The High-throughput Solid-Phase Extraction in the Field of Synthetic Biology: Applications for the Food Industry and Food Managements

  • The field of synthetic biology has emerged in response to the ongoing progress in the life sciences. Advances have been made in medicine, farming, eating, making materials, and more. Synthetic biology is the exploration of using living organisms to create new organisms. By manipulating specific genes to express targeted proteins, proteins can be created that are both productive and cost-effective. Solid-phase extraction (SPE) and liquid-liquid extraction (LLE) are employed for protein separation during the production process involving microorganisms. This study centers on Scanning Probe Microscopy (SPM) to showcase its utility in the food industry and food management. SPE is predominantly utilized as a pretreatment method to eliminate impurities from samples. In comparison to LLE, this method presents benefits such as decreased time and labor requirements, streamlined solvent extraction, automation capabilities, and compatibility with various other analytical instruments. Anion exchange chromatography (AEC) utilizes a similar methodology. Pharmaceutical companies utilize these technologies to improve the purity of biopharmaceuticals, thereby guaranteeing their quality. Used in the food and beverage industry to test chemical properties of raw materials and finished products. This exemplifies the potential of these technologies to enhance industrial development and broaden the scope of applications in synthetic biology.

LC-MS/MS를 이용한 혈청 중 finasteride 분석 (Determination of finasteride in human serum by LC-MS/MS)

  • 본 연구는 LC-ESI-MS/MS를 이용하여 혈청 중의 피나스테라이드의 분석법을 확립하고, 밸리데이션을 통하여 분석방법의 타당도를 검증하였다. 혈청에 내부표준물질로 베클로메타손을 첨가한 후 methyl tert-butyl ether (MTBE)을 사용하여 액체상추출법(liquid-liquid extraction, LLE)으로 전처리하였다. LC-MS/MS의 양이온 모드에서 피나스테라이드와 베클로메타손의 MRM (multiple reaction monitoring) mass transition은 각각 m/z 373.2${\rightarrow}$305.2, m/z 409.3${\rightarrow}$391.2 이었으며, 머무름 시간은 각각 5.81분, 5.46분이었다. 피나스테라이드의 정량한계는 0.1 ng/mL로 나타났으며, 0.1~20.0 ng/mL의 농도 범위에서 검량선은 우수한 직선성($R^2$=0.9997)을 확인할 수 있었으며, 회수율은 80~83% 범위이었다. 피나스테라이드의 일내에 대한 정밀도는 6.3~10.6%, 정확도는 97.3~103.6% 범위이었으며, 연속 3일간 수행한 일간 정밀도는 0.8~5.2%, 정확도는 99.8~102.5%로 나타났다.

  • Several studies investigating the prevention and treatment of external parasites in farmed olive flounder Paralichthys olivaceus have found that the anticoccidial agent toltrazuril sulfone is an effective antiparasitic. Prior to undertaking a full-scale study, we developed analytical methods to detect the levels of toltrazuril and toltrazuril sulfone in farmed flounder samples using liquid chromatography-electrospray ionization tandem mass spectrometry (LC-MS/MS). This analysis showed that LC-MS/MS changed the mobile phase and collision energy of toltrazuril and toltrazuril sulfone. This was validated using established conditions. Sample pre-treatment for this process involved extraction with dichloromethane and purification by liquid-liquid extraction in formic acid, acetonitrile, and h-hexane, followed by determination of all compounds by LC-MS/MS. Separation was achieved within 10 min by gradient elution using a Capcell Pak C18 ($3.0{\mu}m$, $100{\times}2.0mm$) analytical column (Shiseido UG 120V) with a mixture of 0.1% (v/v) formic acid and acetonitrile. Multiple reaction monitoring was used for selective detection of toltrazuril and toltrazuril sulfone. This method yields satisfactory results for linearity, precision, and limits of quantification. Therefore, the method established in our study will serve as a basis for further research on parasite control by toltrazuril and toltrazuril sulfone.

Exreaction of Thiosulfinates from Garlic Using Gas-in-Liquid-Dispersion

  • Extraction yields of thiosulfinates from garlic were studied by using gas-in-liquis-dispersion(GLD) system to maxmize the thiosulfinate extraction. Extracted thiosulfinates were spectrophotometically quantified using synthesized dially1 thiosulfinate. The conditions for maximum extraction were optimized by response surface methodology. The volatile components extracted by various methods, such as hexane extraction, simultaneous steam distillation and GLD system, were compared by using gas chromatography. The results indicated that the thiosulfinate yield was increased by incresaing temperature and nitrogen gas flow rate, while the effects of bubble sizes on thiosulfinate extraction were not significant at the ranges tested. Application of GLD system resulted in extraction of more volatile components than other extraction methods, Therfore, it was suggested that GLD system was one of the efficient extraction methods among the ever introduced ones, for thiosulfinate extraciton.

Measurement of Spray Deposit Amount Using Spectrophotometer and Food Dye as Tracer

  • Measurement of spray deposit is necessary for evaluation of a chemical application technology. However it is not easy and time consuming. A simple method for measuring the deposition amount of spray using a tracer and a spectrophotometer was developed. Various materials were tested to determine an adequate tracer. Food dye was selected as a tracer, because it was cheep and easily treatable. Using NIRS(Near Infrared Reflection Spectrophotometer), a regression curves between maximum absorbance of a solution and concentration of the tracer were obtained. Yellow food dye solution showed a peak of spectrum at 452 nm, and absorbance of peak showed a tendency to increase as concentration increased. Green or pink food dye were tested and judged to be good tracers. However, tracer concentration should not exceed certain limits in order to measure maximum absorption. Using spraying liquid with known tracer concentration and known amount of washing liquid, spray deposit amount on real targets on leaves could be estimated at less than 13% error level.

