Histopathological Studies on Experimental Nitrate Poisoning in Rabbits (질산염중독(窒酸鹽中毒)에 관한 병리조직학적연구(病理組織學的硏究))

  • In order to clarify the histopathological changes resulting from nitrate poisoning, rabbits were experimentally poisoned by the oral administration of $KNO_3$ or $NaNO_2$ and examined clinically and histopathologically. In addition, the quantitative changes of glycogen level in hepatic cells were histochemically observed. The results obtained were summarized as follows: 1. Clinical symptoms observed from the acute cases which died within 2 hours after the administration were severe cyanosis of visible mucosa, frequent urination, and dyspnea. However, in chronic cases administrated daily with $KNO_3$ for 43, 50 and 74 days respectively, no marked symptoms were observed. 2. Macroscopic changes observed in acute cases were severe methemoglobinemia, cloudy swelling of hepatic cells, hemorrhage and hyperemia of gastric mucosa, and hyperemia of other organs. In chronic cases there were marked hyperemia, dark-red coloring and increasing of consistency in liver and kidney, and swelling of spleen. 3. Microscopic changes observed in acute cases were hemorrhage and hyperemia of various organs, cloudy swelling and centrilobular necrosis of hepatic cells and necrosis of convoluted tubular epithelium in kidney. In chronic cases there were round cell infiltration of the interlobular connective tissue and epithelial proliferation of interlobular bile ducts in the liver, and necrosis of the convoluted tubular epithelium and proliferation of interstitial connective tissue in kidney, thickening of alveolar septa of lungs, activated hemopoiesis of bone marrow, and myeloid metaplasia of sqlenic pulp. 4. Glycogen storage in liver cells was decreased in acute cases, on the contrary, increased in chronic cases.

Effects of Chrysanthemum boreale M. Water Extract on Serum Liver Enzyme Activities and Kupffer Cells of Carbon Tetrachloride-Induced Rats

  • Effects of water extract obtained from Chrysanthemum boreale M. (CE) on serum enzyme activities and Kupffer cells of carbon tetrachloride ($CCl_4$)-induced rats were investigated. Thirty-two healthy male Sprague-Dawley rats were divided into normal (N), $CCl_4$-induced (T), CE-supplemented (C), and $CCl_4$-induced and CE-supplemented (TC) groups. $CCl_4$ injection significantly increased aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, and alkaline phosphatase activities in serum. Significant increases in total cholesterol and triglyceride concentrations were also observed in $CCl_4$-induced rats. Oral administration of CE at 300 mg/kg body weight significantly decreased serum enzyme levels and suppressed $CCl_4$ hepatotoxicity-induced lipid profile changes. Histological findings showed fatty change, fibrosis and increased number of Kupffer cells in T group. Electron microscopic examination showed increased lysosome content and dilation of rough endoplasmic reticulum within Kupffer cells in T group, whereas CE supplement attenuated liver injury in $CCl_4$-induced liver. These results indicated CE could significantly alleviate CC4-induced hepatotoxicity injury.

Pathological Changes in Rats Fed petasites japonicus Maxim II. Immunohistochemical Localization of Cytochrome P4502E1 and GST-P in Liver

  • We investigated metabolism and carcinogenesis in livers of Sprague-Dawley rats fed juices and pelleted diets containing Korean native plants petasites japonicus Maxim by evaluating cell localization and expression of cytochrome P450s and GST-P. Anti-cytochrome P450s application in liver sections revealed three to four times increased expression of cytochrome P450E1 immunoreactivity in degenerative hepatocytes when compared to histologically normal hepatocytes. Anti-GST-P in showed positive pren plastic foci as well as in individual hepatocytes randomly scattered throughout all liver sections examined. Additionally GST-P was evident in proliferative endothelial cells and biliary epithelial cells in exposed rat livers. These results suggested that the increased level of cytochrome P4502E1 in affected hepatocytes was a direct consequence of Petasites japonicus toxicity. Further immunoreactivity to anti-GST-P in hepatocytes endothelial cells and biliary epithelial cells indicated a possible preneoplastic effects of Petasites japonicus in Sprague-Dawley rat.

