The combined effects of iron and selenium status on glutathione peroxidase (GSHPx) activity, cytochrome P-450 activity, and lipid peroxidation in the liver and intestinal mucosa of rats were investigated. In experiment one, four experimental groups (+Se+Fe, -Se+Fe, +Se++Fe, -Se++Fe) were manipulated for 3 weeks with intramuscular administration of irondextran (++Fe) and/or normal diet (+Fe) and deionized water (-Se) and/or selenium-supplemented deionized water (+Se). In experiment two, 2% dietary carbonyl iron (instead of the parenteral administration) was fed for 3 weeks to rats. Body weight of rats was significantly decreased in both parenterally and orally iron-overloaded groups (p<0.01), regardless of Se supplement. Serum iron was significantly increased in parenterally iron-overloaded groups but it was marginally increased in orally iron-overloaded groups. There was no significant difference in hemoglobin content among experimental groups in either experiment one or two. Total iron in the small intestine, intestinal mucosa, and livers was significantly high in both parenterally and orally iron-overloaded rats, regardless of selenium status. In the liver and intestine, GSHPx activity was significantly higher in all selenium-supplemented groups, compared to Se-deficient groups (p<0.01) and lipid peroxidation was significantly enhanced in both parenterally and orally iron-overloaded groups, compared to iron-adequate groups. There was no significant difference in cytochrome P-450 activity in the livers between groups in both experiment one and two. These results indicated that GSHPx activity in liver and intestinal mucosa was depended on selenium status, regardless of iron status, and iron-overload enhances lipid peroxidation in liver and intestinal mucosa by increasing the tissue iron content.

Effects of dietary lipid level and source (squid liver oil being rich in n-3 HUFA, soybean oil being rich in 18:2n-6, and linseed oil being rich in 18:3n-3) in fishmeal-based diet on growth and body composition of grower sunshine bass raised in seawater were investigated. Fifteen grower (an initial weight of 146.8$\pm$0.23 g) sunshine bass were randomly distributed into 27 of 250 L fiber reinforced plastic flow-through tanks. Fish were hand-fed to satiety twice daily for 6 days a week throughout the feeding trial. Survival was over 97% and not significantly affected by either dietary lipid level or lipid source (n-3 highly unusaturated fatty acid, HUFA). Weight gain of fish tended to improve with dietary n-3 HUFA level up to 2.9%, but sharply decreased at 3.5%. The best weight gain was obtained in fish fed the diet supplemented with 6% squid liver oil and 3% soybean oil. FER and PER were not significantly affected by either dietary lipid level or dietary lipid source. The lowest moisture content of the whole body was observed in fish fed the diet supplemented with 12% squid liver oil and highest for the diet supplemented with 9% linseed oil, respectively. Protein content of fish was not significantly affected by either dietary lipid level or dietary lipid source. However, lipid content of the whole fish tended to increase with an increase of either dietary lipid level or dietary n-3 HUFA level, except for fish fed the diet supplemented with 9% linseed oil. Ash content of fish fed the diet with no supplementation of oil was highest and lowest for the diet supplemented with 9% soybean oil, respectively. Significant differences in saturated fatty acids (16:0, 18:0 and 24:0), monoene (18:1n-9), 18:2n-6, 20:5n-3 and sum of n-3 HFUA of fish were observed. In considering these results, it could be concluded that supplementation of 9% oil combined with 6% squid liver oil and 3% soybean oil into fishmeal-based diet was the most recommendable for growth of grower sunshine bass raised in seawater.
The present study was conducted to investigate te effect of sea tangle and hypoglycemic agent(Metformin) on lipid peroxidation and antioxidants levels in normal and diabetic rats. Male Sprague-Dawley rats were fed AIN-76 based experimental diets containing 5% (w/w) cellulose or 15%(w/w) sea tangle for 3 weeks, and then rats of diabetic groups were rendered diabetic by intramuscular injection of streptozotocin(STZ, 45mg/kg BW). After induction of diabetes Metformin(350mg/kg BW) was given once a day using a feeding tube for 5 days. Blood glucose level in diabetic rats fed sea tangle was significantly lower than that of diabetic rats fed cellulose. Metformin feeding resulted in further lowering blood glucose. Plasma and liver microsomal levels of lipid peroxides were increased significantly in diabetic rats as compared to normal rats, and the plasma level tended to be decreased by sea tangle feeding. Plasma level of retinol was reduced by STZ treatment, but it was increased by Metformin feeding in diabetic rats fed sea tangle. The liver contents of retinyl plamitate were reduced in diabetic rats but recovered by sea tangle feeding to some extent. Liver contents of total vitamin A were increased significantly by sea tangle in diabetic rats. Although difference in either plasma or liver level of $\alpha$-tocopherol by diet and STZ treatment was not significant, $\alpha$-tocopherol levels were the highest in the group with simultaneous feeding of sea tangle and metformin. Liver contents of zinc and copper were not influenced by either STZ treatment or sea tangle feeding. The present study indicates that the lowering blood glucose level could be attained by simultaneous trial of sea tangle diet and hypoglycemic agent and the increased oxidative stress caused by STZz treatment could be relieved by sea tangle feeding.
