BACKGROUND/OBSECTIVE: Airway inflammation by eosinophils, neutrophils and alveolar macrophages is a characteristic feature of asthma that leads to pathological subepithelial thickening and remodeling. Our previous study showed that oxidative stress in airways resulted in eosinophilia and epithelial apoptosis. The current study investigated whether glutathione-containing dry yeast extract (dry-YE) ameliorated eosinophilia, goblet cell hyperplasia and mucus overproduction. MATERIALS/METHOD: This study employed $2{\mu}g$/mL lipopolysaccharide (LPS)- or 20 ng/mL eotaxin-1-exposed human bronchial epithelial cells and ovalbumin (OVA)-challenged mice. Dry-YE employed in this study contained a significant amount of glutathione (140 mg in 100 g dry yeast). RESULTS: Human bronchial epithelial cell eotaxin-1 and mucin 5AC (MUC5AC) were markedly induced by the endotoxin LPS, which was dose-dependently attenuated by nontoxic dry-YE at 10-50 ${\mu}g$/mL. Moreover, dry-YE inhibited the MUC5AC induction enhanced by eotaxin-1, indicating that eotaxin-1-mediated eosinophilia may prompt the MUC5AC induction. Oral supplementation with 10-100 mg/kg dry-YE inhibited inflammatory cell accumulation in airway subepithelial regions with a reduction of lung tissue level of intracellular adhesion molecule-1. In addition, ${\geq}50$ mg/kg dry-YE diminished the lung tissue levels of eotaxin-1, eosinophil major basic protein and MUC5AC in OVA-exposed mice. Alcian blue/periodic acid schiff staining revealed that the dry-YE supplementation inhibited goblet cell hyperplasia and mucus overproduction in the trachea and bronchiolar airways of OVA-challenged mice. CONCLUSIONS: Oxidative stress may be involved in the induction of eotaxin-1 and MUC5AC by endotoxin episode and OVA challenge. Dry-YE effectively ameliorated oxidative stress-responsive epithelial eosinophilia and mucus-secreting goblet cell hyperplasia in cellular and murine models of asthma.
Two trials were conducted to study the effects of two Chinese herbal polysaccharides, Astragalus polysaccharides (APS) and Achyranthes bidentata polysaccharides (ABPS), and one Chinese herbal saponin, Acantbepanax senticosus saponin (ASS), on the immunity and growth performance of weaned pigs. Experiment 1 was a 14-day growth assay, in which 32 weaned pigs were randomly allocated to one of four dietary treatments: i) 0.05% talcum powder control; ii) 0.05% APS; iii) 0.05% mixture of APS and ASS in a 1:1 ratio by weight; and iv) 0.05% mixture of APS, ASS, and ABPS in a ratio of 1:1:1 by weight. Blood samples were collected on day 14 to determine plasma parameters. Feed intake, body weight gain, and feed efficiency were also determined. Experiment 2 was a 21-day immunity assay, in which 16 weaned pigs were randomly allotted to one of two dietary treatments: i) 0.05% talcum powder control; and ii) 0.05% mixture of APS and ASS in a 1:1 ratio by weight. On day 21, pigs were challenged with lipopolysaccharide (LPS) and 3 h later blood samples were collected and analyzed for lymphocyte proliferation as well as interleukin 6 (IL-6), insulin-like growth factor 1 (IGF-1), growth hormone (GH), and cortisol levels. In Experiment 1, feeding Chinese herbal polysaccharides and saponin increased growth performance of the pigs. The effects of the mixture of APS and ASS were especially notable, as there was a significant improvement in growth performance compared with the control (p<0.05). The plasma concentration of immunoglobulin G (IgG), nitric oxide (NO), and nitric oxide synthase (NOS) were increased in all treatments groups, with the mixture of APS and ASS increasing the level of IgG and NOS significantly (p<0.05), compared with the control. There was no difference in the NO level between the control and treatment groups (p>0.05). In Experiment 2, Chinese herbal polysaccharides and saponin showed immunostimulating effects. The level of cortisol, GH, and IGF-I were significantly increased (p>0.05), and the level of IL-6 showed a significant decrease (p<0.05) in the APS and ASS treatment after the LPS challenge. The mixture of APS and ASS could stimulate the blood lymphocyte proliferation significantly whether the LPS was injected or not (p<0.05). These results show that Chinese herbal extracts can improve growth performance and stimulate immunity of weaned pigs. A mixture of APS and ASS, compared with APS alone, could be a new kind of immunostimulant for weaned pigs, which could result in greater positive effects on their growth performance and immunity.
