Gas-liquid chromatography에 의한 한국(韓國) 주요식품(主要食品)의 아미노산(酸) 함량측정(含量測定) (Gas-Liquid Chromatographic Determination of Amino Acids in Some Korean Foods)

  • 우리나라 주요 식품들의 단백질 아미노산 함량을 최근에 와서 크게 실용화되고 있는 GLC 방법에 의해 분석함에 있어서 GLC의 기술적 재평가를 하고자 시험 하였든바 1. 우선 여러식품들의 단백질 질소함량을 조사하여 GLC 분석에 필요한 시료량을 정하였으며 2. GLC에 주입한 17종 아미노산들이 대하여 조사한 결과 Arginine, Cystine, Histidine, Tyrosine을 제외한 13종만이 주입한 량에 비례하여 detector response 를 얻었다. 3. 13종 아미노산에 대한 RMR 치(値)를 얻었다. 4. 아미노산에 대한 recovery 검정(檢定)을 하였든바 거의 100% 가까이 recover되었으며, 가수분해가 recovery에 미치는 영향을 조사하였든바 별 영향이 없었음을 알았다. 5. GLC와 ion-exchange chromatography 분석법을 비교시험하였든바 백어포 시료에 있어서는 Threonine과 Asparagine만이 두 방법에서 거의 같게 나왔고 다른 아미노산들은 GLC에서 보다 많은량이 나타났다. 6. 각식품들에 대한 단백질 아미노산함량을 측정하였든바 식품종류에 따라 그리고 보리 및 대두에 있어서는 품종에 따라서도 아미노산 함량이 다르게 나타났다.

상수도 정수장 오이중 Alum회수 및 재활용에 관한 연구 (A Study on Alum recovery and reuse from the sludge in water treatment plant.)

  • Alum recovery has recently gained more attention because many water utilities need to improve their sludge handling and disposal practices. As part of an overall sludge management program recovery can reduce the amount of solids and allow for reuse of the recovered Alum as a coagulant. This study was examined the effectiveness of Alum recovery from the Sludge at the D water treatment plant in Kwangju city. The results were summarized as follows 1. Alum recovery was obtained sufficiently acidification(An optimum condition was pH2-3) With $H_{2}SO_{4}$ to settled sludge. In this case recovered liquid Alum from sludge of 2.1% solids concentration at pH 2.1 was contains Aluminum $1,602mg/{\ell}$(as $Al_{2}O_{3}$ 0.3% ) and other metal of low level. 2. It was an optimum condition to all reuse of recovered Alum as a coagulant that rate of Commercial Alum:Recovered Alum=$14{\mu}{\ell}/{\ell}{\;}:{\;}200{\mu}{\ell}/{\ell}$ In a result of Jar Tests. 3. It was a result of Alum recovery from sludge, the reduction effect of amount of solids was about 57.4%. 4. If all recovered Alum were reused the reduction effect of solid wastes disposal cost and chemical drug's cost was about 22%.

Al 스크랩으로부터 금속회수에 관한 연구 (A Study of the Metal Recovery from the Aluminium Scrap)

  • 본 연구에서는 Al 스크랩으로부터 재생지금 제조시 시료의 예비처리, 용제첨가 및 용해분위기가 Al 회수율에 미치는 영향을 조사하였다. 실험결과에 따르면 Al 드로스는 용탕표면에서의 산화반응에 의해 발생하였다. 예비처리의 영향에 다르면 탈지하지 않고 압착한 칩 bale 시료의 경우에는 압착하지 않은 칩 시료에 비해 약 14%의 회수율이 증가하였으며, Al seed 용해공법을 채택하는 경우에는 탈지하지 않고 단지 세편과 압착만을 행하여도 97%의 높은 회수율을 얻을 수 있었다. Al 스크랩 용해시 7wt%까지 첨가된 염에 의해 회수율은 최대 95%까지 증대되었으며, 탄소 및 질소분위기에서도 역시 회수율은 증가하였으나, 염과 탄소의 혼합분 첨가시 과잉 첨가된 탄소는 오히려 회수율을 감소시켰다.