Inhibitory effects of the methanol extract, hexane extract, methanol soluble fraction (MSF) and juice from 3 weeks fermented Kimchi on the tumor formation in sarcoma-180 cell transplanted mice were studied. Effects of the solvent extracts and juice of the Kimchi on the levels of lipid peroxide, glutathione, and the enzyme activities of the liver were also investigated in normal and sarcoma-180 cell transplanted mice. At 32 days following trans-plantation, MSF reduced the tumor formation by 54% compared with the control group, resulting in the smallest tumor weight. Lipid peroxided content in liver increased by the transplantation of sarcoma-180 cells. However, it decreased when MSF of Kimchi was treated to the mice. MSF also suppressed xanthine oxidase activity in cytosol of the liver cells in mice transplanted by sarcoma-180 cells. Kimchi extracts had no inhibitory effect on hepatic aminopyrine-N-demethylase activity in sarcoma-180 cell transplanted or normal mice. Methanol extract and hexane extract of Kimchi slightly increased hepatic glutathione contents in sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice. The injection of MSF from Kimchi markedly increased glutathione levels in the liver of sarcoma-180 treated mice compared to the controls. The MSF recovered the activities of hepatic glutathione reductase and glutathione S-transferase that decreased by the injection of sarcoma-180 cells. These results showed that MSF of Kimchi could suppress the growth of tumors, inhibiting lipid peroxide production and xanthine oxidase activity, in mice. We also suggested that Kimchi extract might play an important role in the prevention of cancer by enhancement of the glutathione level itself as well as via glutathione reductase and glutathione S-transferase.
Certain indigestible oligosaccharides may benefit gastrointestinal tract via fermentation and proliferation of desirable bacterial species. The purose of this study was to elucidate the effect of selected oligosaccharides, such as fructooligosaccharides(FOS), soybean oliosaccharides(SOE), and highly concentrated branched oligosaccharides(HiBOS), on fecal micorflora proliferation, lipid concentration, lipid peroxide formation and antioxidant enzymes acitivies in plasma and liver of the rats. Thirty two male Sprague-Dawley rats were randomly assigned to one of four treatments ; 1) control diet(AIN-93G diet); 2) control diet +5% FOS ; 3) control diet + 5% SOE ; 4) control diet + 5% HiBOS. The duration of the study was 4 weeks. Fecal bifidobacteria concentration were significantly higher (p<0.05) in the HiBOS group compared with the control after 4 weeks of dietary treatment. FOS and SOE groups also had higher fecal bifidobacteria levels than control, but statistical significance was not found. The concentration of plasma total lipid was decreased by oligosaccharide consumption, especially in HiBOS group(p<0.05). The concentration of plasma total triglyceride was significantly lower in all of the oligosaccharide containing groups compared with the control(p<0.05). The plasma total cholesterol concentration tended to be lower in the oligosaccharide consuming groups than control. The concentrations of hepatic total lipid, triglyceride and total cholesterol were not affected by consumption of oligosaccharides. Thiobarbituric acid reactive substance(TBARS) concentrations and antioxidant enzyme activities in plasma and liver were not affected much by experimental diets. There results suggest that dietary oligosaccharides may be beneficial for increasing intestinal bifidobacteria and lowering plasma lipid levels.
This study was performed to investigate effect of dietary fibers in rice and barley on glucose, lipid and cadmium (Cd) metabolism in the rat. Fifty-six male Spague-Dawley rats weighing 244.6$\pm$2.7g were blocked into eight groups according to body weight and raised for four weeks with diets containing 0 or 0.04%(w/w) CdCl2 and four different carbohydrate sources, starch, rice flour, barley flour and mixture of rice and barley flour(7 : 3, w/w). Total dietary fibers and $\beta$-glucan contents of barley were about three times higher than those of rice (10.75% vs. 3.94%, 3.11% vs. 1.06%, respectively). Food intake, weight gain, food efficiency ratio, liver and kidney weights were lower in Cd exposed groups, and barley group among Cd exposed animals showed highest weight gain, food efficiency ratio and organ weights. Fasting serum glucose levels were not significantly different among groups, Serum cholesterol level was lowest in Cd exposed barley group. Serum HDL-cholesterol level was higher in none-Cd exposed starch and barley groups, and HDL-cholesterol : total cholesterol ratios were higher in none-Cd rice and mixed flour groups than other groups. Liver total lipid and triglyceride levels were lowest in barley groups regardless of Cd administration. Fecal total lipid, cholesterol and triglyceride excretions were high in barley and mixed flour groups. Liver Cd concentrations were low in Cd exposed barley and mixed flour groups. In Cd exposed barley group, fecal weight and Cd excretion were highest and Cd retention ratio was lowest among groups. Small intestine metallothionein(MT) concentration was highest in Cd exposed rice group, and kidney MT concentration was highest in Cd exposed barley group. In conclusion, cereals showed different effects on lipid and Cd metabolism that might be mediated by dietary fibers in cereals. Especially $\beta$-glucan-rich barley group showed greatest lipid and Cd lowering effects by increasing fecal lipids and Cd excretions.
