Vitamin A 유도체로 인한 간의 약물대사효소 변동 (The Effect of Vitamin A Derivatives on the Activity of Drug-metabolizing Enzyme in Rat Liver)

  • It has been known that retinoids are intrinsically of critical importance for control of premalignant epithelial cell differentiation. In the absence of retinoids, normal cellular differentiation and growth does not occur in epithelia such as those of trachea and bronchi. Furthermore, it was also reported that retinoid deficiency enhanced susceptibility to chemical carcinogenesis in the respiratory system, in the bladder, and in the colon of the experimental animal. In 1974, Bollag examined the effects of synthetic retinoids in prevention of development of cancer and demonstrated synthetic retinoids to have more favorable therapeutic index than retinoic acid for causing regression of skin papilloma in mice. Therefore, it was assumed that this anticarcinogenic effect of vitamin A derivatives could be due to modification of the metabolism of the carcinogenic polycyclic hydrocarbon, which must first be activated to exert their effect. Hill and Shih reported that vitamin A compounds and analogs had inhibitory effect on drug metabolizing enzyme from liver and lung tissue of mouse and hamster. Lucy suggested that the chemoprevention effect of vitamin A derivatives is due to reaction with molecular oxygen, and it is possible that inhibition of hydroxybenzpyrene formation is a result of this property. On the other hand, butylated hydroxytoluene which is a potent antioxidant strongly inhibited the formation of mammary tumor induced by dimethylbenranthracene. Also, it was observed that this antioxidant inhibited cancer induction in rats by N-2-fluo-renylacetamide. The purpose of this experiment was to investigate the effect of vitamin A derivatives such as retinoic acid and retinoid on drug-metabolizing enzyme and to determine whether riboflavin tetrabutylate or vitamin E could prevent of modify any changes induced by vitamin A delivatives in the rats. The results obtained were as followings. 1) Body weight was significantly reduced by retinoic acid, but not by retinoid. 2) Retinoic acid markedly increased liver weight while retincid showed no effect on liver weight. Treatment of riboflavin tetrabutylate did not affect retinoic acid-induced change in both body weight and liver weight. 3) Both retinoic acid and retinoid remarkably decreased the activity of aminopyrine demethylase. Pretreatment of riboflavin tetrabutylate, however, prevented inhibitory effect of retinoic acid on the enzyme activity. 4) No significant effect of vitamin E on aminopyrine demethylase was observed in both groups treated with retinoic acid and retinoid.

산화된 지방식이를 급여한 흰쥐에서의 머루즙 투여가 지질대사와 항산화에 미치는 영향 (Effect of Wild Grape Juice on Lipid Metabolism and Antioxidaive Activity in Rat Fed High Oxidized Lipid)

  • This study is to find out the antioxidative effect and serum and liver lipid composition of wild grape juice in vivo. Forty 6-week-old white Sprague Dawley rats were divided into 4 groups such as normal lipid group, normal lipid group with wild grape juice, oxidized lipid (basic diet plus 10% of oxidative lipid) group and oxidized lipid group with wild grape juice, and 2ml juice was provided everyday. After 4 weeks of feeding with experimental diet each groups were examined for the antioxidant enzyme activity in blood and liver microsome and their serum and liver lipid composition. Glutanthione peroxidase activity in blood was significantly higher in oxidized lipid group with wild grape juice than in oxidized lipid group. Glutanthione peroxidase activity showed no difference depending on wild grape juice supplementation. Glutanthione peroxidase activity in liver was significantly higher in the groups with wild grape juice than in the groups supplemented only with oxidized lipid. Glutanthione reductase activity showed no difference depending on the supplementation of wild grape juice. Serum triglyceride level in the group supplemented with oxidized lipid diet and wild grape juice showed similar value to the normal lipid group and the normal lipid group with wild grape juiceoxidized fa6. Liver total lipid in the group supplemented with oxidized lipid and wild grape juice showed similar value to the normal lipid group and the group supplemented with normal lipid and wild grape juice. And it was lower than that of oxidative lipid group without juice. The liver triglyceride level in the group supplemented with normal lipid and wild grape juice was lower than that in the oxidative lipid group, but it was as low as in the group supplemented only with normal lipid.

알코올 전처치한 흰쥐에 Cyclohexane 투여가 Cytochrome P-450 dependent aniline hydroxylase 활성에 미치는 영향 (Effect of Cyclohexane Treatment on the Liver Cytochrome P-450 Dependent Aniline Hydroxylase Activity in Alcohol-pretreated Rats)

  • To evaluate the effect of cyclohexane(CH) treatment on the liver cytochrome P-450 dependent aniline hydroxylase(CYPdAH) activity in alcohol-pretreated animals, CH(1.56 g/kg body weight) was intraperitoneally administered to Sprague-Dawley male rats, which had been drunk 15% alcohol in distilled water for 1,3 and 6 weeks. CH was injected to rats 4 times every other day and the animals were sacrificed at 24 hours after injection of CH. In the alcohol-pretreated rats, liver injuries were not demonstrated on the basis of the liver weight per body weight, the levels of serum alanine aminotransferase and hepatic microsomal glucose-6-phosphatase activities. By the CH treatment, alcohol-pretreated animals showed the significantly increased activity of hepatic microsomal CYPdAH. Concomitantly $V_{max}$ value in CYPdAH was more increased, whereas $K_{M}$ value more decreased in alcohol-pretreated animals by the treatment of CH. In conclusion, the increasing cause of microsomal CYPdAH in CH-treated rats pretreated with alcohol may be due to induction of enzyme protein in rat liver.r.r.