Chemical Inhibition of Cell Recovery after Irradiation with Sparsely and Densely Ionizing Radiation

  • The dependence of cell survival on exposure dose and the duration of the liquid-holding recovery (LHR) was obtained for diploid yeast cells irradiated with ionizing radiation of different linear energy transfer (LET) and recovering from radiation damage without and with various concentrations of cisplatin - the most widely used anticancer drug. The ability of yeast cells to recover from radiation damage was less effective after cell exposure to high-LET radiation, when cells were irradiated without drug. The increase in cisplatin concentration resulted in the disappearance of this difference whereas the fraction of irreversible damage was permanently enlarged independently of radiation quality. The probability of cell recovery was shown to be constant for various conditions of irradiation and recovery. A new mechanism of cisplatin action was suggested according with which the inhibition of cell recovery after exposure to ionizing radiations was completely explained by the production of irreversible damage.

  • 이산화탄소 분리 및 메탄 회수를 동시에 수행하기 위한 배출구가 두 개로 분리된 내부순환식 기포탑 반응기를 이용하여 $CO_2$ 분리용 흡수제인 모노에탄올아민(MEA)의 최적조건을 도출하였다. 5 wt% MEA 수용액에서 이산화탄소의 흡수 및 탈기 반응 시 pH 변화, 공기주입량에 따른 이산화탄소 탈기량 및 메탄 함량변화를 관찰하였다. 또한 액상수위 및 초기 MEA 흡수액 온도변화에 따른 이산화탄소 흡수 및 메탄 회수율을 측정하였다. 최적반응조건은 정상상태에서 액상수위 40 mm, 공기주입량은 1.5 L/min, 흡수액의 온도 $25^{\circ}C$이었다

탄산리튬 분말 제조에 있어서 초음파 에너지를 적용한 기액반응에 관한 연구 (A study on the fabrication of lithium carbonation powder by gas-liquid reaction using ultrasonic energy)

  • 기존 연구에서 본 연구자들은 여러 가지 리튬 함유 용액으로부터 탄산리튬 분말 제조에 대하여 보고하였으며, 이중에서 수산화리튬 용액과 CO2 가스와의 반응이 열역학적으로 탄산리튬 생성이 가능하고 89.4 %의 회수율을 나타내었다. 본 연구에서는 수산화리튬 용액과 CO2 가스와 기액반응에 있어서 초음파에너지를 가하여 탄산리튬 분말을 제조하는 실험을 실시하였다. 실온에서 탄산리튬 생성 반응에 초음파를 가하였을 경우에 리튬의 회수율은 83.8 %이었으며, 60℃에서 초음파를 가하면 99.9 %로 회수율이 급격하게 증가되었다. 또한 초음파 에너지를 가하지 않을 경우의 입자 크기는 D50에서 약 87.7 ㎛이었으며, 초음파를 가한 경우에는 D50이 약 8.4 ㎛로 급격하게 감소하는 결과를 얻었다.

  • This experiment was carried out to determine the progesterone concentration of bovine plasma by liquid phase double antibody enzyme immunoassay. The optimum conditions of assay-system, enzyme conjugate and gelatin were invested. The sensitivity, recovery rate and reproducibility by this assay were also analyzed. The results obtained were as follows: 1. The suitable supplementation level of gelatin was 0.2%. As the gelatin level increased to 1%, coefficient variation of interassay was shown to be irregular. 2. The optimum dilution rate of enzyme conjugate was 30 times. Enzyme activity was greatly fluctuated depending on the minor condition of enzyme conjugate technique. Therefore, it was considered to be checked when determined. 3. The sensitivity of the assay was 12 pg/tube. 4. Recovery rate was decreased when the amount of sample was too little or too much, but the recovery rate was high as 97.8% when the amount of sample between 50 and $200{\mu}l$. 5. Mean intra-assay and inter-assay coefficient of variation was 4.5% and 5.9%, respectively. By using liquid phase double antibody enzyme immunoassay, progesterone in plasma can be detected, and also this assay system is applicable to study on physiological function of progesterone and to diagnosis of reproductive disorders.