Naegleria fowleri로 면역된 마우스 비장세포를 이입(이입)함으로써 원발성 아메바성 수막 뇌염의 발생을 방어할 수 있는지 즉 방어면역을 피동적으로 전달할 수 있는지를 관찰하였다. 무균 배양한 N. fowleri, ITMAP 359 영 양형 7×l04개를 생후 6주된 ICR 마우스에 감염시켰다. 살아있는 N. fowleri 영양형 106개씩을 1주일 간격으로 3회 복강내로 주입시켜 면역시켰다. 면역시킨 마우스의 비장을 적출하여 107개의 비장세포가 함유된 부유액을 마우스 복강내로 주입시키고 3일 후 N, fowleri를 감염시켰다. 비장세포에 Con. A와 lipopolysaccharide(LPS)를 처리한 후 배자발생 정도를 methyl-[3H]-thymidine을 사용하여 측정하였다. N. fowleri를 감염시킨 마우스의 사망률을 보면 정상 비장세포를 주입시킨 실험대조군에서 84%, 면역 비장세포를 주입시킨 실험군에서 72%로서, 정상 대조군에서의 사망률 100%에 비해 낮음을 알 수 있었다. 면역된 비장세포를 주입시킨 실험군에서 LPS를 처리한 비장세포의 배자발생정도는 감염 7일 후 실험대조군이나 정상대조군에 비해 증가되어 있었고, Con. A처리에 의한 배자발생 정도도 감염 7일 후 증가되어 있음을 관찰할 수 있었다. 혈청내 항체가는 감염 12일 후 정상 대조군에 비해 실험군과 실험대조군에서 높았다.

For centuries, Panax ginseng Meyer (Korean ginseng) has been widely used as a medicinal herb in Korea, China, and Japan. Ginsenosides are a class of triterpene saponins and recognized as the bioactive components in Korean ginseng. Ginsenosides, which can be classified broadly as protopanaxadiols (PPD), protopanaxatriols (PPT), and oleanolic acids, have been shown to flaunt a vast array of pharmacological activities such as immune-modulatory, anti-inflammatory, anti-tumor, anti-diabetic, and antioxidant effects. In recent years, a number of ginseng and ginsenoside researches have increasingly gained wide attention owing to its unique pharmacological properties. Although good efficacies of ginsenosides have been reported, lack of target specific delivery into tumor sites, low solubility, and low bioavailability due to modifications in gastro-intestinal environments limit their biomedical application in clinical trials. As a result to this major challenge, nanotechnology and drug delivery techniques play a significant role to solve this problematic issue. Thus, we reported the preparation of poly-ethylene glycol (PEG) and glycol chitosan (GC) functionalized to ginsenoside (Compound K and PPD) conjugates via hydrolysable ester bonds with improved aqueous solubility and pH-dependent drug release. In vitro cytotoxicity assays revealed that PEG-CK, and PPD-CK conjugates exhibited lower cytotoxicity compared to bare CK and PPD in HT29 cells. However, GC-CK conjugates exhibited higher and similar cytotoxicity in HT29 and HepG2 cells. Furthermore, GC-CK-treated RAW264.7 cells did not exhibit significant cell death at higher concentration of treatment which supports the biocompatibility of the polymer conjugates. They also inhibited nitric oxide production in lipopolysaccharide (LPS)-induced RAW64.7 cells. In addition to polymer-ginsenoside conjugates, silver (AgNps) and gold nanoparticles (AuNps) have been successfully synthesized by green chemistry using different m. The biosynthesized nanoparticles demonstrated antimicrobial efficacy, anticancer, anti-inflammatory, antioxidant activity, biofilm inhibition, and anticoagulant effect. Special interest on the effective delivery methods of ginsenoside to treatment sites is the focus of metal nanoparticle research.In short, nano-sizing of ginsenoside results in an increased water solubility and bioavailability. The use of nano-sized ginsenoside and P. ginseng mediated metallic nanoparticles is expected to be effective on medical platform against various diseases in the future.