The activities of aminopyrine demethylase which is marker enzyme of the microsomal drug-metabolizing system, NADPH-cytochrome a reductase and glutathione peroxidase were measured during the course of liver regeneration after about seventy percent hepatectomy in Wistar rats. In addition, the extent of lipid peroxidation and contents of cytochrome P-450 were also measured. Partial hepatectomy produced a significant depression in aminopyrine demethylase, to reach a minium about 24 hours after operation, but this activity was increased to normal value during regeneration. On the other hand, in sham-operated animals, this showed no change. All the activities of NADPH-chrome P-450 contents of liver microsomes were rapidly decreased at the early stage of regeneration. These values returned to normal after 7 days. By contrast, the activity of glutathione peroxidase was nearly unchanged. According to these results, at the early stage of regeneration, the decrease of cytochrome P-450 and NADPH-cytochrome c reductase activity lead to decrease of lipid peroxidation and drug metabolizing enzyme activity. But these phenomena were not detected after 7 days of regeneration.

The effects of acute and chronic ethanol administration on hepatic and cerebellar glutathione (GSH) statuses and lipid peroxide levels in rats were investigated. In the liver, chronic ethanol feeding (6.9 g/kg, per day) as 10% (v/v) drinking water for 4 weeks produced a slight decrease of total GSH and an increase in the ratio of GSSG/total GSH without change of GSSG (oxidized GSH). Lipid peroxide level however was not modified. Many other studies have shown the acute ethanol loading effect in the rat liver, that is moderate decrease of total GSH and elevation of lipid peroxide level. Relating to this, it was observed that total GSH in the plasma obtained from post. hepatic inferior vena cava was increased by acute ethanol injection (50 mmol/kg, i.p.). This increased hepatic efflux of GSH into blood, in addition to the promoted antioxidative utilization of GSH, could be suggested as one of the possible reasons for the decrease of hepatic GSH induced by ethanol load. In the cerebellum, acute ethanol load did not change the total GSH and GSSG, but increased the lipid peroxidation rate. In the chronic, neither GSH pattern nor lipid peroxidation rate was changed.
The overall purpose of this study was to investigate the effects of Chunggukjang and Greentea-Chunggukjang on the lipid profile, lipid peroxidation and antioxidative enzyme activities of liver tissue in growing male rats fed cholesterol. Twenty seven rats were divided into three treatment groups (Control, Chunggukjang and Greentea-Chunggukjang) and were given experimental diets with 1% cholesterol for 9 weeks. All rats in this study were fed a casein-based diet. Chunggukjang groups were fed diet containing 33.1% Chunggukjang powder. The Chunggukjang and Greentea-Chunggukjang groups showed significantly lower weight gain, food efficiency ratio than the control group regardless of Chunggukjang type. Serum total cholesterol was significantly lower in the Chunggukjang group than in the control group, whereas serum triglyceride and atherogenic index were significantly lower in the Greentea-Chunggukjang group than in the control group. Hepatic triglyceride contents was not significantly different among the diets. However, hepatic cholesterol content was significantly lower in the Greentea-Chunggukjang group than in the control group. Lipid peroxidation of malondialdehyde (MDA) contents was significantly lower in the Chunggukjang and Greentea-Chunggukjang groups than in the control group. Activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) in liver tissue of the Chunggukjang and Greentea-Chunggukjang groups were not significantly different. It can be concluded that Chunggukjang and Greentea-Chunggukjang influence lipid profile and hepatic malondialdehyde contents in growing male rats fed cholesterol.

Effects of kimchi on lipid metabolism and immune function were studied in experiments using 63mals SD rats fed 6 inds of Baechu-kimchi containing diet during 4 weeks. Three kinds of freeze dried kimchi differ in fermentation period (not fermented, 3-, 6-week-fermented at 4$^{\circ}C$) were added at 5%, 10% of the diet containing 15% lard. The levels of serum total lipid and triglyceride and the content of liver total lipid and triglyceride of all kimchi groups were lower than those of a control group. But the levels of serum and liver cholesterol is not affected by kimchi intake. The triglyceride concentration of epididymal fat pad and feces of kimchi groups were higher than those of a control group. The food efficiency ratio, epididymal fat pad weight of 3-, 6-week-fermented kimchi 10% groups were significantly lower than control and not-fermented kimchi groups. Especially 6-week-fermented kimchi groups showed adipocytes, less in number and larger in size than those of other groups. The blastogenesis of spleen lymphocytes to LPS was higher in rats fed fermented kimchi diets than rats fed control and not-fermented kimchi diet. These results suggest that kimchi stimulates lipid mobilization to epididymal fat pad and lipid excretion via feces, so lower serum and liver triglyceride concentration. The fermented kimchi stimulate the proliferation of B cell and lower the lipid accumulation in epididymal fat pad, especially kimchi fermented for 6 weeks at 4$^{\circ}C$ lower the adipose cell number.