Gas-Liquid Chromatography를 이용한 사과 및 배 중의 농약 다성분 잔류분석법 (Analysis of Multiple Pesticide Residues in Apples and Pears Using Gas-Liquid Chromatography)

  • 사과 및 배 시표에서 농약 다성분의 gas chromatography를 이용한 잔류분석법 확립을 위하여 199가지의 농약을 선정하여 retention time 및 검출기에 따라 ECD 5 그룹 및 NPD 5 그룹의 10개 그룹으로 분류하였다. 시료의 종류에 따른 분석조건을 확립하기 위한 회수율 시험은 농약들의 log $P_{ow}$ 값과 화학적 분류에 따라 총 18개 (ECD 11개, NPD 7개)의 농약을 선정하였다. 예비 실험 후 확립된 분석방법에 따라 10개 그룹의 혼합 표준용액으로 사과 및 배에 대한 회수율시험을 행하였다. 그 결과, 총 196가지의 농약의 70%에 해당하는 사과에서 136개, 배에서 133개의 농약들에서 회수율 70에서 120%의 양호한 결과를 나타내었다. 그러나 사과에서 43개, 배에서 45개의 농약들이 70% 미만의 회수율을 보였고, fenvalerate는 120% 이상의 회수율을 나타내었으며, 사과에서 17개 및 배에서 18개의 농약들은 검출이 되지 않았다. 그러나 확립된 분석법은 SOP에 의한 신속하고 수월한 수행으로 농산물 중의 잔류농약을 검출 및 모니터링하는 목적에 적합하다고 사료된다.

Analysis of fatty acid methyl ester in bio-liquid by hollow fiber-liquid phase microextraction

  • Bio-liquid is a liquid by-product of the hydrothermal carbonization (HTC) reaction, converting wet biomass into solid hydrochar, bio-liquid, and bio-gas. Since bio-liquid contains various compounds, it requires efficient sampling method to extract the target compounds from bio-liquid. In this research, fatty acid methyl ester (FAME) in bio-liquid was extracted based on hollow fiber supported liquid phase microextraction (HF-LPME) and determined by Gas Chromatography-Flame Ionization Detector (GC-FID) and Gas Chromatography/Mass Spectrometry (GC/MS). The well-known major components of biodiesel, including methyl myristate, palmitate, methyl palmitoleate, methyl stearate, methyl oleate, and methyl linoleate had been selected as standard materials for FAME analysis using HF-LPME. Physicochemical properties of bio-liquid was measured that the acidity was 3.30 (${\pm}0.01$) and the moisture content was 100.84 (${\pm}3.02$)%. The optimization of HF-LPME method had been investigated by varying the experimental parameters such as extraction solvent, extraction time, stirring speed, and the length of HF at the fixed concentration of NaCl salt. As a result, optimal conditions of HF-LPME for FAMEs were; n-octanol for extraction solvent, 30 min for extraction time, 1200 rpm for stirring speed, 20 mm for the HF length, and 0.5 w/v% for the concentration of NaCl. Validation of HF-LPME was performed with limit of detection (LOD), limit of quantitation (LOQ), dynamic range, reproducibility, and recovery. The results obtained from this study indicated that HF-LPME was suitable for the preconcentration method and the quantitative analysis to characterize FAMEs in bio-liquid generated from food waste via HTC reaction.

Crucible Cover of Multilayer Porous Hemisphere for Cd Distillation

  • The electrorefining process is generally composed of two recovery steps in pyroprocessing - the deposit of uranium onto a solid cathode and the recovery of the remaining uranium and TRU elements simultaneously by a liquid cadmium cathode. The liquid cathode processing is necessary to separate cadmium from the actinide elements since the actinide deposits are dissolved or precipitated in a liquid cathode. Distillation process was employed for the cathode processing. It is very important to avoid a splattering of cadmium during evaporation due to the high vapor pressure. In this study, a multi-layer porous round cover was proposed and examined to develop a splatter shield for the Cd distillation crucible. Cadmium vapor can be released through the holes of the shield, whereas liquid drops can be collected in the multiple hemisphere. The collected drops flow on the round surface of the cover and flow down into the crucible. The crucible cover was fabricated and tested in the Cd distiller. The cover was made with three stainless steel round plates with a diameter of 33.50 mm. The distance between the hemispheres and the diameter of the holes are 10 and 1 mm, respectively. About 40 grams of Cd and about 4 grams of Bi was distilled at a reduced pressure for two hours at $470^{\circ}C$. After the Cd distillation experiment, cadmium was not detected and more than 90 % of Bi remained in the ICP-OES analysis. Therefore the crucible cover can be a candidate for the splatter shield of the Cd distillation crucible. Further development of the crucible cover is necessary for the decision of the optimum cover geometry and the operating conditions of the Cd distiller.