Avian malaria associated with Plasmodium spp. infection in a penguin in Jeju Island

  • Plasmodium spp. in domestic and wild birds are microscopic, intracellular parasitic protozoa within the blood cells and tissues cause avian malaria. A 17-month-old Magellan penguin (Spheniscus magellanicus) with a clinical signs of anorexia, depression, and respiratory distress for 3 days was submitted to the Pathology Department of Veterinary Medicine, Cheju National University in October 2005. It was born and reared in the Jeju Island. Grossly, the liver was enlarged, pale and friable. The spleen was also enlarged with dark red coloration and friable. Histopathologically, the lesions in the liver were characterized by multifocal infiltration of macrophages and lymphocytes especially in perivascular regions. The schizonts of Plasmodium spp. contained up to 30 merozoites were found in numerous infiltrated mononuclear cells. Similarly, histiocytic cells were proliferated in red pulp of spleen and the schizonts were found in these cells. Numerous dark brown pigments were widely distributed in the liver and spleen. The result of the nested polymerase chain reaction clarified the causative agent of this case was Plasmodium spp.. This is the first report for the outbreak of avian malaria caused by Plasmodium spp. in a penguin that was born and reared in Jeju Island in Korea.

Evaluation of apoptosis after ionizing radiation in feeding and starving rats

  • It has been known that $\gamma$-irradiation usually induces cell death in regenerating stem cell in normal tissues like skin, intestine and hematopoietic organ. The experiment were carried out to evaluate the early response of radiation injury in radiosensitive and intermediate radiosensitive tissues in feeding and starving rats with the doses of 3.5 and 7.0 Gy. The results of the study showed that the histological phenomenon was apoptosis in the doses of the radiation as the early response of tissue injury. Apoptosis were showed organ-specific and cellular specific responses suggesting that the selection of apoptosis be exactly focused on highly renewal organs and cells. It was interesting that the rats starved for 72 hours prior to irradiation induced less apoptosis in liver than fed rats. As for cellular responses it appeared that apoptotic cells were mostly distributed in ductal or periportal cells in liver of feeding rats unlikely in liver of Starving rots which showed no difference in zonal distribution. In salivary gland apoptotic cells in fed rats were highly induced in intercalating and ductal cell population than in acinar cell population although unlikely in starved rats. This study showed the value of apoptosis using the detection system of TUNEL for evaluating cellular damage after radiation injury and the diminished effect of starvation on cell damage after ionizing irradiation.

Ex vivo Boosted Immune Cell Therapy for Canine Hepatic Disease

  • A 12-year-old male American Cocker Spaniel was diagnosed with a type of chronic hepatits (CH) called cholangioheaptits. Routine supportive medication was administered to the patient, and ex vivo boosted immune cell (EBI-C) therapy was used for the treatment. A histopathologic examination of the liver 19 months later revealed that the cholangiohepatitis had progressed to cholangiocarcinoma. The medication and immune cell therapy was maintained. Two months after the new diagnosis, the patient's state worsened, and the dog died 635 days after the first visit. EBI-C therapy is a type of immunotherapy, where immune cells are isolated from the patient's peripheral blood mononuclear cells, expanded ex vivo, and then infused into the patient intravenously every two weeks. EBI-Cs (mean: 2.78 × 108 cells) were obtained 38 times and infused every two weeks. Most EBI-C were T-lymphocytes (99.24% of total EBI cells). T-lymphocytes produce large interferon (IFN)-γ, and IFN-γ inhibits liver fibrosis in dogs with CH. Moreover, in bile duct cancer, an increase in T-lymphocytes correlates with decreasing tumor invasion and metastasis. Thus, we propose that EBI-C therapy is applicable as a new supportive therapy for canine liver disease if other treatments like drug medication, surgery, or radiation are unavailable.