본 연구는 매실박과 홍삼박을 사료에 첨가하여 육계 체내 생리활성 효과를 구명하고자 실시하였으며, 이를 위해 고온 스트레스 자극과 LPS 염증 반응을 유도하여 스트레스에 대한 반응을 조사하였다. 이를 위해 고온 스트레스 자극에 따른 생산성 및 혈액생화학 변화를 조사하였으며, LPS로 염증 반응이 유도된 육계에서 immunoglobulin 농도 및 비장 조직내 cytokine mRNA 발현을 조사하였다. 공시계로는 1일령 Ross종 육계수컷 192수를 선별하여 고온 환경(96수)과 일반 환경(96수)으로 구분하였다. 고온 환경은 불쾌지수 86($33^{\circ}C$, 75%)의 고온 스트레스를 자극하여 5주간의 사양 시험을 실시하였으며, 일반 환경은 5주간의 사양 시험 종료 후 LPS를 주입하였다. 환경에 따라 각각 처리당 24수(3수${\times}$8반복)씩 4처리구에 총 96수를 임의 배치하여 실시하였다. 시험구 배치는 무첨가구(negative control; C), 시판 면역 증강제 첨가구(positive control: PC, 25 ppm ${\beta}$-glucan), 매실 부산물 1% 첨가구(PM: plum marc), 홍삼 부산물 3%(RGM: red gingeng marc)로 배치하였다. 고온 스트레스에 대한 혈액생화학 조성에서 매실박 첨가구는 Mg 농도를 높게 유지하여 Ca/Mg 비율을 무첨가구보다 낮게 유지하였으며, 홍삼박 첨가구는 Mg 농도가 무첨가구와 같았지만, Ca 농도를 높여 Ca/Mg 비율을 무첨가구보다 낮게 유지하였다(P<0.05). 따라서 매실박과 홍삼박 첨가구는 고온 환경에서 무첨가구보다 C a/Mg 비율을 낮게 유지하여, Ca/Mg 비율과 정의 상관관계에 있는 cholesterol 수준을 낮추는데 긍정적으로 작용할 것이라 사료된다. LPS 접종 후 IgM 농도는 매실박 첨가구가 무첨가구에 비해 유의적으로 낮은 수준을 나타내었으며(P<0.05), 홍삼박 첨가구는 무첨가구에 비해 낮은 경향을 나타냈지만 유의적인 차이는 없었다. 또한 비장 조직 내 cytokine mRNA 발현에서 홍삼박 첨가구의 경우 공통적으로 IL-1, IL-2 및 IL-6의 mRNA 발현 비율이 무첨가구에 비해 유의적으로 낮았으며, 매실박 첨가구도 IL-1, IL-2에서 무첨가구와 베타글루칸 첨가구보다 낮게 나타났다(P<0.05). 따라서 육계에서 매실박은 LPS 감염 시 IL-1, IL-2 같은 친염증 사이토카인의 발현을 감소시킴으로 염증 반응을 줄여 IgM 상승을 억제하는 것으로 사료된다. 홍삼박 첨가구의 경우 친염증성 사이토카인 억제 효과가 높았지만 항체 농도는 무첨가구와 같은 수준으로 상승하였는데, 생산성, 면역 반응을 개선할 수 있는 첨가 수준에 대한 추가적인 연구가 필요할 것으로 사료된다. 또한 이 실험에서 양성 대조군으로 활용한 ${\beta}$-glucan 전체적으로 긍정적인 효과를 보이지 않았는데, 양성 대조군 활용 시 첨가 수준에 대한 세밀한 검토가 필요할 것으로 사료된다. 결론적으로 매실박과 홍삼박은 고온 스트레스와 외부 병원성 물질 자극에 대한 반응을 최소화하는데 긍적적으로 작용하는 것으로 사료된다.
