce liver damage until the treatment ended. Results: Serum biochemical analysis indicated that GE ameliorated the elevation of aspartate aminotransferase and alanine aminotransferase and albumin decline in $CCl_4$-treated rats. Moreover, $CCl_4$-induced accumulation of hepatic total cholesterol and triglyceride was inhibited. The hepatoprotective effects of GE involved enhancing the hepatic antioxidant defense system including glutathione, glutathione peroxidase, glutathione reductase, glutathione S-transferase, superoxide dismutase, and catalase. In addition, histological analysis using hematoxylin and eosin and Masson's trichrome staining showed that GE inhibited $CCl_4$-induced hepatic inflammation and fibrosis. Furthermore, immunohistochemical staining of alpha-smooth muscle actin indicated that $CCl_4$-triggered activation of hepatic stellate cells was reduced. Conclusion: These findings demonstrate that GE improves $CCl_4$-induced liver inflammation and fibrosis by attenuating oxidative stress. Therefore, GE could be a promising hepatoprotective herbal formulation for future development of phytotherapy.

Alterations of Antioxidant Status and Mitochondrial Succinate Dehydrogenase Activity in the Liver of Wistar Strain Albino Rats Treated with by Ethanol Extracts of Annona senegalensis Pers (Annonaceae) Stem Bark

  • Numerous ethnomedicinal uses have been attributed to different parts of Annona senegalensis (ASE), including its uses as food and food additives. The present study investigated toxicological and antioxidant effects of 28 days administration of ethanol extracts of ASE stem bark to Wistar strain albino rats. Acute toxicity test was done to determine lethal dose in Wistar rats while sub-acute toxicity test was conducted on rats divided into four groups (A - control, B - 50 mg/kg, C - 100 mg/kg, D - 150 mg/kg, respectively and treated for 28 days. Oxidative stress markers in liver and kidney as well as hepatic succinate dehydrogenase activity in the mitochondrial and post mitochondrial fractions (PMF) were evaluated. The $LD_{50}$ value of ASE was > 2,000 mg/kg. White blood cell counts gradually increased, but red blood cell counts and haematocrits level decreased significantly (p < 0.05) by about 50%. Liver enzymes in the serum and mitochondrial succinate dehydrogenase activity increased significantly (p < 0.05). Superoxide dismutase and catalase activities also increased in liver mitochondria and PMF while malondialdehyde (MDA) and reduced glutathione levels increased only in the PMF. Furthermore, only MDA levels increased significantly in the kidney after 28 days extract administration. Histopathological examination showed hepatic necrosis and no obvious signs of nephrotoxicity. Anona senegalensis is relatively safe, but prolonged ingestion could induce oxidative stress and impair ATP synthesis through the modulation of the activity of mitochondrial succinate dehydrogenase.

산화적 스트레스에 의한 간세포의 DNA 손상 및 세포사멸 유도에 미치는 원지 에탄올 추출물의 보호 효과 (The Protective Effect of Ethanol Extract of Polygalae Radix against Oxidative Stress-Induced DNA Damage and Apoptosis in Chang Liver Cells)

  • Objectives: The purpose of the present study was to evaluate the preventive effects of ethanol extract of Polygalae radix (EEPR) against oxidative stress (hydrogen peroxide, $H_2O_2$)-induced DNA damage and apoptosis in Chang liver cells. Methods: Chang liver cells were pretreated with various concentrations of EEPR and then challenged with 0.5 mM $H_2O_2$. The cell viability and apoptosis were assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and flow cytometry analysis, respectively. The levels of reactive oxygen species (ROS), mitochondrial membrane potentials (MMPs) and adenosine tri-phosphate (ATP) contents were measured. Expression levels of Bcl-2 and Bax were also determined using Western blot analysis. Results: The results showed that the decreased survival rate induced by $H_2O_2$ could be attributed to the induction of DNA damage and apoptosis accompanied by the increased production of ROS, which was remarkably protected by EEPR. In addition, the loss of $H_2O_2$-induced MMPs and ATP contents was significantly attenuated in the presence of EEPR. The inhibitory effect of EEPR on $H_2O_2$-induced apoptosis was associated with up-regulation of Bcl-2 and down-regulation of Bax, thus reducing the Bax/Bcl-2 ratio. Conclusions: Our data prove that EEPR protects Chang liver cells against $H_2O_2$-induced DNA damage and apoptosis by scavenging ROS and thus suppressing the mitochondrial-dependent apoptosis pathway.

임신 쥐의 철 과잉섭취가 조직의 산화적 스트레스에 미치는 영향 (Effects of Iron Overload during Pregnancy on Oxidative Stress in Maternal Rats)

