The Effect of Isoflavone and/or Grape Seed Oil Supplementation on Blood Lipid Profiles and Bone Strength in Ovariectomized Female Rats (이소플라본과 포도종실유의 보충이 난소절제 흰쥐의 혈중지질 패턴 및 골격강도에 미치는 영향)

  • The study was performed to investigate the effects of isoflavone and/or grape seed oil supplementation on serum and liver lipid profiles and bone strength in ovariectomized female rats. Female Spraque-Dawley rats were assigned into 4 groups of ovariectomized (O), isoflavone (0.085 mg/100 g b.w/day) in ovariectomized rats, grape seed oil in ovariectomized rats and isoflavone and/or grape seed oil in ovariectomized rats. After 8 weeks, biochemical profiles of serum, liver and bone were analyzed. Total food intakes, body weight gains and FER (food efficiency ratio) were not statistically significantly different among groups. Total cholesterol, triglyceride and LDL-cholesterol levels in serum were decreased by fed of isoflavone and/or grape seed oils. However, crude lipid and total cholesterol contents in liver were not affect of isoflavone and/or grape seed oil. The hepatic glutathione contents were increased by isoflavone and/or grape seed oil fed. The hepatic glutathione-S-transferase activity in isoflavone and/or grape seed oil supplemented groups were higher than that O group. Bone (scapular and femur bone) dry weight, femur of max weight and bending strength were no significant difference among groups. Our finding suggest that isoflavone/grape seed oils might have potential role for serum lipid profiles improvement and bone strength in vivo.

The Effects of Shellfish Hydrolysates on Lipid Metabolism in Rats Fed High-Fat Diet (고지방식이를 급여한 흰쥐의 패류가수분해물 급여 효과)

  • This study was conducted to investigate the effects of shellfish hydrolysate on lipid metabolism in rats fed high fat diet. Male Sprague-Dawley rate weighting approximately 110g were fed basal control diet, high fat diet and high fat diet plus 4 different shellfish hydrolyates for 4 weeks. The shellfish hydrolysates from the different sources, were oyster, hard-shelled mussel, little neck clam and march clam. After 4 weeks, serum GOT, GPT, ${\gamma}$-GTP, triglyceride and total cholesterol was significantly decreased in shellfish hydrolysates supplementation with high fat diet compared to basal control and high-fat group(p<0.05). The total lipid and cholesterol content in liver showed significant decrease(p, 0.05). There were no different in serum GPT, HDL-cholesterol, liver total cholesterol and lipid of rats between basal control diet and high-fat diet. The unsaturated fatty acids, specific components of shellfishes were a little components in shellfish hydrolysate as they were a low and not different among the groups and were most well reflected in liver and plasma. Considering digestive and absorptive process of in human body, it was assumed that the hypolipidemic effect of shellfish was not under the influence of unsaturated fatty acids but the other components, peptides, taurine and betaine and so on was detected in the process of hyperlipidemia induced by high-fat diet.

  • This study investigated the effects of cell cultured Acanthopanax senticosus extract (ASE) supplementation and swimming exercise on lipid profiles and carnitine concentrations in C57BL/6J mice fed high fat diets. Male C57BL/6J mice (n=50), aged 4 weeks, were divided into 5 groups based on exercise and/or ASE supplementation (0.5 g/kg of body weight): normal diet (N-C), high fat diet (H-C), high fat diet non-supplement & exercise (H-NSE), high fat diet supplement & no exercise (H-SNE), high fat diet supplement & exercis (H-SE). Liver nonesterified carnitine (NEC) was significantly higher in the H-SNE group than in the H-C group, and liver total carnitine (TCNE) levels were significantly higher in the H-SNE group than in the H-NSE and H-SE groups. Liver and muscle carnitine palmitoyltransferase-I (CPT-I) mRNA levels tended to be higher with ASE supplementation and/or exercise. These results suggest that supplementation with ASE and/or exercise might have a role in improving lipid oxidation.

Effects of Calcium Intake on Lipid Contents and Enzyme Activity in Rats of Different Ages (연령이 다른 흰쥐에 있어 칼슘이 섭취수준이 체내 지질함량과 효소활성에 미치는 영향)

  • The purpose of this study was to investigate the effect of calcium intake on lipid contents and enzyme activities in rats of different ages. Lipid levels in serum and liver and GOT, CPK and LDH activities in serum were compared in rats of different ages(4 weeks and 10 months) that were fed various levels of calcium(50, 100, 200% of requirement)for 3 weeks. Body weight gain and feed efficiency ratio were significantly higher in young rats than in adults. Serum calcium level was increased by elevation of calcium intake levels were decreased. Liver phospholipid and triglyceride levels in the high-cal-terol and triglyceride levels were decreased. Liver phospholipid and triglyceride levels in the high calcium group were significantly lower than those in other groups. Serum GOT and LDH activities of adults were significantly higher in low-calcium group than those in adequate/high-calcium groups. However, serum CPK activity of adults was significantly higher in high-calcium group than that in low/adequate-calcium groups. The results of this study suggest that adequate calcium intake may have protective effects ont he alteration of lipid and enzyme activity in rats.