Rapid Analysis of Melamine Content in Powdered and Liquid Milk Using Fourier Transform Infrared Spectroscopy

  • Melamine is a chemical intermediate to manufacture amino resins and plastics, which cannot be used as food additive since it can cause kidney stones. A qualitative determination method of melamine residue in powdered and liquid milk was developed using Fourier transform infrared (FTIR) spectroscopic technique. The calibration sets consisted of 21 standard melamine solutions, in which 1% trichloroacetic acid aqueous solution and acetonitrile (3:1, v/v) were used as solvent. The model was validated using 10 standard melamine solutions which were unused to build up the calibration set. Infrared (IR) absorbance peaks specific to almost all chemical groups in melamine molecule were shown in the spectral range between 1,100 and 1,800/cm. Combined partial least squares (PLS)-$2^{nd}$ derivative calibration model coupled with mean centering (MC) mathematical enhancement showed the highest correlation coefficients ($R^2$>0.99). In brief, the FTIR technique can be used for quantitative analysis of melamine in milk samples.

  • Liquid smoke is made by concentration of smoke generated from charcoal of broad-leaved trees, or by refinement of smoke originated from incomplete combustion of wood condensate (Park et al., 1994). Generally, it is well known that phenol and its derivatives have antioxidative effect, while acids and formaldehyde have antimicrobial effect (Park et al., 1994). Meanwhile, some studies (Alonge, 1988; Dungel, 1961) reported that the high incidence of stomach cancer has been associated with the consumption of smoked fishes, and these were investigated to direct intake of fishes treated with smoke, containing polycyclic aromatic hydrocarbons(PAHs)(Moret et al., 1999). Isolation of a Calcium-binding Peptide from Chlorella Protein Hydrolysates

  • To isolate a calcium-binding peptide from chlorella protein hydrolysates, chlorella protein was extracted and hydrolyzed using Flavourzyme, a commercial protease. The degree of hydrolysis and calcium-binding capacity were determined using trinitrobenzenesulfonic acid and orthophenanthroline methods, respectively. The enzymatic hydrolysis of chlorella protein for 6 hr was sufficient for the preparation of chlorella protein hydrolysates. The hydrolysates of chlorella protein were then ultra-filtered under 5 kDa as molecular weight. The membrane-filtered solution was fractionated using ion exchange, reverse phase, normal phase chromatography, and fast protein liquid chromatography to identify a calcium-binding peptide. The purified calcium-binding peptide had a calcium binding activity of 0.166 mM and was determined to be 700.48 Da as molecular weight, and partially identified as a peptide containing Asn-Ser-Gly-Cys based on liquid chromatography/electrospray ionization tandem mass spectrum.

Isolation of Ginsenoside Rh1 and Compound K from Fermented Ginseng and Efficacy Assessment on Systemic Anaphylactic Shock

  • Ginsenosides are responsible for the pharmacological and biological activities of ginseng. In this study, ginsenoside Rh1 and compound K were isolated and purified from fermented ginseng substrate and their anti-allergic effects were assessed in compound 48/80-induced anaphylactic shock model. The fermented ginseng substrate was extracted by methanol and ginsenoside Rh1 and compound K were efficiently purified by preparative high performance liquid chromatography (prep HPLC). Their quality and quantity were analyzed by liquid chromatography-mass spectrometer (LC-MS) and HPLC. Ginsenoside Rh1 showed better anti-allergic effects than compound K in compound 48/80-induced anaphylactic shock model. This study suggested that fermented ginseng extracts with enriched Rh1 may be utilized as a potential biomaterial of functional food for the alleviation of allergic symptoms.

Levels of Perfluorinated Compounds in Liquid Milk Products in Korea

  • 본 연구는 국내 유통되는 액상 유제품에 함유된 17종의 과불화화합물 (PFCs) 함량에 대한 모니터링을 위해 수행되었다. 샘플을 액체-액체 추출을 통해 추출하고 LC-MS/MS로 정량 분석하였다. 시중에 유통중인 98개의 액상 유제품 샘플에서 PFCs의 모니터링을 진행하였으며 그 결과, PFTeDA를 제외한 16종의 PFCs가 검출되었다. 총 PFCs의 함량은 0.9037 ng/mL로 액상 우유에서 가장 높았으며, PFPeA, PFDS, PFHxA도 액상우유에서 0.1 ng/mL 이상의 농도 수준을 보였다. 미국과 이탈리아에서 수행한 연구와 비교했을 때 PFOA와 PFOS의 잔류 패턴은 유사한 것으로 나타났으며, 농도 수준은 0.1 ng/mL 미만이었다. 액상 유제품은 다양한 가공단계(살균, 혼합, 지방제거 등)를 거쳐 총 PFCs의 함량이 감소하는 것으로 보여 지며, 특히 짧은 탄소 사슬을 가진 PFCs가 크게 감소하거나 검출되지 않았다. 이는 식품 안전에서 원료의 PFOS와 PFOA의 잔류정도가 중요하다는 것을 의미한다.