  • 본 연구는 임신기 동안 어미쥐의 철 섭취 수준이 모체의 철 대사지표와 산화적 스트레스 및 임신의 결과에 미치는 영향을 검토하기 위해 비임신쥐를 대조군으로 하여 수행하였다. 10주령 200 g 이상 된 암컷과 수컷 흰쥐를 1 : 1로 교배시켜 임신을 확인한 후, 비임신쥐 (대조군)와 임신쥐 (실험군)에게 식이 중 철 수준이 정상수준 (AIN-93G diet 수준, 35 mg Fe/kg diet), 고수준 (정상의 10배, 350 mg Fe/kg diet) 및 과잉수준 (정상의 30배, 1,050 mg Fe/kg diet)의 3가지 실험식이를 급여하였다. 임신 19일째 되는 날, 비임신쥐와 임신 쥐를 희생시켜 분석한 결과는 다음과 같다. 체중의 증가나 식이섭취량은 철 섭취 수준의 영향을 받지 않았고, 임신의 결과로서 태아의 수, 태아체중 및 태반무게도 철 섭취 수준의 영향은 받지 않았다. 헤모글로빈, 헤마토크릿, 혈청 철 농도 등의 혈액지표들은 철 섭취 수준의 영향은 받지 않았으나 임신에 의한 감소 경향을 보였다. 임신쥐의 철 섭취 수준의 증가에 따라 간과 지라 조직의 철 함량이 유의적으로 증가하였다. 또한 임신쥐의 간에서 페리틴 단백질 수준이 철 섭취의 증가에 따라 현저히 증가하였다. 산화적 손상지표인 지질과산화물 (MDA)은 철 섭취수준의 영향을 받지 않았고, 단백질 산화물 (protein carbonyls)은 비임신쥐와 임신쥐에서 모두 철 과잉 섭취군의 경우 유의적으로 증가하였다. 항산화효소 중에서는 철 과잉섭취군에서 간의 GPx 활성이 유의적으로 감소하였다. 결론적으로, 임신기 동안 어미쥐의 철 섭취수준의 증가는 어미쥐의 혈액지표와 임신의 결과에는 유의적인 영향을 미치지 않았지만, 간 조직 내 철 함량과 페리틴 단백질 수준을 유의적으로 증가시켰으며, 간 조직에서 단백질 산화물인 protein carbonyl 농도를 증가시키고, 항산화효소 중 특히 GPx의 활성 감소를 초래하였다. 또한 간 조직에서 세포사멸을 억제하는 중요한 인자인 Bcl-2 단백질 수준이 임신쥐에서 철 섭취의 증가에 따라 유의적으로 감소하였다. 이러한 영향이 철을 정상 수준의 10배 섭취한 군에서는 약하게 나타났으나, 30배 과잉으로 철을 섭취한 군에서는 유의적으로 차이를 보였다. 이 결과는 임신시 철 과잉 섭취의 해로운 영향이 지금까지 철 대사의 측정도구로 삼아왔던 혈액지표의 변화 보다는 체내에서 일어나는 조직의 산화적 스트레스의 증가나 조직 내 철의 축적 등에 보다 더 민감하게 반영됨을 알 수 있었다. 따라서, 철 과잉 섭취가 모체 뿐만 아니라 태생 조직의 산화적 스트레스에도 영향을 줄 수 있음을 시사하였다.

Endogenous catalase delays high-fat diet-induced liver injury in mice

  • Non-alcoholic fatty liver disease (NAFLD) has become the most prevalent liver disease in parallel with worldwide epidemic of obesity. Reactive oxygen species (ROS) contributes to the development and progression of NAFLD. Peroxisomes play an important role in fatty acid oxidation and ROS homeostasis, and catalase is an antioxidant exclusively expressed in peroxisome. The present study examined the role of endogenous catalase in early stage of NAFLD. 8-week-old male catalase knock-out (CKO) and age-matched C57BL/6J wild type (WT) mice were fed either a normal diet (ND: 18% of total calories from fat) or a high fat diet (HFD: 60% of total calories from fat) for 2 weeks. CKO mice gained body weight faster than WT mice at early period of HFD feeding. Plasma triglyceride and ALT, fasting plasma insulin, as well as liver lipid accumulation, inflammation (F4/80 staining), and oxidative stress (8-oxo-dG staining and nitrotyrosine level) were significantly increased in CKO but not in WT mice at 2 weeks of HFD feeding. While phosphorylation of Akt (Ser473) and $PGC1{\alpha}$ mRNA expression were decreased in both CKO and WT mice at HFD feeding, $GSK3{\beta}$ phosphorylation and Cox4-il mRNA expression in the liver were decreased only in CKO-HF mice. Taken together, the present data demonstrated that endogenous catalase exerted beneficial effects in protecting liver injury including lipid accumulation and inflammation through maintaining liver redox balance from the early stage of HFD-induced metabolic stress.

The effects of physical training on antioxidative status under exercise-induced oxidative stress

  • This study investigated the effect of physical training and oxidative stress on the anti oxidative activity and on plasma lipid profile. Forty eight rats were given either a physical training or no training for 4 weeks and were then subdivided into 3 groups: before-exercise (BE); during-exercise (DE); after-exercise (AE). The antioxidative activity was evaluated with the activities of catalase in plasma and superoxide dismutase (SOD), the ratio of reduced glutathione/ oxidized glutathione (GSH/GSSG) and the level of malondialdehyde (MDA) in liver. The plasma concentrations of triglyceride (TG), total cholesterol (TC), high-density lipoprotein-cholesterol (HDL-C)) were also compared. Compared to those of non-training group. catalase activities of training group were lower before exercise but higher during and after exercise. SOD activities were higher regardless of exercise. GSH/GSSG ratio was higher before exercise but was not significantly different during exercise and even lower after exercise. There were no differences between non-training group and training group in MDA levels regardless of exercise. Compared to those of non-training group, atherosclerotic index of training group was lower after exercise and there were no significant differences before and during exercise. There were no differences between non-training group and training group in HDL-C regardless of exercise. These results suggest that moderate physical training can activate antioxidant defenses and decrease the atherosclerotic index and this beneficial effect is evident under exercise-induced oxidative stress.
