  • The microorganisms producing a lipoxygenase inhibitor were screened from a wide variety of sources. The isolated strain was assigned to genus Penicillium by its cultural and morphological characteristics. The proper medium for the production of lipoxygenase inhibitor was composed of glucose 3.0%, ammonium sulfate 0.4%, and potassium phosphate (dibasic) 0.1%. The cultivation for lipoxygenase inhibitor production was carried out in 500m1 Erlenmyer flasks containing 100m1 of the medium at $30^{\circ}C$ by cultivating reciprocally. The highest lipoxygenase inhibitor production was observed after 8 days of cultivation. The inhibitor was the low molecular weight substance and inhibited specifically soybean origin lipoxygenase.

  • A strain of Penicillium sp. extracellularly produced an inhibitory substance for lipoxygenase. These purification procedures were followed : ethanol treatment, chromatographies on Dowex 50W, Sephadex G-25, silica gel column and HPLC. The inhibitor was stable in pH range from 3.0 to 5.0 at $25^{\circ}C$, and a treatment at 10$0^{\circ}C$ for 2 hours didn't diminish its original activity. The purified inhibitor was charred at temperature near 22$0^{\circ}C$~23$0^{\circ}C$ and decomposed. Molecular weight of the inhibitor was estimated to be approximately 270 by Sephadex G-25 column chromatography. The inhibitor rapidly formed EI complex with lipoxygenase and inhibited enzyme activity.

  • Soybean (Glycine max (L.) Merr.) seed is the main source of protein and oil for human and animal. The use of soybean protein has been expanded in the food industry due to their excellent nutritional benefits. But, antinutritional and allergenic factors are exist in the raw mature soybean. Lipoxygenase, Kunitz trypsin inhibitor (KTI) protein, and ${\alpha}^{\prime}-subunit$ of 7S globulin are main antinutritional factors in soybean seed. Breeding of a new soybean strain with lacking these components is needed. The objective of this research was to select new soybean line with lipoxygenase-free, KTI-free, and ${\alpha}^{\prime}-subunit$ free (lx1lx1lx2lx2lx3lx3titicgy1cgy1 genotype). Total 434 $F_2$seeds were obtained from the cross of cultivar, "Gaechuck#2" and PI506876. Presence and absence of lipoxygenase, KTI protein, and ${\alpha}^{\prime}-subunit$ of 7S globulin was tested by SDS electrophoresis using a partial seed of each $F_2$seed. Only one $F_2$seed with lacking these three components was selected and was planted to $F_2$plant. Absence of lipoxygenase, KTI, and ${\alpha}^{\prime}-subunit$ protein was confirmed on the $F_3$seeds harvested. Selected line with lx1lx1lx2lx2lx3lx3titicgy1cgy1 genotype might be useful for soybean breeding.

  • Object : To verify the effect of the lipoxygenase inhibitor and cycloxygenase inhibitor on meningioma cell proliferation. Method : Using two meningioma cell lines, cell proliferation was determined at 96 hrs after adding inhibitor (AA861, Nordihydroguaiaretic acid(NDGA), Indomethacin, acetyl-11-keto-beta-boswellic acid(AKBA) into medium by methyl tetrazolium salt/phenazine methosulfate(MTS/PMS) non-radioactive cell proliferation assay. We checked optical density with 490nm wavelength UV and this value was used as a proliferative index. The percent of inhibition was also calculated from this value. Conclusion : Indomethacin and NDGA showed no effect on meningioma proliferation. AA861 also showed no significant inhibitory effect, but AKBA demonstrated a significant inhibitory effect on meningioma cell proliferation.

  • Lipoxygenase-deficient soybeans, Jinpumkong (lipoxygenase-2, 3 lacking) and Jinpumkong 2 (lipoxygenase-1, 2, 3 lacking), were breeded for the improvement of beany flavor problem. The objectives of this study were to characterize and to examine the storage stability of two lipoxygenase-deficient soybeans by comparing with Hwangkeumkong having high lipoxygenase activity. The crude protein and crude lipid content of Jinpumkong 2 were lower than those of Hwkangkeumkong and Jimpumkong. All soybean samples were middle-sized and yellow-coated seeds. The rate of water uptake and trypsin inhibitor activity of Jinpumkong were greater than those of others. The cooking rate of Hwangkeumkong was the highest among all. The lipoxygenase activity of Hwangkeumkong was decreased when the soybeans were stored at $40^{\circ}C$ for 96hrs at 90% RH which is the condition of accelerated aging. After accelerated aging, the germination ratio of Hwangkeumkong was not changed but the ratio and speed of germination dropped rapidly in Jinpumkong and Jinpumkong 2.

  • In order to investigate the effect of tannic acid and ${\alpha}$-tocopherol on methyl linoleate autoxidation and on inhibition activity of lipoxygenase in phospholipid, the rate of formation of methyl linoleate hydroperoxide was measured by HPLC. The reaction mixture contained methyl linoleate 70mM, radical initiator AMVN 0.7mM, tannic acid and ${\alpha}$-tocopherol 0.7mM, each, in a mixture of hexane/isopropanol(1 : 1, v/v). Under this reaction condition, tannic acid was good enough antioxidant. The tannic acid and ${\alpha}$-tocopherol for the inhibition activivity of lipoxygenase were measured at the reaction conditions: phospholipid $1{\mu}M$. tannic acid and ${\alpha}$-tocopherol were reacted as an inhibitor of lipoxygenase in phospholipid, especialy in phosphatidy lcholine.

  • In order to get infonnations on the development of alcohol induced cardiovascular disorders, primary cultured vascular smooth muscle cells (PVSMC) were treated with acetaldehyde, one of the most reactive metabolites of ethanol. Acetaldehyde caused the striking release of lactate dehydrogenase (LDH) from PVSMC and it stimulated the prostaglandin synthesis in the same system. But it didn't induce cyclooxygenase activity. lipoxygenase inhibitors-propyl gallate and nordihydroguaiaretic acid could reverse the effect of acetaldehyde, but dexamethasone, a phospholipase $A_2\;(PIA_2)$ inhibitor and cyclooxygenase inhibitors except indomethacin could not protect the cells from acetaldehyde toxicity. These results indicate that enhanced prostaglandin synthesis by acetaldehyde is not a direct cause of cell death, but secondary effect due to the activation of PIAl and also, the roles of the lipoxygenase metabolites and/or $PIA_2$ activity itself might be more important in the cytotoxicity of acetaldehyde.

  • Lipoxygenase inhibitory acitivity of the methanolic extract of 60 different kinds of plant seeds was determined by a spectrophotometric method using a soybean lipoxigenase(SLO) and linolenic acid. Among the extracts examined, the methanolic extract of nutmeg(Myristical fragrans)seed showed the most potent SLO inhibitory activity. To isolate SLO inhibitor, hence, the defatted methanol extract was further partitioned with ether, ehtylacetate , and n-butanol , stepwise. The ether souble fraction was successively chromatographed on silica gel, Sephadex LH-20 and preparative TLC. Three phenolic compounds were isolated , and one of them showing a strong SLO inhibition activity was identified as a 2,6-dihydroxy-9-(3', 4', -dihydroxyphenyl)nonylphenone (IC50a=0.39$\mu\textrm{g}$/ml) by 1H-& 13C0NMR, IR, and MS spectroscopy.