넙치 간에 있어 가수분해 효소의 부분정제 및 특성규명 (Partial purification and characterization of phosphatidylcholine hydrolyzing enzyme from liver membrane of flounder , Paralichtys olivervaceus)

  • 본 실험은 넙치 (Paralichthys olivaceus) 간으로부터 membrane부분에 존재하는 PC 가수분해 효소의 특성에 대해 조사하였다. 먼저 간 조직을 초고속 원심 분리기와 nonion-detergent인 1% triton X - 100을 이용하여 membrane 부분을 분리하였으며, Heprain-Sepharose CL-6B 칼럼과 Heparin-5PW 칼럼을 이용하여 분리정제 하였다. 얻어진 PC 가수분해효소에 대한 반응 ․ 생성물을 확인하기 위해 autoradiography를 실시하였다. 지용성 부분의 결과에서 transphosphatidylation 반응의 결과물인 PEtOH을 형성하는 것으로 보아 PC-PLD임을 알 수 있었다. 얻어진 PC 가수분해효소에 대한 생화학적 특성을 조사한 결과 적정 pH가 6.5이하인 산성 조건 및 $37^\circ{C}$의 배양온도에서 최고 활성을 나타내었으며, 이가 이온들에 대한 영향의 경우 칼슘은 1.67mM 농도에서 최고 활성을 나타냈으나, 마그네슘은 활성에 영향을 미치지 않았다. 각종 세포막 기질에 대한 영향을 조사한 결과 PC는 $0.75\mu{M}$, PIP2는 $2.35\mu{M}$, PE는 $26.8\mu{M}$ 농도에서 최고 활성을 나타내었다. 이상의 결과부터 넙치 간조직의 막층부분에 존재하는 PC를 가수분해효소는 PC-PLD가 존재함을 알 수 있었다.

Effect of Ethanol on Mouse Liver Monoamine Oxidase

  • The effects of ethanol and acetaldehyde on monoamine oxidase activity in mouse liver mitochondrial fraction were studied. In vivo, a single dose of ethanol increased the hepatic monoamine oxidase activity compared to control group, and chronic ethanol consumption also increased the enzyme activity using tyramine, benzylamine or serotonin as substrate. Acetaldehyde, the metabolite of ethanol, significantly increased monoamine oxidase activity more than ethanol did. In contrast to the in vivo results, it was found that the monoamine oxidase activity was inhibited in vitro by ethanol or acetaldehyde in the dose-dependent manner.

알코올 투여후 마우스 간 크산틴 산화효소 활성에 미치는 인삼의 영향 (Effect of Ginseng Saponins on Xanthine Oxidase Activity after Ethanol Treatment in Mouse Liver)

  • A dose, 1g/kg of ethanol produced experimental hyperuricemia in mouse. Ginseng saponins were tested for their ability to alter the hepatic xanthine oxidase activity and the blood level of uric acid in the ethanol-treated mouse. Intraperitoneal injection of ginseng saponin 4mg/kg markedly decreased the xanthine oxidase activity in the ethanol-treated mouse liver. It was also observed that ginseng saponin reduced the blood concentration of uric acid in experimentally induced hyperuricemia by alcohol treatment. In vitro, it was found that a low concentration of ginseng saponin in the reaction mixture incresed the hepatic xanthine oxidase activity, while a high concentration inhibited both enzyme preparations of normal and ethanol treated mice. In contrast with the xanthine oxidase, uricase activity was not influenced by ginseng saponin as well as in vivo. These results suggest there is a possibility that ginseng saponin may have some therapeutic effect on gout and other hyperuricemia syndrome.