Inhibitory Effect of Saengangeonbitang-gasamchilgn on Liver Fibrosis in Rat (생간건비탕가삼칠근(生肝健脾湯加三七根)이 흰쥐의 간섬유화 억제에 미치는 영향)

  • The aim of this study was to investigate the inhibitory effect of Saengangeonbitang-gasamchilgn(SGGBTGSCG) on collagen production in rat hepatic stellate cells(HSC) and on the TAA-induced chronic liver injury model in rats. Methods : 1) HSCs were treated with SGGBTGSCG extract powder(50% EtOH SGGBTGSCG, dw SGGBTGSCG). After the treatment, MTT assay, BrdU assay and procollagen assay were done. In addition, gene expressions of collagen type $1{\alpha}2$, ASMA, TIMP1, and TIMP2, all of which are known to be associated with liver fibrosis, were analyzed by RT-PCR. 2) Liver fibrosis was developed in rats by injection of TAA 3 times a week for 5 weeks. After the SGGBTGSCG-treatment, body weight, liver & spleen weight, liver function test, the complete blood cell count and the change of portal pressure were studied. Results : In MTT assay, SGGBTGSCG significantly decreased the viability of HSCs in a dose-dependent manner. In BrdU assay, SGGBTGSCG significantly inhibited the HSC proliferation in a dose-dependant manner. In procollagen assay, SGGBTGSCG decreased procollagen production by HSC. In the change of rats' liver and spleen weight, TAA+SGGBTGSCG groups showed little difference compared with TAA-only group. In the liver function test, SGGBTGSCG decreased the serum level of ALT, AST, and Alp elevated by TAA. In the complete blood cell count, SGGBTGSCG significantly decreased WBC elevated by TAA and increased RBC and Hct lowered by TAA. In the change of portal pressure, SGGBTGSCG decreased portal pressure elevated by TAA. Conclusions : These results suggest that SGGBTGSCG is beneficial in the treatment of cirrhotic patients as well as for patients with chronic hepatitis.

Effect of Unilatromral Pneumonectomy on the Compensatory Growth of the Residual Lung, Liver, and Kidney, ana Serum Electrophoresis Pattern (일측폐장절제가 잔류폐, 간 및 신장의 대상성 증식과 혈청전기영동상에 미치는 영향)

  • In order to clarify the effect of the unilateral pneumonectomy on the compensatory growth of the residual lung, liver and kidney, and serum electrophoresis pattern, right lung pneumonectomy was performed on rabbits under general anesthesia with pentobarbital sodium. On the fifth day after the surgery, the weight of the residual lung, liver and bilateral kidneys was measured and organ weight-body weight ratio was calculated. And in an attempt to know whether the cells in the liver and the kidney were proliferated by unilateral pneumonectomy, DNA content was determined. The quantity(g/100 ml) of serum protein was determined also and serum electrophoresis was performed on cellulose acetate membrane. The results obtained are summarized as following. The weight of the residual lung and lung weight-body weight ratio was significantly increased respectively. The weight of the liver and organ weight-body weight ratio were not changed but the DNA content of the liver and kidney tissue increased significantly, illustrating that unilateral pneumonectomy caused cellular hyperplasia in the liver and in the kidney as well as in the residual lung. The quantity(g/100 ml) of serum protein was significantly increased and in the analysis of the electrophoregram, there was significant difference between the normal and pneumonectomy group. Taken together, these results indicate that unilateral pneumonectomy caused the compensatory hyperplasia of the liver, the kidney and the residual lung as well as the change of electro-phoretic pattern. And it also suggests that a humoral factor, which proliferates the cells in the residual lung, the liver and the kidney, existed in the pneumonectomized rabbits.

  • The determination of fungal flora in some kinds of cereals have been carried out in other to obtain an appropriate information of the population of fungi and toxin productivity The results were summarized as follow; 1. The predominant genera were Aspergillus, Penicillium, Mucor, Rhizopus, Alternaria, Fusarium. 2. Six of Aspergillus flavus were aflatoxin-producing strains. 3. Sample barleys were found to contain the highest content of aflatoxin. 4. In electron microscopic studies of liver cells from mouse which had been injected with crude toxin, the liver cells showed the cytoplasmic change.

  • Hepatic coccidiosis was occurred in a rabbit farm in Chonbuk province. Clinically, rabbits showed anorexia, diarrhea, dehydration, and depression, subsequently died 3 - 5 days after onset of clinical signs. Grossly, multifocal white spots or lines on the liver suface were observed. Histopathologic lesions included hyperplasia of bile duct epithelium with infiltration of inflammatory cells such as plasma cells and granulocytes, which represents chronic pericholangitis. Different developmental stages of Eimeria stiedae were observed inside the epithelium of biliary system. This is the case of hepatic coccidiosis in rabbits.