뱀장어의 복강내에 Edwardsiella tarda의 FKC, HKC 및 crude LPS를 주사한 후 경시적으로 순환혈액 중 면역세포 수의 변화와 식작용, 항체가를 비교하여 항원 종류별 뱀장어의 면역 반응을 검토하였다. 백신 투여 뱀장어의 순환혈액 중 림프구 수는 백신 투여 후 6시간까지 감소하였으나 이후 증가하기 시작하여 24시간 후에는 정상치로 회복되었다. 이후 모든 면역 실험구에서는 실험기간 동안 대조구에 비하여 림프구 수가 증가된 상태로 지속되는 경향을 보였다. 한편, 호중구의 출현수를 대조구와 비교하면 백신 투여 후 12시간 이내에 최고치를 나타낸 후 서서히 정상 수준으로 회복되는 경향을 나타내었다. 모든 백신 투여 실험구의 혈청중 항체가는 백신 투여 3주째에 128에 이르러 높은 항체가가 지속되었으나, HKC 면역구의 경우에는 6주후 4로 떨어졌다. 호중구의 식작용에 있어서는 LPS 면역구의 항원 주사 6시간 후와 1주후의 반응 6시간째의 식균지수가 각각 28.3과 3.9로 가장 높았다. 항원 주사 후 12시간째의 식균지수가 반응 6시간째 FKC 면역구 18.8, HKC 면역구 10.7, LPS 면역구 28.3 및 대조구 1.2로서, 1 주 후 각 면역구의 식균지수보다 높게 나타났다. 신선 항혈청의 항균작용은 FKC와 LPS 투여구에서만 나타나고, HKC 투여구에서는 세균 수의 감소를 볼 수 없었다. 그리고, 공격시험에서는 HKC, FKC 및 LPS 면역구의 RPS가 각각 10%, 20% 및 30% 이었으므로, 본 세균은 항원의 처리 방법에 따라 숙주의 방어력 증강에 미치는 효과가 다른 것을 알 수 있었다.
본 연구는 홍삼박을 산란계 사료에 첨가하여 고온 환경에서의 적응과 면역 개선 효과를 조사하는 것이 목적이며, 이를 위해 면역 증강 효과를 가진 ${\beta}$-glucan 첨가 사료를 양성대조군으로 설정하여 고온 스트레스 후의 생산성 변화, 혈액생화학 조성, LPS에 대한 면역 반응 및 혈액 항산화 활성을 조사하였다. 52주령 ISA Brown 갈색계를 무첨가구(Negative Control; NC), 시판 면역제제 첨가구(Positive Control; PC, ${\beta}$-glucan 25 ppm), 홍삼 부산물 3% 첨가구(RGM; Red Gingeng Marc)로 구분한 후, 처리당 15수(3수${\times}$5반복)로 배치하여 총 11주간 사양 실험을 2하였다. 8주차에 불쾌지수(THI; temperature humidity index)를 76($27^{\circ}C$, 60%)에서 86($34^{\circ}C$, 60%)으로 상승시키고, 24시간 동안 유지한 후 3주간 회복 시간을 가졌으며, 사양 기간이 종료된 후 LPS(Excherichia coli; SIGMA) 접종을 하였다. 8주간의 일반 사양 실험에서 홍삼박은 산란율을 증대시켰으나(P<0.05), 고온 스트레스 자극 후 혈액 생화학 및 생산성 변화에서 홍삼박의 첨가에 의한 효과는 나타내지 않았다. LPS 접종 시 전체적으로 IgM 농도가 감소되지만 홍삼박 첨가구의 경우 면역제제 첨가구와 동일하게 LPS 주입 후 IgM 농도를 대조구보다 높게 유지하는 것으로 관찰되었다(P<0.05). 또한 혈액 내 SOD 유사 활성을 분석한 결과, 홍삼박 첨가구와 면역제제 첨가구 모두 무첨가구보다 높은 항산화 활성을 유지하는 것으로 관찰되었다(P<0.05). 종합해 보면, 홍삼박은 산란계에서 강한 고온 스트레스 자극에 대한 효과는 관찰되지 않았지만 일반환경에서 산란율을 증대시켜 생산성을 개선하는 효과가 있었다. 또한 ${\beta}$-glucan을 함유한 면역제제와 유사 패턴으로 LPS 접종에 대해 IgM 농도를 높게 유지하고, 혈액 내 항산화 활성을 높이는 효과를 나타내었다. 따라서 홍삼박은 산란계 체내 면역 조절 및 항산화 효과를 가져 산란계 건강과 생산성 증진에 긍정적으로 작용할 것으로 사료된다. 또한 고온 스트레스자극 실험 결과는 앞으로 고온 스트레스 관련 연구에서 고온 환경 설정(불쾌지수, 자극시간)에 유용한 자료로 활용될 수 있다고 사료된다.