  • This paper presents the effect of the supramolecular complex of GA (Glycyrrhizic acid) and Mt(menthol) and GA: Mt (4: 1) obtained on their basis can restore functional dysfunction of the liver mitochondria in alloxan diabetes, ie, inhibit lipid peroxidation. The hypoglycemic activity and mitochondrial membrane stabilizing properties of the supramolecular compound GA: Mt (4: 1) in alloxan diabetes were more pronounced than those of menthol, GA and its GK: Mt (2: 1) and GA: Mt (9: 1) compounds. According to the results obtained, the concentration of GA did not affect the peroxidation of lipid membranes of the liver mitochondria. However, a concentration of 15 μM of GA was found to reduce LPO (lipid peroxidation) formed by the effect of Fe2+ / ascorbate on the mitochondrial membrane by 58.0 ± 5.0% relative to control. Effects of Dietary Cadmium on the Respiratory Burst of Phagocytes and the Antioxidant Defense in Cultured Red Seabream (Pagrus major)

  • To examine effects of cadmium on the respiratory burst of kidney phagocytes and antioxidant defense in liver, juvenile red seabream Pagrus major were fed a cadmium-incorporated diet $(1g\;CdC1_2/kg\;diet)$. The respiratory burst activity measured by chemiluminescence (CL) was significantly reduced by oral intake of cadmium. Lipid peroxidation in liver expressed as thiobarbituric acid reactive substances (TBARS) was significantly higher in the fish fed a cadmium-incorporated diet than that of the fish fed a control diet both on Day 3 and Day 9. Liver Glutathione S-transferase (GST) activitiy was significantly increased both on Day 3 and Day 9 by feeding a cadmium-incorporated diet, when compared with the controls. From the present results, it can be concluded that oral intake of cadmium in red seabream is associated with marked reduction of respiratory burst capacity of kidney phagocytes which can elevate susceptibility of fish against infecting pathogens. Cadmium administration also elicits significant increment of lipid peroxidation in liver, and fish try to detoxify cadmium by increasing GST activity.

Effects of Sayeoksanhap-Pyeongweisan-Gamiban(SPG) on hematological changes in animals damaged by CCl4 (사역산합평위산가미방(四逆散合平胃散加味方)이 사염화탄소에 의한 간손상 모델 동물의 혈액학적 변화에 미치는 영향)

  • Objectives : This study was designed to investigate protective effects of Sayeoksanhap-Pyeongweisan-Gamiban(SPG) on hematological changes in rats damaged by CCl4 Methods : We investigated the effects of SPG on hematological changes in terms of AST/ALT levels, Total cholesterol, HDL-cholesterol and triglyceride in serum and lipid peroxidation in rat liver. Results : Administration of SPG decreased AST level in serum effectively. In addition, lipid peroxidation in liver tissue, which was elevated by injection of CCl4, was decreased by administration of SPG. In contrast, ALT level, Total cholesterol, HDL-cholesterol and triglyceride in serum were not affected by administration of SPG. Conclusion : These results suggest that oral administration of SPG was useful to protect liver against CCl4 by its antioxidant action in liver tissues.

Effect of Potato Polyphenolics on Lipid Peroxidation in Rats (감나 폴리페놀이 흰쥐의 생체내 지질과산화에 미치는 영향)

  • The total contents of polyphenolics in potatoes measured by Folin Denis method were 42~76mg/100g in fresh weight. A major phenolic component contained in potato polyphenolics was identified as chlorogenic acid(3.6~15mg/100g in fresh weight). The antioxidative effects of potato polyphenolics and chlorogenic acid on the lipid peroxidation of liver microsome were studied in vivo and in vitro systems by measuring the formation of thiobarbituric acid reactive substances(TBARS) and the content of urine 8 hydroxy deoxyguanosine(OHdG). The TBARS contents of liver showed an increase in the cholesterol diet compared to those in the normal diet. This trend, however, was minimized when potato polyphenolics and chlorogenic acid were supplemented in the cholesterol diet. On the other hands, urinary 8 OHdG contents showed a marked increase with the supplementation of potato polyphenolics in the cholesterol diet. However, there was a trend of marked decrease by the supplementation of chlorogenic acid. In vitro study, potato poly phenolics and chlorogenic acid effectively inhibited the formation of TBARS in liver microsomal system in a dose dependent manners. These results suggest that potato polyphenolics exerts an antioxidative activity in cholesterol fed rat liver.

  • Objectives: This study was designed to investigate protective effects of Sayeoksanhap-Pyeongweisan-Gamibang(SPG) on hepatological changes in rats damaged by $CCl_4$. Methods : We investigated the effects of SPG on hepatological changes in terms of AST/ALT levels, Total cholesterol, HDL-cholesterol and triglyceride in serum and lipid peroxidation in rat liver. Results : Combination treatment with acupuncture and oral administration of SPG decreased AST and ALT in serum. Combination treatment also decreased lipid peroxidation in liver and triglyceride in serum. In addition, HDL-cholesterol in serum was increased significantly by combination treatment. Contrary, level of total cholesterol was not changed by combination treatment. Conclusion : These results suggest that Combination treatment with acupuncture and oral administration of SPG was useful to protect liver against $CCl_4$ by its antioxidant action in liver tissues.

  • The present study is aimed at finding the effect of B-20 drops, a homoeopathic formulation, in streptozotocin (STZ) induced diabetic rats. B-20 drops comprises of the constituents derived from plants and other natural sources, and are generally prescribed by the homoeopathic physician, in cases of hyperglycemia and diabetes. The elevated levels of fasting blood glucose and pancreatic lipid peroxides observed in rats treated with STZ were significantly reduced by the treatment of B-20 drops. The reduced liver glycogen contents were also brought back to near normal level by B-20 drops treatment in STZ diabetic rats. STZ induced histopathological changes in pancreas and liver was also partially reversed by B-20 drops. The findings indicate that B-20 drops help in improving the glycogen stores in the liver and prevents STZ induced damage through free radicals by decreasing the pancreatic lipid peroxides levels.