절식이 흰쥐의 간과 신장의 Thiobarbituric Acid-Reactive Substance량 및 항산화효소 활성도에 미치는 영향 (Effects of Total Dietary Restriction on the Contents of Thiobarbituric Acid-Reactive Substance and Antioxidant Enzymes in the Liver and Kidney of Rats)

  • The effects of total dietary restriction(100% restriction of energy intake) on thiobarbituric acid reactive substance(TBARS) contents and intracellular antioxidant enzymes activities in the liver and kidney of young male Sprague Dawley rats were studied. The TBARS contents were reduced in both liver and kidney, up to 77% and 79% of the control rats, fed ad libitum, respectively at 7 days after dietary restriction . Superoxide dismutase(SOD) activities in the liver and kidney of rats were increased significantly by total dietary restriction. However, the activity of catalase in kidney was decreased 27% at 6 days after dietary restriction, but this enzyme activity did not change in liver. The changes of glutathione peroxidae(GSHPx) and catalase activities in the liver and kidney of rats with dietary restriction were not significant. These result suggested that dietary restriction reduce the free radical induced by tissue damage, as determined by TBARS content, in both the liver and kidney but the changes of activities of antioxidant enzymes may not be a contributory factor in reducing oxidative damage to tissue.

흰민들레(Taraxacum coreanum)추출물이 급성 수은 중독된 생쥐의 간에 미치는 효과 (Effects of Dandelion (Taraxacum coreanum) Extracts on the Mouse Liver with Acute Toxicated by Mercury Chloride)

  • 본 연구는 급성 수은 독성에 대한 민들레의 효과를 알아보기 위하여 시도되었다. 30 g 내외의 생쥐를 대상으로 대조군, 수은(5 mg/kg) 투여군, 수은 투여 후 민들레(3 g/kg)를 구강투여 한 후 다시 24, 48, 72, 96시간, 1주일군으로 세분하여 간장 손상 억제효과를 알아보기 위하여 생화학적 및 조직학적 실험을 실시하였다. 혈액중 aspartate amiotransferase (AST)와 alanine aminotransferase (ALT)의 수치는 민들레 투여군이 수은 투여군보다 감소되었다. 또한 간조직의 SOD와 catalase활성도 역시 민들레 투여군이 수은 투여군에 비하여 감소하였으나 통계학적 유의성은 없었다. 간 조직의 광학현미경적 관찰에서 심한 조직괴사가 관찰되지만 민들레 투여군에서는 문맥주위의 약간의 괴사와 심한 호중구 침윤현상이 관찰되었다. 전자현미경적 관찰 결과, 간장에서 수은 투여군은 간세포의 핵이 함입되어 불규칙했으며 미토콘드리아와 조면소포체의 수조가 팽대되고, 리보솜의 탈락이 관찰되었다. 민들레 투여군은 핵이 정상적인 상태로 관찰되었고, 전자밀도가 높은 미토콘드리아가 분포되어 있었으며, 리보솜이 부착된 상태로 층판구조를 형성하는 조면소포체가 관찰되었다. 이상의 연구 결과로 보아 민들레가 수은으로 유발된 간손상을 보호하는 효과가 있는 것으로 사료되며 민들레에 대한 다양한 연구가 필요할 것으로 사료된다.

  • 우리나라의 주요 해산양식어종인 돌돔, Oplegnathus fasciatus을 대상으로 30, 60, 120 및 240 mg/kg의 아연을 40일 동안 경구 투여에 따른 아가미와 간의 항산화효소 활성의 변화를 검토하였다. 돌돔 간의 superoxide dismutase(SOD) 활성은 30~240 mg/kg 아연농도, glutathione peroxidase (GPx) 활성은 30~120 mg/kg의 아연농도에서 유의적으로 증가하였다. 돌돔 아가미의 SOD 및 Glutathione(GSH) 활성은 120 및 240 mg/kg 아연농도, GPx활성은 60~240 mg/kg의 아연농도에서 유의한 증가가 관찰되었다.

Effect of Glycyrrhizae Radix on the Expression of UDP-Glucuronosyltransferase-1A1 (UGT1A1) in Rat Liver

  • Licorice has been widely used in combination with other herbs or synthetic drugs for various disorders. In an effort to study the effect of licorice roots (Glycyrrhizae Radix, GR) and glycyrrhizin on the hepatic glucuronidation, we have previously found that the pretreatment of GR or glycyrrhizin for 6 days resulted in a marked increase in the enzymatic activity of 3-methylcholanthrene (3-MC)-inducible hepatic UDP-glucuronosyltransferase (UGT) isozyme that has high affinity toward phenolic substrates (p-nitrophenol form, UGTIA) in Sprague-Dawley rats. As an approach to elucidate the mechanism for the enzyme activation by licorice in rat liver, we examined the levels of hepatocellular mRNAs for UGTIA upon the treatment of GR or glycyrrhizin. The hepatic mRNAs were extracted from Sprague-Dawley rats and Wistar rats after the treatment of the methanol extract of GR (1 g/kg, p.o.), glycyrrhizin (23 mg/kg, p.o.) for 6 days, or 3-MC (40 mg/kg, i.p.) for 3 days. Using the UGT1A1 CDNA as a probe, we found that the mRNAs for the enzyme were induced by 3-MC treatment while those were influenced neither by GR nor by glycyrrhizin in both strains of rats. These results indicate that the activation of rat liver UGTI A by licorice and glycyrrhizin was not due to the induction of mRNAs for the enzyme.