본 연구는 구멍갈파래 급여가 육계 혈액 내 항산화, 면역조절 효과에 미치는 영향을 구명하기 위해 실시 하였으며, 이를 위해 LPS로 염증반응이 유도된 육계에서 혈액 SOD 유사 활성, immunoglobulin 농도 및 비장 조직 내 cytokine mRNA 발현을 조사하였다. 공시계로는 1일령 Ross종 육계수컷 96수를 선별하여, 육계전기(0~3주), 육계후기(3~5주)의 5주 동안 사양시험을 실시하였다. 처리당 24수(3수${\times}$8반복)씩 4처리구에 총 96수를 임의배치 하여 실시하였다. 시험구 배치는 무첨가구(Negative Control ; NC), 시판 면역증강제 첨가구(Positive Control ; PC, ${\beta}$-glucan 25 ppm), 구멍갈파래 분말 3% 첨가구(Ulva pertusa kjellman Powder ; Ulva P) 및 구멍갈파래 추출물 0.3% 첨가구(Ulva pertusa kjellman Extract ; Ulva E)로 배치하였다. 혈액 내 SOD 유사 활성을 분석한 결과 구멍갈파래 첨가구 (Ulva P, Ulva E)는 무첨가구와 면역제제 첨가구보다 높은 항산화 활성을 가지는 것으로 나타났다(P<0.05). immunoglobulin 농도에서 IgA와 IgG 농도는 처리구간의 차이가 나타나지 않았지만 IgM농도에서 구멍갈파래 추출물 처리구가 무첨가구에 비해 유의적으로 낮은 수준을 나타냈다(P<0.05). 이는 구멍갈파래 추출물이 LPS에 대한 면역자극을 조절하여 과잉면역반응을 억제한 것으로 사료된다. 비장 조직 내 cytokine mRNA 발현량을 조사한 결과 IL-1, IL-2 및 IL-6에서 처리구별 공통적인 차이를 나타내었는데 구멍갈파래 처리구(Ulva P, Ulva E)의 mRNA 발현 비율이 무첨가구와 면역제제 처리구에 비해 낮았으며, 구멍갈파래 처리 간 비교에서 구멍갈파래 추출물이 분말보다 더 낮았다(P<0.05). iNOS의 경우 구멍갈파래 분말 첨가구는 무첨가구와 유의적인 차이가 없었지만 구멍갈파래 추출물 첨가구는 모든 처리구보다 iNOS의 발현이 낮았다(P<0.05). 구멍갈파래 분말과 추출물은 LPS 주입에 의한 염증 관련 사이토카인 mRNA 발현을 억제하는 경향을 나타내며, 특히 구멍갈파래 추출물이 발현억제 효과가 더 높은 것으로 확인되었다(P<0.05). 본 연구 결과 육계에서 구멍갈파래 급여는 염증, 질병의 원인인 활성산소 제거에 효과가 있다고 사료된다. 특히 구멍갈파래 추출물은 혈액 내 IgM의 농도를 조절하여 외부항원에 대한 과잉면역반응을 억제하고 염증 관련 cytokine mRNA 발현을 억제하여 육계 면역조절에 긍적적인 영향을 미치는 것으로 사료된다.

거쳐 즉시 처리하여야 합니다. 다만, 즉시 처리가 곤란한 경우는 회원에게 그 사유와 처리일정을 통보하여야
합니다.
제 11 조 (회원의 의무)
① 이용자는 회원가입 신청 또는 회원정보 변경 시 실명으로 모든 사항을 사실에 근거하여 작성하여야 하며,
허위 또는 타인의 정보를 등록할 경우 일체의 권리를 주장할 수 없습니다.
② 당 사이트가 관계법령 및 개인정보 보호정책에 의거하여 그 책임을 지는 경우를 제외하고 회원에게 부여된
ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.
③ 회원은 당 사이트 및 제 3자의 지적 재산권을 침해해서는 안 됩니다.
제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)
① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을
원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
